ABSTRACT

Despite the availability of anti-TB drugs, the proportion of cases of multidrug-resistant tuberculosis (MDR) is increasing. One of the important tasks to identify effective drugs in nature is the study of numerous experiments of traditional medicine. This work is devoted to the study of chemical, biochemical, medical evaluation of the new therapeutic food additive "Majmui Rahmoniy", and its effective effect on Mycobacterium smegmatisand MS Rv2349c gene which responsible for the phospholipase activity.

ABSTRACT

@# T-Vec(talimogene laherparepvec)是由Ⅰ型单纯疱疹病毒(HSV-1)改造而来的一种溶瘤病毒,能够选择性地在恶性肿 瘤细胞中复制而不伤及其他正常细胞。T-Vec在治疗晚期黑色素瘤患者的Ⅲ期临床试验中显示出良好的安全性和肿瘤治疗效 果,已于2015年经美国FDA批准用于治疗晚期黑色素瘤。为了提高T-Vec的疗效,扩大其应用范围,T-Vec联合其他抗肿瘤疗法 以及应用于其他肿瘤的临床试验仍在陆续开展。近期,T-Vec联合免疫检查点抑制剂在治疗晚期黑色素瘤的临床试验中取得新 进展,临床数据显示T-Vec联合疗法具有更强的抗肿瘤活性。此外,T-Vec在治疗头颈癌、胰腺癌、肝癌等肿瘤的临床研究中也取 得了一定进展。本文对近年来T-Vec治疗肿瘤的临床试验的相关研究进展做一综述。

ABSTRACT

@#AIM: To evaluate the efficacy and safety of VEC combined with laser in treatment of retinoblastoma(RB)in children with different ages.<p>METHODS: Totally 63 children with RB intraocular phase in our hospital were selected as subjects. According to their ages, they were divided into 1 year old group(<i>n</i>=23, 36 eyes), >1 year old group(<i>n</i>=40, 66 eyes). All of them were treated with VEC combined with laser therapy. Indirect ophthalmoscope and ultrasound were used to measure the thickness and diameter of tumor and tumor base before treatment and after 6 treatment courses.<p>RESULTS: The remission rate in the 1 year old group was 75.0%, and that in the >1 year old group was 92.4%(<i>P</i><0.05). In the 1 year old group, 5 eyes were developed, and in the >1 year old group, 1 eye was removed. The average tumor thickness and average base diameter of the remaining eyes after treatment were significantly smaller than those before treatment(<i>P</i><0.05). The mean thickness and mean diameter of base of >1 year old group were smaller than those of 1 year old group(<i>P</i><0.05). Two groups of children had mild nausea and vomiting and other gastrointestinal symptoms and hair loss,1 year old group had 1 case of mild myelosuppression and liver function damage, >1 year old group had 2 cases of mild myelosuppression,1 case of mild liver function damage.<p>CONCLUSION:VEC combined with laser is safe and feasible in the treatment of children with intraocular RB. >1 year old group is better than 1 year old group.

ABSTRACT

Aortic valve stenosis is a heart disease prevalent in the elderly characterized by valvular calcification, fibrosis, and inflammation, but its exact pathogenesis remains unclear. Previously, aortic valve stenosis was thought to be caused by chronic passive and degenerative changes associated with aging. However, recent studies have demonstrated that atherosclerotic processes and inflammation can induce valvular calcification and bone deposition, leading to valvular stenosis. In particular, the most abundant cell type in cardiac valves, valvular interstitial cells, can differentiate into myofibroblasts and osteoblast-like cells, leading to valvular calcification and stenosis. Differentiation of valvular interstitial cells can be trigged by inflammatory stimuli from several immune cell types, including macrophages, dendritic cells, T cells, B cells, and mast cells. This review indicates that crosstalk between immune cells and valvular interstitial cells plays an important role in the development of aortic valve stenosis.

