ABSTRACT

OBJECTIVE: To study the dose-time-effect relationship of Tibetan medicine Rannasangpei in cerebral ischemic- reperfusion injury model rats with intragastric administration. METHODS: The rats were randomly divided into sham operation group (normal saline, 10 mL/kg), model control group (normal saline, 10 mL/kg), positive control group (nimodipine, 30      mg/kg), Rannasangpei different dose groups (0.52, 1.04, 2.08, 4.17, 8.33, 16.67, 33.34, 66.68, 133.36, 266.72 and 533.44    mg/kg), with 18 rats in each group. Each group was given relevant medicine intragastrically once; 25 min after intragastric administration, cerebral ischemic-reperfusion injury model was established with suture-occluded method in those groups except for sham operation group. 24, 48, 72 h after cerebral ischemia, neuroethology of rats were graded in each group. The rate of cerebral infraction was detected to evaluate the optimal effective time, the optimal dose (Dmax) and maximal effect (the rate of minimum cerebral infraction, Emax) of Ratnasampil at different periods of cerebral ischemia. Dose-time-effect relationship of Rannasangpei dose with the rate of cerebral infraction was fitted with Thermo Kinetica 5.1 software. The area under curve (AUClast) and retention dose (MRTlast) of dose-effect curve were calculated, and detect the levels of SOD and MDA. RESULTS: Compared with sham operation group, the neurobehavior of model group was significantly abnormal (P<0.05 or P<0.01), and the rate of cerebral infarction was significantly increased (P<0.01); the level of SOD was decreased significantly (P<0.01, 48 h), and the level MDA was increased significantly (P<0.05, 48 h). Compared with model control group, there was no significant change in neurobehavioral abnormalities in the nimodipine group (P>0.05), and the rate of cerebral infraction was decreased significantly (P<0.01, 24, 48 h). The level of SOD in rats were increased significantly (P<0.01, 48 h), while the level MDA decreased significantly (P<0.05, 48 h). Rannasangpei 2.08-33.34 mg/kg could significantly improved neurobehavioral abnormalities (P<0.05, 24 h); 24 h after cerebral ischemia, the rate of cerebral infraction was decreased significantly in Rannasangpei 4.17-133.36 mg/kg group (the lowest is 33.34 mg/kg group, P<0.05 or P<0.01). The level of SOD in rats were increased significantly in 33.34-533.44 mg/kg groups (P<0.05). the level MDA was decreased significantly in 0.52-2.08, 8.33, 33.34, 266.72 and 533.44 mg/kg groups (P<0.05). Dmax was 33.34 mg/kg, Emax was 3.02%, AUClast was 5 141.76 mg/kg and MRTlast was 329.161 mg/kg. 48 h after cerebral ischemia, the rate of cerebral infraction was decreased significantly in Rannasangpei 2.08-133.36 mg/kg groups (the lowest is 66.68 mg/kg group, P<0.05 or P<0.01), while the level of SOD was increased significantly in 1.04-533.44(except for 4.17)mg/kg groups (P<0.05). The level of MDA was decreased significantly in 16.67-66.68, 533.44 mg/kg groups (P<0.05), Dmax was 66.68 mg/kg, Emax was 2.13%, AUClast was    5 219.36 mg/kg and MRTlast was 340.521 mg/kg. 72 h after cerebral ischemia, the rate of cerebral infraction and the level of MDA had no significant decreased in Rannasangpei groups (P>0.05), and the levels of SOD had no significant increase (except for 0.52 mg/kg group, P>0.05). CONCLUSIONS: The optimal effective time of Rannasangpei for the treatment of cerebral ischemia-reperfusion injury in rats is 48 h, and the Dmax is 66.68 mg/kg. The improvement mechanism may be related to increase the level of SOD and decrease the level of MDA.

