ABSTRACT

Abstract Production of lignocellulolytic enzymes by filamentous fungi have a great potential at industrial level due to their widespread applications. Mixed fungal cultures and particularly mixed fungal biofilms constitute a promising fermentation system for an enhanced enzyme production. However, it has not been addressed how much of this enhancement depends on the mixed biomass proportion. In this sense, the aim of this study was to develop a method to specifically and accurately quantify mixed fungal biomass. For this purpose, mixed biofilm cultures composed of Aspergillus niger and Trichoderma reesei, two filamentous fungi used industrially for cellulase production, were collected from 48 to 120 h of growth; mycelia were pulverized, and DNA was extracted for qPCR assays with specific primers for each fungus. Primers were designed from non-conserved regions of sequences of actin and β-tubulin genes of both A. niger and T. reesei. Specificity of these primers was tested in silico and experimentally. A statistically significant correlation was obtained between qPCR-calculated biomass and dry weight biomass data. By this method, it was possible to detect changes on mycelia proportions in biofilms over time, suggesting a competitive interaction between these two fungi. In conclusion, this method allows a specific and accurate quantification of mixed fungal biomass and could be also applied to different mixed culture systems for studying microbial interactions.


Resumen La producción de enzimas lignocelulolíticas por hongos filamentosos tiene un gran potencial a nivel industrial debido a sus diversas aplicaciones. Los cultivos fúngicos mixtos y particularmente las biopelículas fúngicas mixtas constituyen un sistema de fermentación prometedor para una mayor producción enzimática. Sin embargo, no se ha abordado cuánto de esta mejora depende de la proporción de biomasa mixta. En este sentido, el objetivo de este estudio fue desarrollar un método para cuantificar de forma específica y precisa la biomasa fúngica mixta. Para este propósito, se recolectaron cultivos mixtos de biopelículas de 48 a 120 h de crecimiento compuestos por Aspergillus niger y Trichoderma reesei, dos hongos filamentosos utilizados industrialmente para la producción de celulasas; el micelio se pulverizó y el ADN se extrajo para ensayos de qPCR con cebadores específicos para cada hongo. Los cebadores se diseñaron a partir de regiones no conservadas de las secuencias de los genes de actina y β-tubulina de A. niger y T. reesei. La especificidad de estos cebadores se probó in silico y experimentalmente. Se obtuvo una correlación estadísticamente significativa entre la biomasa calculada mediante qPCR y los datos de biomasa en peso seco. Mediante este método, fue posible detectar cambios en las proporciones de los micelios en las biopelículas a lo largo del tiempo, lo que sugiere una interacción competitiva entre estos dos hongos. En conclusión, este método permite una cuantificación específica y precisa de la biomasa fúngica mixta y también podría aplicarse a diferentes sistemas de cultivo mixto para estudiar interacciones microbianas.

ABSTRACT Objective. To measure the in vitro fermentation variables of a cellulolytic bacteria consortium (CBC) isolated from a water buffalo rumen in coculture with total ruminal bacteria (TRB) on two fibrous substrates. Materials and methods. A CBC was isolated from the ruminal fluid of a female water buffalo in selective cellulolytic media. The experimental design was completely random with a 3x2 factorial arrangement; factors were treatments [TRB, CBC, and coculture (TRB + CBC)] and substrates (cobra grass and corn stover). Total gas and methane (CH4) production were measured at different time intervals. At 72 h, measurements were taken of pH, ammoniacal nitrogen (NH3-N), dry matter degradation (DMD), neutral detergent fiber degradation (NDFD) and total bacteria population. Results. Gas production with both substrates was highest (p≤0.05) in the coculture at 3, 6 and 24 h. At 48 and 72 h, gas production in the cobra grass was highest (p≤0.05) in the coculture. The coculture and TRB did not differ (p>0.05) in terms of CH4, DMD and NDFD values at 48 and 72 h. With the cobra grass, NH3-N concentration was higher (p≤0.05) in the coculture than in the TRB. Conclusions. The gas production and dry matter degradation values of the water buffalo rumen cellulolytic bacteria consortia indicate them to be a promising alternative for improving cobra grass structural carbohydrates degradation when in coculture with bovine ruminal bacteria.


