ABSTRACT

Objective: To investigate the clinical efficacy of fecal microbiota transplantation (FMT) for treating steroid-refractory gastrointestinal acute graft-versus-host disease (GI-aGVHD) . Methods: This analysis included 29 patients with hematology who developed steroid-refractory GI-aGVHD after allogeneic hematopoietic stem cell transplantation (allo-HSCT) in Huaian Hospital Affiliated to Xuzhou Medical University from March 2017 to March 2022. Among them, 19 patients underwent FMT treatment (the FMT group) and 10 patients did not (the control group). The efficacy and safety of FMT were assessed, as well as the changes in intestinal microbiota abundance, lymphocyte subpopulation ratio, peripheral blood inflammatory cytokines, and GVHD biomarkers before and after FMT treatment. Results: ① Complete remission of clinical symptoms after FMT was achieved by 13 (68.4%) patients and 2 (20.0%) controls, with a statistically significant difference (P<0.05). Intestinal microbiota diversity increased and gradually recovered to normal levels after FMT and FMT-related infections did not occur. ②The proportion of CD3(+) and CD8(+) cells in the FMT group after treatment decreased compared with the control group, and the ratio of CD4(+), regulatory T cells (Treg), and CD4(+)/CD8(+) cells increased (all P< 0.05). The interleukin (IL) -6 concentration in the FMT group was lower than that in the control group [4.15 (1.91-5.71) ng/L vs 6.82 (2.40-8.91) ng/L, P=0.040], and the IL-10 concentration in the FMT group was higher than that in the control group [12.11 (5.69-20.36) ng/L vs 7.51 (4.10-9.58) ng/L, P=0.024]. Islet-derived protein 3α (REG3α) was significantly increased in patients with GI-aGVHD, and the REG3α level in the FMT group was lower than that in the control group after treatment [30.70 (10.50-105.00) μg/L vs 74.35 (33.50-139.50) μg/L, P=0.021]. Conclusion: FMT is a safe and effective method for the treatment of steroid-refractory GI-aGVHD by restoring intestinal microbiota diversity, regulating inflammatory cytokines, and upregulating Treg cells.

Resumen Introducción: La determinación de las diferentes subpoblaciones de los linfocitos T en las diversas patologías y el monitoreo postratamiento ayuda a que el médico tome decisiones terapéuticas teniendo como referencia la cinética de los linfocitos T localizados en sangre periférica. Métodos: Se realizó la estandarización de un perfil de moléculas de superficie para la caracterización de subpoblaciones de linfocitos T: naïve, activados y de memoria, así como las células natural killer o asesinas naturales (CD3− CD56+) en sangre periférica de individuos clínicamente sanos. Resultados: Se identificaron las subpoblaciones de linfocitos: naïve (CD3+, CD4+ o CD8+, CD45RA+, CD62L+, CCR7+), activados (CD3+, CD4+ o CD8+, CD45RA+ o CD45RO+, CD69+ y/o CRTAM+), efectores (CD3+, CD4+ o CD8+, CD45RA+, CD62L−, CCR7−), de memoria central (CD3+, CD4+ o CD8+, CD45RO+, CD62L+, CCR7+) y de memoria efectora (CD3+, CD4+ o CD8+, CD45RO+, CD62L−, CCR7−) en las poblaciones de linfocitos T CD4+ y CD8+. Se integraron los datos obtenidos con estadística descriptiva (valores mínimos, valores máximos, media, mediana). Conclusiones: Este panel será de gran utilidad para monitorear pacientes en quienes se requiera valorar el estado inmunológico desde el punto de vista celular. Particularmente, puede apoyar en el seguimiento de los pacientes en los que se requiera evaluar la reconstitución inmunológica (componente celular de estirpe T).