ABSTRACT

Objective:To establish an HPLC method for the determination of content and entrapment efficiency of HIV-1 virus in-fection factor Vif inhibitor VEC-5 liposomes. Methods:VEC-5 liposomes were prepared by a method of freeze-drying and reconstruc-tion. The separation of free drug from the liposomes was achieved by ultracentrifugation, and an HPLC method was used to determine the content and entrapment efficiency of VEC-5 liposomes. Results:The linear range of VEC-5 was 20-100 μg·ml-1(r=0. 999 0). The average recovery was 100. 25% and RSD was 0. 93%(n=9). The content of three batches of VEC-5 liposomes was 98. 63%, 100. 43% and 102. 65%, respectively within the range of 90%-110%, and the entrapment efficiency was 94. 89%, 93. 68% and 94. 56%, respectively, which was above 90%. Conclusion:The method is accurate and reliable, which can be used to determine the content and entrapment efficiency of VEC-5 liposomes.

ABSTRACT

Objective To observe the changes of serum sialic acid(SA)and in diabetic patients with chronic kidney disease(CKD) and its relation with vascular endothelial function. Methods 97 T2DM patients were divided into <30 mg/24 h group(n=31) ,30~299 mg/24 h group(n=34) and ≥300 mg/24 h group(n=32) according to the level of UAlb/Cr ,meanwhile 30 healthy subjects were selected as normal control(NC) group. The level of serum SA ,nitric oxide (NO) ,endothelin‐1(ET‐1) in four groups were detected. Results Compared with the NC group ,the level of SA and ET‐1 were increased gradually in <30 mg/24 h ,30 ~ 299 mg/24 h and ≥ 300 mg/24 h group ,and the NO were decreased gradually ( P<0.05) .Pearson correlation analysis showed that the SA level was positively related with ET‐1 and negatively with NO(P<0.05). Conclusion The level of SA is increased gradually in the development of CKD ,and the SA level is correlated with NO and ET‐1.

ABSTRACT

Objective To explore the protective effect of atorvastatin on irradiated endothelium and the thrombomodulin(TM)expression.Methods Cultured human coronary artery endothelial cells(HCAEC)and human umbilical vein endothelial cells(HUVEC)were treated by atorvastatin at the final concentration of 10 μmol/ml for 10 min,and then irradiated with 2 and 25 Gy.Cell cycles status and TM expression were quantitatively measured by flow cytometry 24 hours after irradiation.Protein C activation in endothelial cells was also assessod.Results After administration with atorvastatin for 24 h,the TM expression increased by 77%,59% and 61% in normal control group,2 Gy group and 25 Gy group,respectively(t=27.395,26.420,58.065;P=0.000).The protein C levels decreased by 23% and 34% compared with the normal group post-irradiation to 2 and 25 Gy,but increased by 79% and 76% compared with the irradiated control group after administration with atorvastatin.The rates of cell apoptosis decreased by 6% and 16% in 2 Gy and 25 Gy groups,respectively after administration with atorvastatin for 24 h(t=4.178,17.863;P=0.000).Conclusions Atorva statin can protect endothelia cell from irradiation-induced apeptosis by increasing TM expression and protein C activation.

Objective To investigate the antithrombosis effects of Buyang Huanwu Decoction(BHD) and its active fractions on cultured vascular endothelial cells(VEC) following the stimulation of thrombin and their mechanism.Methods The human umbilicus vein endothelial cells(ECV 304) were cultured in media containing 10 U/mL thrombin and different dosese of drugs.The levels of tPA and PAI-1 were detected by ELISA,the expression of TF,TFPI,and TM mRNA was measured by RT-PCR,and the expression of PKC? was examined by immunohistochemical staining 24 h later.Results Compared to the control without any treatment,the release of tPA was increased evidently(P0.05).Compared to the data after the mere stimulus of thrombin,each dose of BHD increased the release of tPA(P0.05);And glycoside(1.25 mg/mL) increased the release of tPA(P

ABSTRACT

Human vascular endothelial cells(VEC)were isolated from umbilicalyeins by trypsin digestion.Spleen cells from BALB/c mice having been im-munized with the isolated endothelial cells were fused with the mousemyeloma cell line SP2/0.After screening and subcloning,three differentmonoclonal antibodies(EC1,EC2,EC3)against human umbilical VEC wereobtained.These antibodies were proved to belong to immunoglobulin sub-classes(IgG_2,IgG_3,IgG_2).EC1 was shown to bind strongly to the vascularendothelial cell membrane,but not to react with peripheral lymphocytes andmonocytes in microcytotoxicity test.We also explored the possible applica-tion of the monoclonal antibodies in VEC typing.
