ABSTRACT

This work describes the chemical characterization of extracts of Jatropha gossypiifolia (from Amazonia region) concerning the presence of phenolic and triterpenic compounds using Ultra High Perfomance Liquid chromatography coupled to Mass Spectrometry in Tandem (UHPLC-MS/MS) methods. The studied compounds belong to the most frequently found classes in medicinal plants (triterpenes, flavonoids, flavones, hydroxycinnamic acids, coumarins, catechins and stilbenes), which have been analyzed as chemical and bioactive markers in hydroethanolic and aqueous extracts. Several polyphenolic compounds identified herein are unprecedented in the scientific literature for this species. The chemical markers identified and quantified in the studied extracts of J. gossypiifolia were gallic acid, chlorogenic acid, catechin, caffeic acid, vanillic acid, p-coumaric acid, ferullic acid, rutin, quercitrin, 3-acetylcoumarin, trans-cinnamic acid, quercetin, luteolin, apigenin, kaempferol, chrysin. α-amyrin, β-amyrin and lupeol. As the majority compounds, (+)-catechin, p-coumaric acid, ferulic acid, luteolin, α-amyrin and β-amyrin were found to be present at mg kg-1 levels. J. gossypiifolia extracts presented a high in vitro activity against different reactive oxygen species (hydroxyl, peroxyl, and superoxide anion radicals). Several polyphenolic compound data presented herein are unprecedented in the scientific literature for this plant species. As a result, this plant can be a new source of bioactive molecules for therapeutic purposes.

ABSTRACT

Confounding factors such as plant species, its location and other environmental associated chemistry were reported to influence the dynamics of phytochemicals from being beneficial health-wise to metabolites that facilitate toxic induced effects. This study investigated the oral sub-acute toxicity profile of the aqueous leaf extracts (ALE) of Jatropha gossypiifolia collected from Mubi, Adamawa State, Nigeria. Generalized loss of body weight, weaknesses, dizziness, loss of appetite and restlessness were observed in the acute toxicity study with more severe effects and mortality recorded in the groups exposed to higher doses of 1000 and 2000 mg/kg body weight. In the sub-acute toxicity study, the ALE following the oral administration of 240 mg/kg, 450 mg/kg and 583 mg/kg for 28 days was observed to profoundly alter the normal architecture of the liver and the kidney. The pathological lesions were observed to have disrupted the normal concentration of the serum biomarkers. The ALT concentrations were found to increases to 10.28±1.26 U/L at 250 mg/kg, 9.38±0.57 U/L at 450 mg/kg and 9.31±0.77 U/L at 583 mg/kg respectively when compared to the control (5.86±0.34 U/L). The concentrations of AST were observed to increase to 49.07±16.2 U/L at 250 mg/kg, 47.10±15.42 U/L at 450 mg/kg and 53.07±10.32 U/L at 583 mg/kg respectively when compared to the control (46.11±9.21 U/L). The activity levels of ALP further shows an increase of 379.50±11.31 U/L at 250 mg/kg, 624.90±22.46 U/L at 450 mg/kg and 662.73±28.62 U/L at 583 mg/kg respectively when compared to the control (349.97±71.70 U/L). It is evident from this study that the ALE of the J. gossypiifolia species in addition to its health benefits also contains a cocktail of toxic phytochemicals. Thereby redefined the previous conclusions that the aqueous leaf extract of J. gossypiifolia plant is entirely safe. Thus, reinforcing the importance of the uses of indigenous/site-specific experiment, having in mind that some phytochemicals are sensitive to numbers of confounding factors.

ABSTRACT

The action of extracts from the stem, leaves, and fruit of Jatropha gossypiifolia on Biomphalaria glabrata was studied by analyzing survival, feeding capacity and oviposition ability. The extracts were obtained by macerating the plant parts in 92% ethanol, which were then evaporated until a dry residue was obtained and phytochemically studied. The molluscicidal activity on B. glabrata was investigated using the procedures recommended by WHO (1965). The amount of food ingested and oviposition were measured during each experiment. The extract of leaves from J. gossypiifolia was shown to be a strong molluscicidal agent, causing 100% mortality of B. glabrata, even in the lowest concentration tested, of 25 ppm. Regarding the fruit extract, there was variation in the mortality, depending on the concentration used (100, 75, 50 and 25 ppm). The snails that were in contact with the fruit extract had significant reduction in feeding and number of embryos in comparison to the control. The stem extract did not present molluscicidal activity nor had any influence on the feeding and oviposition abilities of B. glabrata, in the concentrations tested. In conclusion, the extracts of leaves and fruits of J. gossypiifolia investigated in this work show molluscicidal effect and may be sources of useful compounds for the schistosomiasis control.


