ABSTRACT

Abstract Piper nigrum (black pepper) is used in Indian traditional medicine and its main alkaloid, Piperine (PIP), presents antioxidant, antitumor and neuroprotective pharmacological properties. This substance is insoluble in aqueous media and can irritate the gastrointestinal tract. Aiming to avoid these inconvenient characteristics and enable PIP oral administration, this study suggested the PIP microencapsulation through the emulsion-solvent evaporation method and the preparation of microparticulated tablets by direct compression. An UV-spectroscopy method was validated to quantify PIP. Microparticles and microparticulated tablets were successfully obtained and the microparticles exhibited excellent flow. The scanning electron microscopy images showed that PIP microparticles were intact after compression. The in vitro release showed a controlled release of PIP from microparticles and PIP microparticles from tablets in comparison to PIP and PIP tablets. The release profiles of PIP microparticles and the microparticulated tablets were similar. Therefore, tablets containing PIP microparticles are promising multiparticulated dosage forms because a tablet allows microparticles administration and the intact ones promote a controlled release, decreasing its irritating potential on the mucosa.

ABSTRACT

A study was conducted to evaluate the effect of Piper nigrum (black pepper) leaf extract on on the growth performance, proximate composition, hematological parameters, and immune response of Labeo rohita fingerlings with an average weight of 22.14 ± 0.98g. After acclimation for two weeks, fish (n=25) were randomly selected and placed in four glass aquaria (T0, T1, T2 and T3) at constant water temperature (30.0 ± 1.0 °C), pH (7.50 ± 0.5) and total hardness (200 ± 2.0 mgL,-¹) for a period of 12 weeks, with three replicates each. Fish were fed with P. nigrum leaf extract supplemented feed @ 0.0%, 1.0%, 2.0% and 3.0% in T0, T1, T2 and T3, respectively. At the end of experiment, five fish were randomly selected from each aquaria for proximate composition, gut and skin microbial load, hematological parameters. Total proteins, albumins, and globulins were also recorded to evaluate immunological memory. The result revealed that fish in T2 showed better growth performance with an average weight gain of 56.11 ± 0.51 g. Thus, it had been concluded that Piper nigrum, a medicinal plant, can also be used to enhance the growth performance and immune response of Labeo rohita as attractive alternatives against antibiotics and vaccines and has shown no negative side effects on fish health as well as on its environment.


Um estudo foi conduzido para avaliar o efeito do extrato da folha de Piper nigrum (pimenta-do-reino) sobre o desempenho de crescimento, composição centesimal, parâmetros hematológicos e resposta imune de alevinos de Labeo rohita com peso médio de 22,14 ± 0,98g. ABSTRACT

Introduction: Carbohydrate and lipid digestive enzymes are instrumental in the absorbability of nutrients associated to diabetes and obesity. This study evaluated hydroethanolic extracts of Piper nigrum leaf and Morinda lucida stem bark for antioxidant capacity and enzymes (carbohydrate and lipid digestive) inhibition. Methods: Colorimetric assays determined enzyme (?-amylase, ?-glucosidase, lipase and cholesterol esterase) inhibition and antioxidant capacity (total phenolic (TPC) and flavonoid (TFC) content, radical scavenging activity (DPPH, ABTS), and ferric reducing antioxidant power (FRAP)) of hydroethanolic ethanolic extracts, ethyl acetate and hexane fractions. Results: At 1 mg/ml extracts of P nigrum and M lucida inhibited ?- amylase (9.82±1.05 - 36.63±0.69 %) and ?-glucosidase (22.47±0.34 - 67.77±0.58 %) activities. At 100 µg/ml extracts and fractions inhibited lipase (56.72±1.11 - 81.61±0.71 %) and cholesterol esterase (18.14 ±0.79 - 36.84±0.70 %) activities. IC50 for ?- amylase (2.20±0.02 - 7.8±1.42 mg/ml), ?-glucosidase (0.16±0.01 - 3.74±0.01 mg/ml), lipase (8.58±2.57 - 53.03±5.20 µg/ml) and cholesterol esterase (172.20±5.12 - 419.80±4.55 µg/ml) were registered. At 4 mg/ml, P. nigrum presented a higher TPC (153.78±8.31 - 354.63±6.33 mg/ml), TFC (21.65±1.14 -33.86±0.00 mg/ml) than M lucida TPC (10.21±0.11 - 169.89±6.54 mg/ml), TFC (ND - 87.32±6.14 mg/ml). P nigrum presented radical scavenging (DPPH and ABTS) activity with IC50 0.12±0.00 - 1.27±0.01 mg/ml compared to 1.31±0.02 - 3.44±0.12 mg/ml of M lucida. The FRAP IC50 values were better for P nigrum (3.38±0.14- 4.48±1.05 mg/ml) than M lucida (3.34±1.32 - 15.4±2.03 mg/ml). Conclusion: P nigrum presented better antioxidant capacity and more effective on lipid digestive enzymes while M lucida was more effective on carbohydrate digestive enzymes.

