ABSTRACT

Abstract The immunomodulatory and growth promoter effects of brown propolis ethanolic extract (PEE) were determined in pacu. Fish (28.4±0.4 g) were randomly distributed into 12 polyethylene circular tanks (250 L, 30 fish per tank) and fed for 60 days with a commercial diet (32% crude protein) supplemented with 0.0, 1.5, 3.0 and 4.5% (v/w) of PEE in a totally randomized experimental design (n=3). Fish organic defense cells numbers such as thrombocytes and neutrophils increased (p<0.05) in fish fed 3.0% dietary PEE. Serum lysozyme concentration also increased (p<0.05) in fish fed 1.5 and 3.0% PEE supplementation when compared to unsupplemented fish. Growth parameters were not influenced (p>0.05) by treatments. Moreover, dietary PEE decreased (p<0.05) fish intestinal muscular thickness when compared to control treatment. Intestine villi height also significantly decreased in fish fed 3.0% PEE. Long term dietary PEE at 3.0% supplementation level modulated fish nonspecific immune system and it is a non-toxic substance for pacu.

ABSTRACT

Background@#The aims of this study were to determine the average length of stay in the Post-anesthesia Care Unit (PACU LOS) in the Philippine General Hospital (PGH) and to create a model that will predict the PACU LOS based on the factors that significantly affect the LOS. Determination and prediction of PACU LOS is essential in resource utilization, and in cost containment and reduction. Addressing the modifiable variables that affect the PACU LOS may lead to an improvement in the LOS of patients in the PACU and, consequently, to better recovery room staffing and a reduced cost for the patients and the hospital.@*Methods@#A prospective chart review of 400 postoperative patients admitted in the PGH PACU was done. Summary statistics were presented. Using the set of variables found to be significant, a regression model was formulated to estimate the PACU LOS.@*Results@#The mean PACU LOS was 4.59 hours. There were significant differences in the mean PACU LOS based on the occurrence of complications. There were also significant differences in the median PACU LOS based on the type and duration of surgery, anesthetic technique, and duration of anesthesia. The multiple linear regression model that best predicted PACU LOS included ASA-PS classification, type of surgery, duration of surgery, anesthetic technique, and occurrence of intraoperative or postoperative complications. @*Conclusions@#The mean PACU LOS of the Philippine Genera Hospital is higher than that of published data. The factors included in the model that best predicts PACU LOS may be studied to improve the PACU LOS.@*@#

ABSTRACT

Objectives:To observe the method of pain assessment and pain management intervention performed by nurses in the PACU.Methodsand Design:A QI prospective observational study was conducted to observe nurse’s pain assessment and management of thirty (30) patients from the time of PACU admission to discharge. The sample size was determined using the sealed envelope power calculator.Data Collection Included:patients demographics, the method and frequency of pain assessments as well as modalities of the pain intervention and the type and average dose of pain medications administered by PACU nurses. Data analysis was done using Microsoft excel. Results:No validated pain assessment tool was used in the PACU. The majority of patients 67%, n=20) had no pain assessments or pain interventions. When performed, the frequency of pain assessments recorded were low, 70% of patients had 1-2 assessments. The principal pain management intervention was pharmacological with the use of opioids, accounting for 96%.Conclusion:Post-operative pain management in the PACU at GPHC does not meet accepted standards of care. More frequent nursing pain assessment using a validated pain assessmenttool is required. Monotherapy with the opioid was the main pain intervention for pain management. Recommendations:Effective pain management begins with the appropriate pain assessment; therefore pain management education programs for health care professionals are essential. Also, the implementation of a standardized pain assessment tool, a standardized post anesthetic order sheet with a multimodal approach to pain management and restructuring the post-anesthetic record to allow for documentation of pain assessment will greatly improve pain management in the PACU

ABSTRACT: The growth of cachama hybrids, cachamoto (Colossoma macropomum ♀ x Piaractus brachypomus ♂) and cachamay (P. brachypomus ♀ x C. macropomum ♂) was compared for 154 days to determine which showed the better productivity. The experiment was carried out in three earth ponds, each divided transversely with a plastic net, yielding six experimental units. Juveniles of cachamoto of 12.95±2.43g and 6.75±0.42cm and cachamay of 16.65±3.64g and 7.41±0.62cm were distributed in each sub-tank of 240m2 at a density of 0.5 fish m-2. Fish were fed twice daily except on days when their weight and length, and the physical and chemical parameters of the water were measured. The following productive variables were evaluated: growth in weight and length, daily weight gain, daily feed intake, apparent feed conversion, specific growth rate, condition factor, productivity and the cost/benefit relationship. None of the analyzed variables presented statistical difference between treatments. Physical and chemical parameters of the water remained within the recommended values for cachama, and survival was 100%. Based on the results obtained, it can be concluded that cachama reciprocal hybrids present a similar productive behavior, so that it is indifferent which one is chosen to be raised under the conditions evaluated.