Estudou-se a ação dos extratos do caule, folhas e frutos de Jatropha gossypiifolia (Pinhão-roxo) sobre Biomphalaria glabrata analisando a sobrevivência, capacidade alimentar e de oviposição. Os extratos foram obtidos pela maceração das partes do vegetal em álcool etílico 92%, evaporados até obter-se um resíduo seco e estudados fitoquimicamente. A atividade moluscicida em B. glabrata seguiu os procedimentos recomendados pela WHO (1965). A medida de quantidade de alimento ingerido e a oviposição foi realizada durante cada experimento. O extrato das folhas de J. gossypiifolia mostrou-se um forte agente moluscicida, promovendo 100% de mortalidade de B. glabrata mesmo na menor concentração testada, 25 ppm. Com o extrato dos frutos houve variação na mortalidade, de acordo com a concentração utilizada (100, 75, 50 e 25 ppm). Os caramujos em contato com o extrato dos frutos apresentaram quando comparado ao controle uma diminuição significativa nos comportamentos alimentares e de número de embriões. O extrato do caule não apresentou atividade moluscicida e nenhuma influência sobre a capacidade alimentar e de oviposição de B. glabrata nas concentrações testadas. Em conclusão, os extratos de folhas e frutos de J. gossypiifolia investigados neste trabalho apresentam efeito moluscicida e possivelmente podem ser fontes de compostos no controle da esquistossomose.

ABSTRACT

A cicatrização constitui processo complexo envolvendo diferentes sistemas biológicos e imunológicos , sendo essencial para manter a integridade do organismo. Três fases bem definidas ocorrem, a inflamatória, a proliferativa e a de maturação. A falha ou prolongamento em uma fase pode resultar em retardo da cicatrização tecidual. A sutura dos tecidos e sua cicatrização é um dos fundamentos básicos da cirurgia e a procura de substâncias que melhorem este processo é um desafio constante. O uso de substâncias de plantas têm sido testados por vários autores. Objetivo - Analisar comparativamente as alterações macroscópicas e histológicas proporcionadas pelo uso do extrato bruto da Jatropha gossypiifolia intraperitonial, na cicatrização de suturas realizadas na bexiga urinária de ratos. Material e método – Sessenta ratos da linhagem Wistar, adultos, machos foram distribuídos em 2 grupos animais. O procedimento experimental constituiu-se em laparotomia mediana infraumbelical, incisão longitudinal de 1cm na parede ventral da bexiga e síntese em plano único com pontos separados de poliglactina 910 5-0 (Ethicon). O procedimento nos animais do grupo controle (ratos 1 a 30) instilou-se na cavidade peritonial água destilada na proporção de 1ml por kg de peso e no grupo Jatropha (ratos 31 a 60) utiilizou-se o extrato bruto de Jatropha gossypiifolia na proporção de 1ml por kg de peso, que representava 200mg do fototerápico, intraperitoneal. Cada grupo foi dividido em 3 subgrupos de 10 animais sendo estes submetidos à eutanásia no 7° e 14º pós-operatório. Foi feita análise macroscópica e histológica comparativa entre os subgrupos . Resultados - No 7° dia foi observado diferença estatisticamente significativa nas variáveis inflamação aguda, neoformação vascular e colagenização, sendo a primeira maior no grupo controle e as duas últimas no grupo Jatropha; no 14° variáveis inflamação aguda e proliferação fibroblástica apresentaram-se mais intensas com significado estatístico ...


