ABSTRACT
Objective: To investigate the effects of Nourishing Kidney-Yin Prescription combined with glucocorticoid on the T lymphocyte subsets and inflammatory cytokines in the mice with systemic lupus erythematosus (SLE), and to elucidate the mechanism of Chinese and Western medicine combination in the treatment of SLE. Methods: Fifty MRL/ lpr mice were randomly divided into model group, Western medicine group, Chinese medicine group, low dose and high dose of combintion groups (n = 1 0) . Ten C57BL/6 mice were selected as control group. The mice in control group and model group were given 0. 5 mL • d_1 normal saline by gavage; the mice in Western medicine group were given 12. 5 mg • kg- 1 prednisone acetate (glucocorticoid) by gavage; the mice in Chinese medicine group were given 25. 0 g • kg- 1 Nourishing Kindey-Yin Prescription by gavage; the mice in low and high doses of combination groups were given 12. 5 and 25. 0 g • kg- 1 Nourishing Kidney-Yin prescription by gavage and 6. 25 mg • kg- 1 prednisone acetate, respectively. After 21 d of continuous administration, the spleen indexes and thymus indexes of the mice in various groups were calculated. The levels of CD3, CD4 and CD8 T lymphocyte subsets in the periphreal blood of the mice in various groups were determined by flow cytometry. The serum levels of interleukin-4 (IL-4), interleukin-6 (IL-6) and y- interferon (IFN-y) were measured by enzyme-linked immunosorbent assay (ELISA) method. Results: Compared with model group, the spleen indexes and thymus indexes of the mice in Western medicine, low dose of combination group and high dose of combination group were decreased obviously (P < 0 . 0 5) . Compared with model group, the levels of CD3 lymphocytes and CD4 T lymphocytes in Chinese medicine group, low dose of combintion group and high dose of combination group were significantly increased (P < 0 . 05). Compared with model group, the CD8 T lymphocyte levels in Western medicine group, Chinese medicine group, low dose combination group and high dose combination group were significantly decreased (P < 0 . 05), the serum IL-4 and IL-6 levels were obviously decreased (P < 0 . 05), and the IFN-y levels were significantly increased (P < 0 . 05); the mesagial cell and mesangial matrix hyperplasia were improved, and the renal intestitial inflammatory cell infiltration was relieved. Conclusion: Nourishing Kidney-Yin Prescription combined with glucocorticoid can effectively regulate the body' s immune balance by reducing the inflammatory cytokines, regulating T lymphocyte subgroup equilibrium, and recovering the deficiency of the immune-suppression therapy by monosaccharide corticosteroids.

Nourishing kidney-yin (NKY) granules and warming kidney-yang (WKY) granules represent one of the prescriptions that prescribed in treating primary osteoporosis (POP) in light of tonifying kidney and nourishing essence principle as well as the theory of "treating both the disease and traditional Chinese medicine (TCM) syndrome".Both granules were created through the systematic analysis of clinic prescriptions by Professor Shi Qi.Consequently clinical investigations have well established that NKY granules significant improved bone mineral density (BMD) as well as relieved the kidney-yin deficiency syndromes in POP patients.Meanwhile,WKY granules relieve kidney-yang deficiency syndrome and the quality of life (QOL).What is more,pharmacological study established the application of common cnidium fruit,and fructus ligustri lucidi alleviated bone loss in OVX-induced mice.In addition,investigation with effective components identified that both NKY and WKY granules play systematic pharmacological effects on bone remodeling by regulating the expression of BMP/Smad,Wnt/β-catenin,RANKL/RANK/OPG axis,and Notch.The drug discovery was performed by the lead of traditional Chinese medicine (TCM) theory.It is one successful transformation investigation based on pharmacological effects,clinical intervention,animal model,cell culture and molecular investigation.

Objective To investigate the effect of herbs for nourishing kidney and strengthening governor vessel on postoperative intervertebral fusion rate in elderly patients with degenerative lumbar spinal stenosis ( DLSS) . Methods A retrospective analysis was carried out for the clinical data of 72 elderly DLSS patients treated with modified transforaminal lumbar interbody fusion (TLIF) during the period from October, 2012 to October, 2013. The patients were divided into two groups, 39 in Chinese medicine group and 33 in control group. Operative efficacy was evaluated with Japanese Orthopaedic Association ( JOA) scores for assessment of cervical myelopathy, postoperative function improvement was evaluated by Oswestry disability index (ODI) scores, and lumbar interbody fusion was evaluated by the modified Brantigan scores. Results (1) During the follow-up period, patients of the two groups had no complications such as screw breaking or loosening, cage displacement, or endplate collapse. (2) Before opera tion and 3 days after operation, the differences of JOA scores were insignificant between the two groups (P>0.05) . (3) Significant difference of intragroup ODI scores were shown 12 months after operation vs 6 months after operation, and between 6 months after operation and pre-operation in the two groups (P<0.01) . ODI scores of Chinese medicine group 6 and 12 months after operation were lower than those of the control group, the difference being significant ( P<0.05 or P<0.01) . ( 4) The results of modified Brantigan scoring showed that the interbody fusion rate 6 months after operation was 71.79% in Chinese medicine group, higher than 48.48% in the control group ( P<0.05) . Twelve months months after operation, the interbody fusion rate was 87.18% in Chinese medicine group, higher than 66.67% in the control group (P<0.05) . Conclusion Herbs for nourishing kidney and strengthening governor vessel may improve the lumbar interbody fusion rate and the quality of life of the elderly DLSS patients treated by modified TLIF.

