ABSTRACT

The objective of the present study is to point out tensions of the theoretical/practical universe that Bioethics is facing in Brazil, in the search for a praxis for the destination of surplus embryos. We consider that Bioethics analyzes the implications of such practices in society and in relationships between individuals. Brief data from other countries were presented to compare the Brazilian situation progressively from the conceptual point of view and the adoption of measures. The research is a scoping review on the main points that have been hindering the progress of discussions on the subject and consequently the respective solution. The legal status of the embryo was described from several perspectives and theories, with the resulting proposals for the destination of surplus embryos in their positive and negative aspects. The tensions of Bioethics were presented in the context of post-modernity and the consequent social and moral plurality, together with the difficulties of identifying a secular bioethical morality. In the end, we conclude the possibility of proclaiming a consensus on the destination of surplus embryos based on secular morality, supported by the figure of the "moral strangers".


El objetivo del presente estudio es señalar las tensiones del universo teórico/práctico que la bioética enfrenta en Brasil, en la búsqueda de una praxis para el destino de los embriones sobrantes. Consideramos que la bioética analiza las implicaciones de tales prácticas en la sociedad y en las relaciones entre los individuos. Se presentaron breves datos de otros países para comparar progresivamente la situación brasileña desde el punto de vista conceptual y de la adopción de medidas. La investigación es una revisión del alcance de los principales puntos que han obstaculizado el avance de las discusiones sobre el tema y, en consecuencia, la respectiva solución. Se describió el estatuto jurídico del embrión desde diversas perspectivas y teorías, con las consiguientes propuestas para el destino de los embriones sobrantes en sus aspectos positivos y negativos. Se presentaron las tensiones de la bioética en el contexto de la posmodernidad y la consecuente pluralidad social y moral, junto con las dificultades de identificar una moral bioética laica. Al final, se concluye la posibilidad de proclamar un consenso sobre el destino de los embriones sobrantes basado en una moral laica, apoyada en la figura de los "extraños morales".


O objetivo do presente estudo é destacar as tensões do universo teórico/prático que a Bioética está enfrentando no Brasil, na busca de uma praxis para a destinação de embriões excedentes. Nós consideramos que a Bioética analisa as implicações de tais práticas na sociedade e nas relações entre indivíduos. Dados resumidos de outros países são apresentados para comparar a situação brasileira progressivamente de um ponto de vista conceitual para a adoção de medidas. A pesquisa é uma revisão de escopo sobre os pontos principais que vem atrapalhando o andamento das discussões sobre o assunto e consequentemente a solução respectiva. O status legal do embrião foi descrito a partir de diversas perspectivas e teorias, com as propostas resultantes para a destinação dos embriões excedentes em seus aspectos positivos e negativos. As tensões da Bioética foram apresentadas no contexto da pós-modernidade e a consequente pluralidade social e moral, juntamente com as dificuldades de identificar uma moralidade bioética secular. Ao final, nós concluímos pela possibilidade de proclamar um consenso sobre a destinação de embriões excedentes baseado na moralidade secular, apoiado pela figura dos "estranhos morais".

ABSTRACT

OBJECTIVE@#To explore the effects of Zishen Yutai Pills (ZYPs) on the quality of oocytes and embryos, as well as pregnancy outcomes in patients with diminished ovarian reserve (DOR) receiving in vitro fertilization-embryo transfer (IVF-ET). The possible mechanisms, involving the regulation of bone morphogenetic protein 15 (BMP15) and growth differentiation factor 9 (GDF9), were also investigated.@*METHODS@#A total of 120 patients with DOR who underwent their IVF-ET cycle were randomly allocated to 2 groups in a 1:1 ratio. The patients in the treatment group (60 cases) received ZYPs from the mid-luteal phase of the former menstrual cycle by using gonadotropin-releasing hormone (GnRH) antagonist protocol. The patients in the control group (60 cases) received the same protocol but without ZYPs. The primary outcomes were the number of oocytes retrieved and high-quality embryos. Secondary outcomes included other oocyte or embryo indices as well as pregnancy outcomes. Adverse events were assessed by comparison of the incidence of ectopic pregnancy, pregnancy complications, pregnancy loss, and preterm birth. Contents of BMP15 and GDF9 in the follicle fluids (FF) were also quantified with enzyme-linked immunosorbent assay.@*RESULTS@#Compared with the control group, the numbers of oocytes retrieved and high-quality embryos were significantly increased in the ZYPs group (both P<0.05). After treatment with ZYPs, a significant regulation of serum sex hormones was observed, including progesterone and estradiol. Both hormones were up-regulated compared with the control group (P=0.014 and 0.008), respectively. No significant differences were observed with regard to pregnancy outcomes including implantation rates, biochemical pregnancy rates, clinical pregnancy rates, live birth rates, and pregnancy loss rates (all P>0.05). The administration of ZYPs did not increase the incidence of adverse events. The expressions of BMP15 and GDF9 in the ZYPs group were significantly up-regulated compared with the control group (both P<0.05).@*CONCLUSIONS@#ZYPs exhibited beneficial effects in DOR patients undergoing IVF-ET, resulting in increments of oocytes and embryos, and up-regulation of BMP15 and GDF9 expressions in the FF. However, the effects of ZYPs on pregnancy outcomes should be assessed in clinical trials with larger sample sizes (Trial reqistration No. ChiCTR2100048441).