Aims@#Piper nigrum L. (black pepper) is an economically important commodity plant in Malaysia, which generated RM 200.95 million from pepper export in the year of 2018. However, the increase in pepper production is restricted by diseases. Fusarium wilt is one of the major diseases of P. nigrum L. The objectives for this study were to isolate Fusarium spp. associated with Fusarium wilt of P. nigrum L. from selected pepper farms in the northwestern region of Sarawak and to characterize the Fusarium spp. isolated morphologically and molecularly.@*Methodology and results@#Fusarium spp. were isolated from diseased root samples. The pathogen was grown on potato dextrose agar (PDA) under dark condition at circa (ca.) 25 °C for morphological characterisation. Molecular characterisation was done by using internal transcribed spacer (ITS). Phylogenetic tree was constructed to study the genetic relationship of the isolates. Fusarium solani, F. oxysporum, F. proliferatum were the three Fusarium species identified. There were variations in morphological characters observed between and among the species, including the colony form, margin, elevation, surface appearance and pigmentation. No distinctive morphological characteristic was specific to a location. In addition, growth rate, macroconidia sporulation rate, and microconidia sporulation rate of the isolates were not correlated. In molecular phylogeny, the three Fusarium species were separated into three distinct clades representing the three identified species. The genetic relatedness between isolates within each species was depicted in the tree. @*Conclusion, significance and impact of study@#Variations were observed among isolates in this study based on morphological and molecular characterization. This study would contribute information on the variations of Fusarium spp. associated with Fusarium wilt of P. nigrum L. from the northwestern region of Sarawak.

ABSTRACT

Objective: To evaluate anti-obesity effect of aqueous P. nigrum extract in a murine model of high-fat diet (HFD) induced obesity. Methods: Male Wistar rats were fed with a high-fat diet (HFD) (20g/day/rat) for a period of 50 d to induce obesity. Aqueous P. nigrum extract (20 mg/kg) administered orally to high-fat diet (HFD) fed rats from 8th day to 50th day (total 42 d). The parameters like gain in body weight, serum lipids, insulin and leptin were measured. Results: The rats treated with extract showed a significant reduction in body weight gain, serum insulin, leptin, lipids as compared to rats fed with only a high-fat diet (HFD). In addition, the extract-treated group showed a considerable rise in high-density lipoprotein (HDL-C) level (29.61±7.68 mg/dl) as compared to the control group (23.23±9.69 mg/dl). Conclusion: The results indicate that aqueous P. nigrum extract possess the potential to reduce obesity markers in a high-fat diet (HFD) fed rats.

ABSTRACT

Piper nigrum L. (Piperaceae), which is a well-known food seasoning, has been used as a traditional medicine for the treatment of vomiting, abdominal pain, diarrhea and anorexia in Korea, China and Japan. Methanol extract from the fruit of P. nigrum was successively partitioned as n-hexane, methylene chloride, ethyl acetate, n-butanol and H₂O soluble fractions. Among those fractions the ethyl acetate soluble fraction showed the most potent DPPH radical scavenging activity, and piperine was isolated from the ethyl acetate fraction. To know the antioxidant activity of piperine, we tested the activities of superoxide dismutase (SOD) and catalase together with oxidative stress tolerance and intracellular ROS level in Caenorhabditis elegans. To investigate whether piperine-mediated increased stress tolerance was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain including CF1553. Consequently, piperine enhanced SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose–dependent manner. Moreover, piperine-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

ABSTRACT

Spices are natural plant products, have been used not only as flavoring and coloring agents, but also as food preservatives and folk medicines throughout the world for thousands of years. Many spices also have been recognized by having both digestive stimulant and carminative actions and also antimicrobial, anti-inflammatory, antimutagenic and anti-carcinogenic potential. Antioxidant and genotoxic potential of species commonly used in Brazil was evaluated. The antioxidant activity was evaluated using different methods, including DPPH radical scavenging activity, ferric reducing power (FRAP), iron ion chelating power, inhibition of lipid peroxidation (TBARS), NO radical scavenging, and oxidative hemolysis inhibition. Furthermore, the antigenotoxic activity was evaluated through mitotic index and chromosome aberration in Allium cepa roots. Quantification of total phenols and flavonoids carried out. The results with the Ocimum basilicum spices in the DPPH test showed activity (82.01%), FRAP (321.12 uM ET and iron chelating activity (94.18) and for the Cinnamomum zeylanicum spice in the TBARS test (18.52%) evaluated by different methods and mechanisms of inactivation of free radicals and according to the evaluation of genotoxicity by the Allium cepa test the spices do not present genotoxic effects.


