ABSTRACT

Background and Objectives@#The benefits of rapid on-site evaluation (ROSE) of endoscopic ultrasound-guided fine needle aspiration (EUS-FNA) of solid masses have not been convincingly shown in large, randomized trials. New equipment using EUS-guided fine needle biopsy (FNB) allows for more material to be acquired that may obviate the need for ROSE. This study aimed to evaluate if EUS-FNB without ROSE was non-inferior to EUS-FNA with ROSE in solid pancreatic masses (SPMs). @*Methods@#Patients with SPMs requiring tissue sampling were randomly assigned to undergo either EUS-FNA with ROSE or EUS-FNB without ROSE. The touch-imprint cytology technique was used to perform ROSE. The primary endpoint was diagnostic accuracy and secondary endpoints were specimen quality, complication rates, and procedure time. @*Results@#Seventy-eight patients were randomized and analyzed (39 EUS-FNA with ROSE and 39 EUS-FNB without ROSE). Non-significantly different diagnostic accuracies were noted in both groups (97% with ROSE and 100% without ROSE, P < 0.371). The bloodiness of histologic samples and complication rates were not significantly different between groups. A significantly shorter mean sampling procedural time was noted for EUS-FNB over EUS-FNA with ROSE (30.4 ± 10.4 vs 35.8 ± 9.8 minutes, P < .02). @*Conclusions@#EUS-FNB demonstrated equal diagnostic accuracy with shorter procedure times in evaluating SPMs compared to EUS-FNA with ROSE. These new-generation FNB needles may obviate the need for ROSE.

ABSTRACT

@#The standardized workflow of computer-aided static guided implant surgery includes preoperative examination, data acquisition, guide design, guide fabrication and surgery. Errors may occur at each step, leading to irreversible cumulative effects and thus impacting the accuracy of implant placement. However, clinicians tend to focus on factors causing errors in surgical operations, ignoring the possibility of irreversible errors in nonstandard guided surgery. Based on the clinical practice of domestic experts and research progress at home and abroad, this paper summarizes the sources of errors in guided implant surgery from the perspectives of preoperative inspection, data collection, guide designing and manufacturing and describes strategies to resolve errors so as to gain expert consensus. Consensus recommendation: 1. Preoperative considerations: the appropriate implant guide type should be selected according to the patient's oral condition before surgery, and a retaining screw-assisted support guide should be selected if necessary. 2. Data acquisition should be standardized as much as possible, including beam CT and extraoral scanning. CBCT performed with the patient’s head fixed and with a small field of view is recommended. For patients with metal prostheses inside the mouth, a registration marker guide should be used, and the ambient temperature and light of the external oral scanner should be reasonably controlled. 3. Optimization of computer-aided design: it is recommended to select a handle-guided planting system and a closed metal sleeve and to register images by overlapping markers. Properly designing the retaining screws, extending the support structure of the guide plate and increasing the length of the guide section are methods to feasibly reduce the incidence of surgical errors. 4. Improving computer-aided production: it is also crucial to set the best printing parameters according to different printing technologies and to choose the most appropriate postprocessing procedures.

