ABSTRACT

Objective:To establish a method to determine the content of lidocaine hydrochloride in buccal adhesive tablets contai-ning carbomer as an excipient by HPLC. Methods:Calcium chloride was used to precipitate carbomer in buccal adhesive tablets. The HPLC analysis was performed on a Welch C18 column(250 mm × 4. 6 mm,5 μm)with the column temperature of 40℃. The mobile phase was the mixture of 0. 05% sodium acetate solution(added 50 ml acetic acid into 930 ml distilled water,adjusting pH to 3. 40 with 1 ml·min-1 sodium hydroxide solution)and acetonitrile(70:30). The flow rate was 1. 0 ml·min-1 ,the detection wavelength was at 254 nm and the injection volume was 20 μl. Results:Within the range of 0. 10-2. 00 mg·ml-1 ,there was a good linear rela-tionship between the concentration and the peak area of lidocaine hydrochloride(r=0. 999 9). The average recovery was 100. 2%( RSD=1. 4%,n=9). Conclusion:The established method is simple,accurate and reproducible,which can be applied to determine the content of lidocaine hydrochloride in buccal adhesive tablets.

ABSTRACT

OBJECTIVE:To optimize the formulation of nisoldipine buccal adhesive tablets. METHODS: The formulation was optimized by orthogonal design with the dosage ratio of carbopol to chitosan in drug-containing layer and protection layer, the content of lactose and the tabletting hardness as factors. The bioadhesiveness and in vitro drug release rate of the tablets was measured as well. RESULTS: The optimal formulation was as follows: the ratio of carbopol to chitosan in drug-containing layer and protection layer were 22.5∶22.5 and 33.3∶16.7 mg, respectively; the dosage of lactose was 10 mg, and tabletting hardness was 5 kg. The averge bioadhesiveness of 3 bacthes of optimized preparations was 79.5 g?cm-2, and the average in vitro drug release rate of the tablets in 12 h was more than 95%. CONCLUSIONS: The optimized nisoldipine buccal adhesive tablets enjoy ideal bioadhesiveness and slow-release character.

ABSTRACT

OBJECTIVE:To prepare nystatin buccal adhesive tablets and to study their properties of bioadhesion.METHODS:Nystatin buccal adhesive tablets of different formula matching ratio were prepared with carbomer(CP)and hy?droxy-propyl methyl cellulose(HPMC)as adhesive material and slow-release framework,and the adhesive power,duration of adhesion,swelling percentage and surface pH data of which were determined.RESULTS:There were great differences regarding bioadhesive properties among the adhesive tablets that of different formula matching ratio,the adhesive tablets that prepared in a formula matching ratio of1:1between CP to HPMC were of the best adhesiveness outcome.CONCLUSIONS:The buccal adhesive tablets were prepared successfully,and which can be used for the further study both in vivo and in vitro.
