Introducción: La acuacultura de truchas ha incrementado gradualmente en las tierras altas de Costa Rica. Las aguas residuales de esta actividad son descartadas directamente en los ríos, sin tratamientos previos. Como consecuencia, la actividad truchícola puede contaminar severamente el agua de los ríos con bacterias que pueden afectar la salud humana. Objetivo: Evaluar la contaminación bacteriana por la acuacultura de truchas en el río Savegre, Costa Rica. Métodos: Contamos los coliformes totales y Escherichia coli de muestras mensuales (2015-2018) en tres secciones del proyecto de acuacultura más grande de la cuenca alta del río. Recolectamos las muestras en la entrada de los estanques para las truchas, a la salida, y 200 m hacia abajo. Resultados: Encontramos menos coliformes totales y E. coli en el agua recolectada justo en la salida del agua de los estanques. El número de coliformes totales fue mayor en el 2016 y 2017, y de E. coli en el 2016. Conclusiones: Conteos de coliformes y de E. coli es muy alto en el río, pero inesperadamente, su número disminuye en el agua residual descartada de los estanques. Podría ser que el mucus producido por las truchas o sustancias liberadas del musgo que cubre la pared de los estanques reduzca el crecimiento de bacterias, como se ha sido sugerido en otros estudios. La contaminación del río parece venir de otras fuentes.


Introduction: The trout aquacultural activity has gradually increased in Costa Rican highlands. Residual waters from this activity are discarded directly in the rivers without any previous treatment process. Consequently, this activity could severely contaminate the river with bacteria that can affect human health. Objective: To evaluate bacterial contamination from trout aquaculture on Río Savegre, Costa Rica. Methods: We counted total coliforms and Escherichia coli from monthly samples (2015-2018) at three sections of the largest aquacultural development in the upper drainage of the river. We collected samples at the fish ponds entrance, exit and 200 m downwards. Results: We found fewer total coliforms and E. coli in the water collected just at the exit of the fish ponds. We counted more total coliforms in 2016 and 2017, and more E. coli in 2016. Conclusions: Coliform and E. coli counts are high in the river, but, unexpectedly, low in the water discarded from the fish tanks. Perhaps the mucus produced by the trouts or substances released by mosses on the fish tank walls reduce bacterial growth, as suggested by other studies. River pollution appears to come from other sources.

ABSTRACT

BACKGROUND: In fish farming, the plant extracts containing antioxidant compounds have been added to the diet for enhancing pathogen resistance. In vitro studies evaluating the antioxidant effect of herbal extracts on fish cell models have focused on ROS production and the respiratory burst mechanism. However, the effects on enzymatic antioxidant defense on salmon leukocytes have not been evaluated. This study aims to evaluate the enzymatic antioxidant defense and ROS-induced cell damage in Salmon Head Kidney-1 (SHK-1) cell line exposed to polyphenol-enriched extract from Sambucus nigra flowers. RESULTS: Firstly, the Total Reactive Antioxidant Power (TRAP) assay of elderflower polyphenol (EP) was evaluated, showing 459 and 489 times more active than gallic acid and butyl hydroxy toluene (BHT), respectively. The toxic effect of EP on salmon cells was not significant at concentrations below 120 mg/ mL and no hemolysis activity was observed between 20 and 400 mg/mL. The treatment of SHK-1 cell line with EP decreased both the lipid peroxidation and protein oxidation induced by H2O2, which could be associated with decreasing oxidative stress in the SHK-1 cells since the GSH/GSSG ratio increased when only EP was added. CONCLUSIONS: These results suggest that plant extracts enriched with polyphenols could improve the enzymatic antioxidant defense of salmon leukocytes and protect the cells against ROS-induced cell damage