The healing process is quite complex, involving different biological and immunological systems and being essential for maintaining the organism integrity. Three well-defined stages take place: inflammatory, proliferate and that of maturation. Failure or lengthening of a phase could result in a delay of tissue healing. The suture of tissues and their healing process is one of the basic fundamentaIs of surgery, and the research on substances which bring an improvement of the process is an ongoing challenge. The use of plant substances has been tested by several researchers. Study Aim - To comparatively analyze the macroscopic and histological alterations brought about by the use of intraperitoneal Jatropha gossypiifolia gross extract, in the healing process of sutures performed on the bladder of rats. Material and Method – Sixty (60) adult, male, Wister rats were distributed into two animal groups. The experimental procedure constituted of infraumbilical median laparatomy, 1 cm longitudinal incision on the bladder ventral wall, and single plane synthesis with separarate polyglactine 910 5-0 (Ethicon) stitches. The procedure for the control group (1 to 30) was peritoneal cavity instillation of distilled water at a ratio of 1 ml/kg/weight, and a 1 ml/kg/weight of Jatropha gossypiifolia extract for the Jatropha group (rats 31 to 60), which represented 200 mg of the intraperitoneal phytotherapical. Each group was subdivided into 3 subgroups of 10 animaIs, being submitted to euthanasia on days 7, 14 and 21 post-surgery. Comparative histological and macroscopic analysis was undertaken between the subgroups. ABSTRACT

This work presents the observed changes in Wistar rats under long treatment (thirteen weeks) with different oral doses of the ethanolic extract (EE) from Jatropha gossypiifolia L., Euphorbiaceae. The most significant toxic signs indicated a reduction of the activity in the central nervous system and digestive disturbances. The histopathological analysis shows hepatotoxity and pulmonary damages. The lethality was 46.6% among males under the higher experimental dose (405 mg/kg) and 13.3% both in females under the higher dose and among the animals treated with 135 mg/kg of the product. These data show the significant oral chronic toxicity of EE of J. gossypiifolia in rats.

ABSTRACT

The antispasmodic activity of Jatropha gossypiifolia L., Euphorbiaceae, aerial parts was investigated in rodents using the mouse intestinal transit model and acetylcholine (ACh, 10-9 to 10-4 M) and calcium (CaCl2, 10-4 to 10-1 M)-induced contractions of isolated rat jejunum. Similar to atropine (1 mg/kg), oral doses of ethanolic extract (EE) of J. gossypiifolia (500, 1000 and 2000 mg/kg) produced a decrease in intestinal transit (37.6 to 57.1 percent) when compared with control. The ACh-induced contraction in the jejunum was inhibited by EE (0.5, 1.0 and 2.0 mg/mL), chloroformic (CF) and aqueous fractions (0.1 and 0.5 mg/mL) and methanolic subfraction (0.05 and 0.25 mg/mL), suggesting an antimuscarinic mechanism. CaCl2 - induced responses in jejunum were also attenuated in the presence of CF (0.05 and 0.1 mg/mL) implying a direct interference of CF with the influx of calcium ions in the cells. Only the organic fraction of the extract had a calcium-antagonist effect, whereas both chloroformic and aqueous fractions had anticholinergic effect. These results suggest that the antispasmodic effect of J. gossypiifolia may be due a combination of anticholinergic and calcium antagonist mechanisms.

ABSTRACT

A espécie Jatropha gossypiifolia L. (Euphorbiaceae), popularmente conhecida como pião-roxo, entre tantos outros nomes, é um bom exemplo do tênue limiar que separa o efeito terapêutico do tóxico. Apesar de ser classicamente conhecida como planta tóxica possui usos na medicina popular. Alguns desses efeitos têm sido comprovados em estudos experimentais, como os de antimicrobiano, antineoplásico, cicatrizante e hipotensor, sendo possivelmente explicados pela presença de substâncias como alcalóides, terpenóides, flavonóides, lignanas e taninos. Esta revisão aborda aspectos importantes, com ênfase na toxicidade crônica dessa espécie, de modo a servir de fonte de informação aos interessados em avaliar a relação risco/benefício do uso terapêutico de Jatropha gossypiifolia L.


The species Jatropha gossypiifolia L. (Euphorbiaceae), popularly known as bellyache bush, among several other names, is an important example of the tenuous threshold that separates the therapeutic from the toxic effect. Although traditionally known as a toxic plant, it has been used in folk medicine. Some of its effects have been proved by experimental studies as antimicrobial, antineoplastic, healing and hypotensive, likely explained by the presence of substances such as alkaloids, terpenoids, flavonoids, lignans and tannins. This review deals with important aspects, focusing on the chronic toxicity of this species, in order to serve as an information source for those interested in evaluating the risk-benefit ratio of the therapeutic use of Jatropha gossypiifolia L.

