ABSTRACT

SUMMARY: This research was to examine the histological and ultrastructural characteristics of prepuce samples, as well as vimentin and S100 protein localization and statistical analysis. Urologists have long struggled with the prepuce, which is used to treat a variety of urethral problems. Skin biopsies were collected from the prepuce at the moment of circumcision and processed for light microscopy, electron microscope examination, immunohistochemical techniques, and statistical analysis in a total of six boys. Histologically, the prepuce epidermis displayed focal spiky ridges, which are saw-toothed interspersed with sulci, slight hyperpigmentation, looser connective tissue and plentiful vascular components. Immunohistochemically, the existence of melanocytes and Langerhans cells in the epidermis, as well as smooth muscles in the dermis, was stained positively for vimentin. Also, there was a positive reactivity of the Langerhans cells in the epidermis and around Meissner's corpuscles in the dermis for S100 protein staining. Ultrastructurally, the prepuce's intercellular gaps were widened, melanocytes rested on a folded basement membrane, and desmosomal content was reduced, with a prominent active euchromatic nucleus. Cytoplasmic projections were distended and elongated, and the interstitial blood vessels were surrounded by endothelial cells and rested on a basement membrane. There were also minimal collagen fibers in the interstitium. The prepuce's histological and ultrastructural features, as well as immunohistological studies using vimentin and S100 protein as intermediate filaments and statistical analysis, all demonstrated that it is a useful scientific resource.


RESUMEN: El presente trabajo de investigación se realizó para examinar las características histológicas y ultraestructurales de las muestras de prepucio, así como la localización y el análisis estadístico de la vimentina y la proteína S100. Los urólogos han intentado trabajar durante mucho tiempo con el prepucio, que se usa para tratar una variedad de problemas uretrales. Se recolectaron biopsias de piel del prepucio de seis niños en el momento de la circuncisión y se procesaron para microscopía óptica, examen con microscopio electrónico, técnicas inmunohistoquímicas y análisis estadístico. Histológicamente, la epidermis del prepucio mostraba crestas puntiagudas focales, intercaladas con surcos, hiperpigmentación leve, tejido conectivo más laxo y abundantes componentes vasculares. Inmunohistoquímicamente, la existencia de melanocitos y células dendríticas epidérmicas (células de Langerhans), así como músculo liso en la dermis, se tiñeron positivamente para vimentina. Además, hubo una reactividad positiva de las células dendríticas epidérmicas en la epidermis y alrededor de los corpúsculos del tacto (de Meissner) en la dermis para la tinción de la proteína S100. Ultraestructuralmente, los espacios intercelulares del prepucio se ensancharon, los melanocitos descansaban sobre una membrana basal plegada y el contenido desmosómico se redujo, con un núcleo eucromático activo prominente. Las proyecciones citoplasmáticas estaban distendidas y alargadas, y los vasos sanguíneos intersticiales estaban rodeados por células endoteliales y descansaban sobre una membrana basal. También había fibras de colágeno mínimas en el intersticio. Las características histológicas y ultraestructurales del prepucio, así como los estudios inmunohistológicos utilizando vimentina y proteína S100 como filamentos intermedios y el análisis estadístico, demostraron que es un recurso científico útil.

ABSTRACT

This experiment was designed to study the administration of normal doses of one of recent antimalarial drug and coadministration of vitamin E on the kidney tissue. A total twenty-four adult male albino rats were used and divided into four groups: the first one served as a control, the second received artemether orally for three days consecutively. The rats of the third and fourth groups received the same dose of artemether concomitantly with 50 and 100 mg/kg vitamin E orally daily for 2 weeks. After the last dose, the rats were sacrificed and the kidney tissues with blood samples obtained and processed for light, electron microscopic and biochemical analysis. Histologically, artemether treated kidneys showed atrophied glomeruli with widened urinary space and kidney tubules were degenerated with disturbed contour and some vacuoles inside it. Ultrastructurally, the glomeruli of this group showed hypertrophic endothelial cells, irregularity of its basement membrane, disrupted foot processes and filtration slits. The kidney tubule cells showed loss of basal infoldings, cytoplasmic vacuolation, polymorphic damaged swollen mitochondria a loss of its microvilli towards its capillary lumen. Artemether plus vitamin E of the rat kidney groups showed improvement of morphological changes compared to the changes seen in artemether alone. These data were confirmed by biochemical findings with marked improvement of blood urea and creatinine levels and increase of anti-oxidant enzyme activities of glutathione peroxidase and superoxide dismutase in the vitamin E treated groups. The results of this study revealed that vitamins E can improve the adverse changes of artemether of rat renal tissue.