ABSTRACT

With the aim to determine the prevalence and mean parasite intensity in Oncorhynchus mykiss, 120 specimens were analyzed between April and September 2018. The cavity analysis was done by visual inspection, scraping of mucus, and extraction of the branchial arches. In the evisceration process, the intestine, the liver, and the kidney were separated, while the musculature was analyzed using the "candling table" method. All the collected material was preserved in 10% buffered formaldehyde and sent to the Laboratory of Parasitology and Parasitic Diseases, of the Center for Agroveterinary Sciences (CAV) of the University of the State of Santa Catarina (UDESC) for making the slides and identifying the parasites. Parasites were detected only in the analysis of the scraping of body mucus. Only specimens of the genus Trichodina were identified, in 34 of the 120 fish analyzed, in two collections, resulting in a prevalence of 28.33%. In total, 144 specimens of Trichodina were observed. The overall mean intensity was of 4.24 parasites in each fish analyzed. Characteristic lesions of infection by protozoa were not identified. This is the first report of the occurrence of Trichodina spp. in O. mykiss bred in an intensive system in Brazil, with low rates of parasitic infection in the mountain region of Santa Catarina.(AU)

ABSTRACT

Length growth as a function of time has a non-linear relationship, so nonlinear equations are recommended to represent this kind of curve. We used six nonlinear models to calculate the length gain of rainbow trout (Oncorhynchus mykiss) during the final grow-out phase of 98 days under three different feed types in triplicate groups. We fitted the von Bertalanffy, Gompertz, Logistic, Brody, Power Function, and Exponential equations to individual length-at-age data of 900 fish. Equations were fitted to the data based on the least square method using the Marquardt iterative algorithm. Accuracy of the fitted models was evaluated using a model performance metrics combining mean squared residuals (MSR), mean absolute error (MAE) and Akaike's Information Criterion corrected for small sample sizes (AICc). All models converged in all cases tested. Evaluation criteria for the Logistic model indicated the best overall fit (0.67 of combined metric MSR, MAE and AICc) under all different feeding types, followed by the Exponential model (0.185), and the von Bertalanffy and Brody model (0.074, respectively). Additionally, ∆AICc results identify the Logistic and Gompertz models as being substantially supported by the data in 100% of cases. The logistic model can be suggested for length growth prediction in aquaculture of rainbow trout.(AU)


O crescimento em comprimento em função do tempo tem uma relação não linear; por isso, funções não lineares são recomendáveis para descrever essa relação. Seis modelos não lineares foram usados para calcular o ganho em comprimento de truta-arco-íris (Oncorhynchus mykiss) durante 98 dias, na fase final da engorda, submetidas a três dietas diferentes em grupos triplicados. Foram ajustadas as equações de von Bertalanffy, Gompertz, logístico, Brody, função potencial e exponencial a dados individuais de comprimento-idade de 900 peixes. O ajuste foi feito pelo método dos mínimos quadrados, usando-se o algoritmo iterativo de Marquardt. A precisão do ajuste foi avaliada pelo uso de critérios combinados de ajuste: quadrado médio do resíduo (QMR), erro médio absoluto (EMA) e o critério de informação de Akaike corrigido para tamanhos amostrais pequenos (AICc). Todos os modelos atingiram a convergência para cada caso avaliado. Os critérios de avaliação do modelo logístico indicaram o melhor ajuste geral (0,67 vez dos critérios combinados MSR, MAE e AICc) para cada grupo de peixe avaliado, seguido pelo modelo exponencial (0,185) e os modelos von Bertalanffy e Brody, com 0,074, respectivamente. Similarmente, os resultados de ΔAICc identificaram-se ao modelo logístico e ao de Gompertz, com grande suporte das informações em 100% dos casos. Por fim, o modelo logístico pode ser sugerido na predição do crescimento em comprimento de truta-arco-íris cultivada.(AU)

