ABSTRACT

Mercury is used in various industrial. Part of Mercury's industrial waste is discharged into the environment, rivers and their tributaries, thus contaminating aquatic animals. Aim:to evaluate Mercury-induced behavioral changes in Zebrafish (Danio rerio) by the analysis of locomotor activity and parameters related to neurotoxicity and to verify whether ultra-diluted substances can decrease neurobehavioral effects and toxic. Methodology:The fishes were separated into 4 monitoring aquariums with 8 fishes each, with temperature, pH controlled, until the time of the toxicological experiments. 0.5 mL of Mercury 6cH, 30cH and distilled water (positive control) were added per liter of water in each aquarium containing 6 liters of water, then 3 mL of medication per aquarium, the white control received no medication and the toxic agent. After 1 hour the drugs were added, toxic mercury (200 µg/L), 4 mL per aquarium was added and remained so for 24 hours. All the experiment was run in blind, and the drugs identified by codes. The animals were subjected to behavioral tests (Open Field-locomotion; Vertical Open Field for neurotoxicity evaluation and Light and Dark Test), and each stage was recorded for later evaluation of movements and neurobehavioral changes. ANOVA was performed, followed by Tukey test, with p <0.05. Results: Mercury produced an anxiogenic effect in animals that were submitted to it without medication. In the vertical open field, there was an increase in erratic movements (1.25 ± 1.0) and tremors (0.87 ± 0.35) compared to the control (0.12 ± 0.35 and 0.25 ± 0.46 respectively), proving the toxic effect. Fishes which received the medication at 6 cH and 30 ch showed tremors and erratic movements similar to control. Conclusion:200 µg/L mercury in water can cause neurobehavioral disturbances in fishes, and animals receiving Mercurius6 cH and 30 cH ultra-diluted drug did not show neurotoxicity.

OBJECTIVES@#A study was conducted to explore the expression pattern and function of ferritin heavy polypeptide gene (fth1b) in zebrafish pharyngeal teeth development and lay the foundation for subsequent research on teeth development and mineralization.@*METHODS@#The zebrafish embryos were harvested at 56, 72, 96, and 120 h after fertilization. The expression of fth1b in zebrafish pharyngeal teeth development was detected by whole embryo @*RESULTS@#The expression pattern of fth1b gene was very similar to that of the known zebrafish pharyngeal teeth marker dlx2b and was specifically expressed in the zebrafish pharyngeal teeth during development. After the specific knockout of the gene fth1b, the earliest gene that can be detect in zebrafish pharyngeal teeth-pitx2 was expressed normally during early development. The dlx2b expression was not significantly different from that of wild type zebrafish, but the mineralization of pharyngeal teeth in the mutant was weaker than that of wild type zebrafish.@*CONCLUSIONS@#The gene fth1b is specifically expressed in zebrafish pharyngeal teeth and acts on their early mineralization.

ABSTRACT

RESUMO A bacia hidrográfica do córrego Água Boa (Dourados, MS) sofre grande influência antrópica, tais como urbana e industrial. Levando-se em consideração sua importância para a manutenção de fauna, flora, indústria e população local, o estudo teve como objetivo avaliar a toxicidade de suas águas nos períodos seco e chuvoso de 2013, da nascente até próximo à sua foz, por meio de testes de toxicidade aguda com D. similis e D. rerio, além de análises físicas, químicas e de concentrações de metais pesados. Os resultados mostraram as variabilidades espacial e temporal dos parâmetros analisados. Além disso, constatou-se efeito tóxico no ponto 02 para D. rerio em ambos os períodos. As análises comprovaram a crítica situação de degradação do ambiente aquático estudado e alertam para os cuidados que deverão ser tomados. Portanto, sugere-se maior fiscalização por parte dos órgãos ambientais, bem como conscientização da população quanto às emissões de efluentes e resíduos no corpo hídrico, a fim de evitar sua degradação.


