ABSTRACT

Background: NEDD9 plays an important role in cell migration, chemotaxis, apoptosis, and cell cycle. NEDD9 siRNA can effectively reduce the gene expression, promote apoptosis and block tumor cell migration and invasion at different levels. Aims: To investigate the expression of NEDD9 in colon cancer tissue, and the effect of NEDD9 siRNA on proliferation, apoptosis, migration and invasion of colon cancer Caco-2 cells. Methods: Colon cancer tissue and corresponding paracancerous tissue from Jan. 2017 to Jan. 2018 at Yancheng District People's Hospital of Luohe were collected. Colon cancer Caco-2 cells were conventionally cultured, and were transfected by LipofectamineTM 2000. Caco-2 cells were divided into control group, negative control group and siRNA interference group. The mRNA expression of NEDD9 in colon cancer tissue and Caco-2 cells was detected by qRT-PCR. The proliferation of Caco-2 cells was detected by CCK-8 method. The apoptosis of Caco-2 cells was detected by flow cytometry. Caco-2 cell migration and invasion were detected by Transwell assay. The protein expressions of NEDD9, MMP-9, Bcl-2, Bax, TIMP1 were determined by Western blotting. Results: The mRNA expression of NEDD9 in colon cancer tissue was significantly higher than that in paracancerous tissue (P<0.05). Compared with control group, the mRNA expression of NEDD9 in the siRNA interference group was significantly decreased (P<0.05), cell proliferation, migration and invasion were significantly inhibited (P<0.05), cell apoptosis was significantly increased (P<0.05), and the protein expressions of NEDD9, MMP-9 and Bcl-2 were significantly decreased (P<0.05), the protein expressions of Bax and TIMP1 were significantly increased (P<0.05). Conclusions: NEDD9 gene can regulate the proliferation, apoptosis, migration and invasion of colon cancer Caco-2 cells through apoptosis-related genes Bcl-2 and Bax and invasion-related genes MMP-9 and TIMP1.

ABSTRACT

Background: Neural precursor cell expressed developmentally down regulated-9 (NEDD9) is a scaffolding metastatic marker protein in multiple cancer types. Generally, the expression occurs during the embryonic development and depletes in adults. Expression of NEDD9 in adults leads to the progression of tumor which is sufficient for the cellular invasion. Elevated behavior of the gene mediates metastatic movement which includes protease dependent neovessel formation, invasion and migration of tumor cells from the site of origin to the distant tissues.Methods: The current study involves the screening and elucidation of differential expression of NEDD9 in normal and tumor subtypes with various tissues of mice by immunohistochemistry.Results: The validating approaches in the study, low expression of NEDD9 was observed in the normal tissues and predominance in the tumor subsets.Conclusions: The experimental analysis proven that NEDD9 expression is merely associated with tumor progression and the molecular mechanism of NEDD9 is restricted in the establishment of metastatic cascade. NEDD9 association in tumor prognosis which helps in the emergence of diagnostic and therapeutic approaches.

ABSTRACT

Objective To study the expression level and related prognostic significance of protein NEDD9,Ki67,P53 in gastric carcinoma. Methods A total of 30 normal stomach tissues were used as control group. The expression level of NEDD9,P53,Ki67 in early gastric carcinoma of 54 cases and advanced gastric car-cinoma in 51 cases were detected by immunohistochemistry ,respectively. Results (1) The expression level of protein NEDD9,P53,Ki67 in normal stomach tissues,early gastric carcinoma and advanced gastric carcinoma were increased(P<0.05).(2)The expression level NEDD9 protein in gastric carcinoma was associated with the depth of invasion and the degree of differentiation(P < 0.05). The expression of protein Ki67 was associated with clinical staging and the depth of invasion(P<0.05). The expression of protein P53 was associated with degree of differentiation,the depth of invasion,lymph node metastasis and clinical staging(P<0.05).(3)NEDD9 protein expression has a positive correlation with Ki67. The difference has statistical significance(r = 0.216,P = 0.027). Conclusions The expression level of protein NEDD9 ,Ki67 ,P53 increased in normal stomach tissues ,early gastric carcinoma and advanced gastric carcinoma. The expression level were significantly higher in early gastric carcinoma. Combined detection of NEDD9 ,P53 ,Ki67facilitatesearlydiagnosis and malignant degree of gastric carcinoma.