Abstract Background: The knowledge of the participation of different subpopulations of T lymphocytes in various pathologies helps to make therapeutic decisions, having as reference the presence of the different subpopulations of the T lymphocytes associated with the disease. Methods: A profile standardization of surface molecules for the characterization of subpopulations of T cells was conducted: naïve, activated and memory, as well as natural killer (CD3− CD56+) cells in peripheral blood of clinically healthy individuals. Results: Naïve (CD3+, CD4+ or CD8+, CD45RA+, CD62L+, CCR7+), activated (CD3+, CD4+ or CD8+, CD45RA+ or CD45RO+, CD69+ and/or CRTAM+), effectors (CD3+, CD4+ o CD8+, CD45RA+, CD62L−, CCR7−), central memory (CD3+, CD4+ o CD8+, CD45RO+, CD62L+, CCR7+), memory effectors (CD3+, CD4+ or CD8+, CD62RO+, CD62L−, CCR7−) subpopulations were analyzed by flow cytometry. Descriptive statistics parameters were calculated (minimum values, maximum values, mean values, median). Conclusions: This panel can be very useful for monitoring patients in whom the immunological status from a cellular perspective is needed. Particularly, it can support the follow-up of patients who require an immunological reconstitution (T-cell component) evaluation.

ABSTRACT

Objective To assess the value of flow cytometry in the diagnosis of postoperative infection following renal transplantation. Methods According to postoperative imaging findings and laboratory examination outcomes, 51 recipients undergoing the first renal transplantation were divided into the bacteria (n=33), fungus (n=9) and BK virus (n=9) groups. Twenty-eight recipients with stable conditions after renal transplantation were assigned into the stable group. Flow cytometry was adopted to detect the percentage and absolute counting of lymphocyte subpopulation in the peripheral blood of recipients in each group. Renal function, percentage and absolute counting of lymphocyte subpopulation in the peripheral blood were statistically compared among different groups. Receiver operating characteristic (ROC) curve was drawn to evaluate the diagnostic value of the percentage and absolute counting of lymphocyte subpopulation in infectious diseases after renal transplantation. Results Compared with the stable group, the serum creatinine (Scr) and blood urea nitrogen (BUN) levels in the bacteria, fungus and BK virus groups were significantly up-regulated, respectively (P=0.035, 0.007, 0.024; 0.037, 0.006, 0.032). Compared with the stable group, the percentage of CD16+CD56+natural killer (NK) cells was significantly declined in the bacterial (P=0.036) and fungus groups (P=0.015), and the proportion of CD4+/CD8+T cells was dramatically decreased in the fungus group (P=0.004). Compared with the bacterial group, the percentage of CD3+CD8+T cells was significantly elevated (P=0.013 and 0.008), the proportion of CD3+CD4+T cells was considerably declined (P=0.003 and 0.010), and the percentage of CD4+/CD8+T cells was significantly declined (P=0.003 and 0.005) in the fungus and BK virus groups. Compared with the stable group, the quantity of CD3+T cells, CD3+CD8+T cells and CD16+CD56+NK cells was significantly declined in the bacterial, fungus and BK virus groups, respectively (P=0.025, 0.002, 0.003; 0.015, 0.005, 0.006; 0.001, 0.001, 0.031). In addition, the quantity of CD3+CD4+T cells was considerably decreased in the fungus and BK virus groups (P=0.001, 0.003). The quantity of CD19+B cells was significantly reduced in the BK virus group (P=0.019). Compared with the bacterial group, the quantity of CD3+CD4+T cells was considerably lower in the fungus group (P=0.023). ROC curve analysis revealed that the quantity of CD3+CD4+T cells [area under curve(AUC)=0.8492] and CD16+CD56+NK cells (AUC=0.8889) yielded relatively high accuracy in the diagnosis of fungal infection. The quantity of CD3+T cells (AUC=0.8472), CD3+CD4+T cells (AUC=0.8452) and CD19+B cells (AUC=0.8115) yielded relatively high accuracy in the diagnosis of BK virus infection. Conclusions Flow cytometry detection of the lymphocyte subpopulation in peripheral blood can evaluate the immune function of patients. Absolute counting of lymphocyte subpopulation can directly assess the degree of immunity. These two combined parameters provide guiding significance for the diagnosis and differential diagnosis of infectious diseases in recipients after renal transplantation.