ABSTRACT The watershed of Água Boa Stream (Dourados/MS) suffers great anthropic influence, such as urban and industrial. Taking into consideration its importance for maintenance of fauna, flora, industry and local population, this study aimed to: assess the toxicity of its waters during the dry and rainy seasons of 2013, near the source to its mouth, through acute toxicity essays with D. similis and D. rerio, as well as physical, chemical and heavy metal concentration analyses. The results showed spatial and temporal variability of the analyzed parameters. Furthermore, a toxic effect was found at point 02 for D. rerio in both periods. The analyses confirmed the critical situation of degradation of the aquatic environment studied, and warns that precautions should be taken. Therefore, it is suggested that greater supervision by environmental agencies, as well as public awareness of the issue of waste and residues in the water body, in order to avoid its degradation.

ABSTRACT

Decabromodiphenylethane (DBDPE) has been widely used as an alternative flame retardant due to the restriction or phase-out of traditional polybrominated diphenyl ethers (PBDEs), and is of increasing concern regarding its ubiquity, persistence, and potential adverse effects. In the present study, the toxicological effects of DBDPE were evaluated using zebrafish as an in vivo model. Upon being exposed to DBDPE-polluted sediments for a short term, it was found that the mortality and malformation of zebrafish (including edema, bent notochord, and bent tail) were not affected even at the highest concentration tested (1000.0 µg/kg dry sediment). Regarding behavioral responses, it was found that zebrafish larvae of 48 hours post fertilization (hpf) in all groups escaped successfully with a touch to the dorsal fin. However, when exposed to the highest DBDPE concentration, the larvae of 120 hpf exhibited significantly smaller distances as compared to the control. Moreover, the results of the acetylcholinesterase (AChE) activity, the expression levels of two important nerve-related genes, and the cell apoptosis all indicated that DBDPE posed low neurotoxicity in embryo-larval zebrafish. The results in this study shed some light on the potential risks of DBDPE in the real environment and highlight the application of the sediment exposure route in the future.

ABSTRACT

OBJECTIVE:To study the cardiac and skeletal toxicity of retinoic acid (RA) in Danio rerio at early life stage. METHODS:Danio rerio embryos of 24 hours post fertilization(hpf)were used as toxicity model and were exposed under medium with various concentrations of RA(0.1,1,10,25,100 μmol/L). The morphology of embryos and larvae hearts were observed 24,48 h after exposed. LC50 was calculated. Danio rerio larvae of 4 days post fertilization (dpf) were used as skeletal deformity model and were exposed with a series of RA at various concentration(0.1,1,10,25,50μmol/L). They were sacrificed 5 d later, and then Danio rerio skeleton were fixed for staining with alizarin red. The microscopic was used to observe the difference of stained skeleton area. RESULTS:RA caused significant adverse effects on hatching capabilities of Danio rerio embryos,and the ob-vious malformation features were produced during the culture process. 1-100 μmol/L RA could cause heart malformation in Danio rerio embryos and larvae,and the main heart malformation characteristics included heart linearization,pericardial edema,yolksa-cedema,hemocytes accumulation incardiac region. 100 μmol/L RA could inhibit the hatching capabilities of Danio rerio embryos, and caused lethal effects on embryos and larvae. The LC50 were 36.44,23.69 μmol/L after exposed for 24,48 h. 0.1-50 μmol/L RA induced vertebral column sclerotization of Danio rerio embryos and larvae in advance,which was positively associated with the con-centration of RA. CONCLUSIONS:RA can cause cardiac and skeletal toxicity in Danio rerio embryo and larvae,which is positive-ly associated with the concentration of RA.

