Resumen Cuando los alimentos cubren los requerimientos energéticos, el organismo almacena el exceso de calorías como glucógeno en el hígado y el músculo, y los triacilgliceroles en el tejido adiposo. Morfológica y funcionalmente se clasifica en blanco y pardo. El pardo tiene gran cantidad de mitocondrias, almacena los triacilgliceroles en vacuolas y disipa la energía en forma de calor; el blanco almacena energía en gotas lipídicas que ocupan la mayor parte de su volumen. Después de la ingesta de alimentos se libera insulina, lo que hace que externen GLUT4 para absorber glucosa. Los quilomicrones o las lipoproteínas de muy baja densidad (VLDL) transportan los triacilgliceroles a los depósitos de tejido adiposo. Durante el ayuno, por acción del glucagón, se liberan enzimas que degradarán a los tri, di y monogliceroles para liberar a los ácidos grasos. El tejido adiposo libera citocinas pro y antiinflamatorias, así como leptina, adiponectina que regulan el apetito y la saciedad. La proteína cinasa activada por AMP se activa como respuesta a una baja en la cantidad de energía de la célula y le ayuda a mantener un balance energético. En el adipocito promueve la degradación de los triacilgliceroles para liberar a los ácidos grasos que se emplearán como fuente energética. Se requiere de mayor cantidad de estudios para conocer más sobre la función del tejido adiposo como regulador del metabolismo y no solo como almacén de energía.


Abstract When food meets energy requirements, the body stores in the liver and in the muscle the excess of calories as glycogen and triacylglycerols in the adipose tissue. Morphologically and functionally, it is classified into white and brown tissues. Brown tissue has many large mitochondria and stores triacylglycerols in vacuoles and dissipates energy as heat; white tissue stores energy as lipid droplets that occupy most of the adipocyte's volume. After food intake insulin is released, which causes GLUT4 externalization into the cellular membrane to absorb glucose. Chylomicrons or VLDL transport triacylglycerols to adipose tissue depots. During fasting, by the action of glucagon, enzymes are released that will degrade tri-, di- and mono-glycerols to release fatty acids. Adipose tissue releases pro and anti-inflammatory cytokines, as well as leptin and adiponectin that regulate appetite and satiety. AMPK is activated in response to a decrease in the cell's energy and helps it to maintain its energetic balance. In the adipocyte, it promotes the degradation of triacylglycerols releasing fatty acids to be used as an energy source. More studies are needed to learn more about the function of adipose tissue as a regulator of the metabolism and not only as an energy storage.

ABSTRACT

Background: The prevalence of overweight and obesity is progressively increasing in younger and adult population in India. It is a medical problem that increases risk of other diseases and health problems, such as heart disease, diabetes, high blood pressure and certain cancers. One of the causes of dyslipidaemia is obesity. High sensitivity C-Reactive Protein (hs-CRP), is a marker of systemic inflammation and a predictor of type 2 diabetes and cardiovascular disease. Hence it is important to check the relationship of hs-CRP with lipid profiles in obese and non-obese students.Methods: A case control observational study was carried out in 60 students. They were divided in to two groups obese and non-obese based on the BMI ranges. Serum lipid levels, hs-CRP and BMI was estimated in both groups to find out correlation of hs-CRP with lipid profile and BMI.Results: There was a significant rise in serum Total Cholesterol, LDL-C, Triacylglycerol and a significant fall in HDL-C in obese group as compared to non-obese group. Serum hs-CRP and BMI was significantly increased in obese students as compared to non-obese students. There was statistically significant positive correlation found between hs-CRP and total cholesterol, LDL-C, Triacylglycerol in obese students.Conclusions: Significant correlation was found between hs-CRP and lipid profile except HDL-C. Serum hs-CRP levels may decrease by treatment of dyslipidaemia. This would minimize the incidence of atherosclerosis and hence decrease the risk for development of coronary artery disease. Hence, improving the quality of life.

