ABSTRACT

Objective: To establish the quality standard for Jiuwei Zhaxun pills. Methods: Fructus Chebulae, Flos Carthami and Herba Dracocephali Heterophylli were identified by TLC. HPLC was used to determine the content of gallic acid in Jiuwei Zhaxun pills. Results: TLC showed clear sports without any interference from the negative control. Gallic acid showed a good linear relationship (r=0. 999 7) within the range of 0. 028 6-0. 572 0 μg. The average recovery was 97. 74% with the RSD of 0. 95% (n=6). Conclusion:The methods are with strong specificity, high sensitivity and good reproducibility, which can be applied in the quality control of Jiuwei Zhaxun pills.

ABSTRACT

OBJECTIVE: To study the active ingredients of Flos Carthami by investigating the relationship between HPLC spectrum and antioxidant activity. METHODS: The specific chromatogram of Flos Carthami processed by means of classical homothermal acceleration was assayed by HPLC. DPPH, ABTS and FRAP assays were established to determine the antioxidant activity of Flos Carthami. The spectrum-effect correlation was studied by partial least squares (PLS) method. RESULTS: S4, S8, S11, S12, S13, S14, S16 and S17 were characteristic compounds in 27 matching characteristic chromatograms which had positive relationship with the ability of scavenging DPPH free radical. S4, S8, S11, S12, S14, S16, S17 and S18 were significantly positively related to the ability of scavenging ABTS free radical. S4, S8, S11, S12, S13, S14, S16, S17, S18 and S26 were significantly positively related to the ability of restoring Fe3+. Among the chromatographic peaks, S4, S8, S11, S12, S14, S16and S17 were positively related to the abilities of scavenging DPPH free radical and ABTS free radical and restoring Fe3+. S14 was determined as hydroxysafflor yellow A. CONCLUSION: The antioxidant material of Flos Carthami is based on the synergy of a variety of compounds. An as comprehensive as possible analysis of the compounds related to the pharmacological effect can reflect the quality of Flos Carthami. The study of spectrum-efficacy relationship is an effective way to determine the compounds related with pharmacological effect and control the quality of traditional Chinese medicinal materials.

ABSTRACT

To characterize and identify multiple constituents in Danhong injection (DHI), a fast ultra-high performance liquid chromatography coupled to electrospray ionization quadrupole time-of-flight tandem mass spectrometry (UHPLC-ESI-QTOF/MS) method was established and validated in the present study. A total of 63 compounds, including 33 phenolic acids, 2 C-glycosyl quinochalcones, 6 flavonoid O-glycosides, 4 iridoid glycosides, 6 organic acids, 5 amino acids, and 3 nucleosides, were identified or tentatively characterized. In conclusion, the UHPLC-ESI-QTOF/MS method is useful and efficient for in-depth structural elucidation of chemical compounds in complex matrices of herbal medicines such as DHI.

ABSTRACT

Objective:To establish the identification method for Aucklandia Lappa and Picrorhiza kurrooa Royle ex Benth and the content determination of hydroxysaffor yellow A in Liangxue Shiwei San. Methods:The identification was carried out by TLC. The con-tent of hydroxysaffor yellow A was determined by HPLC. The column was Kormasil C18 (250 mm × 4. 6 mm, 5 μm) and the flow rate was 1. 0 ml·min-1. The mobile phase was methanol-0. 5% acetic acid (30∶70). The detection wavelength was 403 nm and the col-umn temperature was 25℃,and the sample size was 10 μl. Results:The TLC spots were clear with high resolution without interference from the negative sample. A good linearity of hydroxysaffor yellow A was within the range of 1.212-48.480μg·ml-1(r=0.999 8). The average recovery was 98. 13%(RSD=1. 6%,n=6). Conclusion:The established TLC and HPLC methods are simple and accu-rate with good reproducibility, which can be used in the quality control of Liangxue Shiwei San.

ABSTRACT

@#Objective To observe the effect of Integrated Chinese-Western Therapy on shoulder-hand syndrome after stroke. Methods 80 stroke patients with shoulder-hand syndrome after stroke were randomly divided into the treating group (n=40) and the control group (n=40). The patients in the treating group accepted occupational therapy and infrared therapy at the acupoints with Flos Carthami Alcohol. The control group accepted occupational therapy only. Results The treating group soothed more pain and improve their ability of motor and daily living. Conclusion Infrared therapy with Flos Carthami Alcohol can facilitate the effects of occupational therapy on should-hand syndrome after stroke.

ABSTRACT

Objective To study the effect of compatibility of Flos Carthami and Semen Persicae in different ratios on the contents of hydroxysafflor yellow A(HSYA) in their water decoction.Methods Reverse-phase HPLC was performed on a Luna C18 column(4.6 mm?150 mm,5?m) at 30℃.The mobile phase consisted of methanol-0.12 mmol?L-1 phosphate solution(28∶72,V/V) at the flow rate of 1.0 mL? min-1.The detector was operated at wavelength of 403nm.Results A good linearity of HSYA was obtained in the range of 4.0~160.0?g?mL-1(Y=25 396X+40 247,r=0.999 7),the average recovery was 94.76% and the RSD(n=6) was 2.93%.The amount of HSYA increased with the ratio of Flos Carthami,but had no correlation with the ratio of Semen Persicae.Conclusion The HPLC method for determination of HSYA is simple,fast and accurate.It can be used for the quality control of Flos Carthami and its compound preparations.Semen Persicae has no effect on the extraction of HSYA.