ABSTRACT

This study was carried out to evaluate the effects of dietary essential oils of ginger (Zingiber officinale), black seed (Nigella sativa) and cone flower (Echinacea angustifolia) on the immune system of cultured rainbow trout, Oncorhynchus mykiss. A total of 360 fish were divided into 4 groups and 3 replicates each containing 30 fish. The fish were fed for 21 days with 1% of the herbal essential oils and with unsupplemented diet as the control group. Haematological and immune parameters including serum lysozyme, number of phagocytic bacteria, Hb, PCV, MCV, MCH, MCHC, number of RBC and WBC were studied. Treatments recorded enhancement in immune parameters compared to the control group. Significantly higher serum lysozyme level and phagocytic germs were detected in the groups fed black seed, ginger and cone flower (P<0.05). However, no significant difference was found in RBC, Hb, PCV, MCV, MCH and MCHC of fish fed the diets containing essential oils and the control group (P>0.05). The results of this study demonstrated that essential oils might strengthen the non-specific immunity of rainbow trout.(AU)


Avaliou-se a eficácia de uma dieta com semente preta (Nigella sativa), gengibre (Zingiber officinale) e Flor-de-cone (Echinacea angustifolia) sobre o sistema imunológico de truta arco-íris (Oncorhynchus mykiss) em cultivo. Um total de 360 peixes foram divididos em 4 grupos com 3 repetições cada, contendo 30 peixes. Os peixes foram alimentados por 21 dias com 1% de óleo essencial herbáceo e com dieta comercial não suplementada como controle. Parâmetros imunes e hematológicos incluindo soro lisozima, número de bactéria fagocítica, Hb, PCV, MCV, MCH, MCHC, número de RBC e WBC foram estudados. Os tratamentos registraram a melhoria em parâmetros imunes comparados ao grupo de controle. Foram detectadas concentrações de lisozima no soro e germes fagocíticos significativamente altos em grupos que receberam semente preta, gengibre e Flor-de-cone (P<0.05). Porém, não foi encontrada diferença significativa em RBC, Hb, PCV, MCV, MCH e MCHC nos peixes alimentados com dietas contendo óleos essenciais e no grupo de controle (P>0.05). Os resultados deste estudo demonstraram que óleos essenciais podem aumentar a imunidade não específica da truta arco-íris.(AU)

ABSTRACT: Body composition (total crude protein, lipid, ash, dry matter and moisture) and fatty acid profiles were compared between wild and farmed brown trout and between farmed rainbow trout. Farmed brown trout contained the highest amount of crude protein (18.39%), whereas farmed rainbow trout contained higher levels of crude lipid (2.35%). Thirty six fatty acids were found, including sixteen saturated fatty acids (SFA), nine monounsaturated fatty acids (MUFA) and eleven polyunsaturated fatty acids (PUFA). The most abundant SFA in all fish were palmitic acid and stearic acid. However, the most abundant fatty acids in all trout samples were MUFA and PUFA. MUFA were the most abundant fatty acid in farmed brown trout because of high abundance of oleic acid in this samples (35.46g / 100g fatty acids). PUFA were predominating in the samples of wild brown trout and of rainbow trout 56.16 and 56.29g/100g fatty acids, respectively). Linoleic acid was the most abundant fatty acid reported in the rainbow trout (47.17g/100g fatty acids). Significantly higher amounts of docosahexaenoic acid, α-linolenic acid, arachidonic acid and eicosapentaenoic acid were observed in the wild trout samples. Wild brown trout contained significantly more docosahexaenoic acid, α-linolenic acid, Eicosapentaenoic acidArachidonic acid. Sensory quality evaluation, by a consumers' panel, revealed all samples were equally well accepted.


RESUMO: A composição corporal (teores de proteína, gordura total, cinzas, matéria seca e humidade) e os perfis de ácidos gordos da truta-fário (selvagem e proveniente de aquacultura) e da truta-arco-íris (cultivada) foram comparados. A truta-fário cultivada continha a maior quantidade de proteína bruta (18,39%), enquanto a truta arco-íris possuía teores mais elevados de lípidos (2,35%). Foram detectados 36 ácidos graxos, incluindo 16 ácidos graxos saturados (SFA), nove ácidos graxos monoinsaturados (MUFA) e 11 ácidos graxos poliinsaturados (PUFA). Os SFA mais abundantes foram o ácido palmítico e o ácido esteárico. No entanto, os ácidos graxos mais abundantes em todas as amostras de truta foram os MUFA e os PUFA. A grande abundância de ácido oleico existente nas amostras de truta-fário cultivada (35,46g/100g de ácidos graxos) faz com que os MUFA sejam os ácidos graxos mais abundantes nesta variedade de truta (42,43g/100g de ácidos graxos) Nas amostras de truta-fário selvagem e nas de truta arco-íris predominaram os PUFA (56,16 e 56,29g/100g de ácidos graxos, respetivamente), sendo o ácido linoleico o mais abundante na truta-arco-íris (47,17g/100g de ácidos graxos). Nas amostras truta-fário selvagem foram observadas quantidades significativamente mais elevadas de ácido docosahexaenóico, ácido α-linolênico, ácido acidoaraquidônico e ácido eicosapentaenóico. Todas as amostras foram igualmente bem aceitas por um painel de consumidores.