Este proyecto fue diseñado para estudiar la administración de dosis normales de uno de los medicamentos antipalúdicos y de la administración de vitamina E en el tejido renal. Se utilizaron 24 ratas albinas machos adultas divididas en cuatro grupos: el primero sirvió como control, el segundo recibió arteméter por vía oral durante tres días consecutivos. Las ratas del tercer y cuarto grupos recibieron la misma dosis de arteméter concomitantemente con 50 y 100 mg / kg de vitamina E por vía oral diariamente durante 2 semanas. Después de la última dosis, las ratas fueron sacrificadas y se obtuvo el tejido renal de cada muestra los cuales fueron procesados para análisis con microscopías de luz y electrónica, además de exámenes bioquímicos. Histológicamente, los riñones tratados con arteméter mostraron atrofia glomerular con espacio urinario ensanchado y túbulos renales degenerados con contorno alterado y algunas vacuolas en su interior. Ultraestructuralmente, los glomérulos de este grupo mostraron células endoteliales hipertróficas, irregularidad de su membrana basal, procesos alterados del pie y hendiduras de filtración. Las células del túbulo renal mostraron pérdida de inflexiones basales, vacuolación citoplasmática, mitocondrias dañadas y pérdida de sus microvellosidades hacia la luz capilar. Arteméter más vitamina E en los grupos de riñón de rata mostraron una mejora de los cambios morfológicos, en comparación con los cambios observados en arteméter solamente. Estos datos fueron confirmados por hallazgos bioquímicos con una marcada mejoría de los niveles de urea y creatinina en sangre y un aumento de las actividades enzimáticas antioxidantes de la glutatión peroxidasa y la superóxido dismutasa en los grupos tratados con vitamina E. Los resultados de este estudio revelaron que la vitamina E puede mejorar los cambios adversos del arteméter del tejido renal de la rata.

ABSTRACT

The objective of this study was to measure the effects of glucose and salt level on white blood cells, red blood cells and platelets (PLTs) in the blood of a leukemic patient by using a white light microscope. Different concentrations of glucose and salt in the range of 0 mM to 500 mM were admixed in the blood sample to prepare blood smear. We revealed that shape of erythrocytes, leukocytes and platelets changes and form aggregates. Increasing concentrations of glucose cause to increases aggregation process of white blood cells, red blood cells and platelets. And the increasing concentration of sodium chloride causes to increase rouleaux formation and aggregation of platelets but dehydration due to increased sodium chloride concentration causes to break the aggregation of white blood cells. Comparison of CBC reports of these samples with and without analytes shows that total leukocyte count (TLC) decreases gradually towards normal ranges of leukocytes which is favorable in the treatment of leukemia but at the same time decreasing level of hemoglobin HGB, mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC) and increasing level of red blood cell (RBCs) causes to reduce oxygen supply which is in favor of cancer growth and anemia. This work provides us the base for translation this in vitro study towards the in vivo case of blood microvasculature as a non-invasive methodology.


O objetivo deste estudo foi medir os efeitos da glicose e do nível de sal nos glóbulos brancos, glóbulos vermelhos e plaquetas (PLTs) no sangue de um paciente leucêmico usando um microscópio de luz branca. Foram misturadas diferentes concentrações de glicose e sal na gama de 0 mM a 500 mM na amostra de sangue para preparar esfregaço de sangue. Descrevemos que a forma dos eritrócitos, leucócitos e plaquetas muda e forma agregados. O aumento das concentrações de glicose aumenta o processo de agregação de glóbulos brancos, glóbulos vermelhos e plaquetas. E a crescente concentração de cloreto de sódio causa o aumento da formação de rouleaux e a agregação de plaquetas, mas a desidratação devido ao aumento da concentração de cloreto de sódio causa a quebra da agregação de glóbulos brancos. A comparação dos relatórios de CBC dessas amostras com e sem analitos mostra que a contagem total de leucócitos (TLC) diminui gradualmente para os intervalos normais de leucócitos, o que é favorável no tratamento da leucemia, mas ao mesmo tempo diminui o nível de hemoglobina HGB, hemoglobina corpuscular média (MCH ), a concentração média de hemoglobina corpuscular (MCHC) e o aumento do nível de glóbulos vermelhos (RBCs) reduz o suprimento de oxigênio, o que é a favor do crescimento do câncer e da anemia. Este trabalho fornece a base para a tradução deste estudo in vitro para o caso in vivo de microvasculatura de sangue como uma metodologia não-invasiva.