Objective To observe the immunological mechanism of glycyrrhizic acid ointment on chronic eczema mouse model. Methods Mice were randomly divided into blank control group,model control group,base material group, dexamethasone acetate cream group,glycyrrhizic acid ointment high,medium and low dose groups.Except for the blank control group,chronic eczema mouse model was established by chronic repeated 2,4-nitrochlorobenzene stimulation.The contents of IL-4, IL-18,IFN-γ in serum and the T lymphocyte subpopulation in the peripheral blood were all determined after 14 days. Results Glycyrrhizic acid ointment in high,medium groups could significantly reduce the contents of interleukin (IL)-2,IL-5,IL-6,IL-18,interferon(IFN)-γ in serum,with significant differences as compared with the model control group(P<0.01 or P<0.05).The content of IL-4 in serum was significantly increased in the glycyrrhizic acid ointment high,medium groups as compared with the model control group (P<0.01 or P<0.05).Compared with model control group,CD+4T lymphocyte percentage and CD+4/CD+8T in peripheral blood were significantly increased,and CD+8T lymphocyte percentage decreased significantly (P<0.01 or P<0.05). Conclusion Glycyrrhizic acid ointment's effect of treating chronic eczema may be related to regulating the balance of Th1 and Th2 and correcting the immune disorders.

A hanseníase é uma doença granulomatosa, infecto-contagiosa causada pelo Mycobacterium leprae. Trata-se de uma infecção crônica com amplo espectro de respostas imunes celulares em humanos. Possui alto poder infectante e baixo poder patogênico. Leprosy is an infectious and granulomatous disease caused by Mycobacterium leprae. The aim of this study was to quantify and compare levels of leucocytes and lymphocyte subpopulations (CD3+, CD3+CD4+, CD3+CD8+, CD19+, CD3-CD16+CD56+) in peripheral blood ofpatients with leprosy and healthy controls. ABSTRACT

Objective To investigate the influence of IVIG on immunologic function and cytokines levels in children with EV71 infection associated high-risk pulmonary hemorrhage. Methods According to the inclu-sion criteria , 64 children were enrolled and randomly divided into two groups: 39 cases in the IVIG treatment group and 25 cases in the general treatment group. The alternations of blood and immune cytokine markers before and after treatment were detected in the patients. Results (1) Before treatment, the peripheral blood T cells, TH and B cells in the IVIG group were higher than those in the general group , but the peripheral blood IgA was lower than that in the general group(P 0.05). Conclusion Disorders of cellular immunity and humoral immunity appeared in children with EV71 infection-re-lated high-risk pulmonary hemorrhage. It has clinical value to use IVIG timely to regulate the immune disorder.

ABSTRACT

To investigate the effects of short-term exposure of beryllium on the human immune system, the proportion of T-lymphocytes such as CD3+, CD4+, CD8+, CD95, and NK cells, andthe proportion of B cells and TNFalpha level in peripheral blood and immunoglobulins in the serum of 43 exposed workers and 34 healthy control subjects were studied. External exposure to beryllium was measured by atomic absorption spectrometer as recommended by the NIOSH analytical method 7300. T lymphocyte subpopulation analysis was carried out with flow cytometer. The working duration of exposed workers was less than 3 months and the mean ambient beryllium level was 3.4 microg/m3, 112.3 microg/m3, and 2.3 microg/m3 in molding (furnace), deforming (grinding), and sorting processes, respectively (cited from Kim et al., 2008). However, ambient beryllium level after process change was non-detectable (< 0.1 microg/m3). The number of T lymphocytes and the amount of immunoglobulins in the beryllium-exposed workers and control subjects were not significantly different, except for the total number of lymphocytes and CD95 (APO1/FAS). The total number of lymphocytes was higher in the beryllium-exposed individuals than in the healthy control subjects. Multiple logistic regression analysis showed lymphocytes to be affected by beryllium exposure (odd ratio = 7.293; p < 0.001). These results show that short-term exposure to beryllium does not induce immune dysfunction but is probably associated with lymphocytes proliferation.

