ABSTRACT

AIM: To explore the way and the feasible time of olfactory ensheathing cells (OECs) transplanting to experimental autoimmune encephalomyelitis (EAE) rats, and to investigate the migration of OECs after transplanted to EAE and the possible mechanism of inducing the protective effect. METHODS: The Lewis rats, which were divided into MOG group and GPSCH group according to the induction by MOG~Igd and GPSCH separately, were used to made EAE model. The animals in MOG group were divided into 3 subsets: OECs blank group (MOG0 group, 4 rats );OECs transplantation by vena caudalis (MOG1 group, 7 rats);OECs transplantation by lateral cerebral ventricle (MOG2 group, 4 rats). The animals in GPSCH group were also divided into 2 subsets: OECs blank group (GPSCH0 group, 4 rats);OECs transplanted by vena caudalis (GPSCH1 group, 4 rats). OECs transplanted through different ways in peak incidence, then the rats were measured to determine whether the symptom was ameliorated. Two weeks after transplantion, the rats were killed and the methods of histofluorescence and histopathology (HE staining and Luxol fast blue staining) were used to examine the distribution of the labeled OECs in EAE rats' bodies and to explore whether there was some amelioration in histology. RESULTS: After OECs transplantation by vena caudalis or lateral cerebral ventricle, the rats' symptom improved. Compared to OECs blank groups, there was significant difference in clinical scores (F=18.470, P<0.01;t=-7.147, P<0.01). No significant difference between MOG1 group and MOG2 group was observed (P>0.05). The labeled OECs entered into the brain through the broken blood-brain-barrier after transplantation by vena caud-alis, the labeled OECs near the subpial area and around the lesions were found. OECs transplantation by the way of lateral cerebral ventricle showed great migration potential, the cells moved towards to the lesions extensively. No significant difference between OECs transplantation group and OECs blank group in histopathology (HE staining and LFB staining) was observed (P>0.05), and the same thing also happened between MOG1 group and MOG2 group (P>0.05). CONCLUSION: The transplantation of adult rats' OECs improves the EAE rats' symptom at some degree by the ways of vena caudalis or lateral cerebral ventricle.

ABSTRACT

Objective To explore the mechanism of IL-6 in EAE. Methods 25 female Wistar rats were randomly divided into 2 groups, EAE group( n=15 ) and normal control group( n=10). The expression of IL-6, TNF-αtand TGF-β of the two groups rots were ob-served by immunohistochemistry staining, and then correlation analysis of the expression of IL-6, TNF-αtandTGF-β was made. Results There was negative correlation between gray scale of IL-6 and symptom scores( r=-0.953, P>0.05). IL-6 and TGF-β had no statistical-ly significant correlation ( r=-0.492, P>0.05). There was negative correlation between gray scale of TNF-α and symptom scores( r=-0.978, P<0.05), and both of the correlation between TNF-α and TGF-β ( r=-0.502, P>0.05), TGF-β and scores( r=0.470,P>0.05 ) were not statistically significant. Conclusions IL-6 may participate in EAE as a inflammatory factor.

ABSTRACT

Objective To study the effects of 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) on experimental allergic encephalomyelitis (EAE) and its immune-modulating mechanisms. Methods 1,25-(OH)2D3 were given to Lewis rats immunized with myelin basic protein (MBP) in complete Freud’s adjuvant (CFA) respectively on the same day of immunization (the prevented group) and the presenting day of symptoms (the treated group). Then clinical symptoms were scored until the 13th day after immunization, when rats were sacrificed. Mononuclear cells (MNC) in draining lymph nodes were collected for the following studies: (1)Total number of MNC in the lymph nodes were counted. (2) Numbers of CD~+4CD~+ 25 cells and CD~+ 86 cells were determinated by flow cytometry. (3) The concentrations of interferon(IFN)-? and interleukin(IL)-4 in cell culture supernatant were measured by enzyme-linked immuno sorbent assay (ELISA). Results 1,25-(OH)2D3 could delay the peak EAE severity in the prevented group while it could relieve the progression of EAE and lower the peak score in the treated group (3.3?0.6 vs 4.0?0.3, P

ABSTRACT

Objective To construct the genes expression profile database of experimental autoimmune encephalomyelitis (EAE) mice model and to screen the high candidate genes with microarray analysis. Methods Polymerase chain reaction (PCR) products on behalf of 4 096 genes from mixed mice tissue library were assembled on the modified Oako glass slide. The general RNA from 6 EAE mice model and 6 normal mice head was transcripted into cDNA by RT-PCR and labelled with cy-3 and cy-5.6 groups each consist one EAE mice and a control. These two probes were mixed and hybridized with the above mentioned gene chips. Scan array 4000 was used for scanning the hybridizing signals and GenePixPro 3.0 for date analysis. Results Of the total 4 096 genes monitored, 43 genes in group one showed changes in expression level of the ratio outside 0.5 and 2, 30 genes differed in group two, 176 genes changed in group three, 76 in group four, 294 in group five and 129 in group six. Conclusions The results showed the consistent different genes expression throughout the EAE mice model. The genes related to immune, cell structure, cell cycle, ion channel, signal transduction, protein synthesis and metabolism are involved in EAE pathogenesis. The results provide deeper insights into the mechanism of EAE and multiple sclerosis.

ABSTRACT

Objective To investigate the change of the concentration of IL-2?IL-6?IL-10 in peripheral blood and cerebral spinal fluid (CSF) in monkey's experimental allergic encephalomyelitis (EAE),as to explaining it's function in mechanism and prognosis of the disease. Methods First set up animal models of monkey's EAE. Then separate the animals by their onset symptoms according to the level and the different stages of disease. At the same time survey the blood and CSF concentrations of IL-2?IL-6?IL-10, finally process the findings statistically. Results IL-2 in the first or the third week in the acute stage of EAE (5.0?0.8 and 5.3?1.2,respectively) was obviously higher than that (0.7?0.3) before the onset,P=0.045,0.041 respectively,both IL-6 and IL-10 in recuperation or chronic stage were higher significantly than before the onset,P=0.004 3,0.006 5 respectively. The findings were significant statistically. Conclusion IL-2 up-regulates immunologically,which accelerates the disease. IL-6 and IL-10 down-regulate immunologically,which reduce the disease. The findings may play an important role in studying immunological mechanism of EAE or MS in the future.

ABSTRACT

AIM:To explore if there is lesion in the auditory path in the brainstem of the Wistar rat with experimental autoimmune encephalomyelitis(EAE).METHODS:EAE was induced by guinea pig spinal cord homogenate(GPSCH),then the evoked potential in both EAE and control groups was examined.The HE staining and myelin staining of the auditory nucleus were conducted in order to corroborate the damage of the auditory path in the brainstem.Through studying the pathological changes using light microscopy and behavior changes of rats,we defined that the model was successful.RESULTS:The latency period of auditory evoked potentials of EAE rats was much longer than that in control group.The pathological pictures manifested that auditory nucleus were invaded by a great number of lymphocytes,demyelinations were discovered in it.CONCLUSION:The auditory path in the brainstem of the Wistar rat with EAE is suffered from inflammation and demyelination.