ABSTRACT

A Jatropha gossypiifolia L. apesar de ser usada na medicina popular com finalidades diversas, é uma espécie classicamente catalogada como tóxica. Este trabalho teve como objetivo a pesquisa de alterações histopatológicas em vísceras de ratos sob tratamento agudo com o extrato etanólico (EE) da Jatropha gossypiifolia L. Ratos Wistar foram tratados por via oral (gavagem) com doses únicas do extrato de até 5 g/kg e observados por 14 dias. Após esse período, os animais foram sacrificados por tração cervical e as vísceras foram coletadas. O coração, o fígado e o rim foram seccionados por incisão sagital e os pulmões submetidos à perfusão via traquéia, com solução de formol a 10 por cento. As secções teciduais foram processadas conforme os métodos habituais, coradas pela hematoxilina-eosina e tricômico e observadas ao microscópio óptico. Apenas nos animais tratados com a maior dose do extrato (5 g/kg) foram observadas alterações em fígado e pulmão evidenciadas por resposta inflamatória e estimulação do sistema imunitário. Estes resultados indicam uma toxicidade aguda oral relativamente baixa; entretanto, corroboram com os indícios de hepatotoxicidade já publicados, expondo ainda um potencial de toxicidade pulmonar do produto, o que ressalta a importância de estudos toxicológicos de longa duração com o EE da espécie Jatropha gossypiifolia L.


Jatropha gossypiifolia L. has been used in folk medicine in Brazil despite its classification as a toxic plant. The aim of this work was the assessment of histopathological a lterations in rats after acute treatment with the ethanol extract (EE) from Jatropha gossypiifolia L. Wistar rats were treated by gavage with single doses of EE until the limit dose of 5 g/kg (w.b.). 14 days after treatment the rats were sacrificed and the viscera were collected. The hard, liver and kidney were sectioned by sagittal incision and the lung submitted to perfusion with phormol (10 percent). The histological sections were processed by the usual methods in hematoxilin-eosin and trichrome staining and were observed through optical microscopy. Only in treated rats with 5 g/kg (w.b.) we observed some alterations in the liver and lung that means inflammatory response and immune activation. These results indicate a low oral acute toxicity, in relative terms, however, it was confirmed the hepatic toxicity already reported and showed the importance of long-term toxicological studies of the EE from Jatropha gossypiifolia L.

ABSTRACT

Este trabalho teve como objetivo a avaliação da toxicidade aguda do extrato etanólico (EE) de partes aéreas de Jatropha gossypiifolia L., espécie vegetal muito usada na medicina popular, apesar de ser catalogada como planta tóxica. Ratos Wistar foram tratados por via oral com doses únicas do extrato (1,2 g/kg; 1,8 g/kg; 2,7 g/kg; 4,0 g/kg e 5,0 g/kg) e observados por 14 dias. Os principais sinais de toxicidade encontrados, em alguns animais, foram: ptose palpebral, perda de peso e paralisia do trem posterior. Outras alterações significativas ocorreram apenas em machos tratados com a dose de 5 g/kg: aumento dos níveis sangüíneos de creatinina, AST, sódio e potássio; diminuição dos níveis de uréia e albumina; leucopenia, além de discretas alterações na coloração e consistência de vísceras. A dose letal mediana (DL50) foi superior a 4,0 g/kg para machos e maior do que 5,0 g/kg para fêmeas. Estes resultados indicam uma toxicidade aguda oral relativamente baixa; contudo, ressaltam a necessidade da realização de estudos toxicológicos de longa duração com o EE de J. gossypiifolia L.


The aim of this work was the assessment of acute oral toxicity of the ethanol extract (EE) of the aerial parts of Jatropha gossypiifolia L. This plant is used in folk medicine despite its classification as a toxic plant. Wistar rats were treated per oralis with single doses of EE [1.2 g/kg; 1.8 g/kg; 2.7 g/kg; 4.0 g/kg and 5.0 g/kg (w.b.)] and observed for 14 days. The most important signs of toxicity were: ptosis; reduction of body weight and hindlimb paralysis. Other sgnificant alterations occurred only in males treated with 5.0 g/kg (w.b.): increase of creatinine, AST, sodium and potassium seric levels; reduction of urea and albumin; leukopenia and small alterations in color and consistence of viscera. The LD50 value was higher than 4.0 g/kg (w.b.) for males and higher than 5.0 g/kg (w.b) for females. These results indicate a low oral acute toxicity, in relative terms, however it shows the importance of long-term toxicological studies of the EE from J. gossypiifolia.