Objective To determine counts of T lymphocyte sub-populations in malignant and tuberculous pleural effusion or ascites and evaluate its significance in difierential diagnosis.Methods T lymphocyte sub-populations in pleural effusion or ascites and peripheral blood were determined in 30 patients with tuberculosis and 31 patients with cancer by flow cytometry.Concentrations of cytokines Th1 and Th2,γ-interferon(IFN-γ),interleukin-12(IL-12)and IL-4 in pleural effusion or ascites were measured by enzyme-linked immunosorbent assay(ELISA).Results Compared to that in peripheral blood,percentage of CD3+ and CD4+ T-celI counts were all higher in both malignant and tuberculous pleural effusion or ascites [(73±6)%and(67±20)%vs.(51±19)%and(48±14)%,P<0.05].Although CD3+T-cells count was higher in tuberculous pleural effusions or ascites,no difference in ratio of CD3+ and CD4+/CD3+ and CD8+ T-cell counts was found between malignant and tuberculous pleural effusions or ascites.However,ratios of IFN-γ and IL-12 to IL-4 were higher in tuberculous pleural effusion or ascites(54±24 and 82±19vs.8±6 and 19±10,t=10.34 and 16.28,respectively,P<0.01).Conclusions CD3+ and CD4+ Tcells can be aggregated in both malignant and tuberculous pleural effusions or ascites,80 nature (tuberculosis or malignancy)of pleural effusion or ascites can not be differentiated by CD4+ and/or CD8+ T-cell counts only,and determination of cytokines Th1 and Th2 can help their differentiation.

OBJECTIVE To know the immunate function state when patients with invasive fungal infection appearing peripheral blood plasmacyte.METHODS Twenty cases of invasive fungal,bacterial infection and normal concentrate blood smear if apper plasmacyte and quantity;adopt flow cytometer technology,nephelometry were used to detect lymphocyte subpopulation,immunoglobulin level.RESULTS Concentrate blood smear find plasmacyte in 15 case invasive fungal infect patient,commonly bacterial infect only 1 case.nomal had not find.invasive fungal infection patien lymphocyte subpopulation CD3+:(50.2?6.1)%,CD4+:(23.6?3.5)%,lower than normal CD3+:(66.8?8.0)%,CD4+:(35.2?5.1)%.But CD8+:(33.9?4.5)%,CD19+:(34.2?7.0)% there were hinger than normal CD8+:(26.5?6.5)%,CD19+:(12.0?4.9)%,CD16/56+ parting(18.8?5.1%,18.2?7.0%).There ware no marked difference in two group of invasive fungal infect patients and normal immunoglobulin level parting:IgG 8.35,7.72 g/L,IgA 1.59,0.99 g/L,IgM 1.20,1.07 g/L.CONCLUSIONS Invasive fungal infect patients when concentrate blood smear find plasmacyte,are found with low immune and blood serum immunoglobulin level with NK cell quantity no obvious change;but CD19+ lymphocyte quantity increase.

Objective To investigate the effects of Modifded DaHuang ZheChong Pill on T lymphocyte subpopulation of S180 tumor-bearing mice.Methods The mice were built to the S180 sarcoma model.The experimental animals were divided into 5 groups:High-dose Modifded DaHuang ZheChong Pill group.middle-dose Modifded DaHuang ZheChong Pill group,low-dose Modifded DaHuang ZheChong Pill group,model group,and normal group.The proportion of the T lymphocyte subpopulation was detected by Flow Cytometer in the different experimental groups.Results Modifded DaHuang ZheChong pill could control radio of CD4/CD8,especially,in the high dosage group.Conclusion Modifded DaHuang ZheChong Pill could remit and improve the immune function in tumor-bearing mice,showing the best results in High-dose Modifded DaHuang ZheChong Pill group.