ABSTRACT

Abstract The phosphorus and nitrogen discharge via effluent of intensive trout farming system was quantified through the use of environmental indicators. The nutrient loads, the mass balance, the estimated amount of nutrients in feed and the amount of nutrients converted in fish biomass were calculated based on the concentrations of phosphorus (P) and nitrogen (N) in the feed and in the water. Of the offered feed, 24.75 kg were available as P and 99.00 kg as N, of these, 9.32 kg P (38%) and 29.12 kg N (25%) were converted into fish biomass and 15.43 kg P (62%) and 69.88 kg N (75%) were exported via effluent. The loads and the mass balance show the excessive discharge of nutrients via effluent, corroborated by the feed conversion ratio (2.12:1) due to the low efficiency of feed utilization, therefore, it is proposed the use of this zootechnical parameter as environmental indicator. In addition, feed management practices are not adequate, highlighting the low frequency of feeding during the day, excessive amount and low quality of feed offered. These results demonstrate the need for adequate feed management and the need for careful monitoring of effluent.


Resumo A descarga de fósforo e nitrogênio via efluente do sistema intensivo de truticultura foi quantificada através da utilização de indicadores ambientais. As cargas de nutrientes, o balanço de massa, a quantidade estimada de nutrientes na ração e a quantidade de nutrientes convertidos em biomassa de peixes foram calculados com base nas concentrações de fósforo (P) e nitrogênio (N) na ração e na água. Da ração oferecida, 24,75 kg estavam disponíveis como P e 99,00 kg como N, destes, 9,32 kg de P (38%) e 29,12 kg de N (25%) foram convertidos em biomassa de peixe e 15,43 kg P (62%) e 69,88 kg N (75%) foram exportados via efluente. As cargas e o balanço de massa mostram a descarga excessiva de nutrientes via efluente, corroborado pela taxa de conversão alimentar (2,12:1), devido à baixa eficiência na utilização da ração, portanto, propõe-se a utilização deste parâmetro zootécnico como indicador ambiental. Além disso, as práticas de manejo alimentar não são adequadas, destacando a baixa frequência de alimentação durante o dia, quantidade excessiva e baixa qualidade da alimentação ofertada. Esses resultados demonstram a necessidade de manejo alimentar adequado e de monitoramento do efluente.

ABSTRACT

A indoleamina 2,3-dioxigenase (IDO) é uma enzima que cataboliza o aminoácido triptofano, levando à inibição da proliferação de linfócitos T, seja pela exaustão desse aminoácido no ambiente, ou pela indução via catabólitos induzindo-os a apoptose. Em mamíferos, esta enzima atua em diversas condições do organismo como a gestação, infecções, inflamações crônicas, transplantes e tumores, atuando na regulação imunológica. Estudos recentes identificaram a presença de moléculas homólogas a IDO em espécies filogeneticamente inferiores, cuja função parece estar restrita ao metabolismo do triptofano como fonte de energia. Este estudo teve por objetivo averiguar a expressão da IDO em células sanguíneas e órgãos hematopoiéticos de truta arco-íris pela imuno-histoquímica, buscando evidências de que a mesma poderia, nesta espécie, estar relacionada ao sistema imune. A expressão de IDO foi observada nos órgãos hematopoiéticos estudados incluindo o rim cefálico que apresentou marcação em células interrenais e leucócitos; baço, na qual a marcação restringiu à alguns leucócitos; no fígado a marcação ficou limitada à apenas algumas células dentro dos vasos sanguíneos e nas extensões sanguíneas pode-se visualizar a marcação de alguns leucócitos como os monócitos, linfócitos e neutrófilos. A predominância da marcação da IDO nesses tecidos pode constituir uma evidência de que a IDO identificada na O. mykiss esteja relacionada ao sistema imunológico nessa espécie...