ABSTRACT Background: Reports of the impact of the 2020 COVID-19 pandemic period/2020 on endoscopic ultrasound (EUS) are scarce. Objective: We analyzed the impact of the pandemic period/2020 on the demographics, indications, and number of diagnostic EUS (D-EUS) and interventional EUS (I-EUS) procedures performed in a high-volume endoscopy unit compared with the previous non-pandemic period/2019. Methods: We retrospectively reviewed the medical records of all patients undergoing D-EUS or I-EUS from March 1, 2019, to February 29, 2020 (non-pandemic period/2019) and from March 1, 2020, to February 28, 2021 (pandemic period/2020). Data compared between the study periods included sex, age, comorbidities, EUS findings and diagnosis, need for interventional procedures during EUS, and adverse events (AEs). Results were significant at P<0.05. Results: EUS procedures decreased from 475 in the non-pandemic period/2019 to 289 in the pandemic period/2020, accounting for a 39% reduction. In non-pandemic period/2019, 388 (81.7%) D-EUS and 88 (18.5%) I-EUS were performed, against 206 (71.3%) D-EUS and 83 (28.7%) I-EUS in pandemic period/2020 (P=0.001). Only 5/289 (1.7%) patients had COVID-19. Fewer patients with comorbidities underwent EUS during pandemic period/2020 due to lockdown measures (P<0.001). D-EUS decreased, whereas I-EUS increased (P<0.001). EUS-guided tissue acquisition (EUS-TA) was the most common I-EUS, performed in 83/289 (28.7%) patients in pandemic period/2020, against 88/475 (18.5%) in non-pandemic period/2019 (P=0.001). AEs did not differ significantly between the study periods. Conclusion: Pandemic Period/2020 had a moderate impact on reducing EUS procedures due to the risks involved. Although I-EUS increased, EUS-related AEs did not. Solid and cystic pancreatic tumors remained a major indication for EUS-TA even during the pandemic period/2020.


RESUMO Contexto: Os dados sobre o impacto da pandemia de COVID-19 2020 na ultrassonografia endoscópica (EUS) são escassos. Objetivo: Analisamos o impacto do período pandêmico/2020 na demografia, indicações e número das EUS diagnósticas (D-EUS) e intervencionistas EUS (I-EUS) realizados em uma unidade de endoscopia de alto volume e os comparamos com o período imediatamente anterior não-pandêmico/2019. Métodos: Revisamos retrospectivamente os prontuários de todos os pacientes submetidos a D-EUS ou I-EUS de 1 de março de 2019 a 29 de fevereiro de 2020 (período não-pandêmico/2019) e de 1º de março de 2020 a 28 de fevereiro de 2021 (período da pandemia/2020). Comparamos os dados entre os períodos do estudo incluímos o sexo, idade, comorbidades, achados e diagnóstico da EUS, necessidade de procedimentos intervencionistas durante a EUS e a ocorrência de eventos adversos (EAs). Os resultados foram significativos com P<0,05. Resultados: O número de ecoendoscopias diminuíram de 475 no período não-pandêmico/2019 para 289 no período pandêmico/2020, representando uma redução de 39%. No período não-pandêmico/2019 e pandêmico/2020 foram realizados 388 (81,7%) D-EUS e 88 (18,5%) I-EUS, contra 206 (71,3%) D-EUS e 83 (28,7%) I-EUS, respectivamente (P=0,001). Apenas 5/289 (1,7%) pacientes tinham COVID-19. Menos pacientes com comorbidades realizaram EUS durante o período pandêmico/2020 devido as medidas de bloqueio (P<0,001). D-EUS diminuiu, enquanto I-EUS aumentou (P<0,001). A EUS associada a aquisição tecidual (EUS-AT) foi a I-EUS mais comum, realizada em 83/289 (28,7%) pacientes no período pandêmico/2020, versus 88/475 (18,5%) no período não-pandêmico/2019 (P=0,001). Os EAs não diferiram significativamente entre os períodos do estudo. Conclusão: O período da pandemia/2020 teve impacto moderado na redução da EUS devido aos riscos envolvidos. Embora o I-EUS tenha aumentado, os EAs relacionados ao EUS não aumentaram. Os tumores pancreáticos sólidos e císticos permaneceram como uma das principais indicações para EUS-AT mesmo durante o período pandêmico/2020.

ABSTRACT

This case study was conducted on the needs of Arabic writing skills AWS for Ugandan Adults Learners UAL using the descriptive analytical method where the sample sized (n=40) was used. The researchers sought to answer, among others, a major research question about how important the AWS from UAL viewpoint are. It attempted to identify the signi?cancy of AWS from UAL viewpoint so that the researcher could engage more relevant design of Arabic writing sub- skills' content alternatives for elementary Arabic learning course EALC. The valid and reliable questionnaire namely, “The Elementary Arabic Learning Analysis Scales EALAS for UAL” with 5 Likert scales was used. On one hand, the descriptive ?ndings assert that UAL ratings for AWS interests are signi?cantly high. On another hand, the correlational ?ndings yielded by this survey indicate that the natural relationships among AWS are signi?cant. They also indicate that there is a positive natural relationship among AWS irrespective of any instructional condition. This is clear manifesto that justi?ed the feasibility of structuring EALC for UAL in which the samples of lessons are presented using AWS.

