ABSTRACT

Type 2 diabetes mellitus(T2DM), a common chronic metabolic disease, is often accompanied by internal heat syndrome. Heat-clearing prescriptions are widely used to treat different heat syndromes of T2DM from the aspects of clearing stagnant heat, excess heat, damp heat, phlegm heat, and heat toxin, demonstrating remarkable effects. The mechanism of blood sugar-lowering agents has always been a hotspot of research. Recently, the basic studies of heat-clearing prescriptions from different perspectives have been increasing year by year. To clarify the mechanisms of heat-clearing prescriptions and find specific mechanisms, we systematically reviewed the basic studies of heat-clearing prescriptions commonly used for the treatment of T2DM in the past decade, intending to provide a reference for related research.

ABSTRACT

Functional dyspepsia (FD) is a common and frequently occurring disease in clinic. With the influence of environmental factors, social factors and dietary factors, the incidence rate of FD in the general population is yearly increasing. Traditional Chinese medicine has a long history and far-reaching influence in the treatment of FD. It can prevent and treat FD in the form of multiple-components, targets and channels, with obvious effect and prominent advantages. This article starts with the common syndrome types of FD, and discusses the research progress of single Chinese medicine, effective ingredients and the mechanism of traditional Chinese medicines in treating FD, in order to provide a theoretical basis for the treatment of FD with traditional Chinese medicines.

ABSTRACT

Aim To observe the protective effect of Gancao Xiexin Decoction (GXD) on intestinal epithelial cells (IECs) under endoplasmic reticulum stress from the perspective of PERK-elF2α-CH0P signaling pathway. Methods Caco-2 cells were cultured and divided into normal control group (NC), model control group(MC), low-dose GXD group(GXD-L), mediumdose GXD group (GXD-M) and high-dose GXD group (GXD-H). Models of the stress epithelial cells were induced by tunicamycin(Tm), and GXD groups were treated with GXD contained serum at the same time. The cell survival rate was assessed by CCK-8 method, the cell apoptotic rate and cell cycle distribution were determined by flow cytometry, and the cell barrier permeability was determined by TEER and FITC-dextran method; the expression levels of core proteins of PERK-elF2α-CH0P signaling pathway were detected by Western blot. Results Compared with MC group, GXD intervention could improve cell survival rate(P < 0. 05), reduce their apoptotic rate(P <0. 01), relieve cell cycle arrest(P < 0. 01), improve cell barrier permeability by increasing cell TEER value (P < 0. 01) and decreasing FITC-dextran concentration (P < 0.05), and the levels of p-PERK, p-elF2α, ATF4 and CHOP in GXD-M and GXD-H groups all visibly descended (P < 0. 01). Conclusions GXD can reduce the excessive apoptosis of IECs and protect intestinal epithelial cell barrier homeostasis by inhibiting the signal transduction of PERK-elF2α-CH0P apoptotic pathway.