Objective To investigate the clinical efficacy of integrated Chinese and western medicine for the treatment of anovulatory dysfunctional uterine bleeding. Methods A total of 108 anovulatory dysfunctional uterine bleeding patients were evenly randomized into treatment group and control group. Both groups were given basic conventional treatment, estrogen-progestogen sequential therapy for 3 menstrual cycles for dysfunctional uterine bleeding patients at puberty and child-bearing period, and Marvelon for patients at menopause transition period after excluding uterine tumor by diagnostic curettage. Additionally, the treatment group was given Chinese herbal decoction of Radix Rehmanniae Preparata, Semen Cuscutae, Fructus Corni, Placenta Hominis, Herba Ecliptae, Fructus Schisandrae Chinensis, Rhizoma Alismatis, raw Rhizoma Dioscoreae, Rhizoma Atractylodis Macrocephalae, Rhizoma Cyperi, Radix Codonopsis, Radix Astragali, Radix Angelicae Sinensis, Radix Paeoniae Alba, Pericarpium Citri Reticulatae, and charred Herba Schizonepetae. Therapeutic effect, bleeding-arresting time, and recurrence rate during 3-month follow-up were observed in the two groups. Results ( 1) The treatment group had a total effective rate of 100.0%, higher than 94.4%in the control group, the difference being significant (P<0.05). (2) Bleeding-arresting time was (45.50 ± 13.0) hours in the treatment group and was (58.0 ± 15.0) hours in the control group, and the difference was statistically significant (P<0.05). (3) During the 3-month follow-up, the treatment group had the recurrence rate of 16.7% ( 9 cases) , lower than 29.6% ( 16 cases) in the control group, and the difference was statistically significant ( P<0.05). Conclusion Integrated Chinese and western medicine has certain effect for the treatment of anovulatory dysfunctional uterine bleeding, showing good prospects for extensive application in gynecological clinic.

Objective To investigate the therapeutic effect of traditional Chinese medicine(TCM)recipe of nourishing kidney and activating blood for treatment of cisplatin induced acute kidney injury(AKI)in mouse model. Methods Twenty-one male BALB/c mice were randomly divided into three groups:control group,AKI model group and treatment group with the above TCM recipe(each n=7). The AKI model was reproduced by a single intraperitoneal injection of cisplatin 20 mg/kg;the TCM recipe with dosage of 10μL · g-1 · d-1 was given to the treatment group,while equal volume of normal saline was given to the model and control groups by gavage,lasting for 6 days in all groups. The changes of each mouse body weight were observed;mouse venous blood serum creatinine(SCr)level was detected,changes of renal tissue weight and its histopathology were observed under light microscope,and the score of acute tubular necrosis(ATN)was calculated. Results Compared to the control group,the body and renal weight in AKI model group were significantly lowered〔body weight(g):17.18±0.29 vs. 19.33±1.43,renal weight(g):0.28±0.01 vs. 0.32±0.11,both P<0.01〕,and SCr and ATN scores were significantly increased in AKI model group〔SCr(μmol/L):86.77±10.97 vs. 14.37±0.81,ATN score:3.33±0.52 vs. 0.17±0.41,both P<0.01〕. Compared to AKI model group,the body weight(g:18.70±0.28)and renal weight(g:0.31±0.01)in the treatment group were markedly increased,and SCr(μmol/L:21.98±5.52)and ATN scores(2.00±0.63)were significantly lower than those of the AKI model group(all P<0.01). Under optical microscope,there were mouse renal tubular epithelial cell edema and necrosis,renal interstitial inflammatory cell infiltration,the glomerular form roughly normal in the AKI model group;compared with the AKI model group,in the treatment group the degree of mouse renal tubular necrosis was significantly reduced and glomerular form was basically normal. Conclusion The TCM recipe of nourishing kidney and activating blood can reduce SCr and improve the pathological changes of renal tissue in cisplatin induced AKI mouse models,thus it has therapeutic effect for treatment of AKI in mice.