Este artigo analisa a singularidade linguística e psíquica de dois bebês, com desfecho de linguagem distintos, aos dois anos. Para análise, utilizaram-se a entrevista inicial e continuada, os Indicadores Clínicos de Referência/Risco ao Desenvolvimento Infantil (IRDI), os Sinais Enunciativos de Aquisição da Linguagem (SEAL), além de filmagens da interação entre mãe e bebê. Os resultados indicaram que em apenas um dos casos houve superação do sofrimento psíquico e atraso na linguagem a partir do segundo ano de vida. Tanto o exercício das funções parentais quanto às condições dos bebês são fatores importantes em sua constituição psíquica e linguística, o que traz reflexões interdisciplinares importantes acerca da singularidade na relação entre esses dois aspectos do desenvolvimento infantil


Este artículo analiza la unicidad lingüística y psíquica de dos bebés con un resultado distinto del lenguaje a los dos años. Para el análisis se utilizó la entrevista inicial y continuada, los Indicadores Clínicos de Referencia/Riesgo para el Desarrollo Infantil (IRDI), los Signos Enunciativos de Adquisición del Lenguaje (SEAL), así como imágenes de la interacción entre madre y bebé. Los resultados indicaron que solo uno de los casos había superado la angustia psicológica y el retraso del lenguaje a partir del segundo año de vida. Tanto el ejercicio de las funciones parentales como las condiciones de los bebés son factores importantes en su constitución psíquica y lingüística, lo que trae importantes reflexiones interdisciplinarias sobre la singularidad de la relación entre estos dos aspectos del desarrollo infantil.


This article seeks to investigate the linguistic and psychic uniqueness of two babies with distinct language outcome at two years. For analysis, the initial and continued interview, the Clinical Indicators of Reference/Risk to Child Development (IRCD), the Enunciative Signs of Language Acquisition (SEAL) were used, as well as footage of the interaction between mother and baby. The results indicated that only one of the cases had overcome psychological distress and language delay from the second year of life. Both the exercise of parental functions and the conditions of babies are important factors in their psychic and linguistic constitution, which brings important interdisciplinary reflections on the uniqueness in the relationship between these two aspects of child development


Cet article cherche à enquêter sur l'unicité linguistique et psychique de deux bébés, en considérant les axes structurels de la constitution du psychisme et la participation de la mère et du bébé dans les protoconversations initiales. Pour l'analyse, l'entretien initial et continu, les indicateurs cliniques de référence/risque pour le développement de l'enfant (IRDE), les signes énonciatifs de l'acquisition du langage (SEAL) ont été utilisés, ainsi que des images de l'interaction entre la mère et le bébé. Les résultats ont indiqué qu'un seuldes cas avait surmonté une détresse psychologique et un retard de langage dès la deuxième année de vie. Tant l'exercice des fonctions parentales que les conditions des bébés sont des facteurs importants dans leur constitution psychique et linguistique, cequi amène d'importantes réflexions interdisciplinaires sur le caractère unique de la relation entre ces deux aspects du développement de l'enfant