Abstract Background Infertility is caused by heterogeneous risks, but most of them are unexplained. The sperm DNA Fragmentation Index (DFI) was increasingly acknowledged as a parameter for the evaluation of male infertility. This study aimed to investigate the association between sperm DFI and laboratory and clinical outcomes in a population with unexplained infertility. Methods The clinical data of an infertile population was collected for the selection of reproductive patients with unexplained infertility. The authors classified the patients with normal sperm parameters in a control group (DFI < 25%) and an observation group (DFI ≥ 25%) and compared the difference in basal characteristics, laboratory, and clinical outcomes between the two groups. The authors conducted a correlation analysis to examine the relationship between DFI and the number of D3 good-quality embryos, as well as the clinical pregnancy rate and live birth rate. A total of 176 cases were enrolled in the retrospective study. Results The observation group (n = 88) showed advanced male age, lower sperm concentration, progressive motility, and morphology assessment than the control group. In addition, lower No. of D3 good-quality embryos, clinical pregnancy rate, and the live birth rate were shown in the observation group. A negative correlation between the DFI and No. of D3 good-quality embryos (rs = -0.347, p < 0.001) or live birth rate (rs = -0.185, p = 0.028) was shown. Conclusions Sperm DFI was a good indicator for the prediction of D3 good-quality embryos in unexplained infertility couples, but it did not provide sufficient information regarding clinical pregnancy outcome but live pregnancy outcome.

ABSTRACT

Background: African bush mango tree is an important fruit plant with high nutritional, medicinal, and commercial values. However, its seedling system remained a deep understanding. This study aimed to evaluate the effect of topophysis and growth regulators on the reactivity of different types of buds and zygotic embryos of wild mango. Methodology: Ripe fruits from two local varieties (Wossro and Sissro) were pulped. The nuts were extracted and dried for one week under greenhouse. Zygotic embryos were excised from nuts and disinfected with the bleach solution (NaClO 10% with 8% active chloride) for 10 min following by three successive rinsing using sterilized water. The second set of nuts was cultivated for under greenhouse in the polybags containing the sand. One month later, buds from different positions (apical, axillary and cotyledonary) were excised and disinfected with NaClO 10% for 10 min follow by the immersion in a mercuric chloride 0.1% added with two drops of Tween 20 especially for axillary and cotyledonary buds for 5 min while 0.01% of mercuric chloride was used for apical buds. The explants were cultured on 糓S and 絎PM media supplemented with BAP, KIN at different concentrations (0.2 mg/L and 3.5 mg/L respectively) and NAA (0.05 mg/L). Results: The best sprouting rate was obtained with the variety Wossro which showed a high bud break rate (26.47%) against (9.88%) for the variety Sissro. The topophysis significantly influenced the response of the buds in tissue culture. 24.48% of axillary buds were sprouted on � MS medium + 3.5 mg/L BAP+ 0.05mg/L NAA. The in vitro germination of embryos was significantly (p? 0.05) influenced by the genotype of the variety. 50.76% of buds were germinated for Wossro while 18.32% were germinated for Sissro. � MS + 0.2 mg/L KIN has significant influenced the plants growth and development. Implication: The findings will help to improve the regeneration rate and plantlets production of African bush mango tree.

Resumen: Objetivo. Analizar las percepciones y prácticas de los clínicos en relación con el manejo del embrión sometido a técnicas de fecundación in vitro. Metodología. Cualitativa (subjetivista y fenomenológico). Se realizaron 15 las entrevistas semiestructuradas aplicando un muestreo por saturación dirigidas a personal clínico que haya participado en procedimientos de fecundación in vitro. Los datos se analizaron con el programa Atlas Ti 8.0®. Resultado. Los clínicos consideran al embrión como un ser humano o futuro ser humano y, además, merecedor de respeto y consideración, proponiendo incluso mejoras en los procesos de manipulación y almacenaje. Conclusión. Los embriones no son considerados como entes susceptibles de recibir daño, desde argumentos no solo técnicos sino éticos. Desde la corriente principialista, se describe la necesidad de promover actitudes de responsabilidad y prudencia para evitar el dogmatismo (objetivismo moral) proponiendo una postura deliberativa.


