ABSTRACT

Objective:To study the effect of curcumin on enterocyte apoptosis and its protective effect on intestinal mucosal barrier in septic rats.Methods:Eighty-seven 3-month male Sprague-Dawley (SD) rats were divided into Sham group, model group and curcumin group by random number table method, with 29 rats in each group. The septic rat model was reproduced by cecal ligation and puncture (CLP). 4 mL dimethyl sulfoxide solution were intraperitoneally injected in both Sham group and model group, 200 mg/kg curcumin dissolved by 4 mL dimethyl sulfoxide solution were intraperitoneally injected in curcumin group 10 minutes after operation. The blood samples (15 rats in each group) were collected 2, 12, 24 hours after operation, and the levels of serum procalcitonin (PCT), tumor necrosis factor-α(TNF-α), D-lactic acid and diamine oxidas (DAO) were tested by enzyme linked immunosorbent assay (ELISA). The ileum tissues were collected 12 hours, 24 hours after operation in three groups, water content was tested by weighting, pathologic structure was observed by light microscope, the enterocyte apoptosis was tested by terminal deoxynucleotidyl transferase-mediated dUTP-nick end labeling method (TUNEL). The 7-day survival rate was observed in three groups (14 rats in each group).Results:The serum levels of PCT, TNF-α, D-lactic acid and DAO were higher in model group at 2, 12, 24 hours after operation than those in Sham group, PCT, TNF-α levels were significantly higher in model group than those in Sham group 2 hours after operation [PCT (μg/L): 1.89±0.17 vs. 0.10±0.02, TNF-α (ng/L): 216.51±1.47 vs. 85.25±8.20, both P < 0.01], D-lactic acid, DAO levels were significantly higher in model group than those in Sham group 12 hours after operation [D-lactic acid (mg/L): 40.53±7.76 vs. 11.29±1.28, DAO (ng/L): 1 120.40±302.35 vs. 330.02±81.28, both P < 0.01]. Compared with model group, the levels of serum PCT, TNF-α, D-lactic acid and DAO were lower in curcumin group 2, 12, 24 hours after operation, the statistical difference appeared from 12 hours after operation [PCT (μg/L): 5.37±0.44 vs. 8.67±0.64, TNF-α(ng/L): 211.12±4.31 vs. 313.30±18.46, D-lactic acid (mg/L): 29.74±1.41 vs. 40.53±7.76, DAO (ng/L): 810.71±201.41 vs. 1 120.40±302.35, all P < 0.05], curcumin group had lower water content in ileum tissues 12 hours, 24 hours after operation [(68.34±0.68)% vs. (70.55±0.87)%, (69.41±0.59)% vs. (71.69±0.87)%, both P < 0.05]. The pathologic structures of intestinal villus were normal in Sham group, however, in model group intestinal villus were atrophic, edematous and shorten 12 hours after operation, it was further exacerbated 24 hours after operation. Compared with model group, the pathologic structures of intestinal villus in curcumin group were relived 12 hours, 24 hours after operation. The number of apoptotic enterocytes were significantly increased in model group compared with Sham group 24 hours after operation (cells: 25.48±6.10 vs. 4.00±2.04, P < 0.05), and the number of apoptotic enterocytes was lower in curcumin group than that in model group at the same time (cells: 15.48±3.75 vs. 25.48±6.10), the difference was statistically significant (both P < 0.05). Seven-day survival rate was significantly lower in curcumin than that in model group [42.9% (6/14) vs. 50.0% (7/14)], however, the difference was not statistically significant ( P > 0.05). Conclusion:Curcumin can protect the intestinal mucosal barrier by inhibiting enterocyte apoptosis in septic rats.

@#【Abstract】 【Objective】To assess effect of calcofluor white and fluorescent dye labeled concanavalin A in Candida albicans growth and adhesion of cells.【Methods】Yeast cells of 20 strains of Candida albicans were labeled by calcofluor white and Alexa fluor 488 conjugated concanavalin A respectively. The growth of labeled Candida albicans were tested by counts of colony forming unit. Then yeast cells of Candida albicans were co-cultured with macrophages and enterocytes for half an hour. The adhesion of labeled Candida albicans to macrophages and enterocytes were observed by fluorescence microscopy.【Results】No difference was observed on the number of colony forming units between calcofluor white-labeled groupand control group(P=0.942). Also,no difference was observed on the number of colony forming units between Alexa fluor 488 conjugated concanavalin A-labeled group and control group. However,either the number of calcofluor white-labeled Candida albicans or Alexa fluor 488 conjugated concanavalin A-labeled Candida albicans that bound to macrophages was less than that in control group(P=0.000,respectively). Either the number of calcofluor white-labeled Candida albicans or Alexa fluor 488 conjugated concanavalin A-labeled Candida albicans that bound to enterocytes was less than that in control group(P = 0.000,respectively). Hyphae were observed in control group but not in calcofluor white group after yeast cells of Candida albicans were co-cultured with cells for half an hour.【Conclusions】Growth of Candida albicans was not changed,while its adhesion to cells was reduced after its labeling by calcofluor white and Alexa fluor 488 conjugated concanavalin A.The germination of Candida albicans was affected when it had been labeled by calcofluor white.

