ABSTRACT

【Objective】 To analyze the causes of chylemia, explore the effects of varying degrees of chylemia on subsequent blood products and the countermeasures. 【Methods】 The information of blood donors in the Central Blood Station of Xinjiang Production and Construction Corps from January 2018 to July 2022 were analyzed retrospectively, and the causes of fatty blood discarding was analyzed in terms of gender, age and times of donation. The effects of chylemia of control group, mild group, moderate group and severe group and corresponding gender and age on the hemolysis rate of suspended red blood cells(RBCs) at the end of storage and residual amount of methylene blue in the inactivated plasma were discussed. 【Results】 The incidence of chylemia in males(0.93%,84/9 047) was much higher than that in females(0.24%,9/3 781) (P<0. 05). The hemolysis rates of WBC-free suspended RBCs of mild, moderate and severe chylemia group at the end of storage(0. 19±0.08,0.28±0.06,0.66±0.25) were higher than that in control group(0.08±0.03), in males significantly higher than those in females(0.08±0.03 vs 0.06±0.02,0.21±0.09 vs 0.15±0.04,0.30±0.05 vs 0.24±0.07), in the age group of 18-30 years significantly lower than those in 31-41 years(0.07±0.02 vs 0.09±0.03) and ≥41 years(0.07±0.02 vs 0.09±0.03), P <0.05. The residual methylene blue in plasma with viral inactivation prepared from mild and moderate chylemia was significantly higher than that in control group(0.12±0.02,0.18±0.06 vs 0.06±0.02) (P<0.05). 【Conclusion】 Chylemia shall be monitored during the whole process of blood collection, and coutermeasures shall be taken to decrease the effects of chylemia on blood quality.

ABSTRACT

【Objective】 To study the protective effect of glycine solution on frozen red blood cell thawing process. 【Methods】 A total of 20 bags of 1 U of leukocytes reduced suspended red blood cells within 6 days were selected for the study. After mixing, each 2 bags of suspended red blood cells were divided into 2 bags and into two groups with 10 bags of 1 U in each group, and were frozen for storage. One group was deglycerolized with sodium chloride solution (control group), and one group was deglycerolized with glycine solution (experimental group). The hemoglobin, free hemoglobin, residual glycerol, total glycerol in red blood cells, adenosine triphosphate (ATP) and 2,3-diphosphoglycerate (2,3-DPG) were detected in the two groups. 【Results】 Compared with the free hemoglobin content (0.90±0.05)g/L and residual glycerol content (1.17± 0.08)g/L in the control group, the final product red blood cell supernatant free hemoglobin content (0.77±0.15)g/L and residual glycerol content (0.79±0.33)g/L in the experimental group were decreased, and the difference was statistically significant (P<0.05). Compared with the ATP content (4.03±0.38)µmol/gHb and 2,3-DPG content (485.65±78.08)µg/L in the control group, the ATP content (4.41±0.35)µmol/gHb and 2,3-DPG content (656.28±116.68)µ g/L in the experimental group were significantly increased, with statistical significance (P<0.05). 【Conclusion】 Using glycine solution instead of sodium chloride solution to prepare frozen thawed deglycerolized erythrocytes achieved the effect of protecting erythrocytes, reduced the hemolysis rate of erythrocytes and glycerin residue, and increased the recovery rate of erythrocytes.

ABSTRACT

【Objective】 To compare the current standards and explore the influencing factors for hemolysis rate of leukocyte-reduced red blood cells at the end of the preservation period, in order to formulate reasonable internal control indicators. 【Methods】 A retrospective analysis was performed on hemolysis rate of 427 samples of leukocyte-reduced red blood cells at the end of the preservation period in Nanning Blood Center from 2015 to 2022. Compared with the current standard for hemolysis rate at the end of the preservation period (GB 18469-2012 Quality Requirements for Whole Blood and Component Blood), the differences were analyzed, and the factors influncing the hemolysis rate were analyzed in terms of different blood donor groups. 【Results】 1) Among the 427 samples, the hemolysis rate of 418 (97.89%) did not exceed 0.4%, all lower than 0.8%; 2)the hemolysis rate of the male group was higher than that of the female group; 3) the hemolysis rate of the 18-29 years old group was lower than that of the 30-39 year old group and the 40-60 year old group, with statistically significant difference; 4) in terms of occupation, the hemolysis rate of students was the lowest, and the differences between groups were statistically significant; 5) no statistical significance was found in ethnicity and blood type. 【Conclusion】 Statistics indicated that gender, age, blood donation volume and occupation of blood donors were the influencing factors of hemolysis rate. The current standard is obviously higher in the qualified range of blood quality control in Nanning. It is advisable to formulate a reasonable quality control strategy with internal control index of hemolysis rate set <0.4%, which is conducive to making accurate evaluation of internal quality control and ensuring blood safety.