Abstract: The objective of this paper is to analyze the perceptions and practices of clinicians in relation to the management of embryos subjected to in vitro fertilization techniques. Methodology is Qualitative (subjectivist and phenomenological). A total of 15 semi-structured interviews were conducted using saturation sampling for clinical personnel who have participated in vitro fertilization procedures. The data is analyzed with the Atlas Ti 8.0® program. Results: Clinicians consider the embryo as a Human being or future human being, in addition, deserving of respect and consideration even proposing improvements in the processes of handling and storage. Conclusion. Embryos are not considered as entities susceptible of damage from not only technical but ethical arguments. From the principialist current, the need to promote attitudes of responsibility and prudence to avoid dogmatism (moral objectivism) is described, proposing a deliberative position.


Resumo: Objetivo. Analisar as percepções e práticas dos médicos em relação ao manejo do embrião submetido a técnicas de fertilização in vitro. Metodologia. Qualitativo (subjetivo e fenomenológico). Foram realizadas 15 entrevistas semiestruturadas por amostragem de saturação para pessoal clínico que participou de procedimentos de fertilização in vitro. Os dados são analisados com o programa Atlas Ti 8.0®. Resultado. Os médicos consideram o embrião como um ser humano ou futuro, além de merecer respeito e consideração, propondo até melhorias nos processos de manuseio e armazenamento. Conclusão. Os embriões não são considerados como entidades suscetíveis a receber danos não apenas de argumentos técnicos, mas éticos. A partir da corrente principialista, descreve-se a necessidade de promover atitudes de responsabilidade e prudência para evitar o dogmatismo (objetivismo moral), propondo uma posição deliberativa.

ABSTRACT

Generation of mutants with clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) is commonly carried out in fish species by co-injecting a mixture of Cas9 messenger RNA (mRNA) or protein and transcribed guide RNA (gRNA). However, the appropriate expression system to produce functional gRNAs in fish embryos and cells is rarely present. In this study, we employed a poly-transfer RNA (tRNA)-gRNA (PTG) system driven by cytomegalovirus (CMV) promoter to target the medaka (Oryzias latipes) endogenous gene tyrosinase(tyr) or paired box 6.1 (pax6.1) and illustrated its function in a medaka cell line and embryos. The PTG system was combined with the CRISPR/Cas9 system under high levels of promoter to successfully induce gene editing in medaka. This is a valuable step forward in potential application of the CRISPR/Cas9 system in medaka and other teleosts.

ABSTRACT

This study aimed to evaluate the ultrastructural morphometry of bovine embryos produced in vitro grown at different concentrations of antioxidants. After in vitro maturation and fertilization, the presumptive zygotes were assigned into five treatments. T1) without the addition of any antioxidants (negative control); T2) addition of 50µM/mL cysteamine; and T3, T4 and T5) adding 2.5µg/mL, 5.0µg/mL or 10.0µg/mL of the antioxidants derived from the oily extract from Lippia origanoides, respectively. On D7 of culture, the embryos in the blastocyst stage were fixed and prepared for electron transmission microscopy. These were evaluated for the proportion of cytoplasm-to-nucleus, cytoplasm-to-mitochondria, cytoplasm-to-vacuoles, cytoplasm-to-autophagic vacuoles and cytoplasm-to-lipid droplets. Blastocysts cultured in media containing oily extract of Lippia origanoides presented morphological characteristics such as high cell:mitochondria ratio and low cell:vacuoles and cell:autophagic vacuole ratio, possibly been morphological indicators of embryonic quality. Inner cell mass (ICM) from blastocysts cultured in media without any antioxidants had the highest cell:vacuole ratio. Similar results were found in the trophectoderm (TE) cells of blastocysts from treatment 2. Embryo culture media supplemented with antioxidants derived from Lippia origanoides oil produced embryos with a higher cytoplasmic proportion of organelles, such as mitochondria. Also, treatments without any antioxidants or with the addition of cysteamine presented cytoplasmic vacuolization, a characteristic related to production of poor-quality embryos.(AU)