ABSTRACT

Chromochloris zofingiensis represents an industrially relevant and unique green alga, given its capability of synthesizing triacylglycerol (TAG) and astaxanthin simultaneously for storage in lipid droplets (LDs). To further decipher lipid metabolism, the nitrogen deprivation (ND)-induced LDs from C. zofingiensis were isolated, purified, and subjected to proteomic analysis. Intriguingly, many C. zofingiensis LD proteins had no orthologs present in LD proteome of the model alga Chlamydomonas reinhardtii. Seven novel LD proteins (i.e., two functionally unknown proteins, two caleosins, two lipases, and one l-gulonolactone oxidase) and the major LD protein (MLDP), which were all transcriptionally up-regulated by ND, were selected for further investigation. Heterologous expression in yeast demonstrated that all tested LD proteins were localized to LDs and all except the two functionally unknown proteins enabled yeast to produce more TAG. MLDP could restore the phenotype of mldp mutant strain and enhance TAG synthesis in wild-type strain of C. reinhardtii. Although MLDP and caleosins had a comparable abundance in LDs, they responded distinctly to ND at the transcriptional level. The two lipases, instead of functioning as TAG lipases, likely recycled polar lipids to support TAG synthesis. For the first time, we reported that l-gulonolactone oxidase was abundant in LDs and facilitated TAG accumulation. Moreover, we also proposed a novel working model for C. zofingiensis LDs. Taken together, our work unravels the unique characteristics of C. zofingiensis LDs and provides insights into algal LD biogenesis and TAG synthesis, which would facilitate genetic engineering of this alga for TAG improvement.

ABSTRACT

Objective • To analyze the related risk factors of decreased heart rate variability (HRV) in type 2 diabetes mellitus (T2DM) patients. Methods • A total of 210 cases with T2DM in Shanghai General Hospital, Shanghai Jiao Tong University School of Medicine from Jan. 2013 to Dec. 2014 were divided into two groups by the levels of urinary albumin creatinine ratio (UACR) and serum triacylglycerol (TAG), and with/without hypertension, respectively. By comparing the differences of clinical characteristics, HRV parameters and other general clinical data between the two groups, the correlations between HRV indexes and their risk factors were explored. Results • The average heart rate of the whole day was higher, and SDNN [normal RR intervals of sinus beats (NN intervals) standard deviation] was lower in the high UACR group than those in the normal UACR group (both P<0.05). The average heart rate of the whole day was higher, while SDNN and pNN50 (percentage of total heart beats with adjacent NN intervals greater than 50 ms) were lower in the hypertriglyceridemia group than those in the normal TAG group (all P<0.05). Minimum frequency domain power hour was lower in the hypertension group than that in the normotension group (P<0.05). Regression analysis showed that the linear correlativity between TAG and SDNN was very prominent (P<0.05), as well as between the diastolic blood pressure (DBP) and the average heart rate of the whole day, SDNN, frequency domain power 24 hours and minimum frequency domain power hour (all P<0.05). Similarly, the age was linearly correlated significantly with the average heart rate of the whole day, SDNN, SDANN (the standard deviation of the mean NN intervals every 5 min), frequency domain power 24 hours, minimum frequency domain power hour and maximum frequency domain power hour (all P<0.05). Conclusion • Age, TAG level and hypertension are the risk factors of decreased HRV in T2DM patients. Thus, early detection of the decrease of HRV in these patients can prevent the cardiovascular events of T2DM.

ABSTRACT Dyslipidemias are associated with the incidence of cardiovascular diseases, obesity, diabetes, hypertension and hepatic steatosis, being the cause of morbidity and mortality. This study investigated the effects of lychee peel flour (PF) on serum levels of total cholesterol (TC), low-density lipoprotein (LDL-c), triacylglycerols (TAG) and various parameters related to obesity, in rats fed a hypercholesterolemic diet. Therefore, 20 male rats were used. In the first 21 days, the animals were fed a hypercholesterolemic diet, except for control group. In the following 21 days, their diets were modified, and they received a standard diet (Control); hypercholesterolemic (Hyper); hypercholesterolemic + 5% PF (PF5) and hypercholesterolemic + 10% PF (PF10). The results revealed that PF intake attenuated weight gain, reduced body mass index, glucose and the levels of TAG, TC, LDL-c, hepatic enzymes and leptin, besides the percentage of hepatic lipids, liver lipid peroxidation and frequency of severe steatosis. Histological studies of the aorta did not show the formation of the atheromatous plaque. These results reinforce its potential to reduce the risk of diseases associated with obesity.