RESUMEN Objetivo. Determinar las variables fermentativas in vitro de un consorcio bacteriano celulolítico (CBC) aislado de una búfala de agua en cocultivo con bacterias ruminales totales (BRT) en sustratos fibrosos. Materiales y métodos. Un CBC se aisló de fluido ruminal de una búfala de agua en medios selectivos celulolíticos. El diseño experimental fue un diseño completamente al azar con arreglo factorial 3x2, los factores fueron tratamientos [BRT, CBC y un cocultivo (BRT + CBC)] y sustratos (pasto pangola y rastrojo de maíz). La producción de gas total y metano (CH4) se midieron a diferentes intervalos de tiempo. Además, se estimó pH, nitrógeno amoniacal (N-NH3), degradación de materia seca (DMS) y de fibra detergente neutro (DFDN), y la población de bacterias totales a 72 h de incubación. Resultados. El cocultivo produjo mayor (p≤0.05) cantidad de gas a las 3, 6 y 24 h en ambos sustratos. A las 48 y 72 h, el cocultivo produjo mayor (p≤0.05) gas en pasto cobra. El cocultivo y las BRT no presentaron diferencias (p>0.05) en la producción de CH4 a 48 y 72 h, y en DMS y DFDN (p>0.05). En el pasto cobra, la concentración de N-NH3 con el cocultivo fue mayor (p≤0.05) que con BRT. Conclusiones. La producción de gas y degradación de materia seca de los consorcios bacterianos celulolíticos procedentes del rumen de una búfala de agua muestran que son una alternativa para mejorar la fermentación de carbohidratos estructurales del pasto cobra cuando se cocultivan con bacterias ruminales bovinas.

ABSTRACT

Abstract Textile industries produce effluent wastewater that, if discharged, exerts a negative impact on the environment. Thus, it is necessary to design and implement novel wastewater treatment solutions. A sequential treatment consisting of ligninolytic co-culture with the fungi Pleurotus ostreatus and Phanerochaete crhysosporium (secondary treatment) coupled to TiO2 /UV photocatalysis (tertiary treatment) was evaluated in the laboratory in order to discolor, detoxify, and reuse textile effluent wastewater in subsequent textile dyeing cycles. After 48 h of secondary treatment, up to 80% of the color in the wastewater was removed and its chemical and biochemical oxygen demands (COD, and BOD5) were abated in 92% and 76%, respectively. Laccase and MnP activities were central to color removal and COD and BOD5 abatement, exhibiting activity values of 410 U L-1 and 1 428 U L-1, respectively. Subjecting wastewater samples to 12 h of tertiary treatment led to an 86% color removal and 73% and 86% COD and BOD5 abatement, respectively. The application of a sequential treatment for 18 h improved the effectiveness of the wastewater treatment, resulting in 89% of color removal, along with 81% and 89% COD and BOD5 abatement, respectively. With this sequential treatment a bacterial inactivation of 55% was observed. TiO2 films were reused continuously during two consecutive treatment cycles without thermic reactivation. Removal percentages greater than 50% were attained. Acute toxicity tests performed with untreated wastewater led to a lethality level of 100% at 50% in Hydra attenuata and to a growth inhibition of 54% at 50% in Lactuca sativa. Whereas sequentially treated wastewater excreted a 13% lethality at 6.25% and an inhibition of 12% at 75% for H. attenuata and L. sativa, respectively. Finally, sequentially treated wastewater was reused on dyeing experiments in which 0.86 mg g-1 adsorbed dye per g of fabric, that is equivalent to 80% of dye adsorption.


Resumen Las industrias textiles producen un efluente que, si es descargado, ejerce un impacto negativo en el ambiente. Así, es necesario diseñar e implementar soluciones novedosas para el tratamiento de estos desechos. Con el fin de decolorar, detoxificar y reutilizar aguas residuales producidas en la industria textil en ciclos de tinturado subsecuentes, se evaluó en laboratorio un tratamiento secuencial, consistente en un cultivo ligninolítico con los hongos Pleurotus ostreatus y Phanerochaete crhysosporium (tratamiento secundario) acoplado con fotocatálisis TiO2/UV (tratamiento terciario). Después de 48 horas de tratamiento secundario, se removió hasta un 80% del color del agua residual y las Demandas Química y Bioquímica de Oxígeno (DQO y DBO5) fueron reducidas en 92 y 76% respectivamente. Las actividades Lacasa y MnP (410 U L-1 y 1428 U L-1, respectivamente) fueron centrales en la remoción de color y en la reducción de DQO y DBO5. Someter muestras de agua residual a 12 horas de tratamiento terciario resultó en una remoción de 86% de color, 73% de DQO y 86% de DBO. La aplicación de un tratamiento secuencial por 18 h incrementó su efectividad, lo cual resultó en 89% de remoción de color, además de 81% y 89% de remoción de DQO y DBO5 respectivamente. Con este tratamiento secuencial se observó una inactivación bacteriana del 55%. Las películas de TiO2 se reutilizaron continuamente durante 2 ciclos de tratamiento consecutivos sin reactivación térmica. Se alcanzaron porcentajes de remoción mayores al 50%. Las pruebas de toxicidad aguda llevadas a cabo con agua residual cruda produjeron un nivel de letalidad del 100% a 50% en Hydra attenuata y una inhibición de crecimiento del 54% a 50% en Lactuca sativa. El agua residual tratada secuencialmente produjo una letalidad del 13% a 6.25% y una inhibición del 12% a 75% para H. attenuata y L. sativa, respectivamente. Finalmente, el agua residual tratada secuencialmente fue reutilizada en experimentos de tinturado en los que la cantidad de colorante absorbido fue de 0.86 mg g-1 por g de tela, lo cual es equivalente a 80% de adsorción.