ABSTRACT

The aim of this paper was to investigate the effect of Banxia Xiexin Decoction(BXD) on inflammatory factors and intestinal flora in a dextran sulfate sodium induced ulcerative colitis(DSS-UC) mouse model, and to explore the mechanism of BXD in treating ulcerative colitis from the perspective of flora disorder. Forty C57 BL/6 J mice were randomly divided into control group, model group and BXD group. A 2.5% DSS-induced ulcerative colitis model was established. On the 8 th day, normal saline, normal saline, and BXD were given daily for 14 days. After 14 days, HE staining was used to observe histopathological changes of the colon. Serum inflammatory factor content was detected by ELISA, and the change of intestinal flora in mice feces was detected by 16 S rRNA sequencing technology. Compared with control group, the colonic tissue of mice in model group was damaged seriously, and the contents of IL-6 and TNF-α in serum were significantly increased(P<0.05). Compared with model group, mice in BXD group had less colonic damage, and the contents of IL-6, TNF-α in serum were decreased significantly(P<0.05). After creation, the richness of Patescibacteria was increased significantly at the phylum level(P<0.05). At the same time, the richness of Faecalibaculum(P<0.01), norank_f_Muribaculaceae(P<0.01) were decreased significantly at the genus level, while the richness of Turicibacter(P<0.01), Romboutsia(P<0.01), Clostridium_sensu_stricto_1(P<0.01) were increased significantly. After the intervention with BXD, the content of Patescibacteria was significantly reduced at the phylum level(P<0.05), and the contents of Lactobacillus(P<0.01), Clostri-dium_sensu_stricto_1(P<0.01), Enterorhabdus(P<0.01), Candidatus_Saccharimonas(P<0.05), Eubacterium_fissicatena_group(P<0.05) were decreased significantly at the genus level, while the contents of Dubosiella, Bacteroides and Allobaculum were increased significantly. Therefore, BXD could significantly improve the symptoms of DSS-UC mice. It not only could reduce the contents of IL-6 and TNF-α, but also could reduce the richness of Patescibacteria at the phylum level, and those of Clostridium_sensu_stricto_1, Candidatus_Saccharimonas, Eubacterium_fissicatena_group at the genus level. Inaddition, BXD could increase the richness of Bacteroides and Bifidobacterium. It suggested that BXD could play a role in the treatment of ulcerative colitis partially through reducing inflammatory factors and regulating the structure of the gut microbiota.

ABSTRACT

OBJECTIVE@#To test the effect of Banxia Xiexin Decoction (, BXD) on the contraction and relaxation of gastric smooth muscle (SM) in diabetic gastroparesis (DGP) model rats, and to explore the mechanism of BXD in the prevention and treatment of DGP through experiments of signal pathway both in vivo and in vitro.@*METHODS@#Sixty Sprague-Dawley rats were divided into 6 groups according to a random number table: control group, model group, high-, medium- and low-dose BXD groups (9.2, 4.6 and 1.8 g/(kg·d), respectively), and domperidone group (10 mg/(kg·d)), 10 rats per group. DGP model was established initially by a single intraperitoneal injection of streptozotocin (STZ), and was confirmed by recording gastric emptying, intestinal transport velocity and gastric myoelectric activity of rats after 2 months. Each group was treated with a corresponding drug for 4 weeks. The mRNA and protein expressions of phospholipase C (PLC), inositol triphosphate (IP@*RESULTS@#Compared with the model group, high- and medium-dose BXD and domperidone significantly increased the expressions of PLC, IP@*CONCLUSIONS@#Treatment with high- and medium-dose BXD significantly attenuated STZ-induced experimental DGP in rats. The therapeutic effect of BXD on DGP rats might be associated with the PLC-IP

ABSTRACT

Objective: HPLC fingerprints of Banxia Xiexin Decoction (BXD) was established to analyze the quality value transmitting relationship between medical herbs-decoction pieces-whole decoction of Scutellariae Radix (SR) and Coptidis Rhizoma (CR). Methods: BXD was prepared by the decoction method recorded in Treatise on Febrile Diseases. BXD HPLC fingerprints was established, and the characteristic peaks were analyzed; The transfer rate, the dry extract rate and the similarity of fingerprint were used as indicators to analyze the quality value transmitting relationship between medical herbs-decoction pieces-whole decoction of SR and CR. Results: The transfer rate of baicalin from medical herbs to decoction pieces was between 81.08% and 119.82%; The transfer rate of baicalin from decoction pieces to BXD was between 44.55% and 59.05%; The transfer rate of berberine from medical herbs to decoction pieces was between 66.65% and 97.51%; The transfer rate of berberine from decoction pieces to BXD was between 7.83% and 22.93%; And the dry extract rate of the whole decoction was between 12.99% and 17.21%. The similarity of fingerprints of 15 batches of BXD ranged from 0.978 to 0.999. Conclusion: The quality evaluation method established by the fingerprint is applicable to the quality control of medical herbs-decoction pieces-whole decoction of SR and CR, which provides some reference for the establishment of the quality evaluation method of famous classic formula.