RESUMO: Comparou-se o crescimento de híbridos de tambaqui e pirapitinga, tambatinga (Colossoma macropomum ♀ x Piaractus brachypomus ♂) e pirambaqui (P. brachypomus ♀ x C. macropomum ♂) durante 154 dias para se verificar qual deles apresentava melhor produtividade. O experimento foi realizado em três viveiros de terra, cada um dividido transversalmente com uma rede plástica, originando seis unidades experimentais. Juvenis de tambatinga de 12,95±2,43g e 6,75±0,42cm e pirambaqui de 16,65±3,64g e 7,41±0,62cm foram distribuídos em cada sub-tanque de 240m2 em uma densidade de 0,5 peixe m-2. Os peixes foram alimentados duas vezes por dia, exceto nos dias em que se mediram peso, comprimento e parâmetros físicos e químicos da água. As variáveis produtivas avaliadas foram: crescimento em peso e comprimento, ganho de peso diário, consumo diário de alimento, conversão alimentar aparente, taxa de crescimento específico, fator de condição, produtividade e relação custo/benefício. Nenhuma das variáveis analisadas apresentou diferença estatística entre os tratamentos. Os parâmetros físicos e químicos da água permaneceram dentro dos valores recomendados para tambaqui e pirapitinga e a sobrevivência foi de 100%. Com base nos resultados obtidos, pode-se concluir que os híbridos de tambaqui e pirapitinga apresentam um comportamento produtivo semelhante, de modo que é indiferente qual deles é escolhido para ser criado nas condições avaliadas.

ABSTRACT

ABSTRACT: Probiotics are a novel alternative to antibiotics as growth factors. Previously, our group isolated, selected and tested in vivo, eight autochthonous strains. They showed no significant effects when administered individually. However, the best doses, stages and ways of administration were combined in a multi strain formula. The aim of this research was to evaluate the effect of this probiotic product on the growth and survival of Piaractus mesopotamicus larvae. The administration was implemented during egg incubation and endogenous feeding period (5), during larvae exogenous feeding period (10) and all along the experiment (15). A group without microorganisms was used as control. The probiotic generates significant increments of mean weight and not significant increases of survival and biomass in two of the three tested stages. These results demonstrate the effectiveness of an autochthonous probiotic formula for the culture of this native fish species.


RESUMO: Os probióticos são uma alternativa inovadora aos antibióticos como fatores de crescimento. Anteriormente, nosso grupo isolou, selecionou e testou in vivo oito cepas autóctones. Estas não apresentaram efeitos significativos quando administrados individualmente. No entanto, as melhores doses, etapas e formas da administração foram combinadas para a formulação de um produto contendo multiples cepa. O objetivo deste trabalho foi avaliar o efeito deste produto probiótico no crescimento e sobrevivência de larvas de Piaractus mesopotamicus. A administração foi implementada durante a incubação de ovos e período de alimentação endógeno (5), durante o período de alimentação exógeno das larvas (10) e ao longo do experimento (15). Um grupo sem microrganismos foi utilizado como controle. O probiótico gera um incremento significativo de peso médio e não significativo de sobrevivência e biomassa em dois dos três estágios testados. Estes resultados demonstram a eficácia de uma fórmula probiótica autóctone para a cultura desta espécie de peixe nativa.

ABSTRACT

This study aimed to evaluate weight and morphometric growth of the products of the complete diallel cross-breeding between pacu and tambaqui during the period from the end of spring to the beginning of winter. Was used 400 pacu, tambaqui, tambacu and paqui. 20 fish from each group were collected, weighed and submitted to morphometric evaluation every 28 days. The Logistic function adjusted better to describe the growth in body weight and the morphometric measures for age on all the genetic groups. The studied groups presented similar behavior of growth for body weight and morphometric measures. There were significant differences between the genetic groups regarding parameters A (asymptotic value) and B (integration constant) for body weight and for all the morphometric measures evaluated. For the parameter K (maturity rate), there were significant difference between groups for body weight, head length and body height. The groups possibly were affected the decrease of temperature. The Tambacu was superior to the Paqui in all the variables evaluated for the parameter A, and was also superior to the pure breed, except for head length and body width.(AU)


Este estudo teve como objetivo avaliar o peso e crescimento morfométrico dos produtos do cruzamento dialélico completo entre pacu e tambaqui. 20 peixes de cada grupo foram coletados, pesados e submetidos à análise morfométrica a cada 28 dias. O modelo que obteve o maior coeficiente de determinação e que melhor se ajuste ao método de Akaike foi verificado. A função logística foi ajustada para melhor descrever o crescimento do peso corporal e as medidas morfométricas para a idade em todos os grupos genéticos. Os quatro grupos estudados apresentaram comportamento semelhante para o peso corporal, bem como para as medidas morfométricas. Houve diferenças significativas entre os grupos genéticos sobre os parâmetros A (valor assintótico) e B (constante de integração) para o peso corporal e para todas as medidas morfométricas avaliadas. Para o parâmetro K (taxa de maturação), houve diferença significativa entre os grupos para o peso corporal, comprimento da cabeça e altura do corpo. Os grupos possivelmente foram afetados pela diminuição da temperatura. O tambacu foi superior ao Paqui em todas as variáveis avaliadas para o parâmetro A, e também foi superior para a raça pura, exceto para o comprimento e largura do corpo de cabeça.(AU)