[Objective] To observe Yishi Oral Liquid effect on treating middle and old aged asthenopia. [Method] Choose 125 cases of middle and old aged asthenopia, and randomly divide them into observation group 65 cases treated wutg Yishi Oral Liquid, and control group 60 cases with Mingmu Rehmannia Pil;meanwhile both groups reduced the time with the eye and made massage on the acupoints around the eyes. [Result] With 2~4 weeks of treatment, in the observation group, 23 cases were cured, 25 had marked effect, 13 effective, 11 had no effect, the total effective rate was 93.85%;for the control one, they were respectively 14,19,16,11 and 81.67%. The comparison showed the observation group was better than control one. [Conclusion] Yishi Oral Liquid treating middle and old aged asthenopia has good clinical effect, can obviously relieve the symptoms of blurred vision, phengophobia, lacrimation and aningerestling eyes, helpful to recovering eyesight.

Objective To observe the effects of nourishing-kidney herbs on rat model with glucocorticoid-induced osteoporosis.Methods 60 Wistar rats male and female half and half were randomly divided into a normal group,a model group and a nourishing-kidney group.By intramuscular injection of dexamethasone (2.5 mg/kg) twice a week to replicate osteoporosis rat model.All groups were treated for 9 weeks.Detected the BMD of femur in vitro and determined the bone metabolism marker in serum by biochemical process.Results The BMD decreased obviously (0.109± 0.007)g/cm2 and the content of TRAP in serum increased evidently (9.96± 1.15) μg/ml in the model group.In the nourishing-kidney group,the BMD was up-regulated (0.116 ± 0.007)g/cm2,and TRAP down-regulated(5.76 ± 0.85)μg/ml.Conclusion Intramuscular injection of dexamethasone can induce GIO rat model,and nourishing-kidney herbs have the effect of anti-osteoporosis.

Objective To study the clinical effect of nourishing kidney, securing essence and cooling blood on incipient diabetic nephropathy. Methods A total of 110 patients with incipient diabetic nephropathy were randomly recruited into a treatment group and a control group. The treatment group was treated with the method of nourishing kidney, securing essence and cooling blood and benazepril hydrochloride tablets. The control group was treated with bcnazepril hydrochloride tablets Exclusively. Urina microalbumin was detected in all patients 24 h before and after treatment. Results The effective rate was 94.7% and 86.8% in the treatment group and the control group respectively, showing significant difference (t= 9.31, P<0.01) . Conclusion The method of nourishing kidney, securing essence and cooling blood is effective to decrease urina microalbumin in incipient diabetic nephropathy.

Objective To observe the effect of applying nourishing kidney and ventilating lung combined with western medicine in treating cough variant asthma (CVA) patients with kidney yang deficiency. Method Eighty CVA patients with kidney yang deficiency were divided into contrast group and therapeutic group, each group with 40 patients. The patients in contrast group were treated with Aminophylline sustained-release tablet, 0.1～0.2 g, 2 times a day, as well as Ketotifen tablet, 1 mg, 1 time a day, p.o. As for the therapeutic group, the patients were treated with Chinese herbs of nourishing kidney and ventilating lung based on the treatment of contrast group. One course was 4 weeks for the two groups, and the evaluation were carried out in the fifth week. Methods Fifty six patients were divided into two groups:the NKD group (30 cases, treated with the NKD), and the Diane-35 group (26 cases, treated with Diane-35). Condition of menstruation, acne, hairiness and side effects were observed. The follicle figures and ovary volume were recorded, and the sex hormone were detected before and after treatment. Objective:To investigate the expression of mmp-2 in mesangial proliferative glomerulonephritis(MsPGN) rat renal tissue and the effects of nourishing kidney and activating blood flow recipe of TCM. Methods: 54 Male SD rats were divided into control group, MsPGN group and treatment group with nourishing kidney and activating blood flow recipe of TCM. Immunohistochemical staining, flow cytometry, western blot and RT-PCR were used to check the protein expression of mmp-2. Results: In MsPGN group, following with the disease progressing, the glomerulus grew graduatly, ECM increased, the basement membrane of glomerulus was thicker. The immunohistochemical staining showed that the chief positive granules were deposited in glomerulus and renal tubular, the expression of mmp-2 was lower than that in control group, but in treatment group the expression of mmp-2 was higher than that in MsPGN group. Conclusion: In MsPGN renal tissue, the expression of mmp-2 was reduced, its activity was weakened. Nourishing kidney and activating blood flow recipe of TCM could induce the expression of MMP-2 and partly recover the activity. Nourishing kidney and activating blood flow recipe of TCM could resist glomerular sclerosis and postpone the course of chronic renal failure.