ABSTRACT

The aim of this paper was to explore the potential molecular mechanism of Banxia Xiexin Decoction in the treatment of colon cancer through pharmacology network and molecular docking methods. The chemical constituents and action targets of 7 herbs from Banxia Xiexin Decoction were collected by using TCMSP database,Chinese Pharmacopoeia and literatures consultation. GeneCards database was used to predict the potential targets of colon cancer. GO biological process analysis and KEGG pathway enrichment analysis of the disease and drug intersection targets were carried out through DAVID database. "Component-target-pathway" network and protein-protein interaction(PPI) network were construction by using Cytoscape and STRING database,and then the core components and targets of Banxia Xiexin Decoction in the treatment of colon cancer were selected according to the topological parameters. Finally, Autodock Vina was used to realize the molecular docking of core components and key targets. The prediction results showed that there were 190 active compounds and 324 corresponding targets for Banxia Xiexin Decoction,involving 74 potential targets for colon cancer. Cytoscape topology analysis revealed 11 key targets such as STAT3,TP53,AKT1,TNF,IL6 and SRC, as well as 10 core components such as quercetin,β-sitosterol,baicalein,berberine,and 6-gingerol.In bioinformatics enrichment analysis, 679 GO terms and 106 KEGG pathways were obtained, mainly involving PI3 K-AKT signaling pathway,TNF signaling pathway and TP53 signaling pathway. The results of molecular docking showed that baicalein,berberine,licochalcone A and 6-gingerol had a high affinity with SRC,STAT3,TNF and IL6. The results suggested that Banxia Xiexin Decoction could play an anti-colon cancer effect by inhibiting cell proliferation, regulating cell cycle, inducing apoptosis and anti-inflammatory function. The study revealed the multi-components,multi-targets and multi-pathways molecular mechanism of Banxia Xiexin Decoction,which could provide scientific basis and research ideas for the clinical application of Banxia Xiexin Decoction and the treatment of colon cancer with compound Chinese medicines.

ABSTRACT

Objective To investigate the effects of the three methods of decocting with deslag, decocting without deslag, and double decocting on the content of nine ingredients baicalin, baicalein, ginsenoside Re, ginsenoside Rb1, monoammonium glycyrrhizinate hydrate, liquiritin, 6-gingerol, berberine hydrochloride, palmatine hydrochloride, and total flavonoids in Banxia Xiexin Decoction (BXD). Methods Nine index components were determined by HPLC. The HPLC analysis was performed on Welch Ultimate XB-C18 column (250 mm × 4.6 mm, 5 μm) with mobile phase of acetonitrile-0.1% phosphate aqueous solution for gradient elution; And carried out at column temperature of 28 ℃, volume flow of 0.9 mL/min, and detection wavelength of 203, 252, 280, and 355 nm. The total flavonoids were determined by colorimetry. Results Nine kinds of ingredients and total flavonoids could be detected in three different decoctions. In the method of decocting with deslag, baicalin, baicalein, ginsenoside Rb1, monoammonium glycyrrhizinate hydrate, and liquiritin increased by 10.01%, 12.88%, 29.09%, 16.75%, and 15.02%, respectively, compared with decocting without deslag; It decreased by 5.54%, 4.15%, 14.49%, 7.85%, and 9.18%, respectively compared with double decocting; Ginsenoside Re, 6-gingerol, berberine hydrochloride, and palmatine hydrochloride increased by 37.90%, 3.78%, 5.33%, and 5.99% compared with decocting without deslag, respectively; compared to the double decocting methods, it increased by 1.07%, 11.57%, 3.41%, and 1.93%. The total flavonoids increased 22.61% higher than decocting without deslag and 6.54% higher than double decocting. Conclusion: The results can effectively reflect the quality difference of different decocting methods. Among the three methods of decoction, the method of decocting without deslag has significantly improved the dissolution of the active ingredients of each component in the decoction, and improve the clinical efficacy of BXD to a certain extent. It provides a good experimental basis for the decocting without deslag method used in Zhang Zhongjing’s Treatise on Febrile Diseases.

