ABSTRACT

OBJECTIVE To comprehensively evaluate the quality of Eriobotrya japonica leaves from different producing areas. METHODS The contents of alcohol-soluble extracts were determined by hot-dipping method using 30 batches of E. japonica leaves from different producing areas as samples. The contents of total flavonoids and total triterpene acids were determined by ultraviolet spectrophotometry. The contents of five kinds of triterpenic acids (euscaphic acid, crataegolic acid, corosolic acid, oleanolic acid and ursolic acid) were determined by HPLC. The quality of E. japonica leaves from different producing areas was comprehensively evaluated by using entropy weight technique for order preference by similarity to an ideal solution (TOPSIS). The bivariate correlation analysis of E. japonica leaves was conducted by SPSS 22.0 software in terms of weight, comprehensive evaluation value, the content of alcohol-soluble extract, the contents of total flavonoids, total triterpene acids and five triterpenic acids. RESULTS The contents of alcohol-soluble extract in 30 batches of E. japonica leaves were (24.56±0.08)%-(34.85±0.13)%; the contents of total flavonoids were (4.69±0.11)-(14.23±0.27) mg/g; the contents of total triterpene acid were (27.58±0.59)- (63.95±1.27) mg/g; the contents of euscaphic acid, crataegolic acid, corosolic acid, oleanolic acid and ursolic acid were (0.728± 0.011)-(6.064±0.063), (0.526±0.013)-(3.245±0.022), (1.222±0.025)-(8.807±0.094), (0.856±0.021)-(2.931±0.075), (4.704±0.087)-(11.806±0.283) mg/g, respectively. The analysis result of entropy weight TOPSIS method showed that the top three samples with comprehensive evaluation values (No.Kjcx-5) were S14 (Huotian Town, Yunxiao County, Zhangzhou,Fujian), S19 (Qinnan District, Qinzhou, Guangxi) and S29 (Guoyang County, Bozhou, Anhui). Comprehensive evaluation 0596-2559522。E-mail:jxrcwxp@163.com of E. japonica leaves was positively correlated with the contents of five kinds of triterpenic acids, such as euscaphic acid, crataegolic acid, corosolic acid, oleanolic acid and ursolic acid (P<0.01). The weight of E. japonica leaves was positively correlated with the comprehensive evaluation value (P<0.01). CONCLUSIONS The qualities of E. japonica leaves from different producing areas are very different. Among them, the qualities of E. japonica leaves from Huotian Town, Yunxiao County, Zhangzhou of Fujian, Qinzhou Qinnan District of Guangxi, and Bozhou Guoyang County of Anhui are relatively better. ABSTRACT

Aim: To investigate antimethicillin-resistant Staphylococcus aureus (MRSA) activity of E. japonica leaf and to elucidate its anti-MRSA mechanism. Methodology: Methanolic extract of dried E. japonica leaf was partitioned in n-hexane, dichloromethane, ethyl acetate (EtOAc) and n-butyl alcohol. Antibacterial activity of extracts was determined by disk diffusion assay and minimum inhibitory concentration.Total RNA isolation was performed by treating MRSA culture with EtOAc sub-fractions. All MRSA isolates were tested for the presence of genes by reverse transcription polymerase chain reaction, followed by Western Blot. Results: The highest MRSA activity was observed from EtOAc sub-fraction 03 of methanolic extract. The minimum inhibitory concentration values of EF03 ranged from 32 to 64 µg ml-1 against methicillin-susceptible and -resistant S. aureus. EF03 fractions inhibited the expression of resistant genes in a dose-dependent manner. Moreover, the results of Western Blot assay indicated that the EF03 fractions inhibited the expression levels of resistant protein, PBP2a in a dose-dependent manner. Interpretation: EtOAc soluble fraction of E. japonica leaf evidenced profound antimicrobial activity, and inhibited expression of resistant genes against MRSA.

ABSTRACT

Eriobotrya japonica locally called as loquat in Kashmir has been studied in various parts of the world but little work has been reported on Kashmiri loquat. The chemical nature of fruits and vegetables offers a great diversity of biological properties and plays an important role in the field of pharmacology. There is a quest for newer drugs with few adverse effects and this poses a challenge for the development of new drugs. The study was undertaken to study the activities of ethanolic extract of Eriobotrya japonica fruits in streptozotocin induced diabetic rats. The phytochemical screening of the plant was also done. The animals were divided into five groups. Normal Control group received only the vehicle. Toxic group included those animals in which diabetes was induced by streptozotocin. The 3rd group was those animals which received streptozotocin and standard antidiabetic drug-glibenclamide. 4th group included those diabetic animals which received 50 mg/kg b.w dose of fruits of Eriobotrya japonica. 5th group animals included those diabetic animals which received 100 mg/kg b.w of the plant extract. The biochemical parameters that were evaluated were blood glucose levels and lipid profile tests. The body weight was also checked. Histopathology of pancreas was also done. The results showed significant decrease in blood glucose levels, lipid profile tests in animals treated with different doses of the plant extracts. Histopathology of pancreas also showed positive results.

