ABSTRACT

Resumen Objetivo: La dispersión transmural de la repolarización ventricular (DTMRV) es un factor de riesgo para muerte en pacientes con síndrome isquémico coronario agudo (SICA). Con el objetivo de conocer el efecto de la realización de angioplastia sobre la DTMRV, se estudió la relación Tp-f/QTc en pacientes con SICA sometidos a angioplastia. Método: Se diseñó un estudio observacional, retrospectivo y descriptivo. Se incluyeron 150 pacientes (N = 150) con diagnóstico de SICA. Se valoró la relación Tp-f/QTc inicial y se evaluó su acortamiento posangioplastia. Como objetivo secundario, se comparó la asociación de dichos cambios en la relación Tp-f/QTc con la mortalidad cardiovascular y los eventos adversos cardiovasculares. Resultados: El promedio en la relación Tp-f/QTc inicial fue de 0.2529, mientras que posangioplastia fue de 0.2397. Por medio de prueba de rangos de Wilcoxon se evidenció un descenso significativo en la relación Tp-f/QTc posterior a la angioplastia, con un valor Z de −2.051 y una p < 0.04. En el análisis secundario se encontró que una Tp-f/QTc ≥ 0.29 posangioplastia es factor de riesgo para presentación de los siguientes eventos adversos: muerte intrahospitalaria (7.4 vs 0%; p < 0.003), nuevo SICA en seguimiento a 1 año (25.9 vs. 18.5%; p < 0.006) y reintervención en seguimiento a 1 año (29.6 vs. 15.0%; p < 0.002). Conclusiones: Existe un acortamiento significativo en la relación Tp-f/QTc posangioplastia en pacientes con SICA. Esta medida de la DTMRV puede servir como un predictor de muerte intrahospitalaria, eventos cardiovasculares y reintervención a 1 año en pacientes con SICA tratados con angioplastia.


Abstract Objective: Transmural Dispersion of Repolarization (TDR) is a Risk factor for Death in patients with Acute Coronary Syndrome (ACS). In order to know the effect of angioplasty on TDR, the Tp-e/QTc ratio was studied in patients with ACS undergoing angioplasty. Method: An observational, retrospective and descriptive study was designed. 150 patients (N = 150) with diagnosis of ACS were included. The initial Tp-e/QTc ratio was assessed and then its post-angioplasty shortening was evaluated. As a secondary objective, we compared the association of these Tp-e/QTc ratio changes with cardiovascular mortality and cardiovascular adverse events. Results: The average in the initial Tp-e/QTc ratio was 0.2529, while post-angioplasty was 0.2397. Through a Wilcoxon rage test, a significant decrease in the Tp-e/QTc ratio after angioplasty was observed, with a Z value of −2.051 and p < 0.04. In the secondary analysis, it was found that a Tp-e/QTc ≥ 0.29 post-angioplasty is a risk factor for presenting the following adverse events: in-hospital death (7.4 vs. 0%; p < 0.003), new ACS in 1-year follow-up (25.9 vs. 18.5%; p < 0.006), and reintervention in 1-year follow up (29.6 vs. 15%; p < 0.002). Conclusions: There is a significant shortening in the Tp-e/QTc ratio post-angioplasty in patients with ACS. This measure of TDR can serve as a predictor of in-hospital death, cardiovascular events and 1-year reintervention in patients with ACS treated initially by angioplasty.

ABSTRACT

Aim To compare high-content cell imaging system and other methods in detecting cell proliferation, including the traditional thymidine ( 3H-TdR) incorporation method, methyl thiazolyl tetrazolium ( MTT ) method and cell counting kit-8 (CCK-8) method. Methods The fibroblast-like synovial cells (FLS) were used as the study object to observe the sensitivity and stability of FLS proliferation in different methods by using the usual proliferative stimulant tumor necrosis factor-α ( TNF-α) and the known proliferation inhibitor methotrexate at different concentrations. Results The 3H-TdR method and high-content cell imaging could detect a significant inhibitory effect of 1 nmol ·L-1 MTX on FLS cell proliferation, MTT assay and CCK-8 method could detect the significant inhibitory effect of 10 nmol· L-1 MTX on FLS cell proliferation. 3H-TdR method was found to have a large degree of discretization in the data set, with a standard deviation of 32.61% ~61.36% , and the MTT method was 11.9% ~ 17.8% , the CCK-8 method was 17.15% ~32.88% , and the high-content cell imaging system method was 12.66% ~26.54%. Conclusion The method of high-content cell imaging system is more accurate and stable for detecting cell proliferation.

