ABSTRACT

Drug repurposing or repositioning has been well-known to refer to the therapeutic applications of a drug for another indication other than it was originally approved for. Repurposing non-oncology small-molecule drugs has been increasingly becoming an attractive approach to improve cancer therapy, with potentially lower overall costs and shorter timelines. Several non-oncology drugs approved by FDA have been recently reported to treat different types of human cancers, with the aid of some new emerging technologies, such as omics sequencing and artificial intelligence to overcome the bottleneck of drug repurposing. Therefore, in this review, we focus on summarizing the therapeutic potential of non-oncology drugs, including cardiovascular drugs, microbiological drugs, small-molecule antibiotics, anti-viral drugs, anti-inflammatory drugs, anti-neurodegenerative drugs, antipsychotic drugs, antidepressants, and other drugs in human cancers. We also discuss their novel potential targets and relevant signaling pathways of these old non-oncology drugs in cancer therapies. Taken together, these inspiring findings will shed new light on repurposing more non-oncology small-molecule drugs with their intricate molecular mechanisms for future cancer drug discovery.

ABSTRACT

Compared with mammals, zebrafish has unique advantages in screening pharmacoactive substance, and has been paid more and more attention in the field of medicine. In recent years, the exploration of zebrafish has been extended and extended to the field of Chinese materia medica (CMM), especially for the screening pharmacoactive substance of the single CMM, the CMM compound and Chinese patent medicines with multi-target, multi-channel and multi-link effects. As a complete animal model, zebrafish can carry out comprehensive and deep research on the effective chemical components that play a role in TCM, and then realize convenient, rapid and high-throughput screening of pharmacodynamics substances in CMM. Combined with the literature reports in recent five years at home and abroad, this paper reviews the latest research progress and unique advantages on the screening of pharmacodynamics substances in CMM in model organism zebrafish, mainly from the screening of cardiovascular drugs, lipid-lowering and liver-protecting drugs, anti-osteoporosis drugs, anti-tumor drugs, anti-inflammatory and other drugs, in order to provide a new idea for the application of model organism zebrafish in CMM and provide reference for the new drugs research of CMM.

ABSTRACT

Objective To conduct a comprehensive evaluation on safety of Honghua Injection through adopting the method of the evidence-based method;To provide reference for clinical reasonable application of Honghua Injection. Methods Computers were used to retrieve some Chinese databases, such as China Biology Medicine, China National Knowledge Infrastructure, Wangfang Database and VIP database. At the same time, other search methods were employed, up to July 2013, including all research types about Honghua Injection. The adverse reactions in the reports of published literature were analyzed by description and statistical analysis. Results Sixty-nine researches on Honghua Injection were included. The total cases of adverse drug reaction (ADR) were 1111, among which male cases were 568 (51%), and female cases were 543 (49%). Thirty-six (52%) papers described ADR of Honghua Injection in detail, and thirty-three (48%) papers just mentioned ADR or did not describe ADR in detail. Skin, skin accessories damage and pathological changes in circulatory system were main contents of ADR. In terms of original diseases, diseases of circulatory system play an important role. Solvent medium was largely in line with its product specification requirements. Most ADR appeared when the drug was used for the first time, from 5 minutes to 5 days. Conclusion The current published literature data show that severe ADR does not happen after the intervention of Honghua Injection.

ABSTRACT

The term decline effect is referred to a diminution of scientifically discovered effects over time. Reasons for the decline effect are multifaceted and include publication bias, selective reporting, outcomes reporting bias, regression to the mean, scientific paradigm shift, overshadowing and habituation, among others. Such effects can be found in cardiovascular medicines through medications (e.g., aspirin, antithrombotics, proton pump inhibitor, beta-blockers, statins, estrogen/progestin, angiotensin converting enzyme inhibitor etc.), as well as with interventional devices (e.g., angioplasty, percutaneous coronary intervention, stents). The scientific community should understand the various dimensions of the decline effects, and effective steps should be undertaken to prevent or recognize such decline effects in cardiovascular medicines.

ABSTRACT

OBJECTIVE: To probe into the status quo of cardiovascular drug-induced ADR and to provide reference for clinical utilization and ADR monitoring. METHODS: A total of 215 ADR cases of our hospital during 2003~2008 collected from electronic reporting system were classified, analyzed and evaluated. RESULTS: 84 kinds of drugs were involved, of which adrenergic agent occupied the dominant place (n=78), followed by calcium antagonists, anticoagulant, fibrinolytic agents and AT1R bloker. 18 cases induced by TCM involved 9 kind of Chinese patent drug. The first 3 drugs that involved in term of frequency of ADR were buflomedil, amlodipine and alprostadil. The main clinic manifestations of ADRs were lesion of skin and its appendants, psychological system and digestive system. CONCLUSION: More attention should be paid to safety of long-term prescription in such chronic disease and cardiovascular drug-induced ADR monitoring.

ABSTRACT

Objective: To study the stability of four kinds of cardiovascular ready-prepared Chinese medicine (Injections of Ciwujia, Shenmai, Dengzhanhuasu and Gegensu) in Betahistine Hydrochlorizel Sodium Chloride Injection. Methods: Four kinds of drugs were added into Betahistine Hydrochloride Sodium Chloride, respectively, and mixed, then observed for 6 hours at 23?C. Results: The appearance, pH value and concentration of four drugs above mentioned were stable in Betahistine Hydrochloride sodium chloride sodium chloride at room temperature (23?C) and in 6 hours. Conclusion: Injections of Ciwujia, Shenmai, Dengzhaihuasu and Gegensu are stable in Betahistine Hydrochlonile Sodium Chloride.