ABSTRACT

BACKGROUND: Although Eriobotrya japonica leaves have been studied as a raw material for various cosmetic products, little is known about the anti-oxidant, anti-inflammatory, and anti-melanogenic activities of Eriobotrya japonica leaf ethanol extract (EJEE). METHODS: This study was conducted to evaluate the anti-oxidant, anti-inflammatory, and anti-melanogenic activities of EJEE using different in vitro models. In addition, we investigated the potential irritation of EJEE to skin and eye using animal alternative tests. RESULTS: The total content of polyphenols, one of the active constituents of EJEE, was analyzed by high-performance liquid chromatography and found to contain 88.68 mg tannic acid equivalent/g. EJEE showed a concentration-dependent 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radical scavenging activity, and a superoxide dismutase-like activity. The anti-inflammatory effect of 0.5% (w/v) EJEE was demonstrated by a reduction in lipopolysaccharide-induced nitric oxide and tumor necrosis factor-alpha levels in RAW 264.7 cells. EJEE also significantly inhibited melanogenesis in melanocyte stimulating hormone-induced B16F1 cells. EJEE did not show any irritation in skin and eye in animal alternative test. CONCLUSIONS: These results indicate that the EJEE possesses anti-oxidant, anti-inflammatory, and anti-melanogenic activities, while it did not induce toxicity or irritation in neither skin nor eye. Therefore, EJEE can be used as a cosmetic ingredient for skin improvement.

ABSTRACT

Objective: To investigate the effects of a herb complex extract (HCE) prepared from Cornus officinalis Sieb. Et Zucc., Eriobotrya japonica Lindley, and olive leaves on immune response of mouse spleen NK cells in vitro and in vivo analysis. Methods: The activity of natural killer (NK) cells was measured in splenocytes and YAC-1 cells. Mice were immunosuppressed using cyclophosphamide (5 mg/kg body weight). Three different doses of HCE (200, 400, and 800 mg/kg body weight) and red ginseng extract (800 mg/kg body weight) which was used as standard immunomodulatory herb were administered orally for 4 weeks. The body weight, dietary, water intake, organs (liver, thymus, and spleen) weight, completed blood count, and cytokines (tumor necrosis factor alpha, interferon gamma, and interleukin-2) production was measured. Results: At the maximum concentration of HCE, the activity of NK cells was increased by 48.5%. HCE increased liver, spleen, and thymus weights without altering numbers of white blood cells, lymphocytes, and neutrophils in a cyclophosphamide-induced immunosuppression rat model. However, HCE recovered the inhibited cytokine expression; HCE (800 mg/kg) increased cytokines levels. The results indicate the immune enhancement potential of this HCE. Conclusion: The HCE enhances immunity by increasing NK cell activity, regulating cytokine levels, and maintaining spleen weight. Therefore, it may be used as a potential immunity enhancer.

ABSTRACT

This study investigated the effects of hydroethanolic E. japonica seed extracts (EJSE) as inhibitors of lipid and protein oxidation on fish pates subjected to refrigerated storage. Five fish pate formulations were developed. These formulations included two control pates (water-control and ascorbic acid-control) and three pates with added EJSE (0.1, 0.2 and 0.4g of seed 100g-1 product, equivalent to 3.4, 6.8 or 13.6mg phenolic compounds kg-1 product), which were then stored under refrigeration for 35 days. Conjugated dienes (CD) and peroxide (PV) values increased along with the storage time; however, these values decreased and were similar among all samples at the end of 35 days of analysis (P<0.05). However, the thiobarbituric acid reactive substances levels (TBARS) did not change along the storage and were not affected by the EJSE. Additionally, there was a linear increase in the protein carbonyl content of fish pates over the storage period (P<0.05), but no effect of EJSE on protein oxidation. The results show that, at the concentrations evaluated, hydroethanolic E. japonica seed extract was unable to inhibit or reduce lipid and protein oxidation in fish pates, but the observed phenolic content emphasizes the need for further studies on the wastes of this fruit.

ABSTRACT

The bagging is an operation of extreme importance in the loquat culture because it provides fruit quality, and allows minimization of chemical products use. However, there is still lack of information, related to the adequate moment to carry through the operation, especially considering the limit of bagging stage, in which there are fruits of marketable quality. Therefore, this study aimed to evaluate the quality of bagged fruits of loquat cultivars in different stadiums of development. The cultivars 'Mizuho', 'Mizumo', 'Mizauto', 'Centennial', 'Crystal Nectar' and selection IAC NE-3 were evaluated in three stages of bagging, that is, when fruits reached an average diameter of 0.5cm, 1.0cm and 1.5cm. After fruits reached the size in accordance to each treatment, the panicles were bagged using newsprint. When mature, the fruits were harvested and evaluated regardless to length, diameter and fruit mass, presence of internal and external damages. The bagging at different developmental stages interfered in the quality of loquat fruit. bagged with diameters between 0.5 and 1.0cm were the ones that had presented the best commercial quality. The IAC NE-3 selection detached e among the loquat fruit due to its dimensions.

