ABSTRACT

Two new sucrose cinnamates(1 and 2) along with nine known compounds(3-11) were isolated from ethanol extract of Polygonum lapathifolium var. salicifolium by silica gel column chromatography, ODS column chromatography and semi-preparative HPLC. Their structures were elucidated by extensive spectroscopic methods including 1 D-and 2 D-NMR experiments, as well as HR-ESI-MS analysis. Eleven compounds(7 sucrose cinnamates, 3 phenylpropanoids and 1 lactone) were obtained and their structures were identified as(1,3-O-di-p-coumaroyl)-β-D-fructofuranosyl-(2→1)-α-D-glucopyranoside(1),(1,3-O-di-p-coumaroyl)-β-D-fructofuranosyl-(2→1)-(6-O-acetyl)-α-D-glucopyranoside(2),(3-O-feruloyl)-β-D-fructofuranosyl-(2→1)-(6-O-p-coumaroyl)-α-D-glucopyranoside(3), hydropiperoside(4), vanicoside C(5),(1,3-O-di-p-coumaroyl)-β-D-fructofuranosyl-(2→1)-(6-O-feruloyl)-α-D-glucopyranoside(6), vanicoside B(7),trans-p-hydroxycinnamic acid methyl ester(8), trans-p-hydroxycinnamic acid ethyl ester(9), methyl ferulate(10) and dimethoxydimethylphthalide(11), respectively. Compounds 1 and 2 were two new sucrose cinnamates, and compounds 1-11 were isolated from this plant for the first time. The antioxidant activities of the isolated compounds 1-9 were investigated by an oxygen radical absorbance capacity(ORAC) assay, and all nine compounds were found to show strong antioxidant activities. Among them, compound 6(10 μmol·L~(-1)) was the supreme one in antioxidant activities, with its ORAC value equivalent to(1.60±0.05) times of 50 μmol·L~(-1) Trolox.

ABSTRACT

AIM To study the chemical constituents from Polygonum lapathifolium L..METHODS The ethyl acetate fraction of 80% ethanol extract of P.lapathifolium L.was isolated and purified by silica column,Sephadex LH-20 and ODS,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Fifteen compounds were isolated and identified as 2',4'-di-hydroxy-6'-mehtoxy-chaltone (1),2',6'-di-hydroxy-4'-methoxy-chaltone (2),2',3'-di-hydroxy-4',6'-di-methoxy-chaltone (3),kaempferol (4),quercetin (5),quercetin-3-O-α-L-rhamnoside (6),quercetin-3-O-α-L-rhamnoside (7),kaempferol-3-O-α-L-rhamnoside (8),kaempferol-3-O-β-D-glucoside (9),quercetin-3-O-β-D-galactoside (10),quercetin-3-O-β-D-glucoside (11),kaempferol-3-O-β-D-galactoside (12),quercetin-3-O-(2"-O-galloyl)-rhamnoside (13),quercetin-3-O-(2"-O-galloyl)-glucoside (14),quercetin-3-O-[(6"-O-transferuloyl)-β-D-galactoside] (15).CONCLUSION Compound 3-8 are isolated from this plant for the first time.

ABSTRACT

Polygonum lapathifolium is an invasive plant spread worldwide, which has been used in traditional medicine for its biological activities. We studied chemical profile of the aerial part using HPLC/DAD/ESI-MS. Eight flavonoids, two chalcones and gallic acid were isolated, identified nad tested for their activity in seven experimental models (for antioxidant, alpha/beta-glucosidase inhibitory, antimicrobial, anticholineserase activity). Polygonum lapathifolium es una planta invasora cuya presencia se extiende por todo el mundo y que ha sido utilizada en la medicina tradicional por sus actividades biológicas. Analizamos el perfil químico de la parte superficial utilizando HPLC/DAD/ESI-MS. Ocho flavonoides, dos chalconas y ácido gálico fueron aislados, identificados y analizados en relación a sus actividades en siete modelos experimentales (de actividad antioxidante, alfa/beta-glucosidasa inhibitoria, antimicrobiana, anticholineserasa). ABSTRACT