ABSTRACT

Objective To evaluate the effects of the classical prescriptions Dahuang-Huanglian-Xiexin decoction combined with conventional western medicine on the lipid metabolism disorder and changes of intestinal microflora in patients with type 2 diabetes mellitus (T2DM). Methods A total of 120 patients who met the inclusion criteria were divided into the two group according to the treatment order, with 60 patients in each group. Both groups were given conventional hypoglycemic therapy, the control group was given bifidobacteria lactobacillus triple viable tablets, and the treatment group was given Dahuang-Huanglian-Xiexin decoction on the basis of the control group. Both groups were treated with 14 days for one course and 4 courses in total. The number of gram-positive coccus (G+c), gram-negative coccus (G-c), gram-positive bacillus (G+b) and gram-negative bacillus (G-b) in each 500 bacteria were observed under oil microscope, and the cocci/bacillus ratio were calculated. The TC and TG were detected by oxidase method, and HDL and LDL were detected by automatic biochemical analyzer. The FPG, 2 hPG and HbA1c were detected by automatic biochemical analyzer. TCM symptom scores were calculated. Results After treatment, the abdominal distension, congestion discomfort, and constipation score were significantly lower than the control group (t values were 3.685, 3.551 and 3.708, respectively, all Ps<0.05); Serum TG, TC and LDL-c levels were significantly lower than the control group (t values were 3.602, 3.581 and 3.421, respectively, all Ps<0.05); HDL-c level was significantly higher than that of the control group (t=3.358, P=0.046); Serum FPG, 2 hPG and HbA1c levels were significantly lower than the control group (t values were 3.502, 3.271 and 3.708, respectively, all Ps<0.05). After treatment, the G-c (271.47/500 ± 22.63/500 vs. 266.81/500 ± 22.36/500, t=3.792), G+b (81.26/500 ± 6.52/500 vs. 73.19/500 ± 6.94/500, t=3.511), G-b (183.76/500 ± 16.19/500 vs. 164.37/500 ± 15.83/500, t=3.306) levels in the treatment group significantly higher than those in the control group (P<0.05). After treatment, both c/b levels was (0.65 ± 0.13 vs. 0.87 ± 0.21, t=3.325) decreased significantly (P<0.05). Conclusions Dahuang-Huanglian-Xiexin decoction combined with conventional western medicine can improve the clinical symptoms of T2DM patients, reduce blood glucose and lipid levels, and inhibit the body mass and improve insulin resistance by adjusting the structure of intestinal flora, increasing probiotics and reducing pathogenic bacteria.

ABSTRACT

Objective To investigate the effect of warm acupuncture combined with Banxia-Xiexin decoction on diabetic gastroparesis (DGP).Methods A total of 86 DGP patients who met the selected criteria were divided into two groups by random number table method.The control group was treated with warm acupuncture.The observation group added Banxia-Xiexin decoction on the basis of the control group.Both groups were treated for 4 weeks.The clinical efficacy of motilin (MTL),somatostatin (SS) and leptin (LP) were evaluated by ELISA.Results The total effective rate of the observation group was 97.7% (42/43) and the control group was 88.4% (38/43).The difference between the two groups was statistically significant (x2=9.492,P<0.01).The post-treatment serum MTL (340.82 ± 51.32 ng/L vs.422.42 ± 64.51 ng/L,t=43.184) and LP (1.98 ± 0.24 ng/ml vs.2.78 ± 0.27 ng/ml,t=5.330) of the observation group were significantly lower than those of the control group (P<0.01),while the serum SS (47.62 ± 7.48 ng/L vs.33.26 ± 5.09 ng/L,t=8.494) was significantly higher than that of the control group (P<0.01).Conclusions The warm acupuncture combined with Banxia-Xiexin decoction can obviously relieve the clinical symptoms of DGP patients,reduce the level of leptin,and there is no obvious adverse reaction.