This article put forward the ‘heart-kidney centric theory’ on basis of analyzing the physiologic function, pathomechanism, onset rules and measures of prevention and treatment of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency. According the new theory, a new TCM drug, the Tongmai Qiangshen Wine, was developed.In human body, kidney produces essential qi and produces energy source for human body; heart transports the essential qi and is the physiologic mechanism of transport of energy source. Deficiency of essential qi will result in stasis of blood vessel, on the other hand, stasis of blood vessel can cause defi ciency of essential qi. The opposition, mutual rooting, waxing and waning, mutual convertibility of them reflect the development rules of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency, and determin the vital process of birth, growth, adult, aging and death. The construction of heart-kidney centric theory and the development and application of Tongmai Qiangshen Wine not only provide new theory and model for prevention and treatment of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency, but also provide new thoughts for preparation of new TCM drugs from aspects of how to maintain the feature of TCM, how to inherit and create on basis of combing holism with disease, principle, method, medicines.

Objective:To investigate the expression of matrix metalloproteinase(MMP) and tissue inhibitor of matrix metalloproteinase(TIMP)in mesangial proliferative glomerulonephritis(MsPGN) rat renal tissue and the effects of nourishing kidney and activing blood recipe of traditional Chinese medicine.Methods:Chose 54 male rats and separated them into control group, MsPGN group and treated group with nourishing kidney and activing blood recipe of traditional Chinese medicine.Chose six rats per group at the time of the second,the fourth and the eighth week.The sections were treated with HE and PAS stains and the routine pathologic study was carried.Immunohistochemical staining was used to check the expression of MMP-2,TIMP-2, membrance type metalloproteinases-1(MT1-MMP) and transforming growth factor-?1(TGF-?1).Flow Cytometry was used to check the protein expression of MMP-2,TIMP-2,MT1-MMP and TGF-?1 in rat renal tissue.Results:In MsPGN group,following with the disease progressing,the glomerulus grew graduatly,ECM became more and more,the basement membrane of glomerulus was thicker.The immunohistochemical staining showed the expression of MMP-2 and MT1-MMP was lower compared with control group,but in treated group the level was weaker,the expression of TIMP-2 and TGF-?1 was higher compared with control group,but in treated group the level was weaker too. 【Objective】To observe the effect of Sicaotang Decoction which is mainly composed of kidney-nourishing,toxicity-removing and blood-activating herbal medicines on parameters associated with mesenteric proliferation,fibronectin(FN),and Bcl-2 gene expression in lupus nephritis(LN).【Methods】 Sixty LN patients were equally randomized into two groups.The two groups received treatment with hormones and cyclophosphamide,and group Areceived Sicaotang Decoction additionally.The changes of Bcl-2 and Fas expression in CD4+ and CD8+ of T lymphocyte subtypes,serum fibronectin(FN),serum and urine collagen Ⅳ(CⅣ) as well as score of blood stasis were observed.【Results】Sicaotang Decoction reduced the expression of Bcl-2 and Fas expression in CD4+ and CD8+,relieved the proliferation of mesenteric cells,and decreased Bcl-2 positive rate,serum FN level,serum and urine CⅣ levels and score of blood stasis(P

Objective To evaluate the effect of Jianpi Bushen Formula(Formula for Invigorating Spleen and Nourishing Kidney) combined with moxibustion on quality of life(QOL) and maturation degree of vaginal exfoliate cells(MDVEC) in premature ovarian failure(POF) patients.Methods Totally 55 POF patients were randomized into treatment group(33 cases,treated by Jianpi Bushen Formula with moxibustion) and control group(22 cases,treated by Premarine).Menopause-specified quality of life(MENQOL) was adopted to evaluate QOL of the patients.Maturation index(MI) and maturation value(MV) were applied to evaluate MDVEC.Results The scores of MENQOL of both groups were improved and the effect of the treatment group(except sexual life dimension) was superior to that of the control group(P

Objective To explore the antagonistic effect of Ziyin Xiehuo Recipe(ZYXHR,Recipe for nourishing kidney yin and purging ministerial fire) on estrogen-like activity of environmental endocrine disruptors,nonylphenol(NP) and mixture of NP and bisphenol A(BPA). Methods Totally 30 three-week-old female SD rats were randomly divided into control group(fed with corn oil),exposed group A (feeding NP100 mg / kg ),exposed group B (feeding NP 50 mg/kg+ BPA 200 mg/kg ),treatment group A (NP 100 mg/kg + ZYXHR),treatment group B (NP 50 mg/kg+ BPA 200 mg/kg + ZYXHR).