ABSTRACT

Abstract Nicotine addiction leads to in a huge burden on public health and the economy worldwide. Resveratrol (3,5,4'-tetrahydroxystilbene) is the most well-known polyphenolic stilbenoid. Resveratrol was shown to exhibit positive effects on numerous mechanisms that are important for drug and substance addiction. Thus, this study aimed to examine the effect of resveratrol on nicotine addiction. Intraperitoneal (i.p.) treatment with nicotine (0.5 mg/kg) significantly enhanced time spent in the nicotine-paired compartment. Resveratrol (50 and 75 mg/kg, i.p.) and varenicline (2 mg/kg, i.p.) co-administered with nicotine during the 3-day conditioning period effectively diminished the acquisition of nicotine-induced conditioned place preference (CPP). On the other hand, the administration of resveratrol (50 and 75 mg/kg, i.p.) and varenicline (2 mg/kg, i.p.) decreased the low dose (0.1 mg/kg, i.p.) nicotine-induced reinstatement. The results suggest that resveratrol and varenicline inhibit the acquisition and reinstatement of nicotine's reward properties. Resveratrol displayed similar results in the CPP phases as obtained with the reference drug varenicline. In conclusion, resveratrol could be beneficial as an adjuvant pharmacotherapy for nicotine addiction; however, more investigation is needed to completely explain this property.

ABSTRACT

Monitoring of host cell proteins(HCPs)during the manufacturing of monoclonal antibodies(mAb)has become a critical requirement to provide effective and safe drug products.Enzyme-linked immunosor-bent assays are still the gold standard methods for the quantification of protein impurities.However,this technique has several limitations and does,among others,not enable the precise identification of pro-teins.In this context,mass spectrometry(MS)became an alternative and orthogonal method that de-livers qualitative and quantitative information on all identified HCPs.However,in order to be routinely implemented in biopharmaceutical companies,liquid chromatography-MS based methods still need to be standardized to provide highest sensitivity and robust and accurate quantification.Here,we present a promising MS-based analytical workflow coupling the use of an innovative quantification standard,the HCP Profiler solution,with a spectral library-based data-independent acquisition(DIA)method and strict data validation criteria.The performances of the HCP Profiler solution were compared to more con-ventional standard protein spikes and the DIA approach was benchmarked against a classical data-dependent acquisition on a series of samples produced at various stages of the manufacturing process.While we also explored spectral library-free DIA interpretation,the spectral library-based approach still showed highest accuracy and reproducibility(coefficients of variation<10%)with a sensitivity down to the sub-ng/mg mAb level.Thus,this workflow is today mature to be used as a robust and straightforward method to support mAb manufacturing process developments and drug products quality control.

ABSTRACT

Data-independent acquisition (DIA) is a high-throughput, unbiased mass spectrometry data acquisition method which has good quantitative reproducibility and is friendly to low-abundance proteins. It becomes the preferred choice for clinical proteomic studies especially for large cohort studies in recent years. The mass-spectrometry (MS)/MS spectra generated by DIA is usually heavily mixed with fragment ion information of multiple peptides, which makes the protein identification and quantification more difficult. Currently, DIA data analysis methods fall into two main categories, namely peptide-centric and spectrum-centric. The peptide-centric strategy is more sensitive for identification and more accurate for quantification. Thus, it has become the mainstream strategy for DIA data analysis, which includes four key steps: building a spectral library, extracting ion chromatogram, feature scoring and statistical quality control. This work reviews the peptide-centric DIA data analysis procedure, introduces the corresponding algorithms and software tools, and summarizes the improvements for the existing algorithms. Finally, the future development directions are discussed.

ABSTRACT

【Objective】 To conduct a case-control study on precocious puberty as an example to introduce the establishment and design of the electronic Data capture and management platform using Research Electronic data Capture (REDCap) system and support the development of clinical research. 【Methods】 Based on the clinical REDCap system, the case-control research project of precocious puberty was created, the case report forms were designed, the user rights were set, and the data quality control rules were formulated. 【Results】 We established the electronic data capture and management platform for our research, which had 15 case report forms, to collect the data of the participants, including sociodemographic information, time for rest and activities, diet, exposure to environmental internal-secretion interfering-substances, physical examination and biochemical indicators. We conducted project management by setting up features such as user permissions and workgroups, and added data quality verification rules to control data quality. The data could be exported in various file formats for analysis and sharing. 【Conclusion】 The application of REDCap to establish the data capture and management platform of precocious puberty case-control study has promoted the efficient implementation of clinical research, which can be further popularized and applied to clinical researches in other fields.