Este estudo teve como objetivo avaliar a morfometria ultraestrutural de embriões bovinos produzidos in vitro e cultivados em diferentes concentrações de antioxidantes. Após a maturação e a fertilização in vitro, os possíveis zigotos foram divididos em cinco tratamentos: T1) sem adição de antioxidantes (controle negativo); T2) adição de 50µM/mL de cisteamina; e T3, T4 e T5) adição de 2,5µg/mL, 5,0µg/mL ou 10,0µg/mL dos antioxidantes derivados do extrato oleoso de Lippia origanoides, respectivamente. No D7 de cultivo, os embriões em estágio de blastocisto foram fixados e preparados para microscopia eletrônica de transmissão. Estes foram avaliados para a proporção entre citoplasma e núcleo, citoplasma e mitocôndria, citoplasma e vacúolos, citoplasma e vacúolos autofágicos e citoplasma e gotículas lipídicas. Blastocistos cultivados em meio contendo extrato oleoso de Lippia origanoides apresentaram características morfológicas como alta relação célula:mitocôndria e baixa relação célula:vacúolos e célula:vacúolo autofágico, possíveis indicadores morfológicos de qualidade embrionária. A massa celular interna (MCI) de blastocistos cultivados em meio sem quaisquer antioxidantes teve a maior razão célula:vacúolo. Resultados semelhantes foram encontrados nas células do trofectoderma (TE) de blastocistos do tratamento 2. Portanto, o meio de cultivo embrionário suplementado com antioxidantes derivados do óleo de Lippia origanoides produziu embriões com maior proporção citoplasmática de organelas, como mitocôndrias. Além disso, tratamentos sem antioxidantes ou com adição de cisteamina apresentaram vacuolização citoplasmática, característica relacionada à produção de embriões de baixa qualidade.(AU)

ABSTRACT

As vantagens dos animais transgênicos têm sido demonstradas em diferentes aplicações, entretanto muitas metodologias usadas para gerar animais geneticamente modificados (GM) apresentam baixas taxas de eficiência. O objetivo deste estudo foi avaliar a entrega dos vetores lentivirais (VLs) em zigotos durante a fertilização in vitro (FIV), para gerar embriões GM, com o gene da proteína verde fluorescente (GFP) ou do fator IX de coagulação humana (FIX). Vetores lentivirais com os genes GFP (pLGW-GFP-LV) ou FIX (pLWE2-FIX-LV) foram utilizados na FIV ou na cultura de embriões in vitro (CIV). A coincubação de pLWE2-FIX-LV com espermatozoides e complexos oócitos-células do cumulus (COCs) durante a FIV diminuiu (P<0,05) as taxas de clivagem e de blastocistos, enquanto com pLGW-GFP-LV diminuiu (P<0,05) a taxa de blastocisto quando se comparou ao controle sem VL. A coincubação de pLWE2-FIX-LV e pLGW-GFP-LV com presumíveis zigotos durante a CIV não afetou (P>0,05) o desenvolvimento embrionário. A expressão da proteína GFP não foi detectada em embriões após a coincubação de FIV ou CIV, embora as células do cumulus expressassem a proteína até o dia oito de cultivo in vitro. Reações em cadeia da polimerase (PCR) não detectaram os genes GFP ou FIX em embriões, mas ambos foram detectados em células do cumulus. Assim, a coincubação de VL com espermatozoide bovino e COCs não é eficaz para produzir embriões geneticamente modificados por meio de FIV.(AU)

ABSTRACT

Objective@#To investigate the developmental toxicity of 2,4-dinitrochlorobenzene in zebrafish embryos.@*Methods@#AB wild-type male and female zebrafish were selected to mate and spawn, then the eggs were cultured with Holt buffer solution. Six dose groups ( 0.4, 0.8, 1.0, 1.2, 1.6, 2.0 μg/mL ), a solvent control group and a cosolvent control group, were set up with 20 embryos each. Malformations and death of embryos were observed at 48, 72 and 96 hpf ( hours post fertilization ), the mortality and 50% lethal concentration ( LC50 ) were also calculated. @*Results@#At 48, 72 and 96 hpf, the LC50 of 2,4-dinitrochlorobenzene on zebrafish embryos were 1.668, 1.043 and 0.895 μg/mL, respectively, with a downward trend. After 72 hpf, when the concentration reached 2.0 μg/mL, all the zebrafish died. In the range of 0.4-2.0 μg/mL, the mortality of zebrafish at 48, 72 and 96 hpf increased with the increase of 2,4-dinitrochlorobenzene concentration ( all P<0.05 ); the malformation rate of zebrafish embryos at 48 hpf increased with the increase of 2,4-dinitrochlorobenzene concentration ( P<0.05 ). Zebrafish embryos exposed to 2,4-dinitrochlorobenzene led to yolk sac edema, pericardial edema and spinal curvature. @*Conclusion@#2,4-dinitrochlorobenzene can affect the development of zebrafish embryos, which will lead to lethal and teratogenic effects.

ABSTRACT

The family of paired box (Pax) genes encodes the transcription factors that have been emphasized for the particular importance to embryonic development of the CNS, with the evidence obtained from various animal models. Human embryos have rarely been available for the detection of the expression of Pax family members. In this study 32 human embryos of Carnegie (CS) stages 10-20 were investigated to find the differences in the expression of Pax6 and Pax7 proteins in different regions of the neural tube and the caudal spinal cord. The expression of Pax6 and Pax7, as determined by immunohistochemistry, showed a tendency to increase in the later stages of the development both in the spinal cord and the brain. Significantly weaker expression of Pax6 and Pax7 was observed at CS 10 as compared to the later stages. At CS 10-12 weak expression of Pax6 was noticed in both dorsal and ventral parts of the developing spinal cord, while the expression of Pax7 was restricted to the cells in the roof plate and the dorsal part of the spinal cord. At CS 14-20 in the developing spinal cord Pax6 and Pax7 were detected mostly in the neuroepithelial cells of the ventricular layer, while only weak expression characterized the mantle and the marginal layers. At the same stages in the developing brain Pax6 and Pax7 were expressed in the different regions of the forebrain, the midbrain and the hindbrain suggesting for their involvement in the differentiation of neurons in specific parts of the developing brain.