ABSTRACT

A paca (Cuniculus paca), um dos maiores roedores da fauna brasileira, possui características inerentes à sua espécie que podem contribuir como uma nova opção de animal experimental; assim, considerando-se que há crescente busca por modelos experimentais apropriados para ortopedia e pesquisas cirúrgicas, foram analisados e descritos em detalhes a anatomia microscópica e ultraestrutural do joelho desse roedor. Os ligamentos colaterais são constituídos por feixes de fibras colágenas arranjadas paralelamente e com trajeto ondulado. Os fibroblastos formavam fileiras paralelas às fibras colágenas; quanto aos ligamentos colaterais, estes apresentaram citoplasma imperceptível à avaliação por microscopia de luz, entretanto, em análise ultraestrutural verificou-se vários prolongamentos citoplasmáticos. Microscopicamente, as estruturas presentes no joelho da paca assemelham-se às dos animais domésticos, roedores e lagomorfos.(AU)


Paca (Cuniculus paca), one of the largest rodent of the Brazilian fauna, has characteristics inherent to the species that can contribute as a new experimental animal; so, considering that there is a growing search for experimental models suitable for orthopedic and surgical research, it was analyzed and described in detail the microscopic and ultrastructural anatomy of the stifle in this rodent. The collateral ligaments are composed of bundles of collagen fibers arranged in parallel and in wavy path. Fibroblasts formed parallel rows to the collagen fibers; concerning the collateral ligaments, they presented imperceptible cytoplasm at light microscopy, but at ultrastructure analysis they presented several cytoplasmic processes. At the microscopic level, the stifle of paca resembles the domestic animals, rodents and lagomorphs.(AU)

ResumenLas técnicas histoquímicas hoy en día permiten seleccionar áreas de tejido y generar información confiable sobre la distribución de reservas energéticas en los moluscos bivalvos durante su ciclo de vida. Mensualmente se examinaron las gónadas y la glándula digestiva (GD) de 15 individuos recolectados entre abril 2012 y febrero 2013 por técnicas histológicas e histoquímicas de microscopia de luz, para relacionar el ciclo gametogénico y la disponibilidad de reservas energéticas con los parámetros ambientales. En el ciclo gametogénico, la proporción mensual de organismos maduros fue mayor en los machos entre agosto (40 %) y noviembre (53 %), mientras que las hembras tienden a presentar un ciclo más corto y sincronizado de liberación de gametos (septiembre 67 % y octubre 60 %). Los períodos intensos de desoves coinciden en ambos sexos (octubre-enero). Entre abril-agosto 2012 y enero-febrero 2013, se observan los valores más altos del IGl (índice de glúcido), mientras que en septiembre disminuyen y alcanzaron valores mínimos entre octubre y diciembre. El IL (índice de lípidos) presentó valores máximos en abril-2012 y febrero-2013, con un valor intermedio en agosto. Los resultados indican que las reservas de la GD presentan un patrón de movilización en relación inversa con la proliferación de los gametos de ambos sexos, vinculado directamente con la disponibilidad de nutrientes como la clorofila a y el seston orgánico.


