ABSTRACT

Aim To investigate the effect and mechanism of Eucommiae ulmoides and Radix Dipsaci in the treatment of osteoporosis.Methods The active ingredients of Eucommiae ulmoides and Radix Dipsaci were obtained by TCMSP,BATMAN-TCM platform and the literature.Osteoporosis related genes were collected by GeneCards and OMIM.The compound target network was constructed with Cytoscape 3.7.2 software.The protein interaction network was constructed with STRING online website.DAVID was used to perform gene ontology(GO)enrichment analysis and kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.The synergistic effect of Eucommiae ulmoides and Radix Dipsaci was tested by osteo-blast proliferation,alkaline phosphatase expression and mineralized nodule formation experiments.The protein expressions of BMP2 and Wnt signaling pathway were investigated by Western blotting.Results A total of 44 active components of Eucommiae ulmoides and 17 active components of Radix Dipsaci were screened,and 80 targets were intersected by osteoporosis.GO analysis showed that the action mechanism of Eucommiae ulmoides and Radix Dipsaci in the treatment of osteoporosis was involved with 52 items of biological process,8 items of cell composition and 16 items of molecular function.The KEGG enrichment pathway was dominated by 6 major signaling pathways.Compared with the control group,the osteoblast proliferation,alkaline phosphatase expression and mineralized nodule formation significantly increased in Eucommiae ulmoides or Radix Dipsaci group(P<0.01).Western blotting showed that Eucommiae ulmoides mainly regulated the expression levels of Wnt signaling pathways(P<0.05 or P<0.01),and Radix Dipsaci up-regulated the expression levels of BMP2 signaling pathways(P<0.01).Meanwhile,Eucommiae ulmoides and Radix Dipsaci significantly enhanced these effects,respectively(P<0.01).Conclusions Although Eucommiae ulmoides and Radix Dipsaci have different active components,their main targets and pathways are the same in the treatment of osteoporosis.Eucommiae ulmoides and Radix Dipsaci can regulate the function of osteoblasts through BMP2 and Wnt signaling pathways,and the combination of the two drugs in the treatment of osteoporosis has synergistic effect.

ABSTRACT

OBJECTIVE:To compare the total flavonoids content and antioxidant activity in the barks,leaves,male flowers and seeds of Eucommiae ulmoides. METHODS:UV spectrophotometry was used to determine the total flavonoids content in differ-ent parts;tests was conducted to clear 2,2′-nitrilobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS+),1,1-diphenyl-2-trinitro-phenylhydrazine(DPPH)radicals and the reducing ability of Cu2+,using half clear/reduction concentration value(IC50)as evalua-tion indexes,and vitamin C was regarded as positive control. RESULTS:The total flavonoids content of the E. ulmoides from hight to low was as follows as leaves>male flowers>barks>seeds,except there was no significant difference in barks and seeds (P>0.05),the other parts had significant differences (Pmale flowers>seeds>barks,except there was no significant difference in the indicators of leaves and male flowers(P>0.05),the other parts had significant differences(Pmale flowers>barks>seeds,there was significant difference in leaves and males flowers with barks and seeds (P<0.05). CONCLUSIONS:The content of total flavonoids in leaves and male flowers is high,and the antioxidant activity is strong,which has a great prospect of exploitation and utilization to make up for deficiencies in barks of E. ulmoides.

ABSTRACT

Objective: To investigate the anti-inflammatory and analgesic activity of Eucommiae ulmoides seed total glycoside(EUG). Methods: Dimethyl benzene (DB)-induced mouse ear edema model and carrageein-induced rat paw edema model were employed for the anti-inflammatory activity experiment. The experiment for analgesic activity was done using hot plate test, photoelectricity-induced test and acetic acid-induced writhing method. Results: EUG at 110, 220 mg/kg showed an obvious inhibitory effect against DB-induced mouse ear edema volume, and at 55, 110, 220 mg/kg it showed an obvious analgesic effect against carrageein-induced rat paw edema rate 4 hours after oral administration. At 220 mg/kg EUG notably raised the pain threshold in the mice stimulated by hot plate and photoelectricity. At 110, 220 mg/kg EUG greatly reduced the number of acid-induced writhing. Conclusion: The result suggests that EUG has potential anti-inflammatory and analgesic activity.

ABSTRACT

OBJECTIVE: To evaluate the effect of Eucommiae ulmoides on the microstructural and ultrastructural change of femur and lumber vertebrae in ovariectomized rats.METHODS: 80 4-month-old female SD rats were randomized to 4 groups: sham operation group,model group,alfacalcidol group,E.ulmoides group.In the sham operation group,the abdominal cavities of the rats were cut open to expose ovaries before being sutured.However,the rats in the other 3 groups were ovariectomized bilaterally.At 10 days after operation the rats were administered intragastrically with corresponding drug.After 3 months,femur and lumber vertebrae were dissected and fixed in 4% glutaral for 3 days,then decalcified in 4% EDTA for 4 weeks,and then the structure of femur and lumber vertebrae was observed under light microscope and transmission electron microscope(TEM).RESULTS: Observed under light microscope,the femur and lumber vertebrae were full of fat in the ovariectomized rats and in which the density of bone trabecula decreased significantly;however,in alfacalcidol-treated rats and E.ulmoides-treated rats,the amount of fat decreased markedly while the density and thickness of bone trabecula increased.Observed under TEM,in the ovariectomized rats,rough endoplasmic reticulum(RER),cytolysosome and mitochondrial were plentiful inside osteoclasts in femur and lumber vertebrae,partial osteoblasts atrophied,the amount of RER and other cellular organ decreased significantly and apoptosis appeared for partial cells;however,in alfacalcidol-treated group and E.ulmoicdes-treated group,the above changes improved significantly.CONCLUSION:E.ulmoides has remarkable beneficial effect on osteoporosis in ovariectomized rats.The mechanism might be attributed to its inhibition on the synthesis of fat and the activity of osteoclasts,promotion on the activity of osteoblasts and inhibition on the apoptosis.