ABSTRACT

OBJECTIVE: To observe the time-effect of stimulation of the embedded poly glycolide-co-lactide (PGLA) suture in Sanyinjiao (SP6) area in normal human body, so as to provide an experimental evidence for clinical application of micro-invasion suture-embedding at an appropriate interval. METHODS: A total of 8 healthy volunteer students (3 boys and 5 girls, ranging in age from 24 to 27 years) were recruited in the present study. A piece of sterilized PGLA suture was implanted into the left SP6 using minimally invasive surgery after strict local skin disinfection. The fat-suppression T2 weighted magnetic resonance images (MRI, displaying local lesion after eliminating interference of fat tissue signals), and T2 mapping 8-echo train images were acquired before and 8 h, 3, 7, 10 and 14 days after PGLA suture embedment by using a MR imaging system. After transformation of the T2-mapping 8-echo train images into T2-mapping images by using a relevant software, the T2 values (meaning the relaxation time of the local muscle) of the left SP6 were measured, followed by analysis of the signal intensity of T2 weighted fat-suppression images and T2 values at different time-points. RESULTS: Before the suture embedding, no abnormal signals were found in the signal intensity of T2 weighted fat-suppression images. After PGLA suture embedment, the local signal intensity of T2WI fat-suppression images was relatively increased at the 8th h, and on day 3, 7, 10 and 14 relevant to pre-embedment, but gradually atte-nuated on day 10 and 14. The T2 values were significantly increased at the 5 time-points of post-embedment (all P0.05), and being markedly lowered on day 14 relevant to day 7 (P<0.01) in spite of being still markedly higher than that of pre-embedding (P<0.01). CONCLUSION: The signal intensity of T2 weighted fat-suppression images and T2 values acquired from PGLA-suture-embedded SP6 acupoint area in healthy subjects may keep at least for 2 weeks, suggesting that the stimulating reaction of suture-embedment persists more than 14 days. Hence, when a micro-invasion embedding with PGLA suture performed, the interval of two weeks would be appropriate.

ABSTRACT

OBJECTIVE: To observe the effect of moxibustion pretreatment at different time on serum hormone levels in diminished ovarian reserve (DOR) rats, so as to explore its protective mechanisms. METHODS: Forty female SD rats were randomly divided into control, model, moxibustion-1 (moxibustion was given 4 weeks before modeling), moxibustion-2 (moxibustion was given 2 weeks before modeling and 2 weeks from the 1st day on after modeling ) and moxibustion-3 (moxibustion was given 4 weeks from the 1st day on after modeling) groups (n=8 rats in each group). The DOR model was established by gavage of Tripterygium Glycosides (75 mg/kg) once daily for 14 days. Grain-moxibustion was applied to "Shenshu" (BL23) and "Guanyuan" (CV4) for 7 cones, 5 times a week for 4 weeks. The body weight and the ovary weight were recorded for calculating the ovarian index. The levels of serum anti-mullerian hormone (AMH), follicle stimulating hormone (FSH), estradiol (E2), androgen (T) and dehydroepiandrosterone (DHEA) were detected by ELISA. RESULTS: After modeling, ovarian index and serum AMH levels were obviously decreased (P<0.05), and the levels of serum FSH, E2, T and DHEA were significantly increased in contrast with the control group (P<0.05). Following intervention and compared with the model group, the serum FSH and DHEA levels of each moxibustion group were significantly reduced (P<0.05), the AMH levels significantly increased and E2 and T contents significantly decreased in the moxibustion-2 and moxibustion-3 groups (P<0.05). The serum FSH, E2 and T contents in moxibustion-2 group were obviously lower than those of the moxibustion-1 and moxibustion-3 groups (P<0.05). CONCLUSION: Moxibustion pre-treatment can improve ovarian reserve function in DOR rats, while the effect is different with different intervention time, and the best intervention time is pre-occurrence and early stage of DOR.

ABSTRACT

OBJECTIVE@#To summarize the research status and influencing factors of the time-effect of acupuncture and to explain and analyze the optimal effect timing of acupuncture, and provide reference for obtaining the best clinical therapeutic effect.@*METHODS@#Literature regarding the studies of the time-effect of acupuncture published from January 2000 to August 2018 was searched, concluded and analyzed in CNKI, WANFANG, VIP and PubMed databases.@*RESULTS@#A total of 43 papers were included. The time-effect relationship of acupuncture was studied from various aspects: immediate effect, post-effect and total time-effect. The influencing factors of time-effect of acupuncture included needle retention time, acupoint types, number of acupoints and frequency of acupuncture treatment.@*CONCLUSION@#Even though preliminary achievements have been made in the study of the time-effect of acupuncture, there are many limitations and deficiency. These studies are mostly isolated and unsystematic, and the observation time of needle retention time and post-effect is not long enough to observe the exact best induction period, half-life period and residual effect period of acupuncture. In the future, we should combine the existing research experience and achievements to formulate a more reasonable research plan and observe the entire period of acupuncture in a longitudinal direction.