Indoleamine 2,3-dioxygenase (IDO) is an enzyme that catabolizes the amino acid tryptophan, leading to inhibition of T lymphocyte proliferation, whether by depletion of this amino acid in the environment, or by induction via the catabolites inducing apoptosis. In mammals, this enzyme acts on various conditions of the body such as pregnancy, infections, chronic inflammation, transplantation and tumors, acting in immune regulation. Recent studies have identified the presence of homologous molecules IDO lower phylogenetically related species, whose function appears to be confined to the tryptophan metabolism as an energy source. This study aimed to investigate the expression of IDO in blood cells and hematopoietic organs of rainbow trout by immunohistochemistry, seeking evidence that it could, this species is related to the immune system. The expression of IDO was observed in hematopoietic organs studied including head kidney that show labeling in interrenal cells and leukocytes; spleen, in which the marking restricted to a few leukocytes in the liver;, labeling was restricted to only certain cells within the blood vessels and the blood extensions can view the marking of some leukocytes including monocytes, lymphocytes and neutrophils. The predominance of IDO marking these tissues may constitute evidence that IDO identified in O. mykiss is related to the immune system in this species...

En este artículo se analizan con parámetros de composición 58 puestas de trucha arcoiris obtenidas, manejadas y monitoreadas individualmente en desempeño desde la fertilización hasta finalizar la etapa de reabsorción de vesícula. Además de los valores reproductivos, el contenido de ácidos grasos en las ovas se determinó en fresco, mediante cromatografía de gases. En el perfil medio se destacan los ácidos palmítico (C16), oleico (C18:1n-9) y docosahexaenoico (C22:6n-3) como los más representativos, con casi el 60% del total y, en general, en concentraciones estables entre las hembras. Tanto para cada ácido determinado como para el conjunto de contenidos integrados de las series n-3 y n-6, y los colectivos de saturados (SAF), monoinsaturados (MUFA) y poliinsaturados (PUFA), se define un patrón de composición similar al reportado para la especie en otros esquemas de manejo y bajo regímenes nutricionales variables. Se analizó la condición conservativa en la incorporación de ácidos grasos al huevo, y se discutió su utilidad como posibles definitorios de calidad, teniendo como referente la alta variabilidad registrada en la supervivencia al final del proceso de incubación...


This article analyzes, with composition parameters, 58 rainbow trout egg-layings obtained, handled and monitored individually in performance from their fertilization until the end of the gallbladder reabsortion phase. Aside from the reproductive values, the content of fatty acids in the eggs was determined in fresh conditions through gas chromatography. The average profile highlights the palmitic acid (C16), oleic acid (C18:1n-9) and docosahexaenoic acid (C22:6n-3), as the most representative, with almost 60% of the total and, in general, in stable concentrations among females. Both for each determined acid and for the suite of integrated series n-3 and n-6, and the groups of saturated (SAF), monounstaturated (MUFA) and polyunsaturated (PUFA), a pattern of similar composition to the one reported is defined for the species in other management schemes and under varying nutritional regimes. The conservative condition was analyzed in the incorporation of fatty acids into the egg and its usefulness as possible quality definers was discussed, using the high variability recorded in survival at the end of the incubation process as reference...