Resumo A indústria têxtil produz águas residuais que causam um impacto negativo ao meio ambiente quando são descartadas. Portanto, é necessário projetar e implementar novas soluções para seu tratamento. Um tratamento sequencial que consiste na co-cultura ligninolítica com os fungos Pleurotus ostreatus e Phanerochaete crhysosporium (tratamento secundário) acoplada à fotocatálise com TiO2/UV foi avaliado no laboratório para descolorir, desintoxicar e reutilizar água residual têxtil em ciclos subsequentes de tintura. Após 48 h de tratamento secundário, aproximadamente 80% da cor foi removida da água residual e sua demanda química e bioquímica de oxigênio (COD, e BOD5) foram diminuídas em 92% e 76%, respectivamente. A atividade de Laccase e MnP foram centrais na remoção da cor e diminuição de COD e BOD5, mostrando valores de atividade de 410 U L-1 e 1428 U L-1, respectivamente. Submeter as amostras de água residual a 12 h de tratamento terciário levou a uma remoção de 86% da cor e uma diminuição de 73% e 86% de COD e BOD5 respectivamente. A aplicação do tratamento sequencial por 18 h melhorou a efetividade do tratamento de águas residuais, resultando em 89% de remoção de cor e uma diminuição de 81% e 89% de COD e BOD5, respectivamente. Com este tratamento sequencial, observamos uma inativação bacteriana de 55%. As películas de TiO2 foram reutilizadas continuamente durante dois tratamentos consecutivos sem reativação térmica. Atingimos porcentagens de remoção acima de 50%. Os testes de toxicidade aguda feitos em água residual sem tratar causaram níveis de letalidades de 100% a 50% em Hydra attenuata e uma inibição do crescimento de 54% a 50% em Lactuca sativa. Enquanto água residual tratada sequencialmente causou uma letalidade de 13% a 6.25% e uma inibição de 12% a 75% para H. attenuata e L. sativa, respectivamente. Finalmente, a água residual tratada sequencialmente foi reutilizada em experimentos de tintura nos quais absorveu 0.86 mg g-1 por g de tecido, o que é equivalente a uma absorção de 80%.

ABSTRACT: The goal of the present study was to evaluate the germination, initial growth, and in vitro co-cultivation of Comanthera curralensis Moldenke, a "sempre viva" native of the Chapada Diamantina state of Bahia. Full strength (MS) and half-strength MS (MS1/2) growth media supplemented with two different sucrose concentrations (15 and 30g L-1) were tested for germination and initial plant growth. Three different plant densities were tested by in vitro culture (8, 10 and 12 plants per container). MS1/2 medium with 15g L-1 sucrose resulted in a higher percentage of germination and plant growth for the in vitro establishment of C. curralensis. The use of 12 plants per container is indicated for cost reduction in C. curralensis in vitro production.


RESUMO: Este trabalho teve como objetivo avaliar a germinação, o crescimento inicial e o co-cultivo in vitro de Comanthera curralensis Moldenke, uma "sempre viva" nativa da Chapada Diamantina-BA. Para germinação e crescimento inicial, foram testados os meios de cultura MS completo e MS1/2 suplementados com duas concentrações de sacarose (15 e 30gL-1); no cultivo in vitro, foram testadas três quantidades de plantas por recipiente (8,10 e 12). A utilização do meio MS1/2 com 15gL-1 de sacarose proporcionou maiores porcentagem de germinação crescimento das plantas no estabelecimento in vitro de C. curralensis , e o uso de 12 plantas por recipiente é indicado para a redução de custos na produção in vitro da espécie.

ABSTRACT

The aim of this work is to implement a biological model of neuromuscular junctions to study the mechanisms involved in intra and inter cellular processes using cell co-cultures. To optimize growth and development of the neuromuscular junction, cells were seeded on plasma polymerized pyrrole which has proven suitable for other types of cell cultures. The cell lines used were motor neuron NG108-15 and skeletal muscle C2C12. Cells were evaluated according to their morphology and electrophysiological characteristics. To observe the expression of specific proteins of the nerve synapse, immunocytochemical techniques were applied using dying antibodies. Proteins localized in nerve terminals were dyed and imaged by fluorescence microscopy. Images of cell co-cultures showed the formation of neuromuscular junctions. The preparation of neuromuscular junctions described in this work will allow the study of the mechanisms involved in their functions.