ABSTRACT

ABSTRACT: The aim of the present study was to evaluate the effect of different irrigation depths on the yield, water use efficiency (WUE), and yield response factor (Ky) of carrot (cv. 'Brasília') in the edaphoclimatic conditions of Baixada Fluminense, RJ, Brazil. Field trials were conducted in a Red-Yellow Argisol in the 2010-2011period. A randomized block design was used, with 5 treatments (depths) and 4 replicates. Depths were applied by drippers with different flow rates, and the irrigation was managed by time domain reflectometry (TDR) technique. The reference (ETo) and crop (ETc) evapotranspiration depths reached 286.3 and 264.1mm in 2010, and 336.0 and 329.9mm in 2011, respectively. The root yield varied from 30.4 to 68.9t ha-1 as a response to treatments without irrigation and 100% replacement of the soil water depth, respectively. Values for WUE in the carrot crop varied from 15 to 31kg m-3 and the mean Ky value was 0.82. The mean values for Kc were obtained in the initial (0.76), intermediate (1.02), and final (0.96) stages. Carrot crop was influenced by different water depths (treatments) applied, and the highest value for WUE was obtained for 63.4% of soil water replacement.


RESUMO: O objetivo deste estudo foi avaliar o efeito de diferentes lâminas de irrigação na produtividade, na eficiência de uso da água (EUA) e no coeficiente de resposta de produção (Ky) da cenoura (cv. 'Brasília') ao déficit hídrico do solo, nas condições edafoclimáticas da Baixada Fluminense-RJ. Ensaios de campo foram conduzidos em 2010 e 2011, em um Argissolo Vermelho-Amarelo, usando um delineamento em blocos casualizados, com 5 tratamentos (lâminas) e 4 repetições. As lâminas foram aplicadas por gotejadores com diferentes vazões e o manejo da irrigação foi realizado com a técnica da TDR. As lâminas de evapotranspiração de referência (ETo) e da cultura (ETc) totalizaram 286,3 e 264,1mm, em 2010, e 336,0 e 329,9mm, em 2011, respectivamente. A produtividade de raízes variou de 30,4 a 68,9t ha-1 como resposta aos tratamentos sem irrigação e com 100% de reposição da lâmina de água no solo, respectivamente. Os valores de EUA no cultivo da cenoura variaram de 15 a 31kg m-3 e o Ky médio foi de 0,82. Foram obtidos os valores médios de Kc nas fases inicial (0,76), intermediária (1,02) e final (0,96). A cultura da cenoura foi influenciada pelas diferentes lâminas aplicadas (tratamentos) e a maior EUA foi obtida para o tratamento com reposição de 63,4% da água do solo.

Objective To investigate the pattern of load transfer in cervical spine treated with Hybrid surgery using total disc replacement (TDR) and spinal fusion, so as to deepen the understanding of Hybrid surgery from the biomechanical view. Methods A finite element model of cervical spine C3-7 (INTACT model) was built to simulate three types of fusion surgeries at C4-6 degenerative segments: upper TDR combined with lower bone graft fusion (TDR45 model), upper fusion combined with lower TDR (TDR56 model), two-level fusion (Fusion456 model). Results In all surgical models, mobility of the fused levels was almost lost, while mobility of the TDR levels increased. Under the axial load of 160 N, the entire cervical motion was less than 4° in the INTACT model, while the motion in the TDR45 model and TDR56 model increased to 8.2° and 8.9°, respectively. In the TDR56 model, the force transferred through the C5 vertebra decreased by 20%, while the force transferred through the facet joint force was 3.8 times larger than that of the INTACT model. The facet contact force in the TDR45 model increased by 50%. The maximal stress in the INTACT model was 0.8 MPa, while the facet contact force in the TDR45 model and TDR56 model were almost 2 times as that in the INTACT model. Conclusions Due to the increased mobility at the TDR levels, the cervical curvature after Hybrid surgery changes greatly under the axial load. The alteration of spinal alignment will result in a decrease in anterior vertebral section force at the operative level, as well as an increase in facet joint force and facet cartilage stress.