ABSTRACT

INTRODUÇÃO:A cicatrização do trato gastrintestinal é de grande importância na história da cirurgia, buscando-se tipos de fios e pontos apropriados e os diversos fatores que a influenciam. O uso de fitoterápicos como aceleradores da cicatrização tem sido feito por muitos anos sendo objetivo atual de pesquisas para a comprovação científica dessas propriedades terapêuticas. OBJETIVO: Avaliar o uso do extrato bruto de Jatropha gossypiifolia L. na cicatrização de gastrorrafias em ratos. MÉTODOS: Foram utilizados 40 ratos Wistar, machos, divididos em dois grupos de 20, denominados de grupos controle e Jatropha. Dez animais de cada grupo foram mortos no 3º dia pós-operatório e denominados subgrupos controle e Jatropha do 3º dia e os 10 restantes de cada grupo foram mortos no 7º dia com a mesma denominação do 7º dia. Em cada animal foi realizado gastrotomia e gastrorrafia em plano único com fio polipropileno 6-0 (Prolene®, Ethicon). Os animais do grupo Jatropha receberam dose única de 200mg/Kg do extrato bruto de Jatropha gossypiifolia L via intraperitoneal no dia do procedimento e os do grupo controle a mesma quantidade em mililitros de solução salina (cloreto de sódio à 0,9%). Foram avaliados os seguintes parâmetros: 1) alterações macroscópicas; 2) a resistência à insuflação de ar atmosférico (pressão de ruptura) da sutura; 3) características histológicas. RESULTADOS: Não houve morte dos animais na evolução clínica, ocorrendo boa cicatrização da parede abdominal, com ausência de sinais de infecção, deiscência, abscessos ou peritonites. A cicatrização da superfície serosa foi considerada boa em todos os animais, não ocorrendo fístulas, porém, as aderências intra-peritoneais ocorreram em sete ratos do subgrupo controle e nove do subgrupo Jatropha do 3º dia pós-operatório e em nove do subgrupo controle e oito do subgrupo Jatropha do 7º dia, não havendo diferenças estatisticamente significantes entre os grupos. A cicatrização da superfície mucosa foi classificada como boa em todos os animais. A resistência das gastrorrafias à insuflação de ar atmosférico demonstrou aumento estatisticamente significante da pressão de ruptura no grupo Jatropha do 3º dia de observação. A avaliação histológica demonstrou diferenças estatisticamente significantes, quanto aos critérios, reação inflamatória aguda menor e coaptação das bordas maior no subgrupo Jatropha do 7º dia pós-operatório em relação ao grupo controle do mesmo período. CONCLUSÃO: O extrato bruto de Jatropha gossypiifolia L. favorece a cicatrização no 3º dia pós-operatório em relação à maior resistência das gastrorrafias à pressão de ruptura, e no 7º dia com melhor coaptação das bordas e reduzindo reação inflamatória aguda à microscopia.