La familia de genes Pax del inglés (Paired box) codifica los factores de transcripción debido a la particular importancia en el desarrollo embrionario del SNC, con la evidencia obtenida de varios modelos animales. Rara vez han estado disponibles embriones humanos para la detección de la expresión de genes de la familia Pax. En este estudio, se investigaron 32 embriones humanos de Carnegie (CS) etapas 10-20 para encontrar las diferencias en la expresión de las proteínas Pax6 y Pax7 en diferentes regiones del tubo neural y la médula espinal caudal. La expresión de Pax6 y Pax7, según la inmunohistoquímica, se observó una tendencia a aumentar en las etapas posteriores del desarrollo, tanto en la médula espinal como en el cerebro. Se observó una expresión significativamente más débil de Pax6 y Pax7 en CS 10 en comparación con las etapas posteriores. En CS 10-12 se notó una expresión débil de Pax6 en las partes dorsal y ventral de la médula espinal en desarrollo, mientras que la expresión de Pax7 se limitó a células en la placa del techo y dorsal de la médula espinal. En CS 14-20 en la médula espinal en desarrollo, Pax6 y Pax7 se observó principalmente en las células neuroepiteliales de la capa ventricular, mientras que expresión débil se caracterizó en las capas marginales. En las mismas etapas en el cerebro en desarrollo, Pax6 y Pax7 se expresaron en las diferentes áreas del prosencéfalo, el mesencéfalo y el mesencéfalo, lo que sugiere su participación en la diferenciación de las neuronas en partes específicas del cerebro en desarrollo.

ABSTRACT

Background: Plant tissue cultures have the potential to reprogram the development of microspores from normal gametophytic to sporophytic pathway resulting in the formation of androgenic embryos. The efficiency of this process depends on the genotype, media composition and external conditions. However, this process frequently results in the regeneration of albino instead of green plants. Successful regeneration of green plants is affected by the concentration of copper sulfate (CuSO4) and silver nitrate (AgNO3) and the length of induction step. In this study, we aimed at concurrent optimization of these three factors in barley (Hordeum vulgare L.), wheat (Triticum aestivum L.), and triticale (x Triticosecale spp. Wittmack ex A. Camus 1927) using the Taguchi method. We evaluated uniform donor plants under varying experimental conditions of in vitro anther culture using the Taguchi approach, and verified the optimized conditions. Results: Optimization of the regeneration conditions resulted in an increase in the number of green regenerants compared with the control. Statistic Taguchi method for optimization of the in vitro tissue culture plant regeneration via anther cultures allowed reduction of the number of experimental designs from 27 needed if full factorial analysis is used to 9. With the increase in the number of green regenerants, the number of spontaneous doubled haploids decreased. Moreover, in barley and triticale, the number of albino regenerants was reduced. Conclusion: The statistic Taguchi approach could be successfully used for various factors (here components of induction media, time of incubation on induction media) at a one time, that may impact on cereals anther cultures to improve the regeneration efficiency

Resumen: OBJETIVO: Comparar la tasa de recién nacido vivo con la de embarazo clínico-transferencia embrionaria, las características clínicas y concentraciones hormonales entre dos esquemas de preparación endometrial para transferencia de embriones desvitrificados: con un agonista de GnRH versus su sustitución con dosis altas de estrógenos. MATERIALES Y MÉTODOS: Estudio observacional, de cohorte histórica, efectuado en pacientes atendidas en el Instituto Nacional de Perinatología en protocolo FIV-ICSI entre enero 2017-marzo 2019. Se compararon dos esquemas de preparación endometrial: grupo A con agonista de GnRH y grupo B con estradiol a dosis de 8 mg al día sin agonista de GnRH. A todas las pacientes se les tomaron muestras de sangre para determinación de FSH, LH, estradiol y progesterona y se dio seguimiento ultrasonográfico durante la preparación endometrial. RESULTADOS: En 99 pacientes entre 23 y 42 años, con embriones congelados, la tasa de recién nacido vivo-transferencia embrionaria fue, respectivamente, de 17.2 y 8.6% (OR 1.98; IC95%: 077-1.53) y la de embarazo clínico de 26.5 y 22.8% (OR1.09; IC95%: 0.77-1.53) en los grupos A y B, respectivamente. La duración total del ciclo, los días con estrógenos y la concentración sérica de estradiol al inicio de la progesterona alcanzaron diferencia significativa entre los grupos. No se detectaron datos clínicos ni de laboratorio sugerentes de ovulación. CONCLUSIONES: La tasa de recién nacido vivo fue mayor con agonistas de GnRH sin alcanzar significación; sin embargo, la preparación endometrial solo con estrógenos es un esquema seguro, de menor costo y más amigable para la paciente y el médico.