ABSTRACT

Objective To analyze biomechanical properties of cervical spine after anterior cervical discectomy and fusion (ACDF) and total disc replacement (TDR) surgery. Methods Twelve cadaveric cervical spines (C2-T1) were adopted, and the motion and load distributions of the cervical segments under intact state and after ACDF and TDR surgery were tested using a three-dimensional (3D) optoelectronics measurement system. All the tests were carried out with displacement control in directions of flexion (Flex), extension (Ext), left bending (LB), right bending (RB), left rotation (LR) and right rotation (RR). Motion characteristics of the normal cervical spine and the implant were also discussed. Results In TDR-treated specimens, range of motion (ROM) was well preserved and could restore to the normal ROM distributions, especially in Flex/Ext and LR/RR direction. While in ACDF-treated specimens, ROM presented a large decrease as much as to 73.41% under the same condition compared with TDR, and ROM distributions were also changed obviously in other motions for the segments. Significant changes of ROM in LB/RB direction occurred in both TDR and ACDF group, which were up to 45.92% and 108.06%, respectively. The experimental data indicated that the normal motion of cervical spines was a 3D coupled motion, especially in LB/RB direction, where a 35% rotation around X-axis existed. The cervical spine could recover close to normal coupled motion after TDR surgery. Conclusions TDR surgery can restore the physiological motion of cervical spines more close to the normal state, especially in Flex/Ext and LR/RR direction. The study provides a theoretical basis and quantitative reference for TDR and ACDF surgery in clinic.

ABSTRACT

Objective To determine whether specific angiotensin-conventing enzyme inhibitor with ramipril would affect ventricular arrhythmia generation in rabbits after myocardial infarction and discuss the mechanism of its antiarrhymic efficacy.MethodsTwenty-four New Zealand rabbits (Wuhan Laboratory Animal Research Center) were separated into 3 groups:sham-operated (SHAM) group (n =8 ),myocardial infraction (MI) group ( n =8) and myocardial infraction with ramipril (RAM) group ( n =8).SHAM group received a median sternotomy without left ventricular coronary artery ligation.MI and RAM groups' rabbits received a median sternotomy followed by left coronary artery ligation. The successful anterior MI was confirmed by elevation of the ST segment with more than 0.2 mV in lead Ⅱ and Ⅲ.After MI,RAM group rabbits were fed with ramipril [ 1mg/ ( kg · d) ]by intragastric administration for 12 weeks.Before and after MI 12 weeks in three groups.Ventricular tachycardia or fibrillation episodes and the monophasic actionpotential duration in epicardium,mid-myocardium and endocardium cadiocytes were recorded.The statistical technique was t-test and ANOVA.Results Ventricular tachycardia or fibrillation episodes were markedly decreased in RAM group than that in MI group after 12 weeks [ (2.6 ± 0.8) vs.(12.4 ± 2.9),P <0.05 ].After MI 12 weeks,the action potential duration of repolarization 90% (APD90) of three-tier ventricular myocytes in MI group was prolonged than that before MI [ (258.2 ±21.1 ) vs.(230.1 ±23.2),( 278.0±23.8 ) vs.(245.8±25.4),(242.6±22.7) vs.(227.0±21.7),P<0.05]; however,it was not significant difference between before and after MI 12w in RAM group (P > 0.05 ).Moreover,the transmural dispersion of repolarization(TDR) was markedly increased after MI 12w in MI group than in SHAM and RAM group [ (36.2 ± 10.2 ) vs.( 18.7 ± 6.2 ),(24.9 ± 8.7 ),P < 0.05 ]; but the TDR was not significant difference between RAM and SHAM group ( 18.7 ± 6.2 ) vs.( 24.9 ± 8.7 ),P > 0.05].ConclusionsRamipril significantly reduced the malignant arrhythmia incidence in rabbits after MI.Mended the abnormal TDR was the mechanism for ramipril to therapy.