ABSTRACT

The aim of this study was to evaluate the effect of parenteral administration of dexamethasone and levamisole on the accumulation of macrophages and formation of giant cells in chronic inflammation by foreign body and blood parameters in pacu (Piaractus mesopotamicus). It was used 50 mg/kg of levamisole and 2.0 mg/kg of dexamethasone and the combination of both drugs. Coverslips were implanted under the skin. After 2, 7, and 15 days postimplantation (DPI), fish were anesthetized for removal of coverslips and the number of macrophages and giant cells were count. It was also collected blood from the caudal vessel to evaluate: red blood cell count, hematocrit, hemoglobin concentration, leukocytes count, thrombocytes count, MCV, and MCHC. We observed that dexamethasone affects negatively the formation of giant cells in the chronic inflammation for foreign body. Levamisole, despite being immunostimulatory in several species, showed limited action. However, it was enough to counteract the effect of dexamethasone; the association of the drugs did not interfere significantly in erythrocyte and leukocyte number in most of the treatments and times studied. In dexamethasone group there was a reduction in the number of erythrocytes and hemoglobin associated with increased mean corpuscular volume, suggesting slight macrocytic anemia. At 15 DPI, most groups showed the recovery of hematologic response. As in mammals, dexamethasone affects negatively the inflammatory response. Levamisole showed little effect by itself. However, in some parameters the association of both drugs causes similar response to control and naïve groups, showing the antagonistic effect of these drugs.


O objetivo deste estudo foi avaliar o efeito da administração parenteral de dexametasona e levamisol na acumulação de macrófagos e formação de células gigantes na inflamação crônica por corpo estranho e avaliação de parâmetros sanguíneos em pacu (Piaractus mesopotamicus). Utilizou-se 50 mg/kg de levamisol e 2,0 mg / kg de dexametasona e a combinação de ambos fármacos. As lamínulas foram implantadas sob a pele. Depois de 2, 7 e 15 dias pós-implantação (DPI), os peixes foram anestesiados para a remoção das lamínulas e contagem do número de macrófagos e células gigantes. Foi coletado sangue da veia caudal para realizar contagem de células vermelhas, leucócitos, trombócitos, concentração de hemoglobina, MCV e MCHC. Observou-se que a dexametasona afeta negativamente a formação de células gigantes na inflamação crónica por corpo estranho. O levamisol, apesar de ser considerado imunoestimulante em várias espécies, mostrou ação limitada. No entanto, foi suficiente para neutralizar o efeito da dexametasona; a associação das drogas não interferiu significativamente no número de eritrócitos e de leucócitos na maioria dos tratamentos e períodos estudados. No grupo da dexametasona, houve redução do número de eritrócitos e concentração de hemoglobina associado ao aumento do volume corpuscular médio sugerindo leve anemia macrocítica. Aos 15 DPI, a maioria dos grupos mostrou recuperação na resposta hematológica. Como em mamíferos, a dexametasona afeta negativamente a resposta inflamatória. O levamisol mostrou pouco efeito por si só. No entanto, em alguns parâmetros a associação das duas drogas provoca resposta similar ao grupo controle e naive, mostrando o efeito antagonista de estas drogas.

ABSTRACT Knowledge on fasting heat production (HEf) of fish is key to develop bioenergetics models thus improving feeding management of farmed species. The core of knowledge on HEf of farmed, neotropical fish is scarce. This study assessed the effect of body mass and water temperature on standard metabolism and fasting heat production of pacu, Piaractus mesopotamicus, an omnivore, Neotropical fresh water characin important for farming and fisheries industries all through South American continent. An automated, intermittent flow respirometry system was used to measure standard metabolic rate (SMR) of pacu (17 - 1,050 g) at five water temperatures: 19, 23, 26, 29 and 33 °C. Mass specific SMR increased with increasing water temperature but decreased as function of body mass. The allometric exponent for scaling HEf was 0.788, and lied in the range recorded for all studied warm-water fish. The recorded van't Hoff factor (Q10) for pacu (2.06) shows the species low response to temperature increases. The model HEf = 0.04643×W0.7882×T1.837 allows to predict HEf (kJ d-1) from body mass (W, kg) and water temperature (T, °C), and can be used in bioenergetical models for the species.

ABSTRACT Objective. This study evaluated the cell kinetic and formation of granuloma during chronic inflammation induced by Bacillus Calmette-Guérin (BCG) in the skeletal muscle of Piaractus mesopotamicus, as a histopathology model to study innate immunity. Materials and methods. Sixty fish were divided in two groups: BCG-inoculated and non-inoculated fish and the inflammatory response analyzed 3, 7, 14, 21 and 33 days post-inoculation (DPI) by histopathology after hematoxylin-eosin and Ziehl-Neelsen staining. Results. 3 DPI of BCG showed a diffuse inflammatory reaction mostly composed by mononuclear cells. The inflammation continued diffuse 7 DPI initiating the cellular organization surrounding the inoculum and have continued at 14 DPI with discrete presence of epithelioid-like type cells with acidophilic cytoplasm and floppy chromatin. Higher cellular organization (21 DPI) surrounding the granuloma with intense peripheral mononuclear inflammatory infiltrate and nevertheless, an increase in the number of fibroblasts and macrophage-like cells was observed. The inflammatory process became less diffuse 33 DPI with formation of small amount of granuloma surrounded by the same type of reaction found in bigger granuloma. Both the young and old granuloma presented typical characteristic around the inoculum composed by a layer of epithelioid-like type cells, besides macrophages, some lymphocytes and abundant fibroblasts. Conclusions. This study showed the feasibility in the use of pacus to study chronic granulomatous inflammatory response induced by BCG, characterized by changes in the kinetics of inflammatory cells in skeletal muscle classifying as immune-epithelioid type, similar to granulomatous inflammation caused by M. marinum in teleost fish.