Abstract: OBJECTIVE: To compare the rate of live newborn with clinical pregnancy-embryo transfer, clinical characteristics and hormonal concentrations between two endometrial preparation schemes for devitrified embryo transfer: with a GnRH agonist (aGnRH) versus its replacement with high doses of estrogens. MATERIALS AND METHODS: Historical cohort observational study conducted in patients treated at the National Institute of Perinatology under IVF-ICSI protocol between January 2017-March 2019. Two endometrial preparation schemes were compared: group A with GnRH agonist and group B with estradiol at 8 mg per day without aGnRH. All patients had blood samples taken for FSH, LH, estradiol and progesterone determination and ultrasonographic follow-up during endometrial preparation. RESULTS: in 99 patients between 23 and 42 years of age, with frozen embryos, the live birth-embryo transfer rate was 17.2 and 8.6%, (OR 1.98; CI95%: 077-1.53) and the clinical pregnancy rate was 26.5 and 22.8% (OR1.09; CI95%: 0.77-1.53) in groups a and b, respectively. total cycle length, estrogen days and serum estradiol concentration at the beginning of progesterone reached significant difference between groups. no clinical or laboratory data suggesting ovulation were detected. CONCLUSIONS: The rate of live newborn was higher with GnRH agonists without reaching significance; however, endometrial preparation with estrogens alone is a safe, lower cost and more patient- and physician-friendLy scheme.

Introduction: The description given in various textbooks and literature on development of nephrons in humankidney doesn’t include details of chronology of nephrogenic events at various fetal ages. Though several studieswere reported the knowledge on development of kidney especially on the nephrogenesis are limited. The studiesemphasize the relationship between prenatal development of kidney and adult onset of renal diseases. Hence, anattempt was made in this study to obtain information by observing the serial sections of kidney of embryos andfoetuses of different gestational ages for better understanding of nephrogenic events.Material and methods: Thirty-five aborted embryos and dead fetuses of 5 weeks gestational age to full term wereutilized for this study. The specimens were subjected to routine tissue processing and haematoxylin and eosin(H&E) staining. 5 embryos of less than 8 weeks gestational age were processed as a whole and were seriallysectioned. The histological sections of 5 microns thickness were observed for the time of appearance of variousnephrogenic components and photographed.Results: Differentiating pronephric, mesonephric and metanephric components in different weeks i.e. 05 – 12, 13– 24, 25-36 were studied. In 06 – 12 weeks group a delay in the appearance of Pro and mesonephric, Meso andmetanephric ducts were observed that appeared during the 6th week. Differentiation of other components havenot completed by 6th week when compared with literature. In 13 – 24 weeks also there is delay in corticomedullary differentiation that was observed at 16 wks. at which time the morphologically recognizable Nephronswere also observed. Major part of development occurred between 16-28 weeks instead of 16-24weeks as statedin the literature. Ampulla division continued beyond 24 weeks. Increased number of mature nephrons wereobserved between 24-28 weeks instead of 16-20 wks., nephron arcades were observed during 24-28 weeksinstead of 14-22weeks.Conclusion: Detailed findings of this study could aid the embryologists, neonatologists and nephrologists tounderstand the chronology of nephrogenic events and related consequences of developmental abnormalities.

ABSTRACT

OBJECTIVE: To determine the clinical pregnancy (CP) and live birth (LB) rates arising from frozen embryo transfers (FETs) that had been generated under the influence of in vitro fertilization (IVF) adjuvants given to women categorized as poor-prognosis.METHODS: A registered, single-center, retrospective study. A total of 1,119 patients with first FETs cycle include 310 patients with poor prognosis (109 treated with growth hormone [GH], (+)GH group vs. 201 treated with dehydroepiandrosterone, (–)GH group) and 809 patients with good prognosis (as control, (–)Adj (Good) group).RESULTS: The poor-prognosis women were significantly older, with a lower ovarian reserve than the (–)Adj (Good) group, and demonstrated lower chances of CP (p<0.005) and LB (p<0.005). After adjusting for confounders, the chances of both CP and LB in the (+)GH group were not significantly different from those in the (–)Adj (Good) group, indicating that the poor-prognosis patients given GH had similar outcomes to those with a good prognosis. Furthermore, the likelihood of LB was significantly higher for poor-prognosis women given GH than for those who did not receive GH (p<0.028). This was further confirmed in age-matched analyses.CONCLUSION: The embryos cryopreserved from fresh IVF cycles in which adjuvant GH had been administered to women classified as poor-prognosis showed a significant 2.7-fold higher LB rate in subsequent FET cycles than a matched poor-prognosis group. The women with a poor prognosis who were treated with GH had LB outcomes equivalent to those with a good prognosis. We therefore postulate that GH improves some aspect of oocyte quality that confers improved competency for implantation.