ABSTRACT

Objective: To observe the effects of the Banxia Xiexin Decoction (BXD) on the intestinal flora and inflammatory factors of diabetic gastroparesis (DGP) rats. Methods: The high fat and sugar feeding combined with intraperitoneal injection of STZ were used for the preparation of DGP rats model, which were randomly divided into model group, the metformin (1.80 g/kg) group, the low (2.55 g/kg), medium (5.10 g/kg), and high (10.20 g/kg) dose group of BXD, and the blank group with 10 rats in each group. Four weeks of continuous ig administration of fasting blood glucose (FBG), gastric residual rate, intestinal propulsion rate, serum D-xylose level, intestinal flora, serum endotoxin, immunoglobulin A, and inflammatory factors were detected. Results: Compared with the blank group, the body weight, intestinal propulsion rate, flora (bacteroides, bifidobacterium, enterococcus, and lactobacilli) amount, IgA, and IL-10 in DGP model were significantly reduced (P < 0.01); FBG, gastric residual rate, serum D-xylose, enterobacter number, endotoxin, and TNF-α were significantly increased (P < 0.01); Compared with model group, high dose BD group of body weight, intestinal propulsion rate, flora (bacteroides, bifidobacterium, enterococcus, and lactobacilli) amount, IgA, and IL-10 level in the low, medium, and high dose group of Banxia Xiexin decoction were increased significantly (P < 0.05, 0.01), FBG, gastric residual rate, serum D-xylose, enterobacter number, endotoxin and TNF-α were significantly decreased (P < 0.05, 0.01). The FBG, intestinal propulsion rate, bacteroides number, and IgA level in the low-dose group were less affected. Conclusion: BXD can effectively adjust the various indexes of DGP model rats, including improving the intestinal flora proportion, reducing endotoxin, inhibiting inflammation factors, and improving the intestinal barrier function, and the potential mechanism may be related to regulation of intestinal bacteria and inflammation factors.

OBJECTIVE: To investigate the effects of different dose compatibility of Scutellaria baicalensis-Rheum palmatum-Coptis chinensis based on Sanhuang Xiexin decoction (shorted for Xiexin decoction) on dissolution rate of baicalin and baicalein, and to provide reference for studying chemical mechanism of TCM compound dose composition compatibility. METHODS: HPLC method was used to determine the contents of baicalin and baicalein. By fixing the dose of S. baicalensis (3 g), using the dose of R. palmatum and C. chinensis as factors, dissolution rate of baicalin and baicalein as response value, two-factor and five-level central composite test was designed. The optimal dose compatibility of S. baicalensis-R. Palmatum-C. chinensis was optimized by response surface method, and compared with the dissolution rate of baicalin and baicalein in classic dose Xiexin decoction (S. baicalensis 3 g, R. Palmatum 6 g, C. chinensis 3 g). RESULTS: When the doses of S. baicalensis, R. palmatum, C. chinensis were 3, 1. 76, 0. 17 g, total dissolution rate of baicalin and baicalein was the highest in extract. The average total dissolution rate of baicalin and baicalein in validation test was 21. 89% (RSD=0. 46%, n=3),and the relative error was 2. 88% with the predicted value of 22. 54%. Compared with the classical dose of Xiexin decoction, the total dissolution rate of baicalin and baicalein in the optimum dose of S. baicalensis-R. Palmatum-C. chinensis in extract was increased by 47. 21%. CONCLUSIONS: The different dose compatibility of S. baicalensis-R. Palmatum-C. chinensis based on Xiexin decoction influence the dissolution of baicalin and baicalein to certain extent. When the doses of S. baicalensis, R. palmatum, C. chinensis are 3, 1. 76, 0. 17 g, dissolution rate of baicalin and baicalein are higher than that of classic dose.