RESUMEN Objetivo. Este estudio evaluó la cinética celular y la formación de granuloma durante la inflamación crónica inducida por el Bacilo Calmette-Guérin (BCG) en el músculo esquelético de Piaractus mesopotamicus, como modelo histopatológico para estudiar la inmunidad innata. Materiales y métodos. Sesenta peces fueron divididos en dos grupos: peces inoculados con BCG y no inoculados y la respuesta inflamatoria analizada en 3, 7, 14, 21 y 33 días post-inóculo (DPI) por medio del análisis histopatológico y tinciones de hematoxilina-eosina y Ziehl-Neelsen. Resultados. 3 DPI de BCG se observó reacción inflamatoria difusa principalmente formada por infiltrado celular mononuclear. Al 7° DPI la inflamación continuaba difusa con inicio de organización celular alrededor del inoculo, que se observó hasta el 14° DPI con discreta presencia de células de tipo epiteliodes con citoplasma acidófilo y cromatina laxa. Para el 21° DPI se observó alta organización celular alrededor del granuloma con intenso infiltrado mononuclear periférico e incremento en el número de fibroblastos y macrófagos. El proceso inflamatorio se tornó menos difuso a los 33 DPI con formación de pequeños granulomas contenidos dentro de uno más grande. Los granulomas formados más rápidamente así como los formados tardíamente, presentaron características típicas alrededor del inóculo compuesta por una camada de células tipo epitelioides, macrófagos, linfocitos y fibroblastos. Conclusiones. Este estudio mostró la viabilidad del uso del P. mesopotamicus para estudiar la respuesta inflamatoria crónica granulomatosa inducida con BCG, caracterizado por la evolución de la cinética de células inflamatorias en el músculo esquelético clasificándolo como de tipo inmune-epitelioide, similar a la inflamación granulomatosa causada por M. marinum en peces teleósteos.

ABSTRACT

Wound healing and tissue repair are necessary to ensure survival and health of any organism. The aim of this study was to investigate the impact of supplementation with chromium carbochelate (CC) and Saccharomyces cerevisiae (SC) on wound healing in tropical teleost fish Piaractus mesopotamicus. Thus, fish were distributed into four groups: a) control (without supplementation); b) supplemented with 18 mg/kg of chromium carbochelate; c) supplemented with 0.3% of S. cerevisiae and d) supplemented with an association of both supplements. After 105 days of feeding, full-thickness skin incisions (2.0 x 1.0 x 0.25 cm) were performed removing epidermis and dermis. Macroscopic and histologic observations were carried out at 1, 3, 7, 14, 21, 28, and 35 days after wounding to monitor the healing rate. Opposing fronts advanced gradually and faster each day demonstrating a progressive increase in the healing process over time. The inflammatory process was exacerbated and expansive, with an increase in mucous cells and chromatophores. Although no significant differences were observed between groups on wound retraction and microscopic parameters, fish supplemented with CC and SC showed faster re-epithelialization, greater degree of organization of collagen fibers, and higher neovascularization. We concluded that supplementation with S. cerevisiae and chromium carbochelate improves specific aspects of cutaneous healing process in pacu.(AU)


A cicatrização e reparação de tecidos são mecanismos essenciais para garantir a sobrevivência e saúde de qualquer indivíduo. O objetivo deste trabalho foi avaliar o impacto da suplementação com carboquelato de cromo (CC) e Saccharomyces cerevisiae (SC) sobre a cicatrização no peixe tropical Piaractus mesopotamicus. Para isto, os peixes foram distribuídos em quatro grupos: controle (sem tratamento), suplementados com 18 mg/kg de carboquelato de cromo, 0,3% de S. cerevisiae e associação de ambos os suplementos. Após 105 dias de alimentação, foram realizadas incisões na pele de espessura completa (2,0 x 1,0 x 0,25 cm) removendo epiderme e derme. Avaliações macroscópicas e microscópicas foram realizadas 1, 3, 7, 14, 21, 28 e 35 dias após a indução das feridas, para monitorar a taxa de cicatrização. As bordas opostas das feridas avançaram gradualmente a cada dia, demonstrando o aumento progressivo do processo de cicatrização ao longo do tempo. O processo inflamatório foi exacerbado e expansivo, com aumento no número de células mucosas e cromatóforos. Apesar deste processo, não foram observadas diferenças significativas na retração das feridas e nos parâmetros microscópicos entre os grupos. Peixes suplementados com CC ou Sc apresentaram rápida reepitelização, maior grau de organização de fibras colágenas e de neovascularização inicial. Concluiu-se que a suplementação com S. cerevisiae ou carboquelato de cromo melhora aspectos específicos do processo cicatricial no pacu.(AU)

Objective To compare the postoperative extubation time and length of PACU retention between patients who had undergone retroperitoneal laparoscopic unilateral adrenal pheochromocytoma resection and other adrenal tumor resection.Methods A retrospective study of 57 hypertensive patients who had undergone retroperitoneal laparoscopic unilateral adrenal tumor resection under the general anesthesia was conducted.Nineteen patients with adrenal pheochromocytoma were assigned to group S,and other 38 patients were assigned to group D.We recorded and compared the gender ratio,ASA grade,age,weight,height,duration of operation and anesthesia,dosage of anes thetics,extubation time,and length of PACU retention between the two groups.Results There were no significant differences in gender ratio,ASA grade,age,weight,height,duration operation and anesthesia,as well as doses of propofol,midazolam,fentanyl and muscle relaxants between the two groups.As to extubation time and length of PACU retention,those of group S were both significantly longer than those of group D [30.0(25.0-42.5)min vs 20.0(15.0-20.0)min;70.0(57.5-82.5)min vs 45.0(33.5-55.0) min,P<0.05,respectively].Conclusion Retroperitoneal laparoscopic unilateral adrenal pheochromocytoma resection might lead to postoperatively longer extubation time and length of PACU retention.It does not relate to the dosage of anesthetics used perioperatively.