Objective:Taking zebrafish embryos as research model, to investigate the toxic effect of different polar fractions of Euphorbiae Pekinensis Radix before and after processing with vinegar on heart. Method:Zebrafish embryos with normal development at 12 h after fertilization were treated with petroleum ether, dichloromethane and ethyl acetate extracts of Euphorbiae Pekinensis Radix before and after processing with vinegar for observation of cardiac development and function at 72 h. Result:Various polar fractions of Euphorbiae Pekinensis Radix before and after processing with vinegar had the cardiotoxicity on zebrafish embryos in a dose-dependent manner. In addition, the cardiotoxicity of different polar fractions was followed by petroleum ether, dichloromethane and ethyl acetate. The cardiotoxicity was mainly manifested as slow cardiac development, pericardial edema, decrease of heart rate and apoptosis of cardiac cells. Compared with the corresponding polar fraction of raw products, the cardiotoxicity of the same polar fraction of vinegar-processed products with similar doses decreased. Conclusion:Euphorbiae Pekinensis Radix has cardiotoxicity to zebrafish embryos and the cardiotoxicity is reduced after processing with vinegar, which can provide some experimental basis for further elucidation of the detoxication mechanism of Euphorbiae Pekinensis Radix processed with vinegar.

ABSTRACT

To study the effects of different fraction of Euphorbiae Pekinensis Radix before and after processing with vinegar on liver and gastrointestinal toxicity of zebrafish embryos,the zebrafish embryos after fertilized 12 h(12 hpf) were exposed to different concentrations of solution until 96 h(96 hpf),for observation of the toxicity response of the liver and gastrointestinal of individual zebrafish embryos. The results showed that toxicity increased in a dose-dependent manner. The liver and gastrointestinal toxicity of the zebrafish embryos in various polar fractions of Euphorbiae Pekinensis Radix before and after processing with vinegar was mainly manifested as slow liver development,smaller liver area,edema of yolk sac,delayed absorption,slowing of gastrointestinal motility,abnormal function of gastrointestinal goblet cell secretion. In addition,the toxicity of different polarity was followed by petroleum ether,dichloromethane,ethyl acetate. The above results indicated that the toxicity was reduced after processing with vinegar,and the fractions of petroleum ether and methylene chloride were the main sites responsible for liver and gastrointestinal toxicity.

Abstract The influence of silver nitrate (AgNO3), benzyladenine (BAP), and indole-3-acetic acid (IAA) on low frequency somatic embryogenesis (LFSE) induction in Caturra and Catuaí arabica coffee was evaluated. For the Caturra cultivar, the production of somatic embryos was significantly increased by adding AgNO3 to the semisolid culture medium. The highest average number of somatic embryos for this cultivar was obtained using 6.6 μM BAP, 2.85 μM IAA, and 40 μM AgNO3. In contrast, for the Catuaí cultivar, the highest average number of somatic embryos was obtained using semisolid medium supplemented with 8.8 μM BAP, and 2.85 μM IAA. Using these protocols, somatic embryos were directly induced using leaf sections of in vitro plants of both coffee cultivars within 8 weeks. The somatic embryos developed into rooted plants with a 100% survival rate upon transfer to the greenhouse.

ABSTRACT

The introduction of new strains of mice in specific pathogen-free (SPF) animal facilities should be performed carefully to avoid breaking sanitary barriers. To meet this need, animals should be rederived to reduce infection risk and thus avoid research interference caused by loss of animal health status and welfare. The objective of this study was to implement mice embryo transfer in the laboratory mouse facility of the Department of Immunology at the Institute of Biomedical Sciences/University of São Paulo, Brazil. Embryo transfers were performed to rederive genetically modified mouse strains with undefined sanitary status, received from different research and educational institutions. Fertilized eggs at two-cell stage were obtained by natural means and transferred into the oviducts of SPF pseudo-pregnant female mice. All surgical procedures were performed under aseptic conditions. A total of 625 embryos were transferred into the recipients. 148 pups were born, of which 140 were reared. Viruses, bacteria and intestinal protozoa were eliminated using this technique. The improvement in the microbiological status of mice allowed their expansion in our SPF facility. With these results, we can stimulate the use of embryo transfer technique between rodent facilities in Brazil and thus encourage the distribution of better models to our scientific community.(AU)