INTRODUCTION: The healing process of the gastrointestinal tract is of huge importance in the surgical field history, as well as the search for appropriate types of threads and suture techniques and the many factors that influence it. The use of phytotherapic drugs as accelerators of the healing process has been done for many years and is one of the current objectives of scientific researches trying to prove its therapeutic properties. PURPOSE: To evaluate the use of Jatropha gossypiifolia L. extract on the healing process of gastrorraphies in rats. METHODS: Forty wistar male rats were divided in 2 groups of 20 rats, named control and Jatropha groups. Ten animals of each group were killed in the third day post-surgery and were named control and Jatropha groups of the 3rd day. The remaining 10 animals of each group were killed in the seventh day and were named accordingly for the 7th day. In each animal, gastrostomy and gastrorraphy were performed in a single plane using polypropylene thread 6-0 (Prolene). The animals from the Jatropha group were given a single dose of 200 mg/kg of the Jatropha gossypiifolia L. extract intraperitoneally on the same day of the procedure and the ones from the control group were given the same quantity in milliliters (ml), but of saline solution (sodium chloride 0.9%). The following parameters were evaluated: 1) macroscopic alterations; 2) the suture's resistance to atmospheric air insufflation (pressure of rupture); 3) histologic characteristics. RESULTS: No animal died during the clinical follow-up and optimal healing of the abdominal wall was seen without any signs of infection, dehiscence, abscesses or peritonitis. Healing of the serous surface was considered good in all animals, without occurrence of fistulas; however, intraperitoneal adhesions occurred in 7 rats of the sub-group control and 9 of the sub-group Jatropha on the 3rd day post-operative and in 9 of the sub-group control and 8 of the sub-group Jatropha on the 7th day, but the differences were not statistically significant between the groups. Healing of the mucous surface was classified as good in all the animals. The resistance of the gastrorraphies to the atmospheric air insufflation showed statistically significant increase of the rupture pressure in the Jatropha group during the 3rd day of observation. Histologic evaluation showed differences that were statistically significant, considered the criteria, as well as reduced acute inflammatory reaction and better coaptation of the edges in the sub-group Jatropha of 7th post-surgery, when comparet to the sub-group control of the same period. CONCLUSION: The raw extract of Jatropha gossypiifolia L. aids the healing on the 3rd day post-surgery, concerning the enhanced resistance of the gastrorraphies to pressure of rupture, and on the 7th day, presenting better coaptation of the edges and reducing acute inflammatory reaction by microscopic analyses.

ABSTRACT

INTRODUÇÃO: A sutura dos tecidos e sua cicatrização é um dos fundamentos básicos da cirurgia e a procura de substâncias que melhorem este processo é desafio constante. O uso de substâncias retiradas de plantas tem sido testado há muitos anos, contudo sem ainda terem comprovação científica consolidada. OBJETIVO: Comparar as alterações macroscópicas e histológicas proporcionadas pelo uso do extrato bruto da Jatropha gossypiifolia L. intraperitonial, na cicatrização de suturas realizadas na bexiga urinária de ratos. MÉTODOS: Quarenta ratos da linhagem wistar, adultos, machos foram distribuídos em dois grupos, experimento e controle. Incisão longitudinal de 1cm na parede ventral da bexiga e síntese em plano único com pontos separados de poliglactina 910 5-0 (ethicon) foi realizada nos dois grupos. No controle, instilou-se na cavidade peritoneal água destilada na proporção de 1ml/kg de peso, enquanto no grupo Jatropha utilizou-se o extrato bruto de Jatropha gossypiifolia L. na proporção de 1ml/kg, que representava 200mg/kg de peso do fitoterápico. Cada grupo foi subdividido em dois subgrupos de 10 animais, sendo estes submetidos à eutanásia no 3° e 7° dia pós-operatório. Foi feita análise macroscópica e histológica comparativa entre os subgrupos. RESULTADOS: No 3° dia foi observada diferença estatisticamente significante nas variáveis inflamação aguda, neoformação vascular e colagenização, sendo a primeira, maior no grupo controle e as duas últimas no grupo Jatropha; no 7° dia as variáveis inflamação aguda e proliferação fibroblástica apresentaram-se mais intensas, com significado estatístico, no grupo controle. CONCLUSÃO: Não se observou efeito favorecedor cicatrizante do extrato bruto da Jatropha gossypiifolia L., intraperitonealmente aplicado, na bexiga urinária de ratos.


INTRODUCTION: The suture of tissues and their healing process is one of the basic fundamentals of surgery, and the research on substances which could improve the process is an ongoing challenge. The use of plants has been tested but till nowadays without scientific demonstration. PURPOSE: To compare the macroscopic and histological alterations done by the use of intraperitoneal Jatropha gossypiifolia L. gross extract, in the healing process of sutures performed on the bladder of rats. METHODS: Forty adult, male, Wister rats were distributed into two animal groups. One centimeter longitudinal incision on the bladder ventral wall, and single plane synthesis with separated polyglactine 910 5-0 (Ethicon) stitches was done in all the animals. In the control group an intraperitoneal cavity instillation of distilled water at a ratio of 1ml/kg/weight, and a 1ml/kg/weight of Jatropha gossypiifolia L. extract for the Jatropha group was done representing 200 mg of the substance. Each group was subdivided into two with 10 animals in each, being submitted to euthanasia on 3th and 7th post-operative days. Comparative histological, macroscopic and statistical analysis were undertaken between the subgroups. RESULTS: Statistical significant difference was observed in the acute inflammation changes, vascular neoformation and bonding on day 3; the first one was greater in the control group, and the latter two greater in the Jatropha group; acute inflammation variables and fibroblastic proliferation presented to be more intense on day 7, with statistical significance favoring the control group. CONCLUSION: No favorable healing effect was observed with the administration of single intraperitoneal dose of Jatropha gossypiifolia L. gross extract on the sutures of bladder in rats.