GAP exhibited a 1.46:1 mannose: galactose ratio, and high molar weight. Both GAP enriched food decreased glycaemia, total cholesterol and triacylglycerol. GAP didn't interfere on food intakes or body weight, although it increased water intake. Furthermore, the relative liver weight indicated toxic galactomannan effects on the histopathological changes of the pancreas in STZ induced diabetes.Conclusions:It is concluded that GAP is a natural product that contains potent galactomannan and is useful in preventing and treating diabetes.

Sudden sensorineural hearing loss (SSHL),which is a common and frequently encountered disease,is considered to be a medical emergency in otolaryngology.The prevalence of SSHL is increasing in China.The pathogenesis of SSHL is not clear yet.Microcirculatory disorder of inner ear is considered as one of the most important causes of SSHL.In recent years,several reports have found the levels of serum lipids were changed in patients affected by SSHL.The relationship between SSHL and serum lipids was reviewed to provide new ideas for the diagnosis and treatment of SSHL.

Objective Long-chain triacylglycerol (LCT) by three producers,each mixed with the same medium-chain triacylglycerol (MCT),were compared with the brand MCT/LCT in causing focal necrosis of hepatocytes in beagle dogs (a bioequivalence evaluation).Methods 21 beagle dogs (male,0.7-1.5 years old,10-15 kg) were used in this study.According to the sources of the LCT,the animals were divided into Group A (LCT made in China),Group B (LCT made in Japan),Group C (LCT made in Germany),and the control group (the brand 10% MCT/LCT).Central venous port was placed via the lumber vein of the animals under general anesthesia.After 2 weeks of rehabilitation,MCT/LCT was administered through this port for 28 days at 9 g/ (kg · d) [while the routine dose used clinically was 1 g/ (kg · d)].The laboratory indexes and the pathomorphism of the liver and kidney were studied single blindly.Results Laboratory tests,including liver and kidney function,blood coagulation function and lipid metabolism,did not identify differences among emulsions with different sources of LCT.Liver biopsy at day 28 showed no focal necrosis in Group C and the control group;there was minor damage in Group B;and Group A had obvious liver necrosis.and the pathological findings of other organs are similar.No significant difference was observed in biopsies of other organs.Conclusions Emulsions with different sources of LCT varied in their damage to the liver.Generics with LCT of higher quality were equivalent to the brand MCT/LCT in terms of safety.

Esta revisão teve como objetivo apresentar e discutir os achados mais recentes do efeito dos ácidos graxos monoinsaturados (AGMI) sobre marcadores plasmáticos do metabolismo lipídico em estudos pós-prandiais e de intervenção clínica nutricional. Realizou-se busca em diferentes bases de dados entre 2010 e 2014, usando os seguintes termos de indexação: MUFA, Lipemia, Lipid Metabolism, Triglycerides e Postprandial. O consumo de refeição com alto conteúdo de AGMI tem demonstrado efeito benéfico na resposta lipidêmica pós-prandial, mas se essa resposta pode ser alterada em indivíduos com excesso de peso e/ou outras doenças crônicas após consumo de AGMI, ainda não está totalmente elucidado. De modo geral, após a intervenção com AGMI, os fatores de risco cardiovascular diminuíram, além de haver melhora no perfil lipídico. Em conclusão, os estudos recentes têm demonstrado um efeito benéfico do consumo de AGMI em curto e longo prazos, mediante aumento/manutenção das concentrações de HDL colesterol e diminuição do LDL colesterol.