El objetivo de este trabajo es implementar un modelo biológico de unión neuromuscular para el estudio de los mecanismos involucrados en los procesos intra e intercelulares empleando co-cultivos celulares. Con el fin de optimizar el crecimiento y desarrollo de las uniones neuromusculares, las células se cultivaron sobre superficies de polipirrol obtenidas mediante polimerización por plasma que han mostrado ser adecuadas en otros tipos de cultivos celulares. Las líneas celulares que se emplearon fueron los modelos de motoneurona NG108-15 y muscular C2C12. Las células se evaluaron de acuerdo a su morfología y características electrofisiológicas. Para observar la expresión de proteínas clave de la sinapsis, se aplicaron técnicas inmunocitoquímicas utilizando anticuerpos específicos para la marcación de proteínas localizadas en las terminales nerviosas adquiriendo imágenes con microscopía de fluorescencia. Las imágenes de los co-cultivos celulares mostraron la formación de uniones neuromusculares. El método de preparación de uniones neuromusculares que se describe en este trabajo permitirá estudiar los mecanismos involucrados en sus funciones.

ABSTRACT

There were no significant differences in the distribution of embryos reaching to 2- cells, 4- cells, morula or blastocysts culturing on human endometrial stromal cells (Secretory or proliferative phases). The percent of morula in stage A (without fragmentation), stage B (<25% fragmentation), stage C (25-50% fragmentation) and stage D (>50% fragmentation) and did not showed significant differences between two coculture groups. Thus, the phase that the endometrial stromal cells were in thereby did not affect on the quality of embryos.


No hubo diferencias significativas en la distribución de los embriones en los cultivos que llegan a las 2 y 4 células, mórula o blastocistos sobre las células del estroma endometrial (fases proliferativa y secretora). El porcentaje de mórulas en etapa A (sin fragmentación), etapa B (<25% fragmentación), etapa C (25-50% de fragmentación) y etapa D (>50% fragmentación), y no mostraron diferencias significativas entre los dos grupos de co-cultivo. Así, la fase en la que se encontraban las células estromales endometriales no afectaron la calidad de los embriones.

ABSTRACT

The present work was carried out to evaluate the in vitro development of bovine embryo co-cultured in granulosa, oviduct, BRL or VERO cells co-cultures, supplemented with 5% or 10% of Fetal Calf Serum (FCS). Cummulus oocyte complexes were aspirated, matured and fertilized in vitro. Embryonic structures were divided into eight treatments. They were placed in culture media TCM 199 containing granulosa, oviduct, BRL or VERO cells, each of them added with 5% or 10% FCS. The conditions for the co-culture were 38.5 ºC, 5% CO2 in air and high humidity for ten consecutive days. Cleavage, blastocyst and hatching rates did not differ (p > 0.05) in co-culture with primary cells (granulosa and oviduct) when FCS concentration increased from 5 to 10%. However, in continuous cells co-culture (BRL and VERO), when FCS concentration increased from 5% to 10%, the blastocyst development rate decreased significantly (p < 0.05) from 33.6 to 16.3% and from 40 to 16.5% in embryo co-culture with BRL and VERO cells, respectively.


O presente trabalho foi realizado com o objetivo de avaliar o desenvolvimento in vitro de embriões bovinos co-cultivados em células da granulosa, do oviduto, BRL e VERO, suplementados com 5% ou 10% de Soro Fetal Bovino (SFB). Os complexos cummulus oócitos foram aspirados, maturados e fecundados in vitro. As estruturas embrionárias foram divididas em oito tratamentos: co-cultivo em TCM 199 contendo células da granulosa, do oviduto, BRL ou VERO adicionadas com 5% ou 10% de SFB. As condições de cultivo foram 38.5 ºC, 5% CO2 em ar e alta humidade por dez dias consecutivos. Os índices de clivagem, blastocisto e eclosão não diferiram (p > 0,05) no co-cultivo com células primárias (granulosa e oviduto) quando a concentração de SFB aumentou de 5 para 10%. Entretanto, no co-cultivo com células de linhagens contínuas (BRL e VERO), quando a concentração de SFB aumentou de 5% para 10%, os índices de blastocistos diminuíram significativamente (p < 0,05) de 33,6 para 16,3 % e de 40 para 16,5% nos embriões bovinos co-cultivados com células VERO e BRL, respectivamente.