ABSTRACT

Objective:To investigate the effect of small molecule antibacterial agent Halicin against Staphylococcus aureus. Methods:The minimum inhibitory concentration (MIC) and minimum bactericidal concentration of Halicin against S. aureus were detected by the microbroth dilution method. The time-kill assay of Halicin against S. aureus was detected by agar plate dilution method. Micro checkerboard dilution method was used to determine the synergistic antibacterial activity between Halicin and conventional antibiotics. Crystal violet staining method was used to assess the biofilm inhibitory and eradicating activity of Halicin. Hemolysis rate was used to detect the mammal cell toxicity of Halicin. Through the mouse skin abscess model, take the skin tissue around the abscess to grind and dilute the colony to detect the antibacterial effect of Halicin in vivo. Results:Halicin showed significant bacteriostasis effects against S. aureus with the minimum inhibitory concentration of 2-4 mg/L. Halicin could significantly reduce the average CFU counts of S. aureus about 5.5×10 6 CFU/ml in a concentration-dependent manner after 8 h treatment at the concentration of 16 mg/L. The fractional inhibitory concentration value between Halicin and ampicillin was 0.5, showing a synergistic antibacterial efficacy. Halicin effectively inhibited the formation of biofilms at the concentration of 4 × MIC, reducing the total biofilm biomass ( A570) from (2.89±0.09) to (1.35±0.17) ( t=11.12, P<0.05). However, there was no eradication effect against preformed biofilms. In addition, Halicin had almost no hemolytic activity on red blood cells even at the concentration up to 128 mg/L. It showed that 20 mg/kg Halicin reduced bacterial burden about 3.0×10 7 CFU/ml in vivo. Conclusion:Halicin had a strong antimicrobial activity against S. aureus with no hemolytic activity.

ABSTRACT

The biocompatibility of nanomaterials has attracted much attention. Graphene oxide (GO) is a nanomaterial widely used in biomedicine, but its toxicity can not be ignored. In this study, the effect of GO on the blood system (the hemolysis rate, the fragility of erythrocyte, and acetylcholinesterase activity) was systematically investigated. The results showed that the hemolysis rate of erythrocytes was lower than 8% when the GO concentration was below 100 μg/mL (P5 μm (LGO) increased the activity of acetylcholinesterase by 42.67% (P<0.05). Then molecular dynamics simulation was used to study how GO interacted with acetylcholinesterase and increased its activity. The results showed that GO was attached to the cell membrane, thus may provide an electronegative environment that helps the hydrolysate to detach from the active sites more quickly so as to enhance the activity of acetylcholinesterase.

ABSTRACT

Objective To comprehensively analyze the effect and various detection indicators of leucocyte depletion suspended red blood cells(RBC) prepared by different storage time and different manufacturers of leucocyte depletion instrument to provide some decision-making basis for peers .Methods Two hundreds and forty bags of suspended RBC(1 ,1 .5 ,2 U) prepared from whole blood by 4 manufacturers were randomly collected and preserved in (4 ± 2)℃ dedicated refrigerator according to different specifica-tions .Then the sterile quick-release connector was used to take 240 leucocyte depletion suspended RBC samples of same specifica-tions and different shelf lives of 0 ,7 ,14 ,21 ,28 ,35 d by different manufacturers .Their capacity ,hematocrit ,leukocyte residues ,plas-ma free hemoglobin content and hemoglobin content were detected ,and the hemolysis rate at the end of shelf life was calculated .Re-sults The effect of leucocyte depletion suspended RBC by the same manufacturer had no statistical difference among different spec-ifications (P>0 .05);the capacity and plasma free hemoglobin content in the same specification had statistical difference among difference among different manufacturers (P<0 .05);the free hemoglobin and hemolysis rate at the end of shelf life in the same manufacturer had statistical difference among different preserving time (P<0 .05) .Conclusion By comprehensive considering the results of indexes of capacity ,hematocrit ,filter time ,free hemoglobin content and hemolysis rate ,our station chose the Shandong Weigao and Fresenius disposable leucocyte depletion plastic bags for ensuring the leucocyte depletion effect ,and safety and efficacy of RBC transfusion .

ABSTRACT

Objective: To prepare Danshensu loaded polycaprolactone (DSS-PCL) nanofiber scaffold by using electrospin technology. Methods: Using dichloromethane-N, N-dimethyl formamide (CH2Cl2-DMF) 8:2 as solvent, nanofibers containing danshensu was prepared by electrospinning, fiber morphology was observed by SEM, danshensu in vitro release rule from drug loaded nanofiber scaffold was investigated, and the security of nanofiber was assessed by MTT and hemolysis test. Results: By using CH2Cl2-DMF as solvent, drug and materials could be well dissolved and successfully electrospun into nanofiber, the average fiber diameters were 210 and 190 nm, the in vitro release experiment showed that the drug release after a quick release into the slow release. MTT test indicated that PCL and DSS-PCL nanofiber showed no influence to the cell viability, and hemolysis test showed hemolysis ratio of nanofiber was under 5%. Conclusion: DSS-PCL nanofibers is easy to prepare by electrospinning.

ABSTRACT

Objective To discuss the feasibility of Shuyisha as hemostasis and repair material for liver wound. Methods Hemolysis rate, acute toxicity and eytotoxicity of Shuyisha were measured. A hemorrhage model was established by making an open wound (5 mm× 3 nun ×2 mm) on the left liver lobe of mice. Hemostasis was performed with Shuyisha in experimental group and with Surgicel in control group, when the hemostatic time and total blood loss (TBL) were accurately recorded and regular macro-scopic and histological observation carried out. Results The hemolysis rate of Shuyisha was 2.33%, with maximum tolerance does of over 0.48 g/kg and the eytotoxicity at zero. The hemostatie time of Shuy-isha was (5.00 ±0.00) s, with total blood loss of (0.88±0.18) g/kg, better than Surgicel (P< 0.05). Shuyisha was degraded completely within 14 days, with the wound healed within 21 days in ex-perimental group, much better than Surgieel. Conclusions The hemolysis rate, acute toxicity and cy-totoxicity of Shuyisha are up to the requirement of biomedical materials. Shuyisha has effective hemosta-sis, which may be related to its molecular structure and adhesion.