The objective of this review is to present and discuss the most recent findings related to the effects of monounsaturated fatty acids (MUFA) on plasma markers of lipid metabolism observed in postprandial studies and clinical nutritional intervention studies. Searches were conducted on several different databases for publications from 2010 to 2014 using the following keywords: MUFA, Lipemia, Lipid Metabolism, Triglycerides and Postprandial. High-MUFA meal has presented beneficial effect on postprandial lipidemia response, but it is not yet completely clear whether this response to MUFA intake may be different in people with excess weight and/or other chronic diseases. In general, cardiovascular risk factors were reduced and lipid profiles improved after interventions with MUFA. In conclusion, recent studies have demonstrated that consuming MUFA has beneficial effects at short and long time by increasing/maintaining HDL-cholesterol concentrations and reducing levels of LDL cholesterol.

ABSTRACT

OBJECTIVE: Atrial fibrillation (AF) is a common arrhythmia which is generally more prevalent in the elderly population. However, the new onset of AF is frequently found in young populations. In order to identify putative prognostic biomarkers for detection of young-onset AF, we purified and characterized lipoproteins in terms of oxidative and inflammatory properties. METHODS: Male patients with AF (46±7 years of age, n=19) were recruited. Their serum and individual lipoproteins were analyzed and compared with healthy controls (48±9 years of age, n=17). RESULTS: The patients with AF revealed hypertriglyceridemia, hyperuricemia with mild obesity, elevated levels of CRP, and a normal level of cholesterol. All lipoproteins from patients with AF demonstrated higher levels of TG and advanced glycated end products, and decreased particle size than controls. AF-LDL showed an increased extent of oxidation with increased atherogenic macrophage phagocytosis. AF-HDL showed impaired antioxidant ability and a lower level of apoA-I expression. CONCLUSION: These data suggest that lipoprotein properties are severely modified in young AF patients, which were correlated with increased oxidation and inflammation.

ABSTRACT

OBJECTIVE: To develop a method suitable for rapid and accurate analysis of triacylglycerols in medium/long chain structured triacylglycerols by ultraperformance convergence chromatography combined with a quadrupole time-of-flight mass spectrometry. METHODS: The separation was performed at 28℃ on a Waters Acquity UPCC system by anHSS C18 SB column (3.0 mm×150 mm, 1.8 μm) with gradient elutionof CO2 and methanol acetonitrile (7:3, V/V), the flow rate was 1.2 mL·min-1, the back pressure was 117.2 MPa, and methanol containing 0.1% formic acid and 2 mmol·L-1 ammonium formate was used as supplement solvent. Positive ion electrospray ionisation (ESI) mode and MS technology were used in which two separate scan functions were programmed for the MS acquisition. The qualitative analyses were carried out by using precise mass information of parent ions and product ions. RESULTS: A total of 52 triacylglycerols were obtained and their stuctures were identified by analyzing the precise mass information of their parent ions and product ions respectively. CONCLUSION: The method is simple, highly accurate, and environmental friendly with good resolution, and can be used for rapid and accurate analysis of medium/long chain triacylglycerols excipients.

ABSTRACT

Lipolysis is defined as the sequential hydrolysis of triacylglycerol (TAG) stored in cell lipid droplets. For many years, it was believed that hormone-sensitive lipase (HSL) and monoacylglycerol lipase (MGL) were the main enzymes catalyzing lipolysis in the white adipose tissue. Since the discovery of adipose triglyceride lipase (ATGL) in 2004, many studies were performed to investigate and characterize the actions of this lipase, as well as of other proteins and possible regulatory mechanisms involved, which reformulated the concept of lipolysis. Novel findings from these studies include the identification of lipolytic products as signaling molecules regulating important metabolic processes in many non-adipose tissues, unveiling a previously underestimated aspect of lipolysis. Thus, we present here an updated review of concepts and regulation of white adipocyte lipolysis with a special emphasis in its role in metabolism homeostasis and as a source of important signaling molecules.