ABSTRACT

This paper presents results from a study of albumin from pacu (Piaractus mesopotamicus, Holmberg 1887) and the catfish pintado (Pseudoplatystoma corruscans, Spix & Agassiz, 1829), two neotropical fish species inhabitants of Brazilian rivers, comparing their molecular mass and discussing their secondary structures based on spectropolarimetric (circular dychroism) measurements. Genetic controlled specimens were obtained from two fish hatcheries, located in Mococa (pacu) and in São João da Boa Vista (pintado), both in São Paulo State, Brazil. After a period of adaptation in holding tanks, fish blood samples were taken by punctioning their abdominal aorta. Purified albumin was obtained by gel filtration. SDS-PAGE electrophoresis was performed for the molecular mass estimation. Circular Dichroism spectra were registered for albumins of the two fish species over the range of 190-250 nm (far-UV), which shown two negative bands at 217 and 208 nm, a positive peak at 196 nm and a crossover at 200 nm. This profile is compatible with proteins that content predominantly alpha-helix structure.


Este artigo apresenta os resultados de um estudo sobre as albuminas de pacu (Piaractus mesopotamicus, Holmberg 1887) e pintado (Pseudoplatystoma corruscans, Spix & Agassiz, 1829), duas espécies neotropicais de peixes nativas do Brasil, determinando as suas massas moleculares e discutindo suas estruturas secundárias, com base em medidas de espectropolarimetria (dicroísmo circular). Espécimes controlados geneticamente foram obtidos de duas diferentes pisciculturas, uma localizada na cidade de Mococa (pacu) e a outra, na cidade de São João da Boa Vista (pintado), ambas no Estado de São Paulo, Brasil. Após um período de adaptação em tanques apropriados, amostras de sangue foram coletadas por punção da aorta abdominal dos peixes. Albumina pura foi obtida por gel filtração dessas amostras e as massas moleculares foram determinadas a partir dos dados da eletroforese SDS-PAGE. Espectros de dicroísmo circular das albuminas dos peixes foram registrados na região de 190-250 nm (far-UV), os quais mostraram duas bandas negativas, a 217 e 208 nm, um pico positivo a 196 nm e um crossover a 200 nm; perfil este compatível com proteínas que contem predominantemente estrutura alfa-hélice.

ABSTRACT

Introduction of fish species is a globally widespread practice that causes losses of native species and homogenization of diversity within and across continents. Diet assessments are important tools to depict the ecological function of species introduced into novel ecosystem and possible direct and indirect ecological effects. In this study, we compare the diet of Piaractus brachypomus, a mainly frugivorous Neotropical fish, introduced into the Sepik-Ramu River Basin (Papua New Guinea) nearly two decades ago, to that of similar size individuals from Neotropical populations in the Amazon and Orinoco River basins (South America). In contrast to native populations that feed mainly on terrestrial plants and invertebrates, the diet of introduced P. brachypomus is mainly composed of fish remains and aquatic plants, while terrestrial plants are frequently consumed but in relatively smaller amounts. These findings show that P. brachypomus has an inherently plastic diet that can be adjusted when displaced to a novel geographic area. While trophic plasticity increases the likelihood of a species to establish breeding populations after its introduction, it also reduces our ability to predict negative effects on native species.


La introducción de peces es una práctica que se extiende globalmente y que causa perdida de especies nativas y homogenización de la diversidad dentro y entre continentes. Los estudios de dieta son herramientas útiles para definir la función ecológica de una especie introducida en un ecosistema nuevo y los posibles efectos directos e indirectos. En este estudio describimos la dieta de Piaractus brachypomus, una especie de pez Neotropical principalmente frugívora, que fue introducida en el río Sepik (Papua Nueva Guinea) hace casi dos décadas; y la comparamos con aquella de individuos de tamaño corporal similar de poblaciones nativas en las cuencas de los ríos Amazonas y Orinoco (América del Sur). En contraste con las poblaciones nativas que se alimentan principalmente de frutas y semillas, plantas e invertebrados terrestres, la dieta de individuos de P. brachypomus introducidos se compone principalmente de restos de peces y plantas acuáticas, mientras que plantas terrestres son consumidas frecuentemente pero en pequeñas cantidades. Estos resultados indican que P. brachypomus tiene una dieta plástica que puede adaptarse cuando la especie se desplaza a un área geográfica nueva. A pesar de que la plasticidad trófica aumenta la probabilidad de que una especie establezca poblaciones reproductivas después de la introducción, también reduce nuestra habilidad de predecir los efectos negativos sobre especies nativas.