In the field of studies of acute toxicity induced by bacterial agents, Shiga toxins have been relevant due to the severity of the extra-intestinal diseases they cause. Numerous studies have shown that Shiga toxin induced apoptosis in different cell types; however, this important process has been little studied in vivo experimental models. In this study, the effects of excretion products of Shigella dysenteriae, in which Shiga toxin is present, were investigated on early larval stages of Zebrafish, an animal model with many advantages over other in vivo experimental models traditionally used. Both the collection of eggs and larvae of Zebrafish, and the product from excretion from Shigella dysenteriae (SdyEP) were performed according to laboratory standards. Also, toxicity bioassay, larvae treatment with pure and diluted solution, 10-1, 10-2, 10-3 , 10-4 and 10-5, v/v SdyEP and cell death in vivo using Acridine Orange (AO) and Ethidium Bromide (EB) were applied. The excretion product of Shigella dysenteriae (SdyEP) effect was expressed in terms of larval mortality and dependent dilution rather than incubation time. The larval population surviving treatment with Shigella excretion product presents severe morphological effects. The larval population generally presents notable severe morphological damage, the necrosis state is represented by the opacity of the larvae after being treated for 24 h (b) compared to control. Other changes associated with larval anatomy were also observed; particularly the caudal end curvature was significant into 10%. The use of AO/EB revealed a distribution pattern from fluorescence into green and orange in surviving larvae SdyEP poisoning, there was a large population of dead cells around the anal and caudal region as evidenced by the presence of orange nuclei in greater numbers as controls in the larvae. The results support the application of coloring AO/EB in Zebrafish experimental models for the evaluation of the toxic action of new molecules and new products with therapeutic potential.


En el campo de los estudios de toxicidad aguda inducida por agentes bacterianos, las toxinas Shiga resultan relevantes debido a la severidad de las enfermedades extra-intestinales que causan. Numerosos estudios han demostrado que la toxina Shiga induce la apoptosis en diferentes tipos de células, sin embargo, este importante proceso ha sido poco estudiado en modelos experimentales in vivo. En este estudio, fueron evaluados los efectos del productos de excreción de Shigella dysenteriae (PESdy), sobre estadios larvarios de pez cebra (Danio rerio), un modelo animal con muchas ventajas sobre otros modelos experimentales in vivo utilizados tradicionalmente. Tanto la recolección de los huevos y larvas de pez cebra, así como la obtención del producto de la excreción, se realizaron de acuerdo a los estándares de laboratorio. Poblaciones larvarias, fueron tratadas con distintas soluciones; pura y diluidas, 101, 10-2, 10-3, 10-4 y 10-5, v/v de PESdy. La muerte celular in vivo, usando naranja de acridina (NA) y bromuro de etidio (BE) fue evaluada. El efecto del SdyEP, se expresó como dependiente de la concentracion y del tiempo de exposición. La población de larvas sobrevivientes, presentaron curvatura troncal en un 10%, en relación a los controles. La necrosis se puso en evidencia a través de la opacidad de las larvas después de 24 h. El uso de NA/BE reveló un patrón de distribución de la fluorescencia en verde y naranja en larvas sobrevivientes al tratamiento. Una granpoblación de células muertas, alrededor de la región anal y caudal, se ponen en evidencia por la presencia de núcleos de naranja en mayor número que en los controles. Los resultados apoyan la aplicación de la coloración NA/BE en modelo experimental de pez cebra, para la evaluación de la acción tóxica de nuevas moléculas y nuevos productos de excreción bacterial con potencial terapéutico.