ABSTRACT

Objective To explore the relationship between blood lipids level with creatinine clearance rate(Ccr)in patients with heart failure(HF). Methods A total of 955 patients who were diagnosed with heart failure(cardiac function NYHAⅡ~Ⅳclassification)upon discharge from the Department of Cardiology of the Second Hospital of Tianjin Medical University, between January 2010 to June 2013 were enrolled as HF group. Healthy adults (n=200) with normal cardiac function which approximately matched basic condition with HF group were selected as control group. The HF group was fur?ther divided intoⅡ,Ⅲ,Ⅳclassification according to their cardiac function(NYHA classification). HF group was also divid?ed into normal renal function group, mild renal injury group and moderate-severe renal injury group based on their Ccr. Ef?fect of gender and lipid parameters were also compared. Binary logistic regression was used to analyze factors influencing renal function in patients with HF. Results Compared with people in the control group, the levels of triacylglycerol(TG), total cholesterol(TC), low density lipoprotein cholesterol(LDL-c)and non high density lipoprotein cholesterol(non-HDL-c)in patients of HF group were increased while Ccr and high density lipoprotein cholesterol(HDL-c)were decreased. Ccr and lipids were obviously decreased in patients with HF of Ⅳclassification. TG and HDL-c were decreased in moderate-severe renal injury group. Females had a higher lipid levels than males in HF group(P<0.05 or P<0.01). Advanced age, coronary heart disease and hypertension were all risk factors for renal impairment in patients with HF by binary logistic re?gression. On the other hand, weight gain and HDL-c were the protection factors for renal function in HF patients. Conclu?sion Dyslipidemias may lead to renal insufficiency in patients with HF. It was important to control lipids and improve re?nal function in patients with HF.

PURPOSE: In epidemiologic and animal studies, a high fat diet (HFD) has been shown to be associated with lower bone mineral density (BMD) and a higher risk of osteoporotic fractures. Meanwhile, consuming a HFD containing diacylglycerol (DAG) instead of triacylglycerol (TAG) is known to offer metabolically beneficial effects of reductions in body weight and abdominal fat. The purpose of this study was to investigate the effects of a HFD containing DAG (HFD-DAG) on bone in mice. MATERIALS AND METHODS: Four-week-old male C57BL/6J mice (n=39) were divided into three weight-matched groups based on diet type: a chow diet group, a HFD containing TAG (HFD-TAG) group, and a HFD-DAG group. After 20 weeks, body composition and bone microstructure were analyzed using dual energy X-ray absorptiometry and micro-computed tomography. Reverse transcription-polymerase chain reaction (PCR) and real-time PCR of bone marrow cells were performed to investigate the expressions of transcription factors for osteogenesis or adipogenesis. RESULTS: The HFD-DAG group exhibited lower body weight, higher BMD, and superior microstructural bone parameters, compared to the HFD-TAG group. The HFD-DAG group showed increased expression of Runx2 and decreased expression of PPARgamma in bone marrow cells, compared to the HFD-TAG group. The HFD-DAG group also had lower levels of plasma glucose, insulin, total cholesterol, and triglyceride than the HFD-TAG group. CONCLUSION: Compared to HFD-TAG, HFD-DAG showed beneficial effects on bone and bone metabolism in C57BL/6J mice.

Introduction: A recently published meta-analysis showed that each additional serving of rice increased risk of type 2 diabetes mellitus (DM) by an alarming 11%. We investigated whether this phenomenon is seen in the Malaysian population by studying the effect of rice intake and added sugar consumption on fasting plasma glucose (FPG) and fasting triacylglycerol (TAG). Methods: Ninety subjects (60 females, 30 males, aged 30-70 years), adequate to detect a weak-to-moderate Pearson correlation of r=0.26 at a=0.05 and power= 0.80, were recruited by convenience sampling from six communities in the Klang Valley, Malaysia. Fasting blood samples were collected by finger-prick and analysed for FPG (AccuCek, Roche) and TAG (Accutrend, Roche). Macronutrient intakes, including rice, were obtained by a single interview using a previously-evaluated food frequency questionnaire (FFQ) and quantitated as grams by the DietPLUS V2 programme. Added sugar intakes by subjects were estimated using an Added Sugar Intake excel programme. Results: Rice contributed to 85% of dietary carbohydrates, accounting for 41.8 % kcal of the average 1750- kcal diet. Rice intakes or added sugar consumption did not have a significant correlation (p>0.05) with FPG nor fasting TAG. Added sugar consumption, which averaged 44g/person/day (5% kcal) was markedly lower than the 137g/person/day reported elsewhere for the Malaysian population. Conclusion: High consumption of rice as a risk factor of type 2 DM was not indicated in the present study. Since white rice consumption varied 10-fold in the present subjects, the reduction in daily intake of this staple food represents a feasible option for cutting back on calorie intake for overweight or obese individuals.