AbstractHistochemical techniques today allow you to select areas of tissue and generate reliable information on the distribution of energy reserves in bivalve molluscs during their life cycle. The main objective of this study was to describe and relate the gametogenic cycle with the availability of energy reserves and the environmental parameters. For this, we sampled and examined the gonads and digestive glands (DG) of 15 individuals collected monthly during April 2012 and February 2013. We processed and analyzed the samples by standard histological and histochemical light microscopy techniques. Our results showed that for the gametogenic cycle, the monthly proportion of mature organisms was higher for males, between August (40 %) and November (53 %), while the females tend to have a shorter synchronized cycle and release of gametes in September (67 %) and October (60 %). The intense spawning periods in both sexes was the same (October to January). Between the periods April-August 2012 and January-February 2013, we observed the highest values of IGl and glucide index (instead, a decrease was observed in September, reaching minimum values during the period October-December). Besides, the maximum values of IL, lipid index, were observed in April-2012 and February-2013, with an intermediate value in August-2013. The results indicated that the reserves of the GD have a pattern of mobilization inversely related to the proliferation of gametes in both sexes; this was directly linked to the availability of nutrients such as chlorophyll a and the organic seston. ABSTRACT

Objective: Early carious lesions in bovine and human enamel developed in vitro using a pH cycling regimen were compared. Material and Methods: Fifteen central bovine incisors and fifteen recently extracted human third molars were randomly divided into two groups: ten for the cross-sectional microhardness test (MT) and five for polarized light microscopy (PLM) analysis. Enamel blocks measuring 5 x 5 mm were made from the buccal face of the teeth. The blocks used for the MT were sliced into two halves: “A” and “B”. “A” slices were embedded in acrylic resin, with the face of the dentin-enamel junction left exposed for the MT prior to pH cycling. “B” slices and whole blocks were coated with acid-resistant varnish, except a 3 x 3 mm central window, and submitted to the pH cycling regimen (demineralizing solution for 3 h and remineralizing solution for 21 h) over five consecutive days. The “B” slices were then submitted to the MT and the whole blocks were processed for the PLM study. Results: The PLM analysis revealed shallow, extensive lesions in the bovine enamel, hardly showing the superficial, dark and translucent zones, as well as deep cavity lesions in the human enamel, with the body of the lesion and the dark zone evident. The MT revealed a significant decrease in microhardness in the superficial levels of the bovine enamel caries and at all depth levels of the human enamel caries. Conclusion: The pH cycling regimen adopted led to the development of deeper and more demineralized carious lesions in human enamel than bovine enamel


Objetivo: Comparar lesões incipientes de cárie em esmaltes humano e bovino desenvolvidas in vitro através de ciclagem de pH. Material e Métodos: Quinze incisivos centrais bovinos e quinze terceiros molares humanos inclusos recém-extraídos foram aleatoriamente divididos em dois grupos: dez para o teste de microdureza tranversal (MT) e cinco para a análise por microscopia de luz polarizada (MLP). Blocos de esmalte com dimensões de 5 x 5 mm foram confeccionados a partir da face vestibular dos dentes. Os blocos utilizados para MT, a qual foi determinada em cinco níveis de profundidade, foram seccionados em duas metades, designadas por “A” e “B”. As fatias “A” foram submetidas ao teste de MT previamente à ciclagem de pH. Já as fatias “B” e os blocos de esmalte foram recobertos por verniz ácido-resistente, exceto por uma janela de 3 x 3 mm, e então submetidos à ciclagem de pH (soluções desmineralizadora por 3 h e remineralizadora por 21 h) por 5 dias consecutivos. As fatias “B” foram então submetidas a MT e os blocos foram processados para a MLP. Resultados: A análise por MLP revelou lesões rasas e extensas no esmalte bovino, com zonas superficial, escura e translúcida pouco visíveis; no esmalte humano, as lesões apresentaram-se profundas e cavitadas, com o corpo da lesão e a zona escura evidentes. A MT revelou significante diminuição na microdureza nos níveis superficiais do esmalte bovino e em todas as profundidades avaliadas do esmalte humano após a ciclagem de pH. Conclusão: A ciclagem de pH levou ao desenvolvimento de lesões de cárie mais profundas e com maior desmineralização no esmalte humano em comparação ao esmalte bovino.