ABSTRACT

Monogeneans are the parasites mostly found on the body surface and gills of fish and can cause large losses in farmed fish. Some studies demonstrate elevated parasitic levels causing hematological alterations. But few of them relate the effects of parasitism on the hematology and histopathology of native freshwater farmed fish. This study evaluated the host-parasite relationship in pacu (Piaractus mesopotamicus) parasitized by the monogenean Anacanthorus penilabiatus. Hematological and parasitological assessments were obtained in 60 fish captured in a fish farm located in Dourados, State of Mato Grosso do Sul, Central Brazil. Fish were analyzed in different categories of parasite number: class I (n=13; 0-200 parasites), class II (n=17; 201-1200 parasites); class III (n=7; 1201-2200 parasites); and class IV (n=23; more than 2200 parasites per host). The highest levels of parasitism caused significant decrease (p<0.05) in the hematocrit, red blood cells (RBC), mean hemoglobin concentration (MCHC) and basophils number. Thrombocytes, mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), monocytes, eosinophils, neutrophils and LG-PAS did not present significant difference among the parasitic levels. In contrast, increased number of total leukocytes and lymphocytes were found in highly-parasitized fish. A positive linear correlation (p<0.01) was found between the amount of parasites and fish weight. Histopathology revealed severe hyperplasia, sub-epithelial edema, fusion of the secondary lamellae, focal and multifocal necrosis in highly parasitized fish.


Parasitos Monogenea são principalmente encontrados na superficie corporal e brânquias dos peixes, e podem acarretar grandes perdas em pisciculturas. Alguns estudos demonstram que elevados níveis de infestação parasitária podem alterar os parâmetros sanguíneos. Porém, poucos estudos se direcionam a esclarecer os efeitos do parasitismo sobre as características hematológicas em peixes nativos. Este estudo avaliou a relação parasito-hospedeiro em pacu (Piaractus mesopotamicus) parasitado pelo monogenético Anacanthorus penilabiatus. Avaliações hematológicas e parasitológicas foram obtidas de 60 peixes capturados de uma piscicultura localizada em Dourados, Estado de Mato Grosso do Sul (MS), Brasil Central. Os peixes foram divididos em diferentes categorias de número de parasitos: classe I (n=13; 0-200 parasitos), classe II (n=17; 201-1200 parasitos); classe III (n=7; 1201-2200 parasitos); e classe IV (n=23; mais que 2200 parasitos por hospedeiro). Os níveis mais elevados de parasitismo causaram diminuição significativa (p<0.05) no hematócrito, eritrócitos (RBC), concentração de hemoglobina corpuscular média (CHCM) e número de basófilos. Trombócitos, volume corpuscular médio (VCM), concentração de hemoglobina corpuscular media (CHCM), monócitos, eosinófilos, neutrófilos e LG-AS não apresentaram diferença significativa entre os níveis de parasitismo. Em contraste, o aumento do número de leucócitos totais e linfócitos foram encontrados em peixes altamente parasitados. Houve correlação linear positiva entre a quantidade de parasitos e o peso dos peixes. O exame histopatológico revelou severa hiperplasia, edema sub-epitelial, fusão das lamelas secundárias, necroses focal e multifocal em peixes altamente parasitados.

ABSTRACT

Utiaritichthys esguiceroi is described from the upper portion of the rio Juruena, rio Tapajós drainage, Mato Grosso State, Central Brazil. The new species distinguished from its two congeners, U. sennaebragai Miranda Ribeiro and U. longidorsalis Jégu, Tito de Morais & Santos, by having 99 to 101 perforated scales on lateral line (vs. 69 to 83), presence of 17 to19 prepelvic spines (vs. 9-13 in U. sennaebragai and 28-31 in U. longidorsalis), 20 to 21 postpelvic spines (vs. 15 to 19 in U. sennaebragai, and 14 in U. longidorsalis), and 23 to 25 circumpeduncular scales (vs. 30-48 in U. sennaebragai, and 33-35 in U. longidorsalis). Furthermore, the new species differs from U. longidorsalis by having larger interdorsal width, and adipose-fin base length (11.8-15.6 vs. 7.1-7.9% of SL, and 4.2-5.8 vs. 3.7-3.8% of SL, respectively).

A espécie nova diferencia-se de suas congêneres, U. sennaebragai Miranda Ribeiro e U. longidorsalis Jégu, Tito de Morais & Santos, pela presença de 99 a 101 escamas perfuradas na linha lateral (vs. 69 a 83), presença de 17 a 19 espinhos pré-pélvicos (vs. 9-13 em U. sennaebragai e 28-31 em U. longidorsalis), 20 a 21 espinhos pós-pélvicos (vs. 15 a 19 em U. sennaebragai e 14 em U. longidorsalis), e 23-25 escamas da série circumpeduncular (vs. 30-48 em U. sennaebragai e 33-35 em U. longidorsalis). Em adição, a espécie nova difere de U. longidorsalis por possuir a largura interdosal e comprimento da base da nadadeira adiposa maiores (11,8-15,6 vs. 7,1-7,9% do CP e 4,2-5,8 vs. 3,7-3,8% do CP, respectivamente).