A introdução de novas linhagens de camundongos em biotérios livres de patógenos específicos (SPF) deve ser realizada com critérios para evitar a quebra das barreiras sanitárias. Dessa forma, os animais devem ser rederivados para reduzir os riscos de infecção e evitar as interferências provocadas pela perda do status sanitário e do bem-estar dos animais. O objetivo deste estudo foi implementar a transferência de embriões murinos no Biotério do Departamento de Imunologia do Instituto de Ciências Biomédicas da Universidade de São Paulo, Brasil. As transferências embrionárias foram realizadas para rederivar linhagens de camundongos geneticamente modificadas com status sanitário não conhecido, recebidas de diferentes instituições de pesquisa e de ensino. Os embriões em duas células foram obtidos pelos métodos naturais e transferidos para os ovidutos de fêmeas de camundongos SPF pseudoprenhas. Todos os procedimentos cirúrgicos foram realizados sob condições assépticas. Um total de 625 embriões foram transferidos para as receptoras. Foram obtidos 148 filhotes nascidos vivos, destes 140 foram desmamados. Por meio desta técnica, foram eliminados vírus, bactérias e protozoários intestinais. A melhora no status microbiológico dos camundongos permitiu a expansão destes em nossa colônia SPF. Com esses resultados, podemos promover o uso da técnica de transferência de embriões entre os biotérios brasileiros e assim incentivar a distribuição de modelos mais adequados para a nossa comunidade científica.(AU)

ABSTRACT

This study was carried out in the animal house laboratories of the Department of biology in the Faculty of Education for Girls/University of Kufa from 1/6/2018 until 1/8/2018, for the purpose of demonstrating the possible preventive effects of vitamin C against skeletal malformations in embryos in pregnancy for 20 days (prenatal) and postnatal newborns respectively for pregnant female rats that treated with indomethacin with a concentration of 8.40 mg/kg. In the current study, 40 female rats (aged 12 weeks) with a mean weight of (135.5 g) were employed and 40 fertile males with age (12 weeks). The females were placed with males for matting, and after obtaining the required number of pregnant female rats, which were divided into (4) main groups, each group contained 10 pregnant female rats. The first group was treated with normal saline that was represented the control group, while the second group was administrated indomethacin drug with a concentration of 8.40 mg/kg only, whereas the third group was subjected to vitamin C with concentration of 85 mg/kg only, and the fourth group was submitted to vitamin C with concentration of 85 mg/kg + indomethacin drug with a concentration of 8.40 mg/kg. Moreover, all the females in the four groups were orally administrated with a special administration tube(gavage) from the first day of pregnancy until delivery and by a dose every day, (5) female rats were explained during pregnancy for 20 days(prenatal), while the remaining (5) female rats were left in each major group to postpartum, the weights of prenatal fetuses and neonatal were measured, as well as skeletal structures(bone skeletons) of prenatal fetuses and postnatal were cleared for studying the structural skeletal changes and abnormalities. The results of this study showed that there was a significant decrease (P 0.05) between these two groups. Also, the findings of present study pointed to a significant decrease (p 0.05)were revealed when the groups that treated with vitamin C with a concentration of 85 mg/kg and the groups of vitamin C with a concentration of 85 mg/kg + indomethacin drug with a concentration of 8.40 mg/kg were compared with each other. Furthermore, the data did not show any skeletal malformations of the fetuses during pregnancy for (20) days (prenatal)and postpartum births in the control groups and groups that orally given vitamin C with a concentration of 85 mg/kg, as well as the groups of vitamin C with a concentration of 85 mg/kg + indomethacin drug with a concentration of 8.40 mg/kg respectively in comparison to the other experimental groups submitted to indomethacin drug with a concentration of 8.40.1 mg/kg which showed various skeletal abnormalities were observed in the fetuses during pregnancy for (20) days (prenatal)and postpartum births in these groups when compared with the control groups and the other study groups.

ABSTRACT

The reproductive cloning in buffalo in India has been started using a simplified somatic cell nuclear transfer technique named handmade cloning. Since the birth of first cloned female buffalo in 2009, a number of buffalo clones have been produced in India by utilizing different types of donor cells such as ear cells, embryonic stem cells, semen somatic cells and urine somatic cells. The use of buffalo cloning on a large scale is restricted due to low pregnancy rates and poor calf survival. Considerable attempts have been made to improve the overall buffalo cloning efficiency, particularly by modifying epigenetic reprogramming of cloned embryos. Previous studies have demonstrated that chemical epigenetic modifiers such as trichostatin A and 5-aza-2'-deoxycytidine, m-carboxycinnamic acid bishydroxamide can be used to treat donor somatic cells and reconstructed fused embryos to correct the epigenetic reprogramming to enhance the overall cloning efficiency in terms of live birth rates.