OBJECTIVES: An ecofriendly alternative to chemical pesticides is bio-pesticides, which are derived from natural sources. The interest in bio-pesticides is based on the disadvantages associated with chemical pesticides. METHODS: We conducted acute toxicity assessments of camphor, a major component of bio-pesticides, by using Daphnia magna (D. magna) as well as assessed the morphological abnormalities that occurred in Danio rerio (D. rerio) embryos. RESULTS: The median effective concentration of camphor on D. magna after 48 hours was 395.0 muM, and the median lethal concentration on D. rerio embryos after 96 hours was 838.6 muM. The no observed effect concentration and predicted no effect concentration of camphor on D. magna, which was more sensitive than D. rerio, were calculated as 55.2 muM and 3.95 muM, respectively. Morphological abnormalities in D. rerio embryos exposed to camphor increased over time. Coagulation, delayed hatching, yolk sac edema, pericardial edema, and pigmentation of embryos mainly appeared between 24 and 48 hours. Further, symptoms of scoliosis and head edema occurred after 72 hours. In addition, bent tails, ocular defects and collapsed symptoms of fertilized embryonic tissue were observed after 96 hours. CONCLUSIONS: The camphor toxicity results suggest that continuous observations on the ecosystem are necessary to monitor toxicity in areas where biological pesticides containing camphor are sprayed.

ABSTRACT

Presence of mercury and other heavy metals above permissible levels in water bodies across the globe is posing a serious threat to aquatic biota and public health. Occurrence of mercury above the permissible limits in the aquatic ecosystem of Hyderabad city is well established. In this context, we carried out static- renewal bioassays on the zebra fish, Danio rerio exposed to different concentrations of mercuric chloride, and the 96-h median lethal concentration (LC50) was found to be 0.077 mgl-1. Behavioral manifestations like loss of scales, hyper secretion of mucus, surfacing and darting movements, loss of balance, irregular swimming patterns were noticed in the fish exposed to 0.077 mgl-1. The present study also examined the toxic effects of mercuric chloride on vital biochemical constituent’s total glycogen and total protein. In order to verify the influence of chronic and acute ambient oxygen levels from egg to adult stage of the zebrafish, in vivo oxygen consumption (MO2), critical tensions of oxygen (Pcrit), heart rate (fH) and total body lactate concentration (Lc) were determined for Danio rerio (Hamilton, 1822) raised at 28 °C under normoxic (7.5 mgO2.L-1 or 80 mm.Hg-1) and hypoxic conditions (4.3 mgO2.L-1) and exposed to acute hypoxia during different developmental stages. Our findings confirmed that very early stages do not respond effectively to ambient acute hypoxia. However, after the stage corresponding to the age of 30 days, D. rerio was able to respond to acute hypoxia through effective physiological mechanisms involving aerobic and anaerobic metabolism. Such responses were more efficient for the fishes reared under hypoxia which showed that D. rerio survival capability increased during acclimation to mild hypoxia. Measurements of body mass and length showed that moderate hypoxia did not affect growth significantly until the fish reached the stage of 60 days. Moreover, a growth delay was verified for the hypoxic-reared animals. Also, the D. rerio eggs-to-larvae survival varied from 87.7 to 62.4 percent in animals reared under normoxia and mild hypoxia, respectively. However, the surviving animals raised under moderated hypoxia showed a better aptitude to regulate aerobic and anaerobic capacities when exposed to acute hypoxia.


A influência de diferentes níveis de oxigênio no desenvolvimento (ovos a adulto) do peixe paulistinha Danio rerio (Hamilton, 1822) foi verificada por meio de medidas experimentais de consumo de oxigênio (MO2), tensões críticas de oxigênio (Pcrit), taxa de batimentos cardíacos (fH) e concentração total de lactato nos tecidos (Lc), para os animais mantidos a 28 ºC sob níveis normóxicos de oxigênio (7.5 mgO2.L-1 ou 80 mmHg) e hipóxicos (4.3 mgO2.L-1) e submetidos a hipóxia ambiental aguda, em diferentes estágios de desenvolvimento. Os resultados obtidos indicam que os peixes em estágios iniciais do desenvolvimento não variam suas respostas fisiológicas em função das oscilações ambientais nos níveis de oxigênio, visto que tais respostas iniciaram-se somente no estágio de 30 dias de vida. A partir deste estágio D. rerio apresentou capacidade em responder à hipóxia aguda por meio de mecanismos fisiológicos efetivos envolvendo metabolismo aeróbico e anaeróbico. Tais respostas foram mais efetivas para os peixes mantidos sob hipóxia, o que mostrou que a capacidade de sobrevivência de D. rerio aumentou durante o período de aclimatação à hipóxia moderada. As medidas de massa e comprimento corpóreos mostraram que a permanência dos peixes em hipóxia durante o desenvolvimento não afetou esses parâmetro até os peixes atingirem o estágio de 60 dias. A partir deste estágio foi observado ligeiro atraso no crescimento dos espécimes mantidos sob hipóxia. A taxa de sobrevivência de D.rerio variou de 87,7 a 62,4 por cento para os animais mantidos respectivamente sob níveis normóxicos e hipóxicos. No entanto, os animais mantidos sob hipóxia moderada, que sobreviveram, mostraram maior capacidade em regular seu metabolismo aeróbico e anaeróbico quando expostos à hipóxia ambiental aguda.