ABSTRACT

INTRODUÇÃO: A espécie vegetal Jatropha gossypiifolia L., conhecida também como pião roxo, é utilizada na medicina popular como cicatrizante, anti-hipertensivo, purgativo e diurético. OBJETIVO: Avaliar a influência da administração intraperitoneal da Jatropha gossypiifolia L., na cicatrização de suturas da parede abdominal ventral de ratos, observando-se os seus aspectos macroscópicos, tensiométricos e microscópicos. MÉTODOS: Foram utilizados no procedimento 40 ratos da linhagem Wistar, machos, distribuídos em dois grupos de 20. Após incisão da parede e abertura da cavidade abdominal, foi instilado 1 ml/kg/peso de cloreto de sódio a 0,9 por cento no grupo controle e no grupo Jatropha o extrato bruto etanólico da Jatropha gossypiifolia L., na concentração de 1 ml/kg/ peso. Realizou-se a sutura da parede abdominal com fio de polipropileno, com pontos separados. Os animais foram avaliados na sua evolução pós-operatória e mortos em dois subgrupos, no 3° e 7° dia. Analisou-se a parede abdominal ventral macroscopicamente, mediu-se a força de resistência a tensão e foram estudados os aspectos histológicos do reparo cicatricial. RESULTADOS: No exame macroscópico encontraram-se aderências mais intensas nos subgrupos Jatropha no 3° e 7° dia. A avaliação tensiométrica foi significantemente maior nos subgrupos Jatropha no 3° e 7° dia. A avaliação histológica comparativa entre os subgrupos demonstrou que o processo inflamatório agudo foi significantemente maior no subgrupo Jatropha no 3° e 7° dia; a neoformação capilar foi significantemente maior no 3° dia pós-operatório do subgrupo Jatropha sendo os outros parâmetros histológicos semelhantes. CONCLUSÃO: O uso do extrato bruto de Jatropha gossypiifolia L. intraperitoneal não demonstrou melhora significativa no processo de cicatrização da sutura da parede abdominal ventral de ratos com a dose e concentração utilizadas.


INTRODUCTION: The Jatropha gossypiifolia L., which is used in popular medicine is considered to have good diuretic effect in hypertension and is also used as a laxative drug. It seems to have a healing effect, although not proved till now. PURPOSE: To evaluate the influence of intraperitoneum administration of Jatropha Gossypiifolia L., in suture healing of ventral abdominal wall of rats, through tensiometric measurement, macro and microscopic aspect of post-operative period. METHODS: Forty wistar male rates were allocated in two groups of 20 animals . After the incision and exposure of abdominal cavity 1 ml/kg/weight of 0,9 percent sodium chloride solution was injected in control group, and in the other one the injection was of 1 ml/kg/weight of a gross ethanol extract of Jatropha gossypiifolia L. The suture of the abdominal wall was than performed with polypropylene separated stitches. The animals were followed-up and killed in the third and seventh days. The ventral abdominal wall was macroscopically analyzed, the resistance strength to strain was measured and it was also studied the histological aspects. RESULTS: On macroscopic examination more intense adhesion was found on the group of Jatropha in both third and seventh post-operative days. The strain evaluation was meanly greater on Jatropha group also in third and seventh days. CONCLUSION: The histological comparative analysis between the different groups showed that the acute inflammatory process was meanly greater for the Jatropha group in third and seventh post-operative days. The vascular neoformation was significantly greater in third pos-operative day of Jathopha group; the other histological parameters were just alike. The intraperitoneum injection of Jatropha extract did not have any significant improvement for the wound healing on ventral abdominal wall on the evaluated animals in this study, no matter if analyzed at the third or seventh pos-operative days.
