ABSTRACT
Objective To investigate the potential roles of nucleotide -binding oligomerization domain (NOD)-like receptor in the pathogenesis of asthma.Methods Through rat asthma model,24 rats were randomly divided into three groups on average,named asthma group,control group and dexamethasone group.Expression levels of NOD1 and NOD2 mRNA were detected by Real -time PCR in lung tissues.Results The expression levels of NOD1 mRNA in the asthma group,control group and dexamethasone group were (0.62 ±0.34)RQ value,(1 .00 ± 0.00)RQ value,(0.65 ±0.33 )RQ value respectively.The levels of NOD1 mRNA in the asthma group was significantly lower than that in the control group(F =4.75,P 0.05).Moreover,expression levels of NOD2 mRNA in the asthma group,control group and dexamethasone group were (0.92 ±0.32)RQ value, (1 .00 ±0.00)RQ value,(1 .50 ±0.56)RQ value,respectively.There was no statistically significant difference of NOD2 mRNA level between the asthma group and control group (P >0.05 ),but level of NOD2 mRNA in the dexamethasone group was significantly higher than that in the asthma group(F =5.64,P 0.05).Conclusion Expression of NOD -like receptor subtype was not at the same level,and their reaction to dexamethasone were different either.

Objective: To prepare a novel slow-releasing ketoconazole spraying membrane and to observe its effect on eczema and tinea corporis in guinea pigs. Methods: A novel ketoconazole spraying membrane was prepared with chitosan chlorhydrate as the membrane material. The contents of ketoconazole and desamethasone in the spraying membrane were assayed by HPLC. The separation was performed on Waters Symttry C18 column (250 mm × 4.6 mm, 5 μm), with the mobile phase consisting methanol-potassium dihydrogen phosphate buffer (4:1), at a flow rate of 0.6 ml/min. The dectection was carried out with a UV detector at 239 nm, at (22±2) °C. The quality criteria for the novel ketoconazole spraying membrane was established. The curative effect of the ketoconazole spraying membrane on eczema and tinea corporis was observed in guinea pigs and was compared with those of spraying membrane matrix, desamethasone antipruritics cream and compound ketoconazole cream. Results: The calibration curves of ketoconazole and desamethasone were linear between 0.2-2.0 mg·ml-1 and 0.02-0.20 mg·ml-1, respectively; the recoveries for ketoconazole and desamethasone were 98.41% and 97.84%, respectively. The intra-day and extra-day RSDs for ketoconazole were 1.17% and 1.26%, respectively, and for desamethasone were 1.57% and 1.40%, respectively. Stimulation test showed that the spraying membrane had no irritative effect on the skin of guinea pig. The novel ketoconazole spraying membrane had similar effect on eczema and tinea corporis as those of desamethasone antipruritics cream and compound ketoconazole cream. Conclusion: This novel ketoconazole sprayi ng membrane is easy to prepare and has obvious therapeutic effects on eczema and tinea corporis in guinea pigs, making it favourable for containing combined agents.

Objective:To explore the pretection of small dosage desamethasone to pulmonary function on acute lung injury model.Methods:54 male SD rats were divided randomly into control group,injury group and therapeutic group.Major observed marker consisted of count of cell sum,total protein level and TNF-? level of bronchoalveolar douche solution.Secondary marker consisted of lung coefficient,PAO2,TNF-? level in blood serum and histopathologic examination.Results: Alveolar texture in therapeutic group existed yet,but alveolar interval increased thickness,neutrophilic granulocyte infiltration relieved obviously,and a little bleeding in alveolar cavity.PAO2 in therapeutic group was higher than that of injured group(P

OBJECTIVE:To establish an approach to determination of ketoconazole and desamethasone acetate in novel ketoconazole spraying film.METHODS:Chromatography was performed using a Waters Symmetry C 18 .The mobile phase consisted of methyl-monopotassium phosphate(4∶1).Detection wave was at239nm.The flowing rate was0.6ml/min.RESUL_ TS:Excellent linear correlations were showed when the concentrations of ketoconazole and desamethasone acetate were in the ranges of0.2~2mg/ml and0.02~0.2mg/ml respectively.The mean recoveries of ketoconazole and desamethasone acetate were98.41%(RSD=2.29%,n=6)and97.84%(RSD=2.60%,n=6)respectively.CONCLUSION:The established approach is precise,reliable and therefore applicable to the quality control of novel ketoconazole spraying film.