ABSTRACT

This study was aimed to investigate the effect of Flos Carthami on the atherosclerosis in rabbit induced by cholesterol diet. 24 rabbits were divided by 3 groups; normal control group, CH group, and CH+FC group. The normal control group was fed with the normal pellet diet. The CH group was fed with pellet diet including 4% cholesterol, and the CH+FC group was fed with pellet diet including 4% cholesterol and 4% Flos Carthami dry -extract powder about 100g diet per 1 kg body weight per a day. After 12 weeks, animals were sacrificed and a piece of ascending aorta was collected. Tissue was sectioned 8 micrometer thickness and sections were stained with hematoxylin -eosin, alcian blue pH 2.5, aldehyde -fuchsin, and Van Gieson 's trichrome method. In CH+FC group, atheroma and mucoprotein formation on tunica intima of the ascending aorta was reduced, and lesion of elastic and collagen fibers in tunica media was also attenuated with respect to that in CH group. According to this result, it is considered that Flos Carthami has a preventing effect on atherosclerosis or a control effect on hypercholesterolemia. But distinct mechanism of action is still unclear.

ABSTRACT

Objective To study the synergistic action of Radix Angelicae Sinensis(RAS) and Flos Carthami(FC) on Clopidogrel Bisulfate(CB) counteracting platelet aggregation. Methods Forty-eight Sprague-Dawley(SD) rats were randomized into 6 groups:normal control group,CB group(1 mg? kg-1? d-1,qd),RAS group(0.7 g? kg-1? d-1,bid) and FC group(0.7 g? kg-1? d-1,bid),CB+ RAS group(CB 1 mg? kg-1? d-1,qd,and RAS 0.7 g? kg-1? d-1,bid),and CB+ FC group(CB 1 mg? kg-1? d-1,qd,and FC 0.7 g? kg-1? d-1,bid). The rats received gastric infusion of corresponding drugs for 5 continuous days. Blood sample of SD rats were collected by puncture of the abdominal artery for the observation of platelet aggregation in each group.Another fifty-four rats were used for the observation of inhibitory effect on thrombosis in arteriovenous shunt. Sixty NIH mice were applied for the observation of effect on bleeding time,clotting time and parameters of routine blood examination. Results Platelet aggregation and thombosis in arteriovenous shunt of rats were inhibited,and the bleeding time was prolonged in the medication group. The above effect was much better in the combination groups. The effect on clotting time and parameters of routine blood examination was not obvious. Conclusion Radix Angelicae Sinensis and Flos Carthami can enhance the effect of Clopidogrel Bisulfate on inhibiting platelet aggregation and thrombosis,and on prolonging bleeding time.

ABSTRACT

AIM: To discuss the GC analysis of naphtha of Semen Persicae,Flos carthami and its couplet medicine(Semen Persicae plus Flos carthami). METHODS: To determine the relative content of naphtha by vapour distill,Gas Chromatography-Mass Spectrometry and Chemometric Resolution Method and GC unitary area method. RESULTS: The obtained naphtha of Flos carthami consisted of 54 constitutes such as dodecanoic acid and n-Hexadecanoic acid.The obtained naphtha of Semen Persicas comprised 6 constitutes such as benzaldehyde.And the obtained naphtha of Semen Persicae plus Flos carthami possessed 40 coustitutes,such as dodecanoic acid,n-Hexadecanoic acid and benzaldehyde. CONCLUSION: The constitutes Semen Persicae plus Flos carthami are basical the linear addition of constitutes Semen Persicae and Flos carthami but 21 kinds of constitutes in the absence of both Semen Persicae and Flos carthami.

ABSTRACT

Objective: To establish the method of fingerprint analysis on Flos Carthami by HPLC. Methods: The HPLC method was used, chromatography conditions were ODS column, CHOH 3 0.4%H 3PO 4 solution (50∶50) as mobile phase and 380nm at detective wavelength. Results: The result showed that 9 peaks (RSD≤3%, n=10 ) were common.Conclusion: This method is reliable, simple and provides a reference standard for the quality control of Flos Carthami.

ABSTRACT

Objective:To study the high-performance liquid chromatographic(HPLC) fingerprints of different germplasms of Flos Carthami. Methods: HPLC was applied in chromatographic separation of 36 different germplasms of Flos Carthami collected from 11 provinces of China.The chromatography condition was: Agilent Zorbax C_(18) column(4.6 mm?250 mm,5 mm);mobile phase: CAN,0.5% H_3PO_4(from 1090 to 8020);running time: 60 min;flow speed: 1.0 ml/min;detection wavelength: 265 nm;and temperature: 20℃.The data were processed by Chromatographic Fingerprint Station designed by Department of Pharmaceutical Analysis,School of Pharmacy,Second Military Medical University.Different germplasms of Flos Carthami were subjected to cluster analysis and their similarity was also evaluated.Results: The different germplasms of Flos Carthami in this study were grouped into 2 chemical types.Samples from northwest of China were of high similarity;samples from the middle China were of high similarity to those from South China.The similarity of some germplasms of Flos Carthami was significantly different.Conclusion: The chemical components of different germplasms of Flos Carthami are obviously different from each other;selection of germplasm is very important in quality control of Flos Carthami.