ABSTRACT

In Brazil, some pioneer studies were carried out using quince seedlings (Cydonia oblonga Mill.) as rootstock for loquat (Eriobotrya japonica (Thunb.) Lindl.). The main advantage of this intergeneric grafting use is plant size reduction. The success of using this intergeneric grafting is related mainly to plant size reduction. Therefore, the objective of this work was to study grafting techniques of loquat cultivars using 'Japonese' quince tree (Chaenomeles sinensis (Thouin) Koehne), with new option for rootstock. The one-year old 'Japonese' quince seedlings (with around 110 cm of height and 0.85 cm of diameter at the grafting point, 15 cm from the soil), were kept in plastic bags (30 x 18 cm, 3 liters of capacity). The seedlings were grafted by the cleft grafting and single bud grafting methods, in two different periods: in autumn (April) and winter (July). Five loquat cultivars commercially planted in Brazil were used: 'Mizuho', 'Néctar de Cristal' (IAC 866-7), 'Mizauto' (IAC 167-4), 'Mizumo' (IAC 1567-411) and 'Centenária' (IAC 1567-420). Using the single bud grafting method, there was no sprouted bud when done in autumn, and just two sprouts each of 'Mizauto', 'Néctar de Cristal' and 'Centenária' when done in winter, however, with low vigor. By the cleft grafting method, higher sprout percentage and better scions growth were observed when the seedlings were grafted in winter, especially for 'Mizuho', 'Centenária' and 'Néctar de Cristal' loquat cultivars.

We examined the inhibitory effects of loquat methanol extract on the adhesion, migration, invasion and matrix metalloproteinase (MMP) activities of MDA-MB-231 human breast cancer cell line. Cells were cultured with DMSO or with 10, 25, or 50 microg/ml of loquat methanol extract. Both leaf and seed extracts significantly inhibited growth of MDA-MB-231 cells in a dose-dependent manner, although leaf extract was more effective. Adhesion and migration were significantly inhibited by loquat extracts in a dose-dependent manner. Loquat extract also inhibited the invasion of breast cancer cells in a dose-dependent manner and leaf extract was more effective than seed extract. MMP-2 and MMP-9 activities were also inhibited by loquat extract. Our results indicate that methanol extracts of loquat inhibit the adhesion, migration and invasion of human breast cancer cells partially through the inhibition of MMP activity and leaf extract has more anti-metastatic effects in cell based assay than seed extract. Clinical application of loquat extract as a potent chemopreventive agent may be helpful in limiting breast cancer invasion and metastasis.

ABSTRACT

OBJECTIVE: To analyze the volatile components in the Eriobotrya japonica by headspace solid-phase microextraction-gas chromatography-mass spectrometry(SPME-GC-MS) and to optimize the operating conditions for SPME.METHODS: DB-5 MS elastic silica capillary column(30.0 m?250 ?m?0.25 ?m) was used as the chromatographic column with helium of high purity(99.999%) as carrier gas at a flow rate of 1.0 mL?min-1.The injector temperature was 250 ℃;MS condition: ionization mode was EI with electron energy of 70 eV and the mass scan range of 30~440 amu.RESULTS: After headspace absorption for 30 min at 80 ℃ using 65 ?m polydimethylsiloxane-divinyl-benzene extraction head followed by desorption for 1 min at 250 ℃,the E.japonica sample was separated and identified by GC-MS analysis.A total of 34 peaks were obtained and 29 components were identified which accounted for 97.42% of the total peak area, topping the list were benzaldehyde and 4-methoxybenzaldehyde.CONCLUSION: HS-SPME-GC-MS can be used for the rapid analysis of the volatile components in E.japonica.

ABSTRACT

A folha é hipoestomática, revelando estômatos anomocíticos.

The leaves of Eriobotrya japonica , Rosaceae, popularly known as loquat, are used in folk medicine for their hypoglycemic properties and to treat cutaneous diseases. The aim of this work was to study the leaf morpho-anatomy of the drug for pharmacognostic and taxonomic purposes. The botanical material was prepared for the usual optical and scanning microtechniques. The leaves are simple, alternate and lanceolate, with entire blade and dentate margins. The epidermis is uniseriate and coated by a thick and slightly striated cuticle. Both surfaces are hairy, particularly the lower, with long unicellular non-glandular trichomes. Anomocytic stomata occur exclusively on the lower surface. The mesophyll is dorsiventral and the small bundles are collateral and may present sheath extensions. The midrib has got biconvex shape and the petiole round contour, both showing amphicrival bundles. Idioblasts containing calcium oxalate crystals, mucilage and phenolic compounds were observed.
