ABSTRACT

Objective To explore the effect of Smad7 gene transfer on tubular cell cycle arrest, tubular apoptosis and fibronectin (FN) synthesis by transforming growth factor ? 1(TGF ? 1) induced. Methods Mouse Smad7 gene was transfected into renal tubular cells in primary cell culture by using Tfx 50 cationic liposome. Tubular cell proliferation was measured by MTT and cell cycle was observed by flow cytometry. The level of FN secretion in the supernatant was determined by ELISA. Results At 48 h after administration of 10 ng/ml TGF ? 1 to renal tubular cells, cell proliferation declined, G 0/G 1 arrested in cell cycle, and cell FN secretion increased significantly. These abnormalities were attenuated by liposome mediated Smad7 gene transfection. Conclusion Transfection and expression of Smad7 can markedly inhibit TGF ? 1 responses in renal tubular cells, which is helpful for further studies of Smad7 gene therapy in vivo .

Objective To study the effects of ? carotene on rat liver fibrosis induced by carbon tetrachloride (CCl 4). Methods ① After being treated with CCl 4 for 3 weeks to induce the experimental rat liver fibrosis, rats were treated with ? carotene 250 000 U/kg body weight by oral feeding twice a week. The normal and the CCl 4 control group were also set. ② The liver pathological and ultrastructual change were studied. ABSTRACT

In order to explore the relationship between the expression of TGF ? 1,its receptor and the wound age during the healing process of rats skin incision wound,a preliminary study were performed on the vital skin wound 0 5~168h after injury by using the immunohistochemical and the molecular biological methods.The results were then compared with those of the postmortem skin injury.The results revealed that the expression of the cytokine TGF ? 1 in the epithelial cells were enhanced 0 5h after the antemortem skin incision and the strongest reactions were seen at 24~96h post injury.The expression of TGF ? 1 was also found in the macrophages and the fibroblasts in the granulation tissues.Analysis of the immunoblotting(Western blot)results showed that the peak value of the TGF ? 1 protein was appeared at 168h after the antemortem skin incision and peaked at 96h.TGF ? 1 was detected 0 5~3h after postmorten skin injury by using the immunohistochemical method.No expression of mRNA was found.It is suggested that some regular and characteristic expression of TGF ? 1 in the incised skin wounds were related with the wound age.It might be used for timing of skin wound on rat.

ABSTRACT

Objective:To investigate the effect of glucocorticoid on treating class IV lupus nephritis(LN) by comparing the content of serum transforming growth factor beta(TGF ? 1).Methods:The study consisted of 33 objects,15 health controls as well as 18 cases of IV LN.The level of serum TGF ? 1 was analyzed by ELISA.Results:The level of serum TGF ? 1 was greatly reduced after the patients were treated with glucorcorticoid.Conclusion:Timely and sufficient glucocorticoid may prevent IV LN from developing into glomerulosclerosis.

ABSTRACT

Objective To study the localization of Smad 2 and Smad 4 proteins, which are intracellular signaling molecules of transforming growth factor ? family in adult rat testis. Method Immunohistochemical ABC method with glucose oxidase DAB nickel enhancement technique was used in the present study. Results Smad 2 immunoreactivity was mainly located in the spermatogonia, spermatocytes, spermatids, Sertoli cells in the seminiferous tubule and Leydig cells in the interstitial tissue. The reactive substance distributes in cytoplasm with negative nuclei. While Smad 4 is mainly expressed in cytoplasm of Leydig cells, and Sertoli cells is weak stained. Conclusion Our findings of Smad 2 expression in spermatogenic cells and Smad 4 expression in Leydig cells provide direct evidence for the molecular mechanism of TGF ? action during spermatogenesis.

ABSTRACT

Objective To observe the distributions of epidermal growth factor receptor(EGFR),transforming growth factor ?(TGF ? 1,? 2,? 3)and thyroid transcription factor 1(TTF 1)in developing lungs,and to discuss their functions and relationships each other. Methods The expression of EGFR and TGF ? was examined in mouse fetal lungs by immunohistochemistry.TIGER image analysiser was used in TTF 1 quantitative study. Results 1.In the early developmental phase,EGFR localized in the epithelium of airway surface,TTF 1 was expressed predominantly in the nuclei of distal lung buds and TGF ? was expressed in the bronchioles with localization patterns.2.At the late developmental phase,the positive reactions of EGFR and TTF 1 were mainly detected in alveolar cells.TGF ? expressed in mesenchymal at high levels.3 The average optical density(A A )of TTF 1 in the distal epithelial cells increased gradually during the development,while their volume(VOL)and volume integrated optical density(VI A )all decreased.Conclusion\ The functions of EGFR,TGF ? and TTF 1 might be respectively different at different developmental stages.They not only stimulate the process of branching morphogenesis,but also modulate epithelial maturation and differentiation.\;[