Neste artigo analisam-se com parâmetros de composição, 58 posturas de truta arco-íris obtidas, manejadas e monitoradas individualmente em desempenho, desde a fertilização até a finalização da etapa de absorção de vesícula. Além dos valores reprodutivos, o conteúdo de ácidos graxos nas ovas foi determinado em fresco, mediante cromatografia de gases. No perfil médio destacam-se os ácidos palmíticos (C16), oleico (C18:1n-9) e docosaexaenoico (C22:6n-3), como os mais representativos, com quase 60% do total e, em geral, em concentrações estáveis entre as fêmeas. ABSTRACT

The exploitation of seaweeds in Chile has been carried out for more than 60 years. More recently, seaweeds have been used for the production of alginate, agar and carrageenan, agricultural fertilizers and industrial aquaculture (feed for abalone and sea urchins), increasing the added value of this natural resource. In the Magellan Region (56ºS), the giant kelp Macrocystis pyrifera (L.) C. Agardh presents the most extensive kelp forest, reaching a biomass of approximately 12 kg.m-2. Recent studies have shown potential benefits from adding M. pyrifera- derived flour to salmonid feed. Research is currently underway to evaluate the useof brown algae-derived products for marine aquaculture feed of Oncorhynchus mykiss in tanks. There was no apparent adverse effect on the evaluated parameters that can be attributed to the incorporation of M. pyrifera meal in the diets fed to salmonids. Even when the control diet had numerically the best performance in zootechnical terms, the analysis of variance of all parameters evaluated showed no significant differences with regard to diets containing M. pyrifera meal. These results demonstrated that seaweed meal has important benefits for animal health and nutrition that could be applied or tested in other marine organisms of commercial importance.

Alteration in the relative percentage of survival (RPS) rate of rainbow trout (Oncorhynchus mykiss) exposed to 5, 10 and 25�g ml-1 levamisole for 2 h against Yersinia ruckeri was investigated. The average weight of the 120 fish used in this study was 6.3g. Upon challenge with a virulent strain, the relative survival percentage of respectively 83.3 percent, 86.7 percent and 76.6 percent was recorded. The results suggest that the application of levamisole in fish farms could increase resistance to infection of fish and offer economic benefits.

ABSTRACT

Growth is one of the most important aspects in the genetic improvement of cultured fish species. Consequently, genetic parameters related to this feature and their response to selection have been the focus of most research in this area. Such research indicates that, in general, there is enough additive genetic variance related to growth, justifying the use of selection. Based on the usefulness of cytogenetic and molecular markers in the fish culture, the aim of the present work was to analyze the possible relationships among cytogenetic characteristics, specifically the NOR phenotypes, and the increase in length and weight in specimens of the rainbow trout (Oncorhynchus mykiss), resultant from directed mating between homozygous females and heterozygous males according to their NOR phenotypic patterns. The equations of the relationship between length and weight of the analyzed specimens followed the model Wt = a Lt b, showing b values higher than 3, determinant of a positive allometric growth. The results showed that the different NOR phenotypes were not related with the growth values for length and weight in any statistical test.


O crescimento é um dos mais importantes aspectos considerados no melhoramento genético de espécies de peixes cultivadas. Conseqüentemente, a ênfase das pesquisas na área tem sido avaliar os parâmetros genéticos relacionados com esta característica e sua resposta à seleção. Essas pesquisas indicam, em geral, haver variância genética aditiva suficiente para justificar o uso da seleção. Considerando que a utilização de marcadores citogenéticos ou moleculares pode ser de grande valia para a piscicultura, o presente trabalho teve como objetivo analisar a possível relação entre as características citogenéticas, de modo específico os padrões fenotípicos das NORs e o crescimento em comprimento e em peso dos exemplares de trutas arco-íris (Oncorhynchus mykiss) resultantes de acasalamentos dirigidos entre fêmeas homozigotas e machos heterozigotos, conforme caracterização do padrão fenotípico da NOR. As equações da relação comprimento e peso dos indivíduos analisados seguiram o modelo onde Wt = a Lt b, mostrando valores de b maiores que 3, determinante de um crescimento do tipo alométrico positivo. Os resultados mostraram que os padrões fenotípicos da NOR dos indivíduos homozigotos e dos heterozigotos não apresentaram diferenças estatisticamente significativas entre seus valores de crescimento em comprimento ou peso.