BACKGROUND: Chronic neuropathic pain is often associated with altered immune function and the modulated immune cell response play a role in neuropathic pain by experimental nerve injury. In order to assess the possible changes in lymphocytes function following peripheral mononeuropathy, this study examined the lymphocyte subpopulation of the spleen using the monoclonal antibodies against the membrane surface markers in neuropathic BALB/c mice by a partial transection of sciatic nerve (PST). METHODS: After confirming tactile allodynia by paw withdrawal threshold, the splenic lymphocytes were stained with fluorescein isothiocyanate (FITC)-conjugated anti-mouse CD45R/B220 (B cell) and CD4 (helper/inducer T cell) or with phycoerythrin (PE)-conjugated anti-mouse CD90.2 (total T cell) and CD8 (suppressor/cytotoxic T cell). The proportions of subsets were analyzed using a FACScan laser flow cytometry system on postoperative day 5 and day 18 respectively. RESULTS: PST induced a mechanical allodynia as verified by the von Frey test at both 5 and 8 days postoperatively compared to pre-surgery (P < 0.05). Lymphocyte subpopulation was affected by PST. The proportion of CD4+ subset was significantly larger in the PST group than in the sham operated group on day 5, while the proportion of CD8+ subset was larger on day 18. In the PST group, there were significantchanges in the proportion of CD4+ on day 5 and in the proportion of CD8+ on day 18 (P < 0.05) compared to pre-surgery. There were no significant fluctuations in the proportion of total splenic T cell and B cell subsets of PST group compared to sham operated group. CONCLUSIONS: These results suggest that development of mononeuropathy is responsible for the proportional changes in splenic lymphocyte subsets in mice.

ABSTRACT

Objective To investigate the changes and correlative factors of natural killer(NK)cells and T lymphocyte subpopulations levels in neonates with hypoxic-ischemic encephalopathy(HIE).Methods Peripheral blood samples were collected from 50 neonates with HIE.The percentages of NK cell(CD3~-CD_(16)~+CD_(56)~+) and T lymphocyte subpopulations including CD3~+,CD4~+,CD8~+ and CD4~+/CD8~+ ratio were determined by Flow Cytometr(FCM).Meanwhile,20 healthy neonates were served as controls.Results (Le)-vels of NK cell,CD3~+,CD4~+,CD8~+ and CD4~+/CD8~+ ratio in neonates with HIE were significantly lower than those in control group(all P0.05).Spearman correlation analysis showed that there were negative correlation between the NK cell,CD3~+,CD4~+,CD4~+/CD8~+ ratio and stages of HIE(all P0.05). ABSTRACT

We examined lymphocyte subpopulations of peripheral blood from BLV infected and noninfected Holstein-Friesian dairy cattle reared in Korea by flow cytometry using monoclonal antibodies specifically reactive with bovine leukocyte differentiation marker. Lymphocyte subpopulations expressing BoCD11b, B-B2, CD5, B, MHC II-DP, MHC II-DQ, and MHC II-DR antigens were significantly abundant in the BLV(+) group than the BLV(-) group (p<0.01). On double staining, subpopulation of B-1a(BoCD5+ BoCD11b+) lymphocytes was significantly increased in leukemic group. However, T-lymphocyte lineage expressing BoCD2, BoCD4, BoCD8, and WC1 antigens was significantly lower than in the BLV(+) group (p<0.01). However the absolute number of T-lymphocytes expressing BoCD2, BoCD4, BoCD8, and WC1 antigens in BLV(+) group remained with in the normal range. Furthermore mean ratio of BoCD4/BoCD8 in the BLV(+) groups was higher than that in the BLV(-) group. Taken together, cellular immune responses did not seem to significantly be decreased in the leukemic cattle.