ABSTRACT

Limited evidence was available to support the effect of self-selected activities performed under free-living conditions on postprandial lipaemia, particularly for older adults. Thus, the aim of the present study was to examine the chronic effect of increased physical activity of daily living on postprandial triacylglycerol (TAG) in postmenopausal women. Twenty-eight postmenopausal women, aged 71 ± 4 years (mean ± SD), were randomly divided into two groups: active (n=14) and control (n=14) groups. The participants in the active group were asked to increase their activities above their usual lifestyle levels for 4 weeks; freely deciding the duration and intensity of their chosen activities. The participants in the control group maintained their usual lifestyle for 4 weeks. All participants were asked to wear a uniaxial accelerometer for 4 consecutive weeks. At baseline and after 4 weeks, all participants rested and consumed a standardised breakfast and lunch after a 24-h period of physical activity avoidance. Blood samples were collected in the fasted state (0 h) and at 2, 4 and 6 h after breakfast. After 4 weeks, the participants in the active group increased their step counts by 600 steps/day (from 6979 ± 2057 to 7586 ± 2301 steps/day, p=0.047). There was no difference in the pattern of postprandial TAG response between groups (trial × time interaction, p=0.335). A previous study has suggested that only recent exercise (i.e., 12h before but not 24 h before) appears to facilitate the exercise-induced postprandial TAG lowering effects. Thus, in the present study, postprandial lipaemia was not reduced after performing self-selected activities under free-living in postmenopausal women.

ABSTRACT

Aim: To show that Gymnema sylvestre (Roxb.) Asclepiadaceae not only has antidiabetic propensities, but it most likely works by regeneration of pancreatic  cells which is imperative in anti-obesity-diabetes therapeutic applications of medicinal plants. Study Design: The present study design investigated the effects of G. sylvestre leaves crude aqueous extracts (AEs), traditionally utilized in diabetes treatment, on the pancreatic β-cell MIN6 proliferation and insulin secretion and extrapancreatic dietary carbohydrate and lipid digestion Place and Duration of Study: Faculty of Pharmacy, The University of Jordan, 2008-2012. Results: Comparable to GLP-1 (500 nM) pancreatic proliferative capacity; G. sylvestre AE concentrations (0.01 and 0.1 mg/mL) induced MIN6 monolayers expansion by respective 130.3% and 127.4% (P<0.001 vs. spontaneous control). Like L-alanine (10 mM) insulinotropic efficacy and without exerting cytotoxicity, glucose-stimulated insulin secretion was potentiated by G. sylvestre AEs (5, 10 and 25 mg/mL) (711.0%, 843.0% and 906.5%, respectively, P<0.001 vs. basal control). The potent plants’ insulin secretory bioactivities were abolished in the depleted Ca2+ conditions (P<0.001). Similar to orlistat antilipolytic efficacy, pancreatic lipase IC50 value for G. sylvestre AEs was 106.3±7.2 μg/mL. Unlike acarbose (100 μg/mL) dual inhibition of α-amylase/α-glucosidase, G. sylvestre AE was inactive at used doses. Dissimilar to guar gum (50 mg/mL) diffusional hindrance in a simple dialysis model, G. sylvestre AEs (10, 25 and 50 mg/mL) proved inactive. This in vitro ineffectiveness was mirrored in respective in vivo oral carbohydrate tolerance tests in overnight fasting normoglycemic rats. Conclusion: This evaluation has revealed that G. sylvestre leaves AEs augmented β-cell expansion and potentiated glucose-evoked Ca2+-regulated insulin secretion; combined with impressive antilipolytic activity. These actions depend on the bioactive water soluble phytoprinciples intact absorption in vivo. Future directives may assess the potential of G. sylvestre as a new alternative for anti-obesity-diabetes pharmacotherapy and prevention.