As especiarias são produtos vegetais naturais, que foram utilizados não só como agentes aromatizantes e colorantes, mas também como conservantes de alimentos e medicamentos populares em todo o mundo há milhares de anos. Muitas especiarias também foram reconhecidas por ter estimulantes digestivos e ações carminativas e também potencial antimicrobiano, anti-inflamatório, antimutagênico e anticarcinogênico. O potencial antioxidante e genotóxico das espécies comumente utilizadas no Brasil foi avaliado. A atividade antioxidante foi avaliada utilizando diferentes métodos, incluindo a atividade de eliminação de radicais DPPH, poder de redução férrica (FRAP), poder quelante de íons de ferro, inibição da peroxidação lipídica (TBARS), eliminação de radicais NO e inibição da hemólise oxidativa. Além disso, a atividade antigenotóxica foi avaliada através do índice mitótico e aberração cromossômica nas raízes do Allium cepa. Quantificação de fenóis totais e flavonoides realizados. Os resultados mostraram que as especiarias (Cinnamomum zeylanicum, Origanum vulgare, Piper nigrum, Zingiber officinale e Ocimum basilicum) apresentaram atividade antioxidante avaliada por diferentes métodos e mecanismos de inativação de radicais livres e de acordo com a avaliação de genotoxicidade pelo teste Allium cepa as especiarias não apresentam efeitos genotóxicos.

ABSTRACT

OBJECTIVE:To establish a method for the detection of volatile constituents from the leaves and fruits of Piper ni-grum. METHODS:HS-SPME-GC-MS was used. The chromatographic conditions:column was HP-5 MS quartz elastic capillaries, carrier gas was high purity helium(99.999%),flow rate was 1.0 mL/min,the inlet temperature was 250 ℃,initial temperature of column was 50 ℃(temperature programmed),split injection with split ratio of 10:1. MS conditions:ionization mode was electron impact ion source,ionization energy was 80 eV,ion source temperature was 230 ℃,quadrupole temperature was 150 ℃,trans-mission line temperature was 280 ℃,electron multiplier voltage was 1588 V,mass scanning range was m/z 30-400. The spectra were retrieved using RTLPEST3. L and NIST08. L,and the relative contents of the volatile constituents were determined by area normalization method. RESULTS:There were 28 volatile constituents in the leaves and 15 in the fruits,respectively accounting for 67.13% and 36.85%. The major volatile constituents of leaves were β-caryophyllene (15.72%),limonene (9.39%),3-carene (9.32%),β-pinene(6.80%),α-terpine(4.98%),etc.,the main volatile constituents of fruits were 1,7,7-trimethyl-2-vinylbicyclo [2.2.1]hept-2-ene(10.45%),espatulenol(8.28%),caryophyllene oxide(4.81%),etc. 5 constituents were owned in both. CON-CLUSIONS:The study basically clears the main volatile constituents from the leaves and fruits of P. nigrum,and verifies existing obvious differences.

ABSTRACT

Piper colubrinum Link., a distant relative of Piper nigrum L., is immune to the oomycete pathogen Phytophthora capsici Leonian that causes ‘quick wilt’ in cultivated black pepper (P. nigrum). The osmotin, PR5 gene homologue, earlier identified from P. colubrinum, showed significant overexpression in response to pathogen and defense signalling molecules. The present study focuses on the functional validation of P. colubrinum osmotin (PcOSM) by virus induced gene silencing (VIGS) using Tobacco Rattle Virus (TRV)-based vector. P. colubrinum plants maintained under controlled growth conditions in a growth chamber were infiltrated with Agrobacterium carrying TRV empty vector (control) and TRV vector carrying PcOSM. Three weeks post infiltration, viral movement was confirmed in newly emerged leaves of infiltrated plants by RT-PCR using TRV RNA1 and TRV RNA2 primers. Semi-quantitative RT-PCR confirmed significant down-regulation of PcOSM gene in TRV-PcOSM infiltrated plant compared with the control plants. The control and silenced plants were challenged with Phytophthora capsici which demonstrated that knock-down of PcOSM in P. colubrinum leads to increased fungal mycelial growth in silenced plants compared to control plants, which was accompanied by decreased accumulation of H2O2 as indicated by 3,3'-diaminobenzidine (DAB) staining. Thus, in this study, we demonstrated that Piper colubrinum osmotin gene is required for resisting P. capsici infection and has possible role in hypersensitive cell death response and oxidative burst signaling during infection.