ABSTRACT

Three different reptilian species Psammophis sibilans (Order Ophidia), Tarentola annularis (Order Squamata and Crocodylus niloticus (Order Crocodylia) are used in the present study. Their tongue is removed and examined morphologically. Their lingual mucosa examined under scanning electron microscopy (SEM) as well as processed for histological investigation. Gross morphological studies revealed variations of tongue gross structure being elongated with bifurcated end in P. sibilans or triangular flattened structure with broad base and conical free border in T. annularis or rough triangular fill almost the floor cavity in C. niloticus. At SEM, the lingual mucosa showed fine striated grooves radially arranged in oblique extension with missing of lingual papillae. Numerous microridges are detected above the cell surfaces in P. sibilans. T. annularis exhibited arrangement of conical flattened filiform papillae and abundant of microridges. However in C. niloticus, the lingual mucosa possessed different kinds of filiform papillae besides gustatory papillae and widespread arrangement of taste buds. Histologically, confirmed SEM of illustrating the lingual mucosa protrusion of stratified squamous epithelium in P. sibilans and presence of apical taste buds in conical filiform papillae of T. annularis. The lingual mucosa of C. niloticus revealed different forms of both filiform and gustatory papillae.


En el presente estudio fueron utilizados tres diferentes especies de reptiles Psammophis sibilans (Orden Ophidia), Tarentola annularis (Orden Squamata y Crocodylus niloticus (Orden Crocodylia). Sus lenguas fueron removidas y examinadas morfológicamente. Las mucosas linguales se examinaron bajo microscopio electrónico de barrido (SEM) y también procesadas para la investigación histológica. El estudio morfológico macroscópico reveló variaciones de la estructura de la lengua, siendo éstas alargadas con el extremo bifurcado en P. sibilans o de estructura triangular aplanada con una base amplia y el borde libre cónico en T. annularis o triangular rugosa llenando casi todo el suelo de la cavidad en C. niloticus. En el SEM, la mucosa lingual mostró finas ranuras estriadas dispuestas radialmente en la extensión oblicua con pérdida de las papilas linguales. Numerosas microcrestas fueron detectadas por encima de la superficie de las células en P. sibilans. T. annularis exhibiendo una disposición de papilas filiformes cónicas aplanadas y abundante de microcrestas. Sin embargo, en C. niloticus, la mucosa lingual posee diferentes tipos de papilas filiformes además de papilas gustativas con una extensa disposición de corpúsculos gustativas. Histológicamente, mediante confirmación de SEM se ilustra la protrusión del epitelio escamoso estratificado de la mucosa lingual en P. sibilans y la presencia de corpúsculos gustativos apicales en las papilas filiformes cónicas de T. annularis. La mucosa lingual de C. niloticus reveló diferentes formas de ambas papilas, filiformes y gustativas.

ABSTRACT

Objetivo: avaliar a correlação entre as leituras de descalcificação artificial de esmalte in vitro por meio do DIAGNOdent® e pela microscopia óptica com luz polarizada. Metodologia: foram utilizados 25 dentes bovinos que foram expostos a um meio artificial de cáries por uma hora, duas vezes ao dia, por 35 dias. Em seguida, procedeu-se as leituras das fluorescências utilizando o aparelho laser DIAGNOdent®. Foram registradas as leituras das áreas sem descalcificação (para calibração) e as áreas desmineralizadas artificialmente. Após as leituras da fluorescência, as regiões de leitura por laser foram cortadas em secções de aproximadamente 400μm e avaliadas por meio da microscopia de luz polarizada. Resultados: demonstraram que, embora com algum grau de variação, o sistema DIAGNOdent® foi capaz de detectar as descalcificações semelhantemente àqueles reconhecidos por microscopia de luz polarizada, estabelecendo um padrão razoável de equivalência de leituras. Conclusão: os valores médios encontrados para o DIAGNOdent® demonstraram um coeficiente de correlação de Pearson de 0,63 com relação aos valores das leituras microscópicas.


Aim: evaluate the correlation between the artificial in vitro enamel decalcification through DIAGNOdent® laser and through optic microscopic with polarized light. Methodology: for this research, 25 bovine teeth had been exposed to an artificial decay environment during one hour, two times a day, for 35 days. After wards, DIAGNOdent® laser has been used to read the fluorescence from the enemel areas without decalcification (for calibration) and to evaluate the non-mineralized artificially. After the fluorescence readings regions of laser read were cut into sections of about 400μm and evaluated by polarized light microscopy. Results: the results had demonstrated that, even with some degree of variation, the DIAGNOdent® system was capable of recognizing the decalcification, establishing regular equivalence of standard readings such as light polarized microscopy. Conclusion: the average values found for DIAGNOdent® laser had demonstrated a 0,63 coefficient of correlation of Pearson to the values of microscopically readings.

