ABSTRACT

Objective: To evaluate the protective and curative potential of PHF, a polyherbal formulation, against two experimentally induced hepatotoxicity models in rats. Methods: Hepatoprotective activity of the PHF containing three indigenous medicinal plants extracts Coccinia indica, Sidacordata and Scoparia dulcis, was screened against CCl4 and paracetamol induced hepatotoxicity in rats. Result: Administration of hepatotoxins (CCl4 and paracetamol) shows significant morphological, biochemical and histopathological deteriorations in the liver of experimental animals. Pretreatment with PHF had significant protection against hepatic damage by maintaining the morphological parameters within normal range and normalizing the elevated levels of biochemical parameters (SGPT, SGOT, ALP and total bilirubin), which were evidently showed in histopathological study. Conclusions: The PHF has highly significant hepatoprotective effect at 100 and 200 mg/kg, p.o. on the liver of all the two experimental animal models. The liver protection due to combined action of all plant extracts along with their phytoconstituents.

ABSTRACT

Objectives: To study the morpho-anatomy of the aerial parts of Rivea hypocrateriformi (Convolvulaceae) to increase the knowledge and standardization parameters of these plants and its family. Methods: Morpho-anatomical studies of aerial parts of have been carried out by free hand. The different types of histochemical tests were performed for the identification of micro-chemical by using staining reagents. Preliminary phytochemical and quantitative estimation has been determined along with HPTLC fingerprinting. CAMAG HPTLC system equipped with Linomat V applicator, TLC scanner 3, Reprostar 3 and WIN CATS-4 software was used for fingerprinting. Results: The morpho-anatomical structure of plant revealed the presence of thick wall collenchyma cell and parenchyma cells along with spharaphides and columnar palisade cells. Presense of numerous covering trichomes, parasitic stomata, oval shaped vascular bundles and calcium oxalate crystals were observed. In powder microscopy lignified xylem vessels, and covering trichomes were clearly seen. Physicochemical parameters, ash value, inorganic elements, moisture content and extractive value were determined to develop the stringent pharmacognostic standards. Qualitative standardization and HPTLC fingerprinting of phytochemical revealed the presense of various plant secondary metabolites. Conclusions:These findings will be useful in establishing pharmacognostic and phytochemical standards for identification, as well as assessment of purity and quality of this plant, which definitely gaining the relevance in plant drug research and establishment of plant monograph.

ABSTRACT

Piperine, a characterizing compound present in fruits of Piper nigrum and Piper longum used as bioavailability enhancer. Ingredients of antioxidant, anti-inflammatory activity has been extracted using soxhlet and supercritical fluid extraction technique. It was isolated using column chromatography. Characterization of compound was done by spectroscopic technique. A simple, rapid, accurate and specific HPTLC method developed and validated. The method proposed can be used for the routine analysis of both P.nigrum and P.longum fruit material and its formulations.

ABSTRACT

The most important challenges faced by herbal formulations arise because of their lack of complete evaluation. Evaluation is necessary to ensure quality and purity of the herbal product. For evaluation of capsule containing single herb various parameters were tested. These parameters for raw material include powder characteristic study, organoleptic, physicochemical, phytochemical parameters etc., and assay of active constituent. Parameters for finished product (capsule) include uniformity of weight, pH, moisture content, disintegration time and dissolution study. HPTLC study, heavy metal analysis, microbial analysis and Nutritional value were carried out as a part of evaluation. Results indicate that Ashwagandha capsule has passed through all organoleptic and physicochemical parameters. Active constituent was present in adequate amount in the Ashwagandha capsule. Data of HPTLC finger printing indicates that extract was derived from genuine plant or parts of the plant and also capsule contain the same extract. Concentration of lead, arsenic and cadmium in capsules passed the limit of heavy metal. Mercury was absent in capsule and in their extract. Capsules have <10 cfu/gm total bacterial count. Total yeast and mould was absent in capsules. The pathogens like E. coli, Salmonella, P.aeruginosa and S.aureus were also absent in capsule.Carbohydrate, protein and cholesterol content for Ashwagadha capsule is 84.56%, 8.29% and 2.32% respectively. Data suggested that capsule and its extract were consistent with various identity, quality and purity parameters such as organoleptic characters, physico-chemical parameter, HPTLC fingerprinting, heavy metal and microbial analysis. Nutritional assessment of the each capsule indicates their dietary supplement value.
