ABSTRACT

Objective To investigate the therapeutic effect of Tiaozhi Decoction for the treatment of coronary heart disease induced stable angina complicated with hyperlipemia, and to observe its effect on serum inflammatory factors. Results One hundred qualified patients were evenly randomized into treatment group and control group. Both groups were given conventional western medical treatment with reference to Guide for Diagnosis and Treatment of Chronic Stable Angina, and additionally, the control group was given oral use of Simvastatin and the treatment group was given Tiaozhi Decoction orally. The treatment of the two groups covered 8 weeks. Before and after treatment, blood lipid levels of total cholesterol ( TC) , triglyceride ( TG) , low-density lipoprotein cholesterol ( LDL-C) , high- density lipoprotein cholesterol ( HDL-C) , apolipoprotein A (ApoA) and apolipoprotein B (ApoB) were observed. The frequency of angina pectoris attack and the dosage of Nitroglycerin Tablets per week in both groups were recorded during the treatment. Therapeutic effect on lowering blood lipid and on improving electrocardiogram was evaluated after treatment. Serum levels of hypersensitivity C-reactive protein (hs-CRP), interleukin-6 (IL-6), homocysteic acid (Hcy), adiponectin (APN), and oxidized low-density lipoprotein (ox-LDL) were detected before and after treatment. Results(1) The results of Ridit analysis showed that the treatment group had better therapeutic effect on lowering blood lipid and on improving electrocardiogram than the control group ( P<0.05) . ( 2) After treatment, TG, HDL-C, ApoB and ApoA levels were much improved in the treatment group compared with those in the control group (P<0.01) . ( 3) The frequency of angina pectoris attack and the dosage of Nitroglycerin Tablets per week were reduced in the treatment group compared with those in the control group (P<0.01) . (4) After treatment, the treatment group had lower hs-CRP, IL-6, Hcy and ox-LDL levels, and higher APN level than the control group (P<0.01) . Conclusion Tiaozhi Decoction is effective for the treatment of coronary heart disease induced stable angina complicated with hyperlipemia through improving lipid metabolism and reducing angina pectoris attack, and the mechanism may be related with the regulation of inflammatory factors.

ABSTRACT

Objective To explore the influence of shenmai injection on C-reactive protein(CRP) and homocysteine(Hcy) of patients with non-ST-segment elevation acute coronary syndrome.Methods 96 patients with nonST-segment elevation acute coronary syndrome were randomly divided into the control group(48 cases) and the observation group(48 cases).The patients in the control group were treated through the conventional treatment,while the patients in the treatment group were treated by shenmai injection based on the conventional therapy.They were all treated for 14 days.CRP and Hcy were detected before and after treatment.Results The total effective rate of the observation group was 95.8%,which was significantly higher than 87.5% of the control group (x2 =4.760,P <0.05).The plasma levels of CRP and Hcy were all decreased after treatment in both two groups.The CRP and Hcy in the observation group were significantly lower than those in the control group (t =2.927,2.624,all P < 0.05).Conclusion Shenmai injection can significandy decrease the plasma levels of CRP and Hcy in patients with non-ST-segment elevation acute coronary syndrome.

ABSTRACT

Objective To investigate the changes of homocysteic acid (Hcy) and C-reactive protein (CRP) levels in blood plasma of elderly patients with coronary heart disease and elucidate the risk factors of elderly patients with coronary heart disease.Methods 90 elderly patients with coronary heart disease were divided into 3 groups:stable angina pectoris group (SAP)(n=30),unstable angina pectoris group(UAP)(n=30),acute myocardial infarction group(AMI)(n=30).30 healthy elderly persons elder than 60 years old were enrolled as control group.Venous blood of all persons was harvested during empty stomach and plasma was separated.Then plasma Hcy and CRP were determined.Results The TC and TG levels in the three elderly coronary heart disease groups were significantly higher than those in control group (P

ABSTRACT

L-homocysteic acid (HCA) and other amino acids were conjugated to rat brain material ( extracted rat brain protein) with glutaraldehyde to form HCA- and amino acids-brain material con-jugates. The specificity of monoclonal antibody (McAb) was tested on serial dilution test and ab-sorption test on enzyme- linked immunosorbent assay (ELISA) using these conjugates as antigens instead of amino acids-BSA (bovine serum albumin) conjugates used previously. The characterized McAb was applied for immunohistochemical staining using PAP (peroxidase antiperoxidase) tech-nique in combination with silver enhancement of diamino-benzene (DAB) products. The results in-dicated that McAb to L-HCA reacted with L-HCA-brain material conjugates, but not with other a-mino acids-brain material conjugates so far tested. McAb absorbed with L-HCA-brain material a-bolished or decreased immunoreactivity of L-HCA-brain material with McAb. The antibody selec-tively stained subpopulation of cells and processes in the hippocampus fixed with glutaradehyde. Absorption of McAb with L-HCA-brain material abolished immunohistochemical staining. These results suggested that McAb was specific for L-HCA-brain materials and could be used for imuno-histocytochemistry. This would provide a new tool for immunohistochemical visualization and locali-zation of L-HCA in the nervous system.

ABSTRACT

Objective:To select the differentially expressed vascular smooth muscle cell(VSMC) apoptosis gene induced by icariin,glucose regulated protein 78(GRP78),to observe the expression of GRP78 in atherosclerosis tissue and to the elucidate the mechanism of anti-atherosclerosis of icariin.Methods:The model was established by using halfcystine in viro.The vascular smooth muscle cell was incubated with various concentrations of icariin for 48 hours.The apoptosis was detected by TUNEL assay.GRP78 gene was amplificated by PCR,recombinant plasmid were constructed,transfected into E.coli,positive clonings were selected and conf irmed by restriction endonucleas analysis,and sequence-ed.Tissue situ hybridization was used to observe the GRP78 expression in atherosclerosis tissue.Results:Icariin promoted obviously vascular smooth muscle cell apoptosis.Furthermore,the fragment,648bp,was similar to the gene of GRP78 from VSMC,and the expression level was signif icantly higher in high cholesterol group than in control nroup.Conclusions:The GRP78 plays a key role in inducing atherosclerotic lesions.The mechanism of anti-atherosclerosis of icariin might be partly due to promoting apoptosis of vascular smooth muscle cell.

ABSTRACT

The present study was undertaken to characterize homocysteic acid (HCA)-and cysteic acid (CA)mediated formation of inositol phosphates (InsP) in primary culture of rat cerebellar granule cells. HCA and CA stimulated InsP formation in a dose-dependent manner, which was prevented by the N-methyl-D-aspartate (NMDA) receptor antagonist D,L-2-amino-5-phosphopentanoic acid (APV). CA-, but not HCA-, mediated InsP formation was in part prevented by the metabotropic glutamate receptor antagonist alpha-methyl-4-carboxyphenylglycine ((+/-)-MCPG). Both HCA- and CA-mediated increases in intracellular calcium concentration were completely blocked by APV, but were not altered by (+/-)-MCPG. CA-mediated InsP formation was in part prevented by removal of endogenous glutamate. In contrast, the glutamate transport blocker L-aspartic acid-beta-hydroxamate synergistically increased CA responses. These data indicate that in cerebellar granule cells HCA mediates InsP formation wholly by activating NMDA receptor. In contrast, CA stimulates InsP formation by activating both NMDA receptor and metabotropic glutamate receptor, and in part by releasing endogenous glutamate into extracellular milieu.
