ABSTRACT

Objective To observe the clinical efficacy of butylphthalide in the treatment of acute cerebral infarction and its effect on serum C-reactive protein(CRP).Methods A total of 120 inpatients with acute cerebral infarction were randomly assigned into control group (n =60) and observation group (n =60) by the digital table method. The control group was treated with Xueshuantong injection, inhibiting blood platelet and improving brain metabolism. The observation group was treated with butylphthalide based on the treatment of the control group. The changes in serum CRP and clinical efficacy were compared between the two groups 2 weeks after treatment. Results After treatment, the CRP level was decreased greatly in the observation group, but the CRP level was reduced slightly in the control group compared with before treatment, the CRP level in the observation group was (2.54 ± 0.79) mg/L,which was significantly lower than (5.66 ± 1.78) mg/L in the control group (t =12.41,P < 0.05).The total effective rate in the observation group was 75.00%,which was significantly higher than 53.33% in the control group (x2 =6.13,P < 0.05).The 3-day NIHSS score [(8.62 ± 1.89) points],1-week NIHSS score [(4.21 ± 1.76) points],and 2-week NIHSS score [(5.79 ± 1.36) points] after treatment were significantly lower in the observation group compared with the control group (t =2.84,11.93,6.67,all P < 0.05).Conclusion Based on conventional treatment, butylphthalide injection in the treatment of acute cerebral infarction can remarkably reduce CRP level, alleviate inflammatory reactions, improve cerebral infarction condition and prognosis, exert obvious effect and it is worthy of clinical promotion.

ABSTRACT

Objective:To analyze the impact of NBP on the expression of HSP 70 and TLR4 in the rat with ischemia-reperfusion.Methods:The 144 rats were divided into 3 groups,Sham group,IR group and NBP group. The 3 groups were divided into 4 groups again according to the time of reperfusion (6h,12h,24h and 48h).The intraluminal ischemia-reperfusion model was made by improved Longa. When the model was made successfully ,the rat in NBP group was gave oral NBP immediately while the IR and Sham group was gave oral NS.The neurological deficit scores , number of apoptotic cells , expression of HSP70 and TLR4 were compared during the 3 groups. Results:The neurological function ,apoptosis, the expression of HSP70 and TLR4 in NBP group at any reperfusion time were statistical lower than in IR group. In all the infusion periods, apoptotic cells in IR and NBP group were significantly higher than that in the Sham group. In all the infusion periods ,the positive cells in IR and NBP group were significantly lower than that in the Sham group. Conclusion:NBP can protect the function of neuron by improving the inhibition of HSP 70 and TLR4.

ABSTRACT

Background:Although blood transfusion is a life saving therapy, it is associated with various ill effects, which can cause increased morbidity and mortality in recipients. Testing of all donated blood for transfusion transmitted infections (TTIs) such as HIV I & II, hepatitis B, hepatitis C and syphilis is one of the strategies recommended by WHO to ensure safe blood. However, if the donor is already having an infection, transmissible by blood, the transfusion will be rather hazardous for the recipient. The national blood policy of India 2002 advocates the disclosure of results of transfusion transmitted infections (TTIs) to blood donors. Aim:To assess the attitude of the transfusion transmitted infections (TTIs) reactive blood donor in response to the post-donation calls from blood bank. Material and methods: A total of 20865 blood donors came to the department of IHBT in period of one year from 1St November 2009 to 31St October 2010.All donated blood was screened against HIV I & II , Hepatitis B, hepatitis C and syphilis and malaria. On screening the units, it was found that 391 donation units (1.874 %) were positive for one of the TTIs, namely HIV I & II, HBsAg, HCV or syphilis. As follow-up, these donors were recalled at blood bank by a phone call. Results: out of 391 reactive donors only 236 responded to call (average response rate was 60.36%). Conclusion: The study suggest that authorities should frame some guidelines and rules that can increase the response rate among reactive donors and make them assessable because it enables their future investigation and treatment and the prevention of diseases transmission to the community.

ABSTRACT

OBJECTIVE: To study the tissue distribution of 3-n-butylphthalide (NBP) in rats. METHODS: NBP was analyzed on a Hypersil ODS2 column with the mobile phase consisting of 0.05 moL · L-1 sodium acetate-acetonitrile (50: 50, adjust pH to 4.5 with acetic acid). The flow rate was 1.0 mL · min-1 and the column temperature was 40°C. The wavelength of UV detector was set at 228 nm. A single dose of 360 mg · kg-1 NBP suspensions was administered orally to rats. The tissues of heart, brain, liver and kidney were taken at 1, 2, 4 and 8 h after administration. NBP concentrations in tissues were assayed by HPLC-UV method. RESULTS: The calibration curves for NBP in tissues were obtained over the concentration range of 0.0498 - 9.96 μg · mL-1 with good linearity and recoveries. The concentration of NBP at 1 h after administration was maximum in kidney followed by brain, heart and liver. CONCLUSION: The method is simple, sensitive, specific, accurate and stable. It can be used for the study on tissue distribution of NBP in rats. The results show that NBP is rapidly and widely distributed in rats and can cross the blood-brain barrier. ABSTRACT

Objective To observe the effect of l-3-n-butylphthalide(l-NBP)on the of nuclear factor-?B and I?B-? in primary cultured murine brain glial cells following oxygen-glucose deprivation/ reoxygenation(OGD/R).Methods Cultured mouse brain glial cells in vitro and established OGD/R model.Glial cells were divided into five groups: normal control(NC)group,OGD 24 h/R 24 h group,l-NBP 20 ?mol/L group,l-NBP 100 ?mol/L group,l-NBP 500 ?mol/L group. The protein expression of NF-?B in nuclear and I?B-? in endochylema were analyzed by Western blot. Results Comparing with NC group,OGD 24 h/R 24 h group showed a significantly higher expression of NF-?B(P
