ABSTRACT

Tachykinins are potent mediators of many functions in the airways.The tachykinins substance P and neurokinin A,presenting in human airways,can be recovered from the airways after a series of stimuli.They interact in the airways with tachykinin Neurokinin1,Neurokinin2 and Neurokinin3 receptors to cause bronchoconstriction,plasma extravasation,mucus secretion and to attract and activate immune cells and inflammation cells.They play a important role in the pathophysiology of asthma.However,the application of tachykinin receptor antagonists in clinic has been rather slow and somehow disappointing and needs to be further studied.

ABSTRACT

Objective To investigate the effect of different concentrations of arsenic trioxide (As2O3,ATO) on neurokinin A (NKA) in renal tissue of BXSB mice and explore its clinical value.Methods Fifty BXSB mice (twelve weeks old and weighted 23~26 g) were randomly divided into control group,systemic lupus erythematosus (SLE) group,and therapeutic group of three different concentrations of ATO.All biochemical indicators were analyzed before and after treatment.The pathology of renal tissue was examined by immunohistochemistry.The concentration of NKA in renal tissues was detected by ELISA and the concentration of NKA mRNA was detected by RT-PCR.Results The concentration of NKA in SLE group in renal tissue (299±26) pg/g was significantly higher than that of normal control group (122±7) pg/g (P<0.05).The concentration of NKA in the SLE group in renal tissue was significantly higher than that of three different concentrations of ATO in low-dose group (151±14) pg/g,moderate--dose group (147±9) pg/g and in highdose group (155±14) pg/g (P<0.05).No difference was found between three different dosages of ATO treatment groups and normal control groups (P>0.05).There were no significant differences among three different dosages of ATO treatment group (P>0.05).The side effects in low-dose group were significantly lower than those of moderate and high-dosage groups (P<0.05).Conclustion NKA concentration expressed in the renal tissues in the SLE group is higher than that in the control group.Decreasing the concentrations of NKA mRNA in renal tissues may be one of the important mechanisms of ATO in treating SLE.Low-dosage ATO is safe and effective to treat SLE and has therapeutic potentials.

ABSTRACT

AIM: To study the changes of neurokinin A (NKA) and NKA mRNA in lung tissues of asthmatic guinea pigs exposed to chronic cigarette smoke and investigate the mechanism. METHODS: The model of asthma in guinea pigs was made by exposure to aerosolized ovalbumin and animals were randomly assigned into: ① inhale cigarette smoke (ICS) 2 weeks before provoked; ② ICS 2 weeks after provoked; ③ 2 weeks after provoked (no-ICS); ④ asthma (no-ICS before provoked); ⑤ normal control. AIM: To explore the regulatory mechanism of n erve growth factor (NGF) on neurokinin A in the experimental asthmatic guinea pi g. METHODS: Radioimmunoassay was used to determine the alteration of neurokinin A levels in the lower respiratory tract and visceral sensory afferent sites while NGF was absent (inhalation of NGF antibody through nasal cavity) in the asthmatic guinea pig. ABSTRACT

Objective To explore the expression and change on the neurokinin A(NKA) of rat ileum during the development. Methods PAP immunohistochemistry method and image analysis were used to detect systematically the expression of NKA in rat ileum from E13 to adult stage. Results 1.In rat ileum, the NKA-IR could first be found in the myenteric plexus at E17, and then appeared on the longitudinal muscle layer, circular muscle layer, intestinal villus, intestinal gland, submucosa and deep muscular plexus. On P30, they showed the distribution feature of adult rat.2.The results of quantitative analysis were in correspondence with the developmental change of NKA-IR in ileum.3.The NKA-IR positive cells showed the typical feature of morphology and distribution of gastrointestinal endocrine cells.Conclusion 1. The ontogeny and development of NKA in rat ileum mainly appeare in the last E1 week and P4 weeks, whereas the density of positive fiber in villus is not as same as adulthood until P60. 2.There are two key periods during the development of NKA in rat ileum. The first crucial stage is from the last El week to P1 week. The second important stage is in the end of P4 weeks. 3.The NKA-IR cells may be the endocrine cell which produces NKA in rat digestive tract.4.NKA may play an important role in the development and physiological function of rat ileum.

ABSTRACT

Immunohistochemical study revealed the distributions of substance P, neurokinin A, neurokinin B and calcitonin gene related peptide immunoreactive nerve fibers in the medulla, cortex and capsule of tracheobronchial lymph nodes in 10 guinea pigs. More immunopositive fibers were found to be distributed in the area around the blood vessels and lymph sinuses, less in the area distant to blood vessels. The four kinds of positive immunoreactive nerve fibers were also seen within the wall of the blood vessels. The results provide some morphological basis for the study of neuroimmunomodulation.