The aim of the present study was to determine the effect of the histaminergic precursor L-histidine and the H3 receptor antagonist thioperamide on the learning process of zebrafish submitted or not to confinement stress. On each of the 5 consecutive days of experiment (D1, D2, D3, D4, D5), animals had to associate an interruption of the aquarium air supply with food offering. Non-stressed zebrafish received an intraperitoneal injection of 100 mg/kg L-histidine, 10 mg/kg thioperamide or saline after training. Stressed animals received drug treatment and then were submitted to confinement stress for 1 h before the learning procedure. Time to approach the feeder was measured (in seconds) and was considered to be indicative of learning. A decrease in time to approach the feeder was observed in the saline-treated group (D1 = 141.92 ± 13.57; D3 = 55 ± 13.54), indicating learning. A delay in learning of stressed animals treated with saline was observed (D1 = 217.5 ± 25.66). L-histidine facilitated learning in stressed (D1 = 118.68 ± 13.9; D2 = 45.88 ± 8.2) and non-stressed (D1 = 151.11 ± 19.20; D5 = 62 ± 14.68) animals. Thioperamide inhibited learning in non-stressed (D1 = 110.38 ± 9.49; D4 = 58.79 ± 16.83) and stressed animals (D1 = 167.3 ± 26.39; D5 = 172.15 ± 27.35). L-histidine prevented the increase in blood glucose after one session of confinement (L-histidine = 65.88 ± 4.50; control = 53 ± 3.50 mg/dL). These results suggest that the histaminergic system enhances learning and modulates stress responses in zebrafish.

ZNF230 is a novel zinc finger gene cloned by our laboratory. In order to understand the potential functions of this gene in vertebrate development, we cloned the zebrafish orthologue of human ZNF230, named rnf141. The cDNA fragment of rnf141 was obtained by rapid amplification of cDNA ends (RACE). The open reading frame (ORF) encodes a polypeptide of 222 amino acids which shares 75.65 percent identity with the human ZNF230. RT-PCR analysis in zebrafish embryo and adult tissues revealed that rnf141 transcripts are maternally derived and that rnf141 mRNA has a broad distribution. Zygotic rnf141 message is strongly localized in the central nervous system, as shown by whole-mount in situ hybridization. Knockdown and over expression of rnf141 can induce abnormal phenotypes, including abnormal development of brain, as well as yolk sac and axis extendsion. Marker gene analysis showed that rnf141 may play a role in normal dorsoventral patterning of zebrafish embryos, suggesting that rnf141 may have a broad function during early development of vertebrates.

ABSTRACT

O Danio rerio (zebrafish, paulistinha) é um pequeno peixe de água-doce, que vem sendo utilizado como modelo de animal para o estudo de numerosas doenças humanas. A sua facilidade de manutenção e reprodução e os métodos laboratoriais para sua criação já estão bem estabelecidos. Sua manutenção requer alguns cuidados básicos e a compra de equipamentos específicos, porém estes são de baixo custo. Este peixe é considerado um bom modelo para o estudo de doenças humanas, pode servir como uma relevante plataforma para estudo de eventos moleculares, estratégias terapêuticas e avaliação dos mecanismos fisiológicos de algumas patologias. O Laboratório Experimental de Hepatologia e Gastroenterologia (LEHG), do Centro de Pesquisas do Hospital de Clínicas de Porto Alegre, iniciou em 2008 estudos na área da Hepatologia com o Zebrafish, aqui apresentados.