ABSTRACT

The sugarcane borer, Diatraea saccharalis Fabricius, is a pest to sugarcane and many other crops. This work aims to characterize morphological variability in the epithelial cells (columnar, goblet and regenerative) along the midgut of D. saccharalis larvae. Fragments of the midgut (anterior, middle and posterior regions) were fixed and processed by light and scanning electron microscopy. There are both cytochemical and ultrastructural differences in the morphology of the epithelial cells, depending on their localization along the midgut. The apical surface of columnar cells shows an increase in both number and size of the apical protrusions from the anterior to the posterior midgut regions. There is an increase in the amount of PAS-positive (Periodic Acid-Schiff Reaction) granules detected in the cytoplasm of both the columnar and regenerative cells, from the anterior to the posterior region. The goblet cell apical surface is narrow in the anterior region, and enlarged in the posterior midgut; the chamber's cytoplasm extrusion are small and thin at the apical cavity surface, being thicker, longer and more numerous at the basal portion of the cavity. Our results suggest that the sugarcane borer midgut has two morphologically different regions, the anterior and the posterior; the middle region is a transitional region.


A broca da cana, Diatraea saccharalis Fabricius, é uma praga da cana-de-açúcar e outras plantações. O objetivo deste trabalho foi caracterizar variações morfológicas nas células epiteliais (colunares, caliciformes e regenerativas) ao longo do intestino médio de larvas de D. saccharalis. Fragmentos do intestino médio (anterior, mediano e posterior) foram fixados e processados para microscopia de luz e eletrônica de varredura. Existem diferenças morfológicas citoquímicas e ultra-estruturais nas células epiteliais, dependendo da sua localização no intestino médio. A superfície apical de algumas células colunares exibe projeções citoplasmáticas que aumentam em número e volume da região anterior para a posterior do intestino médio. Existe aumento dos grânulos PAS-positivos (Reação de Schiff) no citoplasma apical das células colunares e regenerativas, da região anterior para a posterior. A câmara das células caliciformes, na região anterior do intestino médio, mostra seu ápice estreito, enquanto que na posterior essa porção da câmara é alargada; as evaginações citoplasmáticas da câmara são pequenas e finas no ápice, sendo numerosas, longas e mais espessas na porção basal. Os resultados sugerem que o intestino médio da broca da cana apresenta duas regiões morfologicamente distintas, a anterior e posterior; a região mediana é uma região de transição.

ABSTRACT

Foram estudados corpúsculos de Herbst da mucosa palatina de avestruz em nível de microscopia de luz. Os corpúsculos compõem-se de uma cápsula externa, cápsula interna e axônio central. A cápsula externa apresentou numerosas lamelas, enquanto que a cápsula interna mostrou estrutura de folhas compactas. Os corpúsculos apresentaram formato ovalado ou circular e circundado por espessos feixes de fibras colágenas. Cada lamela estava composta de uma densa rede de fibras espessas. Os axônios terminais estavam situados ao longo do eixo, terminando em um bulbo terminal. As fibras da cápsula externa, coradas por Picrosirius e examinadas no microscópio óptico sob luz polarizada, revelou a presença de fibras colágenas do tipo I em verde e na região periférica observou-se grande quantidade de fibras colágenas do tipo III. Os corpúsculos apresentaram-se envoltos por células planas e envoltos por fibras colágenas.


Herbst corpuscles of the palatine mucosa of ostrich were studied by light microscopy. The corpuscles are composed of an outer core, inner core and central nerve terminal. The outer core presents numerous lamellae, while the inner core shows compact structure of cytoplasm sheets. The corpuscles are elongate or oval in shape and are surrounded by bundles of collagen fibers. Each lamella is composed of a dense network of thick fibrils. The terminal axons are located along the axis and form a bulb terminal. The fibers of external core stained by Picrosirius and examined by polarized light microscopy revealed to be green in color like type I collagen fibers, and at the periphery is a large amount of collagen type III. The corpuscles are surrounded by flat cells and dense collagen fibers at the periphery.