ABSTRACT

Summary High room temperature reduces production efficiency of broiler chickens. Different factors contribute to this situation: fast growth, physiological variations, and changes in the small intestine mucosa. This review aims to define the concept of heat stress and its effects on several physiological aspects related to the development of the small intestine mucosa and the performance of broiler chickens. Heat stress triggers a corticosteroid increase and a circulating triiodothyronine hormone (T3) reduction, increases respiratory frequency, which triggers respiratory alkalosis, diminishes food intake, and leads to changes in the cellular dynamics of the small intestine mucosa. These changes depend on animal genotype, intensity, and duration of the stressor. Acute heat stress leads to a reduction in enterocyte proliferation and also to a decrease in crypt depth, without affecting villus height or villus/crypt ratio. On the other hand, chronic stress diminishes villus height and jejunum weight. These changes affect the capacity of broilers to digest and absorb the nutrients required for maintenance and production.


Resumen Una alta temperatura ambiente disminuye la eficiencia de la producción de pollos de engorde. Diversos factores contribuyen a esta situación: crecimiento rápido, cambios fisiológicos, y anomalías de la mucosa del intestino delgado. El objetivo de la presente revisión fue definir el concepto de estrés calórico y sus efectos sobre algunos aspectos de la fisiología, en el desarrollo de la mucosa del intestino delgado, y en el rendimiento de pollos de engorde. El estrés calórico desencadena un aumento de los niveles de corticosteroides, reducción de la hormona triyodotironina (T3) circulante, aumento en la frecuencia respiratoria que resulta en alcalosis respiratoria, disminuye la ingesta de alimentos y provoca cambios en la dinámica celular de la mucosa del intestino delgado. Estos cambios dependen del genotipo del animal y de la intensidad y duración de la acción del factor estresante. El estrés calórico agudo causa una disminución en la proliferación de enterocitos, reducción de la profundidad de las criptas sin cambio en la altura de las vellosidades y en la relación vellosidades/cripta. Ya el estrés crónico disminuye la altura de las vellosidades y el peso del yeyuno. Estos cambios afectan la capacidad del pollo de engorde para digerir y absorber los nutrientes para su mantenimiento y producción.


Resumo A temperatura ambiente elevada diminui a eficiência produtiva de frangos de corte. Diferentes fatores contribuem para esta situação: crescimento rápido, mudanças fisiológicas e alterações na mucosa do intestino delgado. Nesta revisão, o objetivo foi definir o conceito de estresse pelo calor e seus efeitos sobre algunos aspectos da fisiologia, no desenvolvimento da mucosa do intestino delgado e o desempenho de frangos de corte. O estresse pelo calor desencadeia aumento nos níveis de corticosteroides e redução do hormônio triiodotironina (T3) circulante, aumenta a frequência respiratória, resultando em alcalose respiratória, diminui a ingestão de alimentos e desencadeia alterações na dinâmica celular da mucosa do intestino delgado. Essas alterações dependem do genótipo do animal e da intensidade e duração da ação do agente estressor. O estresse pelo calor agudo promove diminuição na proliferação dos enterócitos, redução na profundidade das criptas sem alteração na altura das vilosidades e na relação vilo/cripta. Já o estresse crônico diminui a altura das vilosidades e peso do jejuno. Estas alterações afetam a capacidade do frango de corte de digerir e absorver nutrientes para sua manutenção e produção.