ABSTRACT

Objective: To carry out the physicochemical and phytochemical standardization with high performance thin layer chromatography fingerprinting of Piper nigrum L. (P. nigrum) fruits in order to ascertain the standard pharmacognostical parameters of this king of spices. Methods: Many standardization parameters like extractive values, total ash value, water soluble ash value and acid insoluble ash, moisture content, loss on drying and pH values of P. nigrum L. fruits were analyzed. The method of Harborne was adopted for the preliminary phytochemicals screening. Analysis of total phenolic and flavonoid contents, pesticides residues, aflatoxin and heavy metals were also performed. CAMAG-high performance thin layer chromatography system was used for fingerprinting of methanolic extract of P. nigrum L. fruits. Results: The results of phytochemicals testing indicated the presence of carbohydrates, phenolic compounds, flavonoids, alkaloids, proteins, saponins, lipids, sterols and tannins in various solvent extracts. Total phenolic and flavonoid contents in methanolic extract were found to be 1.728 1 mg/g and 1.087 μg/g, respectively. Heavy metals concentrations were found to be within standard limits. Aflatoxins and pesticides residues were absent. Conclusions: The outcome of this study might prove beneficial in herbal industries for identification, purification and standardization of P. nigrum L. fruits.

ABSTRACT

Microbial colonization as biofilm is one of the reasons for the emergence of drug resistant strains. In the oral cavity, drug resistant strains limit the efficacy of oral hygiene practices. Enterococcus faecalis and Staphylococcus aureus have been reported as drug resistant bacteria and producing oral biofilms in oral cavity. In this study we demonstrate the efficacy of aqueous extract of Azadirachta indica, Mangifera indica, Piper betel and Pepper nigrum for antibiofilm activity against E. faecalis and S. aureus. The aqueous extracts were obtained by cold percolation method. The antibiofilm activity of plants extract was evaluated at 30, 15 and 7.5 mg/ml concentration. The percentage yield of extract was maximum in P. nigrum. The aqueous extract of A. indica significantly suppressed E. faecalis and S. aureus biofilm at 7.5 mg/ml at p<0.01 and p<0.001 significance level. P. betel significantly (p<0.001) disintegrated the E. faecalis biofilm at 30 mg/ml and S. aureus at 15 mg/ml (p<0.01). P. nigrum disintegrated E. faecalis and S. aureus biofilm significantly (p<0.05 and p<0.001) at 30 and 15 mg/ml respectively. M. indica significantly (p<0.05) suppressed S. aureus biofilm at 30 mg/ml. These results clearly demonstrate the antibiofilm activity of plants extract against oral pathogens.

Black pepper, the king of spices, the most important and widely used spice in the world is the dried mature berries of P. nigrum L. Inorganic nitrogen is assimilated into amino acids glutamine, glutamate, asparagines and aspartate which serve as important nitrogen carriers in plants. The enzymes glutamine synthetase (GS) EC. 6.3.1.2, glutamate synthase (GOGAT) E.C. 1.4.1.14, glutamate dehydrogenase (GDH) E.C.1.4.1.4, aspartate aminotransferase and asparagine synthetase are responsible for biosynthesis of these amino acids. Although extensive studies have been carried out on the epidemiology of the diseases and agronomical aspects of P. nigrum, very few studies have been made on the enzymological aspects especially that associated with the nitrogen assimilation. A better understanding of the various properties of the enzyme will be valuable in the future studies concerned with Nitrogen metabolism in P. nigrum. Main objective of present study was to standardize the extraction and study the properties of GDH from P. nigrum L. Effect of additives and its optimum concentration were determined, standardized homogenizing medium was formulated, standardized assay system formulated, different properties like pH optimum, Substrate saturation and km value determined; it’s all in agreement with the previously reported values, result indicate NADP+/NADPH is the better substrate i.e. the enzyme activity is contributed by chloroplastic form of GDH than by the mitochondrial form.