ABSTRACT

Objective: To study the time-effect and dose-effect relationships of reducing uric acid of one complex extraction. Methods: Sixty male Kunming mice were randomly divided into blank group, model group, allopurinol group, and three groups of high, medium, and low complex extraction group. In the study of time-effect relationship, the mice were continuously treated with complex extraction for 2, 4, 7, 10, 13, 16, and 20 d, then the rats model was established by ip administration with oxonic acid potassium salt (20 mL/kg); In the study of dose-effect relationship, the mice were continuously treated with complex extraction for 7 d, then the rats model was established by ip administration with oxonic acid potassium salt (20 mL/kg). The mice urine, blood, and liver tissue were collected. Results: In the study of time-effect relationship, after 4 d of administration, the serum uric acid concentration was decreased obviously, and 7 d later, the low, medium, and high dose groups showed the significant activity of lowering uric acid, and the physiological and mental state of mice were very good in 20 d tests. Study on the relationship between dose and effect found that in low dose (100 mg/kg) showed good reduction of serum uric acid value activity (P < 0.05), and the high dose group (400 mg/kg) showed the most significant activity (P < 0.001). There was a certain dose-effect relationship. The high-dose group also showed significant inhibition of liver xanthine oxidase (XOD) activity (P < 0.05). Conclusion: The complex extraction has good reduction activity on uric acid in vivo, and shows certain dose-effect and time-effect relationships. The lowering effect on uric acid has relation with the inhibitory activity on xanthine oxidase.

ABSTRACT

Objective To observe the time-effect relationship between acupuncture treatment and the absorption of cerebral hematoma and edema in the acute stage of cerebral hemorrhage to determine the optimal time for acupuncture intervention in cerebral hemorrhage. Methods Three hundred patients with cerebral hemorrhage in the acute stage were randomly allocated to treatment groups 1, 2, 3 and 4, and control group, 60 cases each. The control group received symptomatic treatment for regulating blood pressure, reducing intracranial pressure by dehydration, maintaining electrolyte balance, nourishing nerves and preventing and treating complications. The treatment groups were given acupuncture in addition. Treatment group 1 received acupuncture at 24 hrs after the attack; treatment group 2, at 72 hrs; treatment group 3, at one week; treatment group 4, at two weeks. Pre-/post-treatment hematoma size parameter difference value and cerebral peripheral edema grade parameters were observed in every group. Results There were statistically significant pre-/post-treatment differences in hematoma size and cerebral edema grade in every group (P<0.01,P<0.05). There were statistically significant post-treatment differences in hematoma size and cerebral edema grade between every treatment group and the control group (P<0.05). There were statistically significant post-treatment differences in hematoma size and cerebral edema grade between treatment group 1, 2 or 3 and treatment group 4 (P<0.05), between treatment group 1 or 2 and treatment group 3 (P<0.05) and between treatment groups 1 and 2 (P<0.05).Conclusions Acupuncture has a positive therapeutic effect in patients with cerebral hemorrhage in the acute stage. The earlier acupuncture intervention in the acute stage of cerebral hemorrhage, the more remarkable the effect.

ABSTRACT

Objective To study the time-and dose-effect of mitochondrial DNA (mtDNA) 4934 bp and 4977 bp deletions in the human peripheral blood irradiated by137 Cs γ-rays,and to evaluate its implication in biological dosimetry. Methods The peripheral blood from five healthy adults was collected and irradiated with γ-rays.The peripheral blood of one healthy adult was irradiated with 5 Gy and cultured for 2,24,48 and 72 h after irradiation. The peripheral blood from the other four healthy adults was cultured for 2 h after 0,0.5,1,2,5 and 10 Gy irradiation. The peripheral blood mtDNA 4934 bp and 4977 bp deletions were detected by real-time polymerase chain reaction and gel electrophoresis. The doseeffect curves were fitted using Curve Expert 1.4 Software. Results mtDNA 4934 bp and 4977 bp deletions were induced at 2 h post-irradiation and the mtDNA 4934 bp deletion had relative high levels at 2 h and 48h after radiation (t =10.782 and 8.966,P < 0.05),and mtDNA 4977 bp deletion reached the highest level at 48 h after radiation (t =7.433,P <0.05).mtDNA 4934 bp (t =2.895-8.105,P <0.05) and 4977 bp deletion (t =3.006-7.715,P <0.05) irradiated at 0.5-10 Gy increased with a dosedependent manner. The incidence of mtDNA 4977 bp deletion was higher than that of 4934 bp deletion for those samples exposed with same dose of irradiation, especially at 10 Gy (t =2.919,P < 0.05),which suggested that 4977 bp deletion might be more sensitive than 4934 bp deletion at high dose. But larger individual differences were found in 4977 bp deletion compared with 4934 bp deletion. The dose-effect equations for 4934 bp deletion and 4977 bp deletion were Y1 =1.178 + 0.1219D (R2 =0.9269) and Y2 =1.2578 +0.1933D (R2 =0.9016),respectively.Conclusions The induction of mtDNA deletion was correlated with radiation dose, and thus it may be a available method for biological dose estimation and prognostic evaluation.