ABSTRACT

Objective To compare the incorporation method of 3H-TdR and 125Ⅰ-UdR on determining the proliferation effect of lymphocytes. Methods The proliferation effects of lymphocyte and Daudi lymphoma cells were estimated by 3H-TdR and 125Ⅰ-UdR incorporation. Results The incorporating fraction of 3H-TdR and 125Ⅰ-UdR into lymphocyte was 20.95% ± 1.06% and 1.00% ±0.04%,respectively, and the incorporating fraction for the lymphoma cells was 29. 94% ± 4. 10% and 6. 02% ±0. 73% ,respectively. The incorporation fractions of 3H-TdR into lymphocyte and lymphoma cells were much higher than those of 125Ⅰ-UdR, but the incorporating fractions of 3H-TdR or 125Ⅰ-UdR into the lymphoma cells were much higher than those of lymphocytes. Conclusions For lymphocytes, 125Ⅰ-UdR cannot substitute 3H-TdR as a tracer agent. But for lymphoma cells, whether 125Ⅰ-UdR could be replace 3H-TdR or not needs further research.

ABSTRACT

Este trabalho tem como objetivos projetar e avaliar o potencial de colunas de solo não-saturado equipadas com sondas de TDR (Time domain reflectometry) e acopladas a um sistema automatizado de aquisição de dados no monitoramento da dinâmica da água e de potássio no solo. O ensaio experimental foi conduzido em uma coluna de acrílico de 35cm de diâmetro interno e 105cm de comprimento preenchida com terra fina seca ao ar. A técnica da TDR possibilitou a obtenção de uma malha espaço-temporal em curtos intervalos de tempo e da umidade e da condutividade elétrica (Ce) nas diferentes profundidades consideradas. Tal resultado pode trazer uma importante contribuição para o entendimento da dinâmica da água e do potássio na terra, auxiliando os trabalhos para proposição e validação de modelos matemáticos que descrevem numericamente esses processos.


The aim of this research was to evaluate the advantages of using unsaturated soil columns equipped with TDR probes (Time Domain Reflectometry) coupled to an automated system of data acquisition for monitoring water and potassium movement in soils. The research was conducted in acrylic columns of 0.35m of internal diameter and 1.05m of length filled with air-dried soil. The use of TDR technique allowed obtaining time-space network at short time intervals, indicating levels of soil moisture and electric conductivity (Ce) at different depths. The results may represent an important help for the works on the proposition and validation of mathematical models that describe numerically these processes, contributing to a better understanding of the dynamics of water and potassium in soils.

ABSTRACT

O crescente uso de substratos na produção agrícola e no cultivo em ambiente protegido, promove produção em grande escala. Nesse sentido, as caracterizações químicas, biológicas e físicas desses materiais se fazem necessárias à proposição e à avaliação de padrões de qualidade que devem preceder a sua comercialização. As propriedades físicas dos substratos influenciam no bom desenvolvimento da plantas, sobretudo, no manejo de irrigação, onde a compreensão da relação retenção de água e aeração é imprescindível. Para tanto, mostra-se necessário que também se determine o volume de água presente nos substratos utilizados. Nesse sentido, o uso da reflectometria no domínio do tempo (TDR) pode representar um avanço em estudos dessa natureza. Dessa forma, baseado na determinação das curvas de retenção de água, neste trabalho, foram feitas as caracterizações físicas de dois substratos orgânicos: casca de pinus e fibra de coco. Também foram ajustadas, para cada um dos substratos avaliados, uma curva de calibração, através da qual, por meio da técnica da TDR, estimou-se o seu conteúdo de água. De maneira geral, com exceção da densidade seca, os substratos em estudo apresentaram características físicas da relação ar-água muito semelhantes. Quando comparado com os valores obtidos pelo método gravimétrico, na faixa de água facilmente disponível, a técnica da TDR apresentou um bom desempenho na estimativa da umidade de ambos substratos apresentando um coeficiente de determinação de 0,9319 para a casca de pinus e de 0,9385 para a fibra de coco.