Through the study of the changing decoction of Banxia Xiexin Decoction in Wu Ju-tong's Wen Bing Tiao Bian, this article explored the inheritance and development of the application of Banxia Xiexin Decoction inShang Han Lun. Wu Ju-tong changed the Banxia Xiexin Decoction for treatment of seasonal febrile diseases, who learned ZHANG Zhong-jing's academic thoughts, drew YE Tian-shi's prescription essence in Lin Zheng Zhi Nan Yi An, and combined with his own clinical experience, which widened the treatment scope, enriched the specific treatment methods and sublimated its application theory. ABSTRACT

The most common manifestations of eosinophilic gastroenteritis (EG) are abdominal pain and diarrhea. Glucocorticoid is currently used in the treatment for EG, but the adverse reactions are relatively large. Banxia Xiexin Decoction is the famous Zhongjing prescription can be used to treat many kinds of gastrointestinal diseases. This article introduced. Banxia Xiexin Decoction combined with Shaoyao Gancao Decoction in the treatment of abdominal pain predominant based EG, as well as Banxia Xiexin Decoction combined with Gegen Qinlian Decoction in treating diarrhea predominant based EG have achieved satisfactory efficacy.

ABSTRACT

<p><b>OBJECTIVE</b>To examinie the synergistic effects of Banxia Xiexin Decoction (, Known as Banhasasim-tang in Korean) extract (BXDE) on cisplatin-induced cytotoxicity in the A549 human lung cancer cell lines.</p><p><b>METHODS</b>A549 cells were treated with varying concentrations (50-200 μg/mL) of cisplatin and BXDE alone or in combination for 96 h. We used 1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan assay and flow cytometry to analyze cell viability and apoptosis, respectively.</p><p><b>RESULTS</b>The exposure of cells to cisplatin and BXDE alone or in combination decreased cell viability dose- and time-dependently (P<0.05), which was found to be mediated by the apoptotic pathway as confirmed by the increase in the annexin V/propidium iodide- stained cell population and a ladder pattern of discontinuous DNA fragments. Furthermore, the apoptosis was inhibited by the pan-caspase inhibitor, benzyloxycarbonyl-Val-Ala-Asp (OMe) fluoromethylketone (z-VAD-FMK).</p><p><b>CONCLUSIONS</b>BXDE significantly potentiated apoptotic effects of cisplatin in A549 cells. Moreover, apoptosis induced by BXDE might be the pivotal mechanism mediating its chemopreventative action against cancer.</p>

ABSTRACT

AIM To observe the therapeutic effects of Shengjiang Xiexin Decoction (Zingiberis recens Rhizoma,Zingiberis Rhizoma,Coptidis Rhizoma,etc.) on irinotecan (CPT-11)-induced delayed diarrhea in colorectal carcinoma mice and to discuss its possible action mechanism.METHODS The AOM/DSS-induced female colorectal carcinoma mice were randomly divided into normal group,model group and Shengjiang Xiexin Decoction group.The Shengjiang Xiexin Decoction group was intragastrically administered with Shengjiang Xiexin Decoction,the normal group and the model group were intragastrically administered with normal saline.The diarrhea index and rectum pathologic morphology were measured,and the β-glucuronide activity,IL-15 content and UGT1A1 expression were detected.RESULTS The diarrhea index of Shengjiang Xiexin Decoction group was significantly lower than that of the model group,which might be related to the significant inhibition of β-glucuronide activity,and sig-nificant improvement of IL-15 content and UGT1A1 expression.CONCLUSION Shengjiang Xiexin Decoction shows therapeutic effects on irinotecan-induced delayed diarrhea in AOM/DSS-induced colorectal carcinoma mice.