The restocking programs are being used more frequently as methods of fish conservation. However, the reduction in the genetic diversity can affect survival of juveniles used in this programs and cause effects on the wild populations. The aim of this study was assess the genetic diversity in three broodstocks (BA, BB and BC) of pacu Piaractus mesopotamicus of a Hydropower plant in São Paulo - Brazil, using in restocking program of Tietê River. Nine RAPD primers were amplified used extracted DNA from 89 fin-clipping samples. Sixty-nine fragments were polymorphic, 15 had frequencies with significant differences (P<0.05), seven were excluded, and six were fixed fragments. High values for polymorphic fragments (47.83% to 71.01%) and Shannon index (0.270 to 0.424) were observed. Most of the genetic variation was found within the groups through the AMOVA analysis, which was confirmed by the results of the identity and genetic distance. Ancestry levels (FST) among the groups values indicated little and moderate genetic differentiation. The estimate of number of migrants by generation (Nm) indicated levels of gene flow. Moderate genetic divergence between groups (0.214 to 0.259) was observed. The results indicate high (BA and BB) and moderate (BC) variability within broodstocks and genetic differentiation among them. The fish stocks analyzed represent a genetic base that will allow the fish technicians to release juveniles without genetic risks to wild populations present in the river. These genetics procedures can be used as models for other migratory species.


Os programas de repovoamento estão sendo usados com mais frequência como métodos de conservação de peixes. No entanto, a redução na diversidade genética pode afetar a sobrevivência dos juvenis usados nesses programas e causar efeitos sobre as populações selvagens. O objetivo deste estudo foi avaliar a diversidade genética de três estoques de reprodutores (BA, BB e BC) de pacu Piaractus mesopotamicus de uma hidrelétrica em São Paulo - Brasil, do programa de repovoamento do rio Tietê. Nove iniciadores RAPD amplificaram-se usando DNA extraído de 89 amostras de nadadeira. Sessenta e nove fragmentos foram polimórficos, 15 tiveram frequências com diferenças significativas (P<0.05), sete foram excluídos e seis foram fragmentos limitantes. Observaram-se altos valores de fragmentos polimórficos (47,83% a 71,01%) e de índice de Shannon. A maior variação genética observou-se dentro dos grupos através da análise de AMOVA, sendo confirmado com os resultados de identidade e distância genética. Os níveis de FST entre os grupos indicaram pequena e moderada diferenciação genética. O número de migrantes por geração (Nm) indicou níveis de fluxo gênico. Observou-se moderada divergência genética entre grupos (0,214 a 0,259). Os resultados indicaram alta (BA e BB) e moderada (BC) variabilidade dentro dos estoques e diferenciação genética entre eles. Os estoques de peixes analisados representam uma base genética que permitirá aos produtores liberar juvenis sem riscos genéticos para as populações naturais presentes no rio. Esses procedimentos genéticos podem ser utilizados como modelos para outras espécies migradoras.

ABSTRACT

Family Trichodinidae comprises ciliate protozoa distributed worldwide; they are considered some of the main parasitological agents infecting cultivated fish. However, the trichodinidae parasitizing important fish species cultured in Brazil are unknown, and more taxonomic studies on this group of parasites are required. This research morphologically characterizes Trichodina colisae Asmat & Sultana, (2005) of pacu (Piaractus mesopotamicus) and patinga hybrid (P. mesopotamicus × P. brachypomus) cultivated in the central and southeast regions of the country. Fresh assemblies were made from mucus scraped from the skin, fins and gills, fixed with methanol and, subsequently, impregnated with silver nitrate and stained with Giemsa for assessment under light microscopy. This research reports not only the second occurrence of T. colisae in the world, but also its first occurrence in South America.(AU)


Tricodinídeos são protozoários ciliados móveis com ampla distribuição mundial; são considerados um dos agentes parasitários que mais acometem peixes cultivados. No Brasil, a maioria dos tricodinídeos que parasitam importantes espécies de peixes cultivados são desconhecidos, o que requer mais estudos taxonômicos com esse grupo de parasitos. Este estudo caracteriza morfologicamente Trichodina colisae Asmat & Sultana, 2005 de pacu (Piaractus mesopotamicus) e do híbrido patinga (P. mesopotamicus × P. brachypomus) cultivados, respectivamente, no Centro-Oeste e Sudeste do Brasil. Foram feitas montagens a fresco do raspado de muco da pele, nadadeiras e brânquias, fixados com metanol e, posteriormente, impregnados com nitrato de prata e coradas com Giemsa para avaliação em microscopia óptica. O presente estudo relata não só a segunda ocorrência de T. colisae no mundo, mas também a primeira ocorrência na América do Sul.(AU)

ABSTRACT

Samples of different organs from intensively-reared Piaractus mesopotamicus were collected and processed using routine histological techniques in order to produce thin sections for staining with hematoxylin-eosin and with the Ziehl-Neelsen method. Through examination under an optical microscope, myxosporidians of the genera Henneguya sp. and Myxobolus sp. were identified, respectivelyin the gills and kidneys of P. mesopotamicus. Plasmodia with immature spores of Henneguya sp. were located along the secondary lamellae, with total length of 30.45±4.84µm and width of 3.52±0.33µm. Spores of Myxobolus sp. were located in the kidneys, with total length of 8.94±0.82µm and width of 5.59±0.39µm. Histopathological analysis of the gills showed plasmodia containing spores of Henneguya sp., at intralamellar and intravascular localities, at different stages of development. Spores of Myxobolus sp. were identified in the kidneys, in the peritubular region and in the interstices and glomerulus, surrounded by melanomacrophages. Focal hemorrhage was recorded in a few cases. Ziehl-Neelsen staining allowed to identify particular features of the spores and facilitated biometry and enabled classification in comparison with hematoxylin-eosin, thus demonstrating its usefulness for histopathological diagnosis of the parasitosis.