The increasing use of substrates on agricultural production and controlled environment has been promoting its large scale production. In this way, chemical, biological and physical characterizations of these materials are necessary to the quality standard proposal and evaluation which must precede its commercialization. High plants development has been influenced by substrates physical properties, mainly on irrigation handling, where understanding about water retention and pore aeration is essential. Water volume determination on substrates used during experiments has been necessary, and in this way the use of the Time Domain Reflectometry (TDR) can represent an advance about this kind of researches. Based on water retention curve determination, this research, has been carried on physical characterizations of two organic substrates: coconut fiber and pine bark substrates. Also, a calibration curve has been adjusted for each tested substrates, and through TDR technique was estimated the water content. In a general, except to the dry density, the tested substrates has showed similar characteristics about its water-air relation. In the range readily available water, for both tested substrates, the TDR technique has showed a good performance on the water content estimative, with a determination coefficient of 0.9319 to the pine barks and 0.9385 to the coconut fiber.

Conduziu-se o presente trabalho com o objetivo de avaliar a distribuição de água no perfil de um Latossolo cultivado com mamoeiro do grupo Formosa, irrigado por dois gotejadores por planta instalados a 0,30 m da mesma. Usou-se gotejamento superficial com emissores de vazão de 3,75 L.h-1 e subsuperficial com emissores de vazão foi 2 L.h-1 a 0,25 m de profundidade. Os valores de umidades foram obtidos por meio de sondas de TDR instaladas e distribuídas em perfis de solo. As leituras foram realizadas em intervalos de dez minutos, durante dois ciclos de irrigação, utilizando um sistema de aquisição de dados composto por uma TDR, um datalogger e quatro multiplexadores. Os dados foram analisados a partir de isolinhas de distribuição de umidade e os resultados mostraram que o sistema de gotejamento subsuperficial apresentou uma menor variação da umidade concentrada na direção do sistema radicular do mamoeiro e o armazenamento de água no sistema subsuperficial foi 17 por cento superior ao superficial.


This work aimed at evaluating water distribution in a Latossol profile cultivated with papaya of Formosa group, irrigated by two drippers per plant installed at 0.30 m from the plant. Surface drip irrigation with emitters of 3.75 Lh-1flow rate and subsurface drip with emitters of 2 Lh-1 at 0.25 m depth were used. The humidity values were obtained by means of TDR probes installed and distributed in soil profiles. The readings were performed in a ten-minute interval during two irrigation cycles, using a data acquisition system composed by a TDR, one datalloger and four multiplexers. The data were analyzed from isolines of humidity distribution and results showed that the subsurface drip system had a smaller variation of humidity concentrated in the direction of papaya root system. The water storage for the subsurface system was 17 percent larger than the one for surface system.

ABSTRACT

PURPOSE: A traumatic diaphragmatic rupture (TDR) is a diagnostic challenge and the associated injuries determine the outcome in those diagnosed early. A TDR has long been considered to be a marker of the severity of injury with an average reported Injury Severity Score (ISS) between 31 and 50. This report reviewed the TDR cases in order to emphasize the method and timing of the diagnosis, associated injuries and the outcome. METHODS: A retrospective analysis was performed on 30 patient treated for TDR between August 1998 and september 2002. RESULTS: The mean age of the patients was 33.4 years and the injury predomiantly affected males (male: female=2: 1). Blunt trauma by TA was the most common cause of the TDR (25 patients). The ruptured sites were on the left in 22 cases and on the right in 8 cases. The most common symptom was chest pain (23 cases) followed dyspnea (21 cases). Liver injuries and a pneumo-hemothorax were the most common associated injuries. The mean CRAMS scale was 6.47 and Injury Severity Score (ISS) was 18.93. Eighteen TDR cases were suspected based on the abnormal chest X-ray findings. Nineteen cases underwent surgery within 6 hours after the trauma (early diagnosis). Although many complications occurred in 11 cases, there were no lethal complications. The mean size of the diaphragmatic rupture was 9.77 cm and an intraabdominal organ herniation had not occurred in ruptures smaller than 6 cm. Surgical repair of the diaphragm was performed via a laparotomy in 20 cases and thoracotomy in 7 cases. A left side TDR was preferred to a laparotomy whereas a right sided TDR was preferred to a thoracotomy. Pulmonary complications (atelectasis, pneumonia, pleural effusion) occurred in 14 cases postoperatively. CONCLUSION: The patient's complaints and physical findings were not a reliable indicator of the diagnosis, but usually a manifestation of the associated injuries. A suspicion and routine chest X-ray was the most reliable diagnostic tool, even though the chest X-ray was normal in 12 cases. A high rate of early diagnosis can be achieved using an aggresive investigation protocol, suspicion and a combined radiologic evaluation in multiple trauma patients. Although pulmonary complications occurred in the early diagnosed cases, lethal complications and long term sequela were directly related to the time of diagnosis. The higher ISS had many complications (11 cases) but there were no lethal complications, long term sequela.