ABSTRACT

Objective To observe the effect of Banxie Xiexin decoction in the treatment of duodenum ulcer complicating HP infection and its influence on IL-6.Methods Retrospective analysis of 80 patients with duodenum ulcer complicating HP infection were conducted.The patients were tested by rapid urease test for Hp infection.All the patients were randomly divided into observation group (40 cases) and control group(40 cases) according to random number table generated by SPSS19.0.The control group was treated with triple therapy(omeprazole,clarithromycin,amoxicillin).The observation group was given Banxie Xiexin decoction on the basis of the control group,14d as a course.Before and after treatment,the Hp eradication rate was calculated.The epigastric burning,belching tunsuan,dry mouth and limbs without temperature score were measured.5mL fasting blood was exsanguinated to test IL-6.The clinical efficacy was compared.Results The eradication rate of Hp in the observation group was 85%,which was significantly higher than that in the control group(55%).The difference was statistically significant(χ2=8.571,P<0.05).The total effective rate of the observation group was 95%,which was significantly higher than that of the control group(75%).The difference was statistically significant(Z=-3.143,P<0.05).The epigastric burning,belching tunsuan,dry mouth and limbs without temperature score of the observation group and the control group after treatment were lower than before treatment(t=7.167 and 5.664,6.919 and 5.713,7.462 and 5.051,7.372 and 5.136,all P<0.05),which of the observation group after treatment were lower than those of the control group(t=4.501,4.464,4.388,4.506,all P<0.01).The IL-6 levels of the observation group and the control group after treatment were (184.3±92.2)pg/mL and (283.6±102.8)pg/mL respectively,which were significantly lower than before treatment(t=14.332,6.325,all P<0.05).The IL-6 level of the observation group after treatment was significantly lower than the control group(t=7.254,P<0.05).Conclusion Banxia Xiexin decoction can improve the clinical curative effect of triple therapy in the treatment of duodenum ulcer complicating HP infection,improve the clinical symptoms,improve the eradication rate of Hp and restrain the local inflammatory factors,so as to relieve mucosal injury.

ABSTRACT

OBJECTIVE:To observe the effect of Banxia Xiexin decoction on nuclear transcription factor κB p65 (NF-κB p65),NF-κB inhibitory protein α(IκB-α)and Toll-like receptor 4(TLR4)in colon tissue of rats with ulcerative colitis(UC),and explore the possible mechanism for UC treatment. METHODS:Rats were randomly divided into normal group (normal saline), model group (normal saline),Sulfasalazine enteric coated tablet (SASP,positive group,0.3 g/kg),Banxia Xiexin decoction low-dose,medium-dose,high-dose groups (3.9,7.8,11.7 g/kg),8 in each group. Except for normal group,other groups were used trinitrobenzene sulfonic acid-ethanol method to reduce UC model. After modeling,they were administrated,ig,once a day, for 3 weeks. After administration,NF-κB p65,IκB-α,TLR4 mRNA and protein expressions in colon tissue of rats in each group were detected. RESULTS:Compared with normal group,NF-κB p65,IκB-α,TLR4 mRNA and protein expressions in colon tissue of rats in other groups were significantly increased(P<0.05). Compared with model group,NF-κB p65,IκB-α,TLR4 mRNA and protein expressions in colon tissue of rats in each administration group were significantly decreased (P<0.05);showing certain dose-dependent,and the decreasing degree of above indexes in colon tissue of rats in Banxia Xiexin decoction high-dose group were higher than SASP group (P<0.05). CONCLUSIONS:Banxia Xiexin decoction can down-regulate the NF-κB p65,IκB-α, TLR4 mRNA and protein expressions in colon tissue of UC rats,which may be one of the mechanisms for UC treatment.

Sanhuang Xiexin Decoction is one of the ancient decoctions, which has thousands years of history and has been still used nowadays. It is widely used in the diseases which are caused by exuberant fire in clinicl. Sanhuang Tablets and Yiqing Granules which are derived from Sanhuang Xiexin Decoction have become standing drugs in Chinese families. This paper reviewed the research reports on Sanhuang Xiexin Decoction for recent 50 years, analyzed the researching characteristics and superiorities of Sanhuang Xiexin Decoction, and found out the key links and core problems. The aim of this paper is to provide the references for the research of modernization and development of Sanhuang Xiexin Decoction.
