ABSTRACT

bjective To optimize the optimal purification technology for total alkaloids from Longzuan Tongbi Recipe (LTR). Methods Five types of macroporous resins were used to adsorb and desorb the total alkaloids in LTR, and the adsorption capacities, adsorption ratios and desorption ratios of total alkaloids were regarded as the indexes to screen out the suitable resin. The purification technology for total alkaloids was optimized from LTR by single factor investigation and central composite design and response surface method. Results The best purification technology of total alkaloid from LTR were determined as follows: the concentration of sample solution of 0.17 g/mL (crude drug) with pH 1.7, sample flow rate at 1 mL/min, washing impurity with 9-column volume of 80% ethanol at a flow rate of 1 mL/min. The mass scores of total alkaloids was 22.23% and the yield of that was 173.27 mg/g. Conclusion HPD100 resin has good purification effect on total alkaloids from LTR, which could provide reference for the further study of pharmacodynamics.

ABSTRACT

Objective: To prepare plumbagin transfersomal (PBG-T) gel and investigate its transdermal penetration characteristics in vitro. Methods: Plumbagin transfersomes were prepared by film-ultrasonic dispersion method. The optimal prescription condition of PBG-T was selected by central composite design and response surface method. The formula of PBG-T gel was optimized by orthogonal test. The Franz diffusion cell was used to investigate transdermal penetration characteristics of PBG-T gel in vitro. Results: The optimal prescription condition of transfersomes was determined as drug 10.0 mg, phospholipids 700.0 mg, Tween-80 91.5 mg, ultrasonication time 13 min. The optimal prescription condition of transfersomal gel was 1% carbomer 940 as gel matrix, and 5% glycerol as the humectant. According to the optimized prescription, the entrapment efficiency, the mean particle size, and Zeta potential of PBG-T were (79.88 ± 2.26)%, (125.64 ± 4.54) nm, and (-30.97 ± 1.13) mV. The cumulative penetration rate of PBG-T gel was 70.0% at 12 h. Conclusion: The optimal preparation technique is stable and feasible. Transfersomal gel features a sustained release in vitro, the transfersomal gel can increase penetration rate of plumbagin through the skin of rats.

ABSTRACT

Objective:To optimize the processing technology of alum by using oven processing combined with central composite design and response surface method to provide basis for processing standardization production of alum. Methods:Alum as the raw ma-terial and on the basis of single factor experiment, the processing temperature, processing time and alum amount were used as the three independent variables factors. A titration method was applied in the content determination of aluminium potassium sulfate in withered alum, and a central composite design and response surface method was used to optimize the processing technology of alum. Results:The optimal processing technology of alum was as follows:the processing temperature was 250℃ with 40-minute processing time and 8 g alum. Conclusion:The optimized alum processing technology by central composite design and response surface method is simple, stable and predictable, which can provide basis for the processing standardization production of alum.

ABSTRACT

Objective:To optimize the ultrasonic extraction process of total alkaloids from Oxytropis falcata bunge. Methods:The independent variables were solvents ratio, extracting time and ethanol concentration, and the dependent variable was content of total al-kaloids. Based on single factor tests, central composite design and response surface methodology was adopted to optimize the extraction technology. Results:The optimal extraction conditions were as follows: extracted twice with 36-fold amount of 72% ethanol ( contai-ning 1% acetic acid) at 60 ℃, and extracted 77 minutes each time. Under the above conditions, the content of total alkaloids was 2. 793 mg·g-1 with the bias ratio less than 2% when compared with the model predictions. Conclusion:Ultrasonic extraction process of total alkaloids from Oxytropis falcata Bunge optimized by central composite design and response surface method is simple, highly pre-cise, reliable and predictable.

ABSTRACT

AIM To optimize the two-phase (ethanol-dipotassium phosphate) aqueous extraction for Astragali Radix by central composite design-response surface method.METHODS With ethanol concentration,dipotassium hydrogen phosphate solution concentration and Astragali Radix feed ratio as influencing factors,together with extraction rates of total flavonoids and total saponins as evaluation indices,the technology optimization was accomplished by central composite design-response surface method.RESULTS The optimal conditions were determined to be 27.97% for ethanol concentration,22.03% for dipotassium hydrogen phosphate solution concentration,and 1 ∶ 58.85 for Astragali Radix feed ratio.The extraction rate of total flavonoids reached 74.13% (concentrated in the upper layer),while that of total saponins reached 81.34% (concentrated in the lower layer).CONCLUSION With good predictability,this simple and accurate method can be used for the extraction of total flavonoids and total saponins in Astragali Radix.