OBJECTIVE@#To evaluate and compare the analgesic and anti-inflammatory activity of pure compound, piperine along with hexane and ethanol extracts of Piper nigrum L. fruit in mice and rats.@*METHODS@#The analgesic activity was determined by tail immersion method, analgesy-meter, hot plate and acetic acid induced writhing test. While the anti-inflammatory activity was evaluated by carrageenan-induced paw inflammation in rats.@*RESULTS@#Piperine at a dose of 5 mg/kg and ethanol extract at a dose of 15 mg/kg after 120 min and hexane extract at a dose of 10 mg/kg after 60 min exhibited significant (P<0.05) analgesic activity by tail immersion method, in comparison to ethanol extract at a dose of 10 mg/kg using analgesy-meter in rats. However, with hotplate method, piperine produced significant (P<0.05) analgesic activity at lower doses (5 and 10 mg/kg) after 120 min. A similar analgesic activity was noted with hexane extract at 15 mg/kg. However, in writhing test, ethanol extract significantly (P<0.05) stopped the number of writhes at a dose of 15 mg/kg, while piperine at a dose of 10 mg/kg completely terminated the writhes in mice. In the evaluation of anti-inflammatory effect using plethysmometer, piperine at doses of 10 and 15 mg/kg started producing anti-inflammatory effect after 30 min, which lasted till 60 min, whereas hexane and ethanol extracts also produced a similar activity at a slightly low dose (10 mg/kg) but lasted for 120 min.@*CONCLUSIONS@#It is concluded from the present study that Piper nigrum L possesses potent analgesic and anti-inflammatory activities.

ABSTRACT

Aim: Identification of anti-Mycobacterium tuberculosis agents of plant origin, against sensitive and multidrug resistant (MDR) strains. Study Design: Assessing anti-M. tuberculosis activity of five Indian medicinal plants, which have been reported in traditional literature for various uses including respiratory ailments. Place and Duration of Study: Mumbai, India; May 2009 – December 2011. Methodology of Study: The reference strain (H37Rv), three susceptible and three MDR clinical isolates of M. tuberculosis were used. Acetone, ethanol and aqueous extracts (prepared sequentially) of Acorus calamus L. (rhizome), Andrographis paniculata Nees. (leaf), Ocimum sanctum L. (leaf), Piper nigrum L. (seed) and Pueraria tuberosa DC. (tuber) were tested at 1, 10 and 100 μg/ml using the Microplate Alamar Blue Assay. The active extracts were assessed for cytotoxicity on the human lung epithelial cell line (A549) using the neutral red assay and a phytochemical analysis was made using High Performance Thin Layer Chromatography (HPTLC). Results: Among the plants tested, the acetone extract of P. nigrum appears promising. It was effective against H37Rv, all susceptible isolates and one MDR isolate at 100 μg/ml. The ethanol extract caused some inhibition of growth, though less than the cut-off of 99%. A combination of acetone and ethanol extracts at 50 μg/ml each was effective against all isolates tested. The known active phytoconstituent of P. nigrum, piperine (also an efflux pump inhibitor), was effective against H37Rv in the presence of suboptimal concentration of Rifampicin, but not against the clinical isolates tested. Presence of piperine in the acetone and ethanol extracts was confirmed by HPTLC. Extracts of P. nigrum and piperine were not cytotoxic to the A549 cell line. Conclusion: Amongst the five plants tested, P. nigrum was active. The acetone extract may have active components in addition to piperine. It is possible that the class and expression of efflux pumps in H37Rv is different from that in the clinical isolates, and hence piperine did not inhibit these isolates. Thus, it is necessary to screen clinical isolates in addition to reference strains. The observation of the increased efficacy of the combination of acetone and ethanol extracts is interesting.

ABSTRACT

Amongst other botanical sources, Annona muricata L., Annonaceae, seeds and Piper nigrum L., Piperaceae, fruits are particularly enriched with acetogenins and piperine-related amides, respectively. These crude ethanolic extracts are potent Aedes aegypti bioactives that can kill Aedes aegypti larvae (dengue fever mosquito). A. muricata displayed a 93.48 µg/mL LC50 and P. nigrum an 1.84 µg/mL LC50. An uncommon pharmacognostical/toxicological approach was used, namely different combinations of both extracts to achieve an improved lethal effect on the larvae. The independence test (χ²) was utilized to evaluate the combination of the two crude extracts. All of the tested combinations behaved synergistically and these novel results were attributed to the completely different biochemical mechanisms of the differentiated chemical substances that were present in the two botanical sources. Besides the two above selected plants, Melia azedarach L., Meliaceae, Origanum vulgare L., Lamiaceae, and Ilex paraguariensis A. St.-Hil., Aquifoliaceae, in order of decreasing toxicity, may also be sought as potential extracts for the sake of synergic combinations.

This review based on the biological role of Piper nigrum can provide that the peppercorn or other parts can be used as crude drug for various diseases while the secondary metabolites such as piperine can be used for specific diseases.