ABSTRACT

Objective To discuss the influence of new weapons in the high-tech war on eye wounds in order to develop the new time-effect treatment strategy. Methods The data of wounds in recent wars were investigated, the causes of eye wounds in high-tech war were analyzed, and the new concept of time effect treatment strategy for eye wounds was proposed. Results The weapons in high-tech war have developed into mass destruction and precisely guided munitions. The main cause of eye wounds is the shrapnel and some new wounds will occur such as eye wounds caused by laser or chemical toxin, radiation cataract. Conclusion In the future high-tech war, the major type of eye-wounds may still be the explosion injury and some new types of eye wounds should be paid more attention. The time-effect treatment strategy should be implemented to meet the demand of improving eye wound therapy and promoting the development of ophthalmology.

ABSTRACT

Objective To analyze the differences of time-effect and pathologic feature in the course of development of liver fibrosis between the two kinds of hepatic fibrotic animal models, which were made by CCl 4 and pig serum (PS) methods. Methods Rat models of liver fibrosis were made by using CCl 4 and PS ten week methods. At 6,10,14 and 20 weeks of model making respectively, pathological features of liver fibrosis were observed and analyzed by Masson staining and computerized image analyzing system. Results In CCl 4-induced model, fibrotic pseudolobuli developed at 6 weeks during model making, became typical at 10 weeks, and got stable thereafter.Ballooning degeneration of liver cells was outstanding, and paralleled with model making course. In PS-induced model, fibrosis occurred at 10 weeks during model making, and still developed though the model making was stopped, but ballooning degeneration of liver cells was not presented. Conclusions There was some different in time-effect relation and pathologic feature of liver fibrosis between the two methods of model making. This indicated that different mechanisms were involved in the pathogenesis of liver fibrosis. The two methods of model making should be combined to study of liver fibrosis. The prophylactic and treatment by anti-fibrotic drugs should last no less than 14 weeks.

ABSTRACT

AIM: To investigate the dose and time-effect relationship of 7 antimicrobial agents in experimental treatment of vibrio vulnificus infection in mice. METHODS: Vibrio vulnificus ( 6.0?10 11 CFU?L -1) was introduced intraperitoneally into the right abdominal cavity. After 1 h, 7 antimicrobial agents were given intraperitoneally using 5 dose levels respectively, and (were given intraperitoneally) using human therapeutic dose after 0.5, 1, 2, 3 h of infection, respectively. 7 antimicrobial agents were imipenem lcilastatin, chloramphenicol, doxycycline hydrochloride, netilmicin sulfate, cefoperazone sodium, piperacillin sodium, and levofloxacin lactate. The numbers of survival mice and the supermicrostructure change of organs were observed. RESULTS: With high dose of the seven agents, blood culture showed negative, and organ supermicrostructure injure recovered. 7 antimicrobial agents produced satisfactory therapeutic effect against experimental vibrio vulnificus infection using human therapeutic dose after 0.5~1 h of infection. Chloramphenicol, netilmicin sulfate, cefoperazone sodium and levofloxacin lactate had the better therapeutic effect after 2 h of infection, and weak effect after 3 h. Imipenem showed a weak effect. CONCLUSION: The treatments with these 7 antimicrobial agents early can obtain satisfactory therapeutic effect against experimental vibrio vulnificus infection in mice.