Amostras de diferentes órgãos de Piaractus mesopotamicus mantidos em criação intensiva foram coletadas e processadas mediante as técnicas histológicas usuais para obtenção de cortes que foram corados com hematoxilina-eosina e pelo método de Ziehl-Neelsen. Ao exame em microscopia de luz foi possível identificar mixosporídeos dos gêneros Henneguya sp. e Myxobolus sp. em brânquia e rim de P. mesopotamicus respectivamente. Plasmódios com esporos imaturos de Henneguya sp. foram localizados ao longo das lamelas secundárias e mensurados (comprimento total 30,45±4,84µm e largura 3,52±0,33µm) e no rim esporos de Myxobolus sp. (comprimento total 8,94±0,82µm e largura 5,59±0,39µm). Na análise histopatológica das brânquias observaram-se plasmódios contendo esporos de Henneguya sp., com localização intralamelar e intravascular, em diferentes estágios de desenvolvimento. No rim identificaram-se esporos de Myxobolus sp., na região peritubular e no interstício e glomérulo, circundados por melanomacrófagos. Em poucos casos foi registrada hemorragia focal. O uso da coloração de Ziehl-Neelsen permitiu identificar particularidades dos esporos, facilitou sua biometria e classificação em comparação com a hematoxilina-eosina, demonstrando sua utilidade no diagnóstico histopatológico da referida parasitose.

Weigh gain, length, and survival of Pacu (Piaractus brachypomus) larvae were analyzed after they were fed one of four diets. At 36 hours post-hatching larvae were fed for the first time and during three consecutive days: Artemia salina nauplii (T1), wild plankton filtered to 200 microns (T2) , concentrated powder with 48% crude protein (T3) or no diet (fasting, T4). Animals were kept at a density of 70 larvae per liter. Statistically significant differences were observed with respect to the final weight for T1, T2, T3, and T4 (2.2 ± 0.3a, 1.7 ± 0.3b, 1.5 ± 0.3bc, 1.5 ± 0.2c mg, respectively) , final length (6.37 ± 0.29a, 0.20b ± 6.16, 5.95 ± 0.27c, 0.26c ± 5.87 mm, respectively), weight gain (0.77 ± 0.39a, 0.31b ± 0.34, 0.16 ± 0.37bc, 0.08 ± 0.21 c mg, respectively) and length gain (0.39 ± 0.17A, 0.18 ± 0.15b, 0.16c ± -0023, -0102 ± 0.15c mm, respectively). The T1 diet had the best results for optimal growth, followed by T2. The survival rate was not statistically different among treatments. These data suggest that the initial diet composition can affect subsequent growth characteristics of Pacu larvae.


Se evaluó la ganancia de peso, la longitud y la supervivencia de larvas de cachama blanca (Piaractus brachypomus) alimentadas con varias dietas. A las 36 horas post-eclosión las larvas se alimentaron por primera vez y durante tres días consecutivos con nauplios de Artemia salina (T1), plancton silvestre filtrado a 200 μm (T2), concentrado pulverizado con 48% de proteína bruta (T3), o ayuno (T4). Los animales se mantuvieron a una densidad de 70 larvas por litro. Se observaron diferencias estadísticas significativas (p<0.05) con respecto al peso final para los tratamientos T1, T2, T3, y T4 (2.2 ± 0.3a; 1.7 ± 0.3b; 1.5 ± 0.3bc; 1.5 ± 0.2c mg, respectivamente), longitud final (6.37 ± 0.29a; 6.16 ± 0.20b; 5.95 ± 0.27c; 5.87 ± 0.26c mm, respectivamente), ganancia de peso (0.77 ± 0.39a; 0.34 ± 0.31b; 0.16 ± 0.37bc; 0.08 ± 0.21c mg, respectivamente) y ganancia de longitud (0.39 ± 0.17a; 0.18 ± 0.15b; -0.023 ± 0.16c; -0.102 ± 0.15c mm, respectivamente). El T1 presentó los mejores resultados, seguido por T2. El porcentaje de sobrevivencia no tuvo diferencia estadística significativa entre tratamientos.


Foram utilizadas larvas de pirapitinga (Piaractus brachypomus) foram mantidas em densidade de 70 larvas/L. Por três dias consecutivos, as larvas eram alimentadas com náuplios de Artemia salina (T1), plâncton selvagem filtrado (200 μm) (T2), ração em pó com 48% de PB (T3) e o tratamento controle era mantido em jejum (T4). Os resultados mostraram diferença significativa (p <0.05) no peso final (2.2 ± 0.3 a; 1.7 ± 0.3 b; 1.5 ± 0.3 bc; 1.5 ± 0. 2c mg.), no comprimento final (6.37 ± 0.29 a; 6.16 ± 0.20 b; 5.95 ± 0.27 c; c 5.87 ± 0.26 mm.), no ganho de peso (0.77 ± Um 0.39 a; 0.34 ± 0.31 b; 0.16 ± 0.37 bc; c 0.08 ± 0.21c mg.) e no ganho de comprimento (0.39 ± 0.17 a; 0.18 ± 0.15 b; -0.023 ± 0.16 c; -0.102 ± 0.15c mm). Os tratamentos T1 e T2 mostraram os melhores resultados em relação aos parâmetros anteriormente citados. Contudo, nenhum tratamento mostrou diferença significativa na porcentagem de sobrevivência.