ABSTRACT

Aim To estimate the anti-uterine cervix cancer effects of the extracts from Lysimachia clethroides Duby in vivo and in vitro. Methods Inhibitory effect on growth was observed by MTT, 3H-TdR and colony-forming units assay. Apoptosis was detected by Hoechst fluorescent staining analysis. The effect on cell migration and morphology was observed by scratch test; Detecting effects of ZE4 on transplant tumor (U14) in mice through observing the tumor weight and organ index. Results ZE4 could inhibit the growth and migration of HeLa cell significantly and induce cell apoptosis. Its half inhibitory concentration (IC50) at 48h was 40.56 mg?L-1 by 3H-TdR assay. The inhibition rate of ZE4 against U14 was 49.9% at the dose of 400 mg?kg-1. Conclusions ZE4 could inhibit the growth of uterine cervix cancer in vitro and in vivo.

Objective:To investigate the effect of human Leptin on the proliferation of human circulating T lymphocytes in vitro and the influence of Leptin on the action of PHA on the proliferation of human circulating T lymphocytes.Methods:PBMC were isolated from hepartinized venous blood of normal donors by density-gradient sedimentation over Ficoll-Hypaque and cultured in the 1640 complete medium.T lymphocytes were then obtained by depleting the monocyte and B cell populations.Cells were then cultured in the 1640 complete medium for 56 h in the presence of PHA.The proliferative effect was assessed by means of incorporation of thymidine.TdR was added and cultured for 16 hours.Cells were then processed in the harvester to measure the incorporation of [()~3H] thymidine.Radioactivity was determined by liquid scintillation counter.Results:The results showed that Leptin alone did not affect the proliferation of T lymphocytes,but it could enhance the effect of PHA on the proliferation of T lymphocytes at the concentrations of PHA from 2-8 ?g/ml.A dose-response studies of T lymphocyte proliferation showed that the maximal effect of Leptin were observed at 10 nmol/L.Leptin did not affect the proliferation of T lymphocytes when the concentrations of PHA were over 8 ?g/ml.Conclusion:The studies demonstrated that Leptin alone did not affect the proliferation of lymphocytes,but it could enhance the action of PHA on cell growth in a dose-dependent manner.

ABSTRACT

Purpose:To study the effect of sodium selenate on the viability and proliferation of PC-3 cell, a kind of human prostate cancer cell. Methods:Sodium selenate was administered to PC-3 cells, and MTT assay and ~(3)H-TdR adulterated assay were used to estimate the viability and proliferation of cell. Results:① When cells were treated with Sodium selenate for 24 h, the optical density (A) of middle-dose group decreased significantly, and the A of the high-dose group decreased dramatically. When cells were treated for 24 h, the A of the low-dose group was significantly lower than that of the control group, while the A of the middle-dose and high-dose groups was much lower than control.② When cells were treated for 24 h, the proliferation of middle-dose group decreased, and that of high-dose group decreased markedly. Conclusions:Sodium selenate can inhibit the viability and proliferation of PC-3 cells, and these actions occur in a dose-dependant manner.