ABSTRACT

Elevated total cholesterol and low-density lipoprotein cholesterol in sera are both well-known risk factors of coronary heart disease. Adequate vitamin D status is important for optimal function of many organs and tissues of our body. There is continuing controversy about the effect of adequate vitamin D consumption on serum lipids and lipoproteins. The present study assessed the effect of vitamin D, calcium and multiple micronutrients supplementation on the lipid profile in Bangladeshi young female garment factory workers who have hypovitaminosis D. This placebo-controlled intervention trial conducted over a period of one year randomly assigned a total of 200 apparently healthy subjects aged 16-36 years to 4 groups. The subjects received daily supplements of 400 IU of vitamin D (VD group) or 400 IU of vitamin D+600 mg of calcium lactate (VD-Ca group), or multiple micronutrients with 400 IU of vitamin-D+600 mg of calcium lactate (MMN-VD-Ca group), or the group consuming placebo (PL group). Serum concentrations of lipid and lipoprotein, 25-hydroxyvitamin D (25OHD) and intact parathyroid hormone (iPTH) were measured at baseline and after one year of follow-up. No significant changes in the serum levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), LDL-C/HDL-C ratio were observed in the supplemented groups compared to the placebo group. Supplementation had a positive effect (p<0.05) on very low-density lipoprotein cholesterol (VLDL-C) and triacylglycerol (TAG). A negative correlation between changes in serum iPTH and HDL-C was observed, which indicated that subjects with the greatest decline in S-iPTH had the greatest increase in HDL-C. The results suggest that consumption of adequate vitamin D with calcium or MMN for one-year may have no impact on serum lipid profile in the subjects studied. Longer-term clinical trials with different doses of supplemental vitamin D are warranted in evaluating the effect of intervention.

O consumidor está cada vez mais consciente da relação entre dieta e doença, que tem impulsionado as pesquisas sobre alimentos funcionais e seus efeitos sobre o corpo. O papel dos óleos e gorduras na nutrição humana tem sido intensamente estudado e discutido por décadas. Tem sido enfatizada a importância da ingestão de ômega-3, ômega-6 e ômega-9 ácidos graxos redução de ácidos graxos saturados e, mais recentemente, controle da ingestão de ácidos graxos trans. Através da mistura e interesterificação química e enzimática de óleos e gorduras, gorduras trans-livre pode ser produzido. Mistura de gordura, foram formuladas por misturas ternárias de estearina de palma, uma gordura láurica (óleo de coco ou óleo de palmiste) e um óleo poliinsaturado (óleo de canola ou azeite de oliva) em diferentes proporções que foram interesterificadas. Neste trabalho, foram produzidos lipídios estruturados por interesterificação química e enzimática. A interesterificação química foi realizada nas seguintes condições: a 88 °C, 60 minutos de reação, 0,4% de catalisador metóxido de sódio, sob agitação e vácuo. A interesterificação enzimática, sendo realizada com duas lipases comerciais Thermomyces lanuginosa e Rhizomucor miehei, com seletividade sn-1,3. A interesterificação enzimática por batelada foi realizado seguindo um planejamento matriz central compósito rotativo em função da temperatura e da composição do meio, estearina de palma, óleo de palmiste e azeite de oliva e catalisado pelas lipases comerciais. O decréscimo do conteúdo de gordura sólida foi observado a 10 e 35 °C após a interesterificação. O biorreator contínuo foi operado nas seguintes condições: mistura de estearina de palma, óleo de palmiste, azeite de oliva (45:30:25), 10 gr de biocatalisador, 65 °C, com tempo de residência de 7 min e por 226 h para Thermomyces lanuginosa e 188 h para Rhizomucor miehei. A atividade do biocatalisador foi avaliada em termos da diminuição do conteúdo de gordura sólida a 35 °C, o qual é um parâmetro chave na produção de margarinas. O perfil de inativação do biocatalisador pode ser bem descrita pelo modelo de desativação de primeira ordem: meia-vida de 88 e 60 h foram estimados quando Thermomyces lanuginosa e Rhizomucor miehei, respectivamente, foram utilizados. Os óleos puros, as misturas originais e interesterificadas foram avaliados quanto à composição de ácidos graxos e triacilgliceróis, distribuição regioespecífica dos ácidos graxos nos triacilgliceróis, ponto de fusão e amolecimento, consistência, conteúdo de gordura sólida, comportamento de fusão e cristalização, estabilidade oxidativa, estrutura cristalina e polimorfismo. A interesterificação química e enzimática promoveram diminuição de triacilgliceróis trissaturados e triinsaturados e aumento dos monossaturados-diinsaturados e dissaturados-monoinsaturados, o que resultou no respectivo decréscimo dos pontos de fusão e amolecimento, consistência e conteúdo de gordura sólida, aumentando a plasticidade das gorduras. As curvas de fusão e cristalização das misturas foram modificadas pela alteração da composição dos triacilgliceróis pela interesterificação química e enzimática. Estabilidade térmica e a temperatura de oxidação da estearina de palma, óleo de coco e óleo de canola e suas misturas foram dependente da composição de ácidos graxos e independente da interesterificação química. Os resultados mostram que a interesterificação química e enzimática oferecem uma ferramenta útil para a concepção de gorduras com sintonizáveis propriedades físico-químicas, melhorando em relação a esse das gorduras de partida