BACKGROUND: Hemophiliacs are known to have higher risk of exposure of hepatitis virus and immunosuppression. The aim of this study is to investigate the positive rate of viral markers for hepatitis and anti-HIV and the changes of lymphocyte subpopulations in Hemophiliacs in Chonnam GwangJu area. METHODS: One hundred four patients who had visited to the Hemophilic Clinic, Chonnam University Hospital from 1999 to 2001 were enrolled. They were checked for type A, B, C hepatitis viral markers, anti-HIV and lymphocyte subpopulations. The prevalence of hepatitis and lymphocyte subpopulation were compared according to severity and age of hemophiliacs. RESULTS: Anti-HAV IgM, anti-HAV IgG, HBsAg, anti-HBs, anti-HCV were positive in 40%(22/55), 65.5%(66/101), 42.3%(42/97) of cases tested. Positivity of anti-HCV showed trends of increase according to the severity of hemophiliacs (P11 years old) (P=0.003, P<0.001, respectively). Although all enrolled patients were negative for anti-HIV, absolute T and B cells counts were decreased in 71.6% and 14.9% of patients, respectively and inversion of CD4/CD8 ratio were found in 65.7%. But there were no statistical difference in not only decrease of T and B cells but also inversions of CD4/CD8 ratio according to age and severity. CONCLUSION: The number of hemophiliacs with previous history of hepatitis B virus infection and seropositivity of anti-HCV were increased according to the age and severity of hemophilia. Active vaccinations of hepatitis B may be required in hemophiliacs. The greater part of hemophiliacs showed decrease in T cell count and inverted CD4/CD8 ratio. The hemophiliacs need a cautiion for infection and follow up tests for immunologic function.

ABSTRACT

Objective To explore the clinical value of lymphocyte sub-population and NK levels in patients with primary lung malignant tumor and patients with chronic obstructive pulmonary disease (COPD). Methods The percentage of CD3 +,CD4 +, CD8 +, CD19 + and NK cells were measured in 60 COPD patients and 50 primary lung malignant tumor patients and 40 normal control subjects using alkaline phosphatase antialkaline phosphatase (APAAP) assay, and CD4 +/CD8 + ratio was calculated. Results The levels of CD3 +,CD4 +,CD4 +/CD8 + and CD19 + cells were significantly lower in the patients with primary lung malignant tumor or COPD than those in the normal control subjects, which CD8 level was significantly higher (P0.05). No difference in the levels of CD3 +, CD4 + and CD8 + cells was found between the primary lung malignant tumor and COPD patients, but NK and CD19 + cells levels had significant difference(P

ABSTRACT

Objective To investigate the dynamic change of T-lymphocyte subpopulations in the patients with SARS to understand the pathogenesis of SARS,and to find the specific early diagnostic and warning indexes for SARS.Methods T-lymphocytes in the peripheral blood were labelled by specific fluorescein antibodies and examined by flow cytometry in 252 cases of patients with SARS.The relationship between the dynamic change of T-lymphocyte subpopulations and the clinical progression was analyzed.Results The CD 3 +,CD 4 + and CD 8 + T cells,especially CD 4 + T cells decreased at the fist two-week of the disease course and reached to the lowest level at the second week,and began to increase from the third week in all patients.The change pattern of CD 3 +,CD 4 + and CD 8 + T cells in the severe type of SARS was the same as that of the mild type of SARS,but with more serious extent and longer time.The CD 4 + cell amount of the first,second and third weeks in the mild type of SARS was 515?444/?l,421?350/?l and 600?389/?l respectively,and was up to normal level at the fourth week.The CD 4 + cell amount of the first,second,third and fourth weeks in the severe type of SARS was 312?158/?l,255?187/?l,435?343/?l and 539?394/?l respectively,and still lower than normal level at the fourth week.There was a significant difference in the CD 3 +,CD 4 + and CD 8 + cell amount among the different weeks of disease course in both types.NK cell amount also decreased during the course of disease.The CD 4 +/CD 8 + ratio did not change much and was within the normal range.Conclusions The decrease of CD 3 +,CD 4 + and CD 8 + T cells is a sensitive index for early diagnosis of SARS.The continuous decrease of them may indicate the severe condition of the patients with SARS.