OBJECTIVE:To study the effect of ligustrazine injection(LTZ)on DNA synthesis of vascular endothelial cell.METHODS:In vitro cultured cell line of human umbilical vein endothelial cells,ECV304,were adopted to measure the ef?fects of LTZ on DNA synthesis by 3 H-TdR incorporation.RESULTS:LTZ could inhibit DNA synthesis of ECV304in a dose-dependent manner.CONCLUSION:The mechanism of anti-angiogenesis of LTS might be associated with inhibiting DNA synthesis of vascular endothelial cell.

ABSTRACT

PURPOSE. To evaluate the effects of interfe-ron-gamma (IFN-?) and interferon-alpha (IFN-?) on human retinal pigment epithelial (RPE) ceils in vitro. METHODS: Cultures of human RPE cells were incubated with either of the IFNs (10IU/ml~10~4 IU/ml) for 3~5 days, and proliferation rates of the cells were assayed by [~3H]-thymidine incorporation and liquid scintillation techniques. RESULTS: There was a slight inhibition on human RPE cells with dosages from 10IU/ml to 10~4IU/ml. The inhibition rates of IFN-? of 10~3IU/ml, 10~4IU/ml were 8%, 15%, 26%, and of IFN-? were 7%, 9%, and 16%, respectively. On the 5th day there was a slight decrease in the inhibition rate than on the 3rd day. CONCLUSION: IFN-? and IFN-? exerted a slight inhibition on human RPE cells lasting 3 days.

ABSTRACT

AIM To analyze the effects of glycyrrhetic acid (GA) on c-fos expression and cell proliferation in rat vascular smooth muscle cell (VSMC), and to figure out the mechanisms of binding GA to angiotensin Ⅱ(AⅡ) receptor. METHODS Primary cell culture of rat VSMC, Northern blot, TdR incorporation DNA assay and MTT assay were used in this study. RESULTS ① GA at both low (1?10 -9 mol?L -1) and high concentration (1?10 -5 mol?L -1) quickly induced c-fos expression in VSMC. Sar-AⅡ(1?10 -5 mol?L -1)inhibited both GA induced and AⅡ (1?10 -5 mol?L -1) induced c-fos expression. GA enhanced AⅡ induced c-fos expression at both low and high concentration in VSMC. ② Low level of GA stimulated the proliferation of VSMC. This stimulatory effect decreased with increasing GA concentration, and changed to be inhibitory at high concentration of GA. Not only did Sar-AⅡ eliminate the stimulatory effect of low concentration of GA on cell proliferation, it also eliminated the inhibitory effect of high concentration of GA. Low concentration of GA enhanced the stimulation of AⅡon cell proliferation, while the inhibitory effect of high concentration of GA on cell proliferation was relieved by adding 1?10 -7 mol?L -1 AⅡ. CONCLUSION This study suggests that GA activates transcription factor c-fos and promotes the proliferation of VSMC. GA may exert its effects on cells through AT 1 receptor since it induces similar changes as AⅡ and its effects can be inhibited by AT 1 receptor antagonist.

ABSTRACT

The activity of colony stimulating factor ( CSF) in the conditioned medium ( CM ) was studied with combination method of 3H-TdR incorporation assay and agar colony assay. These two assays were demonstrated to be replaceable each other. The mice were administered with nontoxic monophosphoryl lipid A(MPLA) which was derived from a Re mutant of Salmonnella Minnesota Re595. The results showed a significant elevation of CSF in the serum and reaching the top at 12th h and returning to normal by 24th h. There is a significant dose-resoonse relationship. The CSF was induced with accompany of formation of colony inhibiting factor (GIF), some of which were heat sensitive factors. It is suggested that the MPLA may be a potent CSF-inducer.

ABSTRACT

The influences of varied concentrations of AG on the 8H-TdR(3H-Thymidine ) incorporation inhibition rate and the release of PGI2 and TXA2 were studied in cultured human and rat lymphocytes . The results indicated that the AG increased the 3H-TdR incorporation inhibition rate on the concentrations of 1.5? 10-7mol/L and 3?10-7mol/L. Using the RIA method, authors find that AG on the concentrations of 4 ? 10-7mol/L, 4 ? 10-3mol/L and 4 ? 10-11mo/L can significantly decrease the cultured lymphocytes TXA2 release while has no clear influences on PGI2 release