Aims: The crude methanolic flower extract of Polygonum lapathifolium (s.l.) (family: Polygonaceae) was to evaluate for its possible phytochemical constituents and selected pharmacological activities (anthelmintic and anti-emetic activity) growing in Bangladesh. Study Design: In vitro anthelmintic and in vivo antiemetic activities were evaluated by Pheretima posthuma model and chick emesis model respectively. Place and Duration of Study: Department of Pharmacy, Noakhali Science & Technology University, Noakhali, September, 2012 to December, 2012. Methodology: Phytochemical screening was done by characteristic color changes using standard procedures. In anthelmintic activity test, the parameters like: time of paralysis (vermifuge) was noted when no movement of any sort could be observed except when the worms were shaken vigorously. Time of death (vermicide) was determined by observing no movement when the worms were shaken vigorously or dipped in warm water (50ºC). The methanol extract was used at the concentration of 20, 40, 60, 80 and 100 mg/ml. Piperazine citrate (10 mg/ml) was used as standard reference. In anti-emetic test, sample extract was administered at a dose of 150 mg/kg orally and emesis was induced by copper sulphate (50 mg/kg orally). Metoclopramide (50 mg/kg .b.w intraperitoneally) was used as a standard drug. The antiemetic activity was determined by calculating the mean decrease in number of retching in contrast with those of control disorders. Results: The phytochemical investigation indicated the presence of alkaloids, phytosterols, diterpins, amino acids and proteins, flavonoids and phenolic compounds. The flower extract exhibited significant anthelmintic activity at a concentration of 40 mg/ml compared with standard reference. The flower extract also showed statistically significant antiemetic effect (85.42% inhibition) compared with reference drug which showed 81.25% antiemetic activity. Conclusion: So, the flower extract exhibited potential anthelmintic and anti-emetic properties, thus provide scientific basis for its use in folk medicine for the management of GI disorders. The plant may further be explored for its various pharmacological activities.

The phytochemical screening shows the presence of phytosterols, diterpens, amino acid & protein, alkaloid and flavonoids, those are responsible for antiviral, antibac-terial, antiallergic, antihypertensive, antiarrythmic, hepatoprotective, anti-inflammatory effects in mammals. In Anthelmintic activity test (using Pheretima posthuma model) five concentrations (20, 40, 60, 80 and 100 mg/ml in distilled water) of extracts were used which involved the determination of time of paralysis (vermifuge) and time of death (vermicidal activity) of the worms, where the concentration of 60, 80 &100 mg/ml showed a significant anthelmintic activity. Observations were comparable with the standard drug at concentration of 10mg/ml of piperi-zine citrate. On the other hand, in anti-emetic test, emesis was induced by the oral administration of copper sulphate 50mg/kg body weight to four days age of young chicks using chick emesis model. The antiemetic activity was determined by calculating the mean decrease in number of retching in contrast with those of control disorders. The extracts (150 mg /kg orally) showed statistically significant antiemetic effect (90.45% Inhibition) compared with reference drug metoclopramide (50mg/kg intraperitoneally) which showed 82.48% Inhibition. From these observa-tions, it was concluded that stems extracts have potential anthelmintic and anti-emetic properties. The plant may further be explored for its various pharmacological activities.

ABSTRACT

OBJECTIVE:To investigate the effect of the extracts of Polygonum lapathifolium on the activity of ?-glucosidase.METHODS:Soxhlet extraction was applied to extract overground part of P.lapathifolium,and the inhibition effect of extracts on ?-glucosidase was determined with 96-microplate-based method,and the inhibitory kinetic characteristics of methanol extract was investigated using Lineweaver-Burk method.RESULTS:The extracts from P.lapathifolium showed good inhibitory activity.The methanol extract had the highest inhibitory activity (IC50=2.13 ?g?mL-1)followed by ethyl acetate extract (IC50=3.826 ?g?mL-1),petroleum ether extract (IC50=230.86 ?g ? mL-1).And they were all higher than that of acarbose (IC50=1 103.01 ?g?mL-1) as positive control.Inhibitory kinetics test indicated that methanol extract was noncompetitive inhibitor.CONCLUSION:The extracts of P.lapathifolium have good inhibition effect on the activity of ?-glucosidase with a good prospect of development and utilization.