ABSTRACT

PURPOSE: Various gastrointestinal factors may contribute to maladaptive behavior in children with autism spectrum disorders (ASD). To determine the association between maladaptive behavior in children with ASD and gastrointestinal symptoms such as severity, intestinal microbiota, inflammation, enterocyte damage, permeability and absorption of opioid peptides. METHODS: This observational cross-sectional study compared children with ASD to healthy controls, aged 2-10 years. Maladaptive behavior was classified using the Approach Withdrawal Problems Composite subtest of the Pervasive Developmental Disorder Behavior Inventory. Dependent variables were gastrointestinal symptom severity index, fecal calprotectin, urinary D-lactate, urinary lactulose/mannitol excretion, urinary intestinal fatty acids binding protein (I-FABP) and urinary opioid peptide excretion. RESULTS: We did not find a significant difference between children with ASD with severe or mild maladaptive behavior and control subjects for gastrointestinal symptoms, fecal calprotectin, urinary D-lactate, and lactulose/mannitol ratio. Urinary opioid peptide excretion was absent in all children. Children with ASD with severe maladaptive behavior showed significantly higher urinary I-FABP levels compared to those with mild maladaptive behavior (p=0.019) and controls (p=0.015). CONCLUSION: In our series, maladaptive behavior in ASD children was not associated with gastrointestinal symptoms, intestinal inflammation (no difference in calprotectin), microbiota (no difference in urinary D-lactate) and intestinal permeability (no difference in lactulose/manitol ratio). ASD children with severe maladaptive behavior have significantly more enterocyte damage (increased urinary I-FABP) than ASD children with mild maladaptive behavior and normal children.

ABSTRACT

Existen dos avances en el manejo de la diarrea, que podrían reducir el número de muertes infantiles; el primero la formulación de Sales de Rehidratación Oral con menor concentración de glucosa y sodio; el segundo la suplementación con zinc para disminuir la duración, severidad y episodios futuros. La OMS/OPS define la diarrea aguda como la eliminación de tres o más evacuaciones intestinales líquidas o semilíquidas en 24 horas. El enterocito, como unidad funcional del intestino, posee una serie de transportadores donde ocurre el proceso de movilización de glucosa y de iones, para mantener el equilibrio eléctrico adecuado. En la evaluación del paciente la anamnesis se realiza con el fin de precisar la severidad de la diarrea y su posible causa. En el examen físico se tendrá en cuenta que esta enfermedad puede ser un signo o síntoma que se relaciona con trastornos extraintestinales. El examen de heces puede ser una herramienta útil en la investigación etiológica. Puede cuantificarse el gradiente osmótico (GAP fecal) y determinar si estamos en presencia de una diarrea osmótica o secretora. La presencia de 5 o más leucocitos fecales sugiere un proceso invasivo siendo de utilidad la realización de un recuento diferencial, si los polimorfonucleares se encuentran en 64% o más, puede ser posible recuperar bacterias en el coprocultivo. Este último tendrá indicación en algunos grupos que por edad o patología se consideran de riesgo elevados por ser no inmuno competentes. La serología para rotavirus permitiría el aislamiento rápido de pacientes infectados para limitar un brote.


Two recent advances in the management of diarrhea which could reduce child deaths are the formulation of Oral Rehydration Salts with a lower concentration of glucose and sodium, and zinc supplementation to decrease the duration and severity of future episodes. Acute diarrhea is defined by the WHO as the passage of three or more loose stools in 24 hours. The enterocyte, as the functional unit of the intestine, has a number of carriers which contribute to the mobilization process of glucose and ions to maintain the proper electrical balance. In the evaluation of the patient, the medical history will clarify the severity of diarrhea and its possible cause. During the physical examination, the fact that this disease may be a sign or symptom associated with extra intestinal disorders should be taken into account. Feces analysis may be a diagnostic tool. Osmotic gradient can be quantified (fecal GAP) to distinguish osmotic from secretory diarrhea. The presence of 5 or more fecal leukocytes suggests an invasive process and if PMNs are found in 64% or more in the differential cell count, it maybe possible to recover bacteria in a stool culture. The latter is indicated in some children considered of high risk for their low immune competence due to their age or to certain pathologies. Serology for rotavirus allows the rapid isolation of infected patients in order to limit an outbreak.

ABSTRACT

Transport of protein in the form of small peptides(di/tripeptides) across the small intestinal wall is a major route of dietary proteins absorption.The H + coupled dipeptide transporter,PepT1,located in the intestine and the kidney,plays an important role in the absorption of di/tripeptide;in addition,it mediates the intestinal absorption of ? lactam antibiotics,angiotensin converting enzyme inhibitors,and other peptide like drugs.The human intestinal cell line Caco 2 is an in vitro model.The dipeptide transporters normally found in the small intestine are present in Caco 2 cells.Caco 2 cells spontaneously differentiate in culture to polar cells possessing microvilli and enterocytic properties.These cells have been evaluated in detail as a model to study both transcellular and paracellular transport of nutrients and drugs in the gut.