ABSTRACT

With the purpose of clarifying the influence of direct moxibustion on the cell count and proportion of leukocytes in peripheral blood, rabbits and humans were treated with direct moxibustion. The moxa cones were the size of a “rice grain”. In the rabbits 10 moxa cones were distributed among four points which were selected on the low back. Humans were treated with the same method except eight points were chosen on the back and legs. Peripheral blood was collected periodically following each treatment. Leukocyte counts and leukocyte differentials were then measured. Lymphocyte subpopulations were also analyzed with flow cytometry in humans.<br>In rabbits, the mean leukocyte count increased transiently after the treatment in both the moxibustion group and the control group. However, the greatest increase was observed in the moxibustion group. In humans, no change was found in leukocyte counts and leukocyte differentials in the moxibustion group. However, the mean T cell percentage in the moxibustion group decreased 24 hours after the moxibustion treatment. Moreover, the mean CD4/CD8 ratio of the moxibustion group increased 2 hours after the treatment, and decreased 24 hours after the treatment. These changes were statistically significant while no significant change was observed in the control group. No change was found in either rabbits or humans after the 4 weeks of treatment.<br>These results suggest that the immune system will be influenced by moxibustion at least transiently.

ABSTRACT

Objective To study the curative effect of praziquantel against juveniles of Paragonimus westermani and the dynamic changes of T lymphocytes and their subpopulations in infected mice after treatment. Methods Mice infected with metacercariae of P. westermani were treated with praziquantel 10 days after infection for examining the curative effect. Results Mice infected orally each with 30 metacereariae of F. westermani were treated with praziquantel at doses of 400,500 and 600 mg/(kg?d) respectively for 3 d, the juvenile reduction rates were 71.01 % ? 10.38% ,82.29% ? 7.92% and 95.83% ? 3.24% respectively; when the mice were infected with 80 metacercariae and treated with a dose of praziquantel 600 mg/(kg?d) for 3 d, the worm reduction rate was 84.49% ?10.91%. T lymphocytes and their subpopulations began to increase at 1 wk and returned to normal at 8 wk after praziquantel treatment. Conclusion The efficacy of praziquantel against juvenile P. westermani was poorer than that against adult worms ; the efficacy was relevant to the drug dosage and the number of metacercariae infected. The T lymphocytes and their subpopulations may be regarded as an index for curative effect.

ABSTRACT

PURPOSE: Radiation-induced alteration in the immune function is well known phenomenon in cancer patients. Our purpose is to evaluate the extent of immune suppression immediately after mediastinal or pelvic irradiation, which include significant volume of active bone marrow in adults. METHODS AND MATERIALS: 48 cancer patients with mediastinal(N=29) and pelvic irradiation(N=19) were the basis of this analysis. Age ranged from 36 to 76 and mean and median value was 57 years, respectively. Sex ratio was 1.3(M:F = 27/21). The immunological parameters were the complete blood cell(CBC) with differenial cell(D/C) count, T cel subset(CD3, CD4, CD8, CD19), NK cell test(CD16,CD56), and serum immunoglobulin (lgG,lgA,lgM) level. RESULTS: The mean value of white blood cell(WBC) was reduced from 7017 to 4470 after irradiation (p=0.0000). In the differential count, the number of lymphocyte, neutrophil, and basophil was markedly reduced with statistical significance(p<0.01) and the number of monocyte was not changed and, on the contrary, that of eosinophil was increased by irradiation.In the lymphocyte subpopulation analysis, the number of all subpopulations, CD3(T cell), CD4(helper T cell), CD8(suppressor T cell), CD16(NK cell), CD19(B cell) was reduced with statistical significance. The mean ratio of CD4 to CD8 in all patients was 1.09 initially and reduced to 0.99 after radiotherapy(p = 0.34), but the proportional percentage of all subpopulations was not changed except CD19(B cell) after irradiation.In the immunoglobulin study, initial values of lg G, lg A, and lg M were relatively above the normal range and the only lg M was statistically significantly reduced after radiotherapy(p=0.02) CONCLUSION: Mediastinal and pelvic irradiation resulted in remarkable suppression of lymphocyte count in contrast to the relatively good preservation of other components of white blood cells. But the further study on the functional changes of lymphocyte after radiotherapy may be necessary to conclude the effects of the radiation on the immunity of the cancer patients.
