ABSTRACT

<p><b>OBJECTIVE</b>To explore the impact of electroacupuncture (EA) on the protein expression of adenosine receptors in the heart of the rats with myocardial ischemia (MI).</p><p><b>METHODS</b>Thirty healthy male SD rats were divided randomly into a control group (=6), a model group (=12) and an EA group (=12). We ligated the left anterior descending artery (LAD) for MI model in the model group and EA group, and exposed the heart after opening the chest without ligation in the control group. EA, 2 Hz /15 Hz and 1.5-2 mA, was applied at bilateral"Neiguan"(PC 6) in the EA group for 20 min, once a day for continuous 5 days. No intervention except grabbing and fixation was used in the control group and model group. We applied 2% TTC staining to observe the infarct size of myocardium, colorimetry to analyze serum lactic dehydrogenase (LDH), creatine kinase (CK), creatine kinase isoenzyme (CK-MB), radio-immunity assessment to detect cardiac troponin T (cTnT), Western blot to evaluate the adenosine A1 receptor (A1AR), A2aAR, A2bAR and A3AR.</p><p><b>RESULTS</b>After treatment, myocardial infarction of (27.56±3.24)% was obvious in the model group; the myocardial infarction in the EA group was (21.04±3.61)%, with statistical significance (<0.05). The expressions of serum LDH, CK, CK-MB and cTnT levels in the model group increased compared with those in the control group (all<0.01), and the expressions of LDH, CK, CK-MB and cTnT levels in the EA group decreased compared with those in the model group (<0.05,<0.01). The A1AR expression in the model group was not different from that in the control group (>0.05), and A2aAR、A2bAR、A3AR expressions decreased (<0.05,<0.01). A2aAR and A2bAR expressions in the EA group increased compared with those in the model group (both<0.01), and there was no statistical significance between A1AR and A3AR expressions (both>0.05). .</p><p><b>CONCLUSION</b>EA may achieve cardioprotective effect by regulating the expressions of A2aAR and A2bAR in myocardial tissue, which induce the corresponding signal cascade for reducing myocardial infarction area.</p>

ABSTRACT

Aim To investigate the protective effect of Glucogon like pep tide-1 (GLP-1 )on H9C2 cardio-myocytes against AGEs-induced apoptosis and the po-tential molecular mechanisms.Methods H9 C2 car-diomyocytes cells cultured in vitro were divided into the following groups:normal control group ,1 0 0 mg · L-1 AGEs group,100 mg·L-1 AGEs+10 nmol·L-1 GLP-1 group,100 mg·L-1 AGEs+5 mmol·L-1 N-acetyl-cysteine (NAC)group.Cell viabillity rate was meas-ured by CCK-8 assay,ROS production was measured by DCFH-DA fluorescent probe;Cells in different groups were stained with Annexin V-FITC/PI and then apoptotic rate was detected by flow cytometry;Nucleus morphology was observed under fluorescence micro-scope after being incubated with Honchest 33258;Bax, Bcl-2 mRNA gene expression was measured using RT-PCR;Western blot was applied to assess the apoptotic components expression including Bax and Bcl-2.Re-sult Compared with control group,cell viability rate in AGEs group was decreased in a dose-dependent manner;cell apoptosis and ROS production in H9 C2 cells were remarkably increased in AGEs group.How-ever,compared with AGEs group,GLP-1 reduced ROS production and ameliorated cell apoptosis caused by AGEs;the expression of pro-apototic proteins Bax was decreased,the expression of anti-apoptotic proteins like Bcl-2 was increased. Conclusion GLP-1 protects H9 C2 cardiomyocytes against AGEs-induced apoptosis, which may be related to the reduction of the active oxy-gen (ROS).