Danio rerio (zebrafish, paulistinha) is a freshwater small fish that has been used as an animal model for studies of several human diseases. Its characteristics of being easily bred and reproduced and its laboratory housing methods are well established. It requires some basic care and the acquisition of specific low-cost equipment. Since this fish has already been considered a good human disease model, it may serve as a powerful preclinical platform for the study of molecular events, therapeutic strategies, and for evaluating the physiological mechanisms of some pathologies. The Laboratory of Experimental Hepatology and Gastroenterology (LEHG) of the Research Center of Hospital de Clínicas de Porto Alegre initiated studies with the zebrafish in 2008, presented here.

ABSTRACT

The objective of this work was to evaluate the toxicity of the sediments of the Monjolinho River (São Carlos - São Paulo/Brazil), through partial chronic toxicity bioassays with juveniles of D. rerio and P. reticulata. Histological analyses of the gills and biometric measurements were conducted to detect the possible deleterious effects that caused the death. In all points the sampled the alterations were found in the gills (hyperplasia, fusion of secondary lamellas and dilation of blood vessels), with the most intense lesions (second stage) occurring at the point two. The biometric analysis pointed to inadequate conditions for the growth of the test-organisms when exposed to the sediment samples. These results showed that the conditions of these environments were not suitable for the survival and growth of these fishes.


Este trabalho teve como objetivo avaliar a toxicidade dos sedimentos do rio Monjolinho (São Carlos - São Paulo/Brasil), por meio de bioensaios de toxicidade crônicos parciais com juvenis de D. rerio e P. reticulata. Foram realizadas análises histológicas das brânquias dos organismos para detectar possíveis efeitos deletérios que antecedam a mortalidade. Em todos os pontos amostrados, foram verificadas alterações nas brânquias (hiperplasia, fusão de lamelas secundárias e dilatação de vasos sanguíneos), sendo no ponto dois foram encontradas lesões mais acentuadas (segundo estágio). A análise biométrica dos indivíduos expostos em amostras de sedimento demonstrou condições inadequadas para o crescimento e desenvolvimentos dos organismos-teste em todo o ambiente estudado. Os sedimentos apresentaram maior toxicidade no período chuvoso, sendo estes resultados confirmados por meio das análises histológicas das brânquias dos organismos expostos, as quais demonstraram lesões mais acentuadas no mesmo período. Tal situação possivelmente decorre dos impactos antrópicos os quais o sistema está suscetível (efluentes domésticos, industriais e agrícolas), que foram constatados pelas análises físicas e químicas dos sedimentos.

ABSTRACT

Toll-like receptors (TLRs) play a central role of innate immunity as well as of enhancing the adaptive immunity. TLR1 interacts with TLR2 to recognize the mycobacterial 19-kD lipoprotein. Danio rerio genomic sequences predicted TLR family genes. Danio rerio TLR1 gene was cloned with RACE-PCR and RT-PCR methods to get more sights of immunity against mycobacterial infection such as leprosy. Cloned TLR1 showed insertion of 22 amino acid residues comparing to NCBI- predicted TLR1, and an open reading frame composing of deduced 795 amino acids. Toll-interlukin1 receptor domain and leucine-rich repeats were in the cloned TLR1 as were noted in NCBI-predicted TLR1. Cloned zebrafish TLR1 had high similarity to human TLR1 with 35% amino acid identities and 57% amino acid positivities across the entire sequence (E=1e-130). The cloned sequence might help to study the innate and adaptive immunity of mycobacterial infection.
