ABSTRACT

OBJECTIVE: To observe the effect of scalp-acupuncture intervention on the expression of Interleukin (IL)-10 mRNA, IL-6 mRNA and tumor necrosis factor (TNF) - α in the parahippocampal gyrus of cerebral ischemia (CI) rats, so as to explore its molecular mechanisms underlying improvement of CI. METHODS: A total of 64 male SD rats were randomized into normal control, model, medication and scalp-acupuncture groups (n=16 rats in each group). The focal CI model was established by middle cerebral artery occlusion. Intraperitoneal injection of Ammonium Pyrrolidine Dithiocarbamate (100 mg•kg-1•d-1) was administrated for rats in the medication group, once a day for 7 days. For rats of the scalp-acupuncture group, the acupuncture needles were rapidly inserted into bilateral Dingnieqianxiexian (MS6), followed by twirling the needles at 100 cycles/min for 1 min, once again every 10 min during 20 min' needle retention. The treatment was conducted once a day for 7 days. The neurologic deficit score (0-4 points, impaired consciousness, death, etc.) and neurological function score (motor, sensory and sensory tests, 0-10 points) were assessed according to Longa's (1989) and Schabitz's (2004) methods, respectively. The expression levels of IL-10 mRNA and IL-6 mRNA were detected by fluorescence quantitative PCR, and the expression of TNF-α was detected by immunohistochemistry. RESULTS: After modeling, the neurologic deficit and neurological function scores and the expression levels of IL-10 mRNA, IL-6 mRNA and TNF-α protein in the parahippocampus were significantly increased in the model group than in the normal control group (P<0.01). Following the intervention, the neurologic deficit and neurological function scores as well as IL-6 mRNA and TNF-α protein expression were significantly down-regulated in both scalp-acupuncture and medication groups (P<0.05), and the expression of IL-10 mRNA was obviously increased (P<0.05) relevant to the model group. CONCLUSION: Scalp-acupuncture can improve neurologic function in CI rats, which is related to its effects in up-regulating the expression of IL-10, then down-regulating the expression of IL-6 and TNF-α (reducing inflammatory response) in the parahippocampal gyrus.

@#Objective To observe the expression of IL-6 mRNA and study the mechanism of acupuncture in focal cerebral ischemia.Methods In situ hybridization was used to detect the expression of IL-6 mRNA in the sham operation group, control group and acupuncture group 2 h, 6 h, 12 h, 24 h after reperfusion in the brain of middle cerebral artery occlusion rats, meanwhile and also observe ultrastructure in cortex of ischemic brain tissue 24 h after reperfusion.Results The sham operation group showed a basic expression of IL-6 mRNA in cortex and striate body. The cells expressing IL-6 mRNA were increased 2 h after ischemic reperfusion in cortex and striate body and peaked 24 h after reperfusion in control group and acupuncture group. The cells expressing IL-6 mRNA in cortex of ischemic area were stronger in acupuncture group than in control group at every research time point besides 2 h after ischemic reperfusion. The cells expressing IL-6 mRNA striate body of ischemic area were stronger in acupuncture group than in control group at every research time point. Acupuncture can lighten the degeneration of cell nucleolus and necrosis of cell, resume the integrity of the nucleolus, improve the function of mitochondrial and endoplasmic, and have significant difference with control group. Conclusion Acupuncture can increase the cells expressing of IL-6 mRNA.

ABSTRACT

Objective To study the effect of Wulong granules on the expression of IL-2 mRNA,IL-4 mRNA and IL-6 mRNA in allergic rhinitis(AR) rats in Spleen Qi deficiency type. To establish the clinic treatment principle-tonify Spleen and reinforce Qi of AR through the lab study. Methods Established AR model with combination of disease and Zheng,checked the expression of IL-2 mRNA,IL-4 mRNA and IL-6 mRNA of nasal mucosa were detected with RT-PCR. Results Wulong granules can adjust the expression of mRNA of AR rats' nasal mucosa. It can descend the ascensus of IL-4 mRNA,IL-6 mRNA and the drop of IL-2 mRNA. Conclusion Wulonggranules can show treatment effect to AR rats through adjusting the expression of cell factors such as IL-2 mRNA,IL-4 mRNA and IL-6 mRNA.

Abstract Objective:The effects of both TM-TGFa and S-TGFa on proliferation were compared to explore the biological characteristics of the two forms of TGF-a.Methods:The proliferation of mouse fibroblast cell line NIH3T3 and its expression of PCNA were determined by the methods of MTT and histochemical technique. The level of IL-6 mRNA in HaCaT cell line was tested by in situ hybridization. ABSTRACT

Objective To investigate the main origin of IL-6 during acute rejection of renal transplantation and provide the experimental foundations for expounding the molecular pathogenic mechanism of acute rejective reaction of renal transplantation Methods 3'IL-6 probe with in situ hybridization technique was used to detect the IL-6mRNA expression in renal graft. Results (1) Renal graft expression of IL-6mKNA in acute rejection was higher than that in CsA toxicosis, stable transplantation and normal control. (2) Expression of IL-6mRNA in tubular epithe-lium of acute rejection graft was higher than that in glomerular cells ,vascular endotheli-um and interstitium. (3) The expression of IL-6mRNA in renal graft of CsA toxicosis was similar with that in stable transplantation and normal control. Conclusion Renal graft cells may produce IL-6 directly during acute rejection. The abnormal expression and activation of IL-6 in renal graft may play an important role in the mechanism of renal acute rejection. Graft tubular epithelium may play a more important role in the cause of immunorejection.