ABSTRACT

Objective: To study whether sodium-hydrogen exchange inhibitor(NHEI) can enhance the protective effect of hyperpolarized preservation solution containing potassium channel opener (PCO), and the effect was compared with that of Celsior solution. Methods: After equilibrated on a crystalloid-perfused Langendorff model for 30 min, rabbit hearts were divided into four groups: (1) Cel group: receiving another 30 min of equilibration, 8 h cold storage with Celsior solution and 1 h reperfusion; (2) Pin group: arrested and stored with pinacidil (a type of PCO) hyperpolarizing cardioplegia; (3) Cel+Car group: receiving the same treatment as Cel group except that 10 μmol/L cariporide (a type of NHEI) was administered during the second equilibration period and reperfusion; and (4) Pin+Car group: receiving cariporide treatment and hyperpolarized arrest and storage. Results: Recovery rate of systolic function in the Pin+Car group was significantly higher than that in the Pin group ([87.4±11.9]% vs [69.0±7.2]%, P0.05). No significant differences were found in any parameters between Cel group and Cel+Car group. There were no differences in coronary flow among all groups. Conclusion: NHEI can greatly enhance the protective effect of PCO hperpolarized preservation solution on the donor heart, and the combination of them has a comparable protective effect to Celsior solution.

ABSTRACT

Objective To investigate the effects of Thomas solution in combination with nitroglycerine of different concentrations on rat cardiac preservation for 8 h. Methods Thirty Wistar rats were selected,and Langendorff heart perfusion models were established.Rats were randomly divided into Thomas solution group,Thomas solution+4?10-6 mol/L nitroglycerine group and Thomas solution+4?10-5 mol/L nitroglycerine group(n=10).After hypothermic preservation(4 ℃)for 8 h,the cardiac function of each group was determined.Tissues of left ventricular wall were obtained,and changes of myocardial ultrastructure were observed by electron microscopy. Results Compared with Thomas solution group,the cardiac function was significantly improved and the myocardial ultrastructure injury was less severe in Thomas solution+4?10-5 mol/L nitroglycerine group after hypothermic preservation(4 ℃)for 8 h,while there was no significant difference in myocardial ultrastructure injury and cardiac function between Thomas solution group and Thomas solution+4?10-6 mol/L nitroglycerine group. Conclusion Thomas solution in combination with 4?10-5 mol/L nitroglycerine can significantly improve the preservation effects for isolated hearts in rats.

ABSTRACT

Objective:To investigate the protective effects of polarizing cardioplegia with Na+ channel inhibitor tetrodotoxin (TTX) on isolated rat hearts.Methods:While Langendorff and Neely perfusion model were steadily established,the rat hearts(10 hearts in each group)were respectively arrested by depolarizing cardioplegia (St.Thomas No.2 solution ,STH-2)in Group Ⅰ(control group),and by polarizing cardioplegia(TTX)in GroupⅡ.These arrested hearts were preserved in corresponding cardioplegia solution respectively at 7℃ for 5 hours and then underwent 30 minute reperfusion.Pre-ischemia and post-ischemia indexes of the rat hearts were studied,including hemodynamic parameters,myocardial enzymology,ATP content,and ultrastructural changes.Results:There were no differences between two groups before myocardial ischemia.But the recovery of myocardial hemodynamic parameters during reperfusoin in GroupⅡwas better than that in GroupⅠ(P

ABSTRACT

Objective:To summarize the clinical experience in CABG,so as to look for the optimal perioperative management procedure and improve the operative outcome.Methods:The data of coronary artery bypass were analyzed in 9 patients with coronary heart disease.Results:CABG of all patients was performed on hypothermia cardiopulmonary bypass (CBP).Myocardial preservation was carried out by cold blood cardioplegia with potassium.The average length of time of CPB was 178(70~222) minutes,and that of aortic cross clamp was 111(70~150) minutes.A total of 23 cases of coronary artery bypass were performed using 8 left internal mammary arteries and 15 long saphenous veins.One patient with rheumatic valvular disease associated with coronary heart disease received coronary artery bypass grafts and double valve replacement at the same operation.One patient died of respiratory function failure and the rest were discharged with angina disappearing.Conclusion:The success of coronary artery bypass lies in obtaining of grafts,identifying adequate target vessel,complete revascularizing ischemic myocardium and refining blood vessel anastomosis.Good myocardial protection and perioperative management are also important.