The consumer is becoming more aware of the relationship between diet and disease, which has driven the research on functional foods and their effects on the body. The role of fats and oils in human nutrition has been intensively studied and discussed for decades. It has been emphasized the importance of intake of omega-3, omega-6 and omega-9 fatty acids, reduction of saturated fatty acids and, more recently, control of intake of trans fatty acids. Through the blend and interesterification of oils and fats, trans-free fats can be produced. Fat blends, formulated by ternary blends of palm stearin, lauric fat (coconut oil and palm kernel oil) and polyunsaturated oils (canola oil and olive oil) were done in different ratios. In this work, were produced by chemical and enzymatic interesterification. Chemical interesterification was performed under the following conditions: at 88°C, 60 minutes reaction times, 0.4% sodium methoxide, under agitation and vacuum. For enzymatic interesterification being carried out with two commercial lipases Thermomyces lanuginosa e Rhizomucor miehei, with selectivity sn-1,3. Batch enzymatic interesterification were performed, following central composite rotatable designs (CCRDs) as a function temperature and media of palm stearin, palm kernel oil and olive oil formulation and catalyzed by a commercial immobilized lipase. A decrease in all SFC values of the blends at 10 °C and 35°C was observed upon interesterification. The bioreactor operated continuously: mixture of palm stearin, palm kernel oil and olive oil (45:30:25, wt %), at 65 °C, at a residence time of 7 min and for 226 h to Thermomyces lanuginosa and 188 h to Rhizomucor miehei.. Biocatalyst activity was evaluated in terms of the decrease of the solid fat content at 35 °C of the blends, which is a key parameter in margarine manufacture. The inactivation profile of the biocatalyst could be well described by the first-order deactivation model: Half-lives of 88 and 60 h were estimated when Thermomyces lanuginose and Rhizomucor miehei, respectively, were used. Pure oil, the original and interesterified blends were examined for fatty acids and triacylglycerols composition, regiospecific distribution of fatty acids in triacylglycerols, melting and softening points, consistency, solid fat content, thermal behavior, oxidation stability, crystalline microstructure and polymorphism. Chemical and enzymatic interesterification caused reduction of trisaturated and triunsaturated and increase in monosaturated-diunsaturated and disaturated-monounsaturated, lowering the initial melting and softening points, consistency and solid fat content, increasing plasticity of fats. Melting and crystallization curves were significantly modified by changing the composition of triacylglycerols by chemical and enzymatic interesterification. The thermal stability and oxidation temperature of palm stearin, coconut oil and canola oil and their blends were dependent on fatty acid composition and independent on chemical interesterification. The results show that the chemical and enzymatic interesterification provides a useful tool to design fats with tunable physicochemical properties, improved compared to that of the starting fats