【Objective】 To investigate the knowledge acquisition status for blood transfusion of transfusion related medical staff in underdeveloped cities in western China and explore its influencing factors. 【Methods】 A questionnaire consisted of blood transfusion laws and regulations, clinical blood transfusion theory and blood transfusion technology was designed, randomly distributed to medical staff and blood transfusion departmenttechnicians of 17 secondary/tertiary hospitals in Wuwei and then collected on the spot. The knowledge acquisition of blood transfusion of each group was compared using statistical description method, and its influencing factors were analyzed by multivariate logistic regression analysis. 【Results】 A total of 507 questionnaires were issued, and 498 valid questionnaires (98.22%) were collected. The scores of transfusion related laws and regulations, blood transfusion theory and blood transfusion technology of doctor group(n=158), nurse group(n=239) and transfusion technician group(n=101)were 4.56-5.97(5.06±0.73)(P<0.01) vs 4.23-5.87(4.98±1.24)(P<0.01) vs 3.71-0.78 (4.15±1.34), 3.67-5.02(4.27±1.02) vs 3.76-5.12(4.06±0.75) vs 4.71-5.98(5.16±0.64)(P<0.01) and 3.41-5.76(3.82±0.56) vs 3.78-5.24(4.01±0.56) vs 3.77-5.46(3.82±0.59). Among the seven departments, blood transfusion department(n=51) won the highest score of above three types of knowledge [4.91-5.97(5.28±0.43) vs 5.03-5.92(5.36±0.59) vs 4.39-5.77(4.97±0.79)(P<0.01) ]. Univariate logistic regression analysis showed that age, occupation, professional titles, training times and hospital grade had an impact on the degree (score) of blood transfusion knowledge acquisition (P<0.05), and multivariate unconditional logistic regression analysis indicated that training times was an important influencing factor(P<0.01). 【Conclusion】 This survey revealed that the level of knowledge acquisition for blood transfusion among medical staff in Wuwei is generally low, and there is a significant difference between staff from hospitals of different grade and different departments. It is urgent to strengthen the training of blood transfusion for medical staff in western China.

ABSTRACT

The gait acquisition system can be used for gait analysis. The traditional wearable gait acquisition system will lead to large errors in gait parameters due to different wearing positions of sensors. The gait acquisition system based on marker method is expensive and needs to be used by combining with the force measurement system under the guidance of rehabilitation doctors. Due to the complex operation, it is inconvenient for clinical application. In this paper, a gait signal acquisition system that combines foot pressure detection and Azure Kinect system is designed. Fifteen subjects are organized to participate in gait test, and relevant data are collected. The calculation method of gait spatiotemporal parameters and joint angle parameters is proposed, and the consistency analysis and error analysis of the gait parameters of proposed system and camera marking method are carried out. The results show that the parameters obtained by the two systems have good consistency (Pearson correlation coefficient r ≥ 0.9, P < 0.05) and have small error (root mean square error of gait parameters is less than 0.1, root mean square error of joint angle parameters is less than 6). In conclusion, the gait acquisition system and its parameter extraction method proposed in this paper can provide reliable data acquisition results as a theoretical basis for gait feature analysis in clinical medicine.

Abstract Objectives: To evaluate the acquisition of surgical skills by otolaryngology residents and established the minimum number of dissections of a lamb's model to be performed before practicing on human patients. Methods: Nineteen second-year otolaryngology residents performed ten dissections each, five on each nasal cavity, always practicing the same three surgical procedures on the lamb model. Each student's training lasted 2-months, and the entire training intervention lasted 4-years, over four generations of residents. All dissections were recorded and were selected at random for examination by two independent otolaryngology surgeons, who were otherwise not involved in the research. Assessment of the 190 dissections used an instrument validated for surgical training of medical residents. Results: To a 1% significance level, statistical analysis revealed increased performance and satisfactory results were observed after the sixth dissection. Furthermore, after the eighth dissection, skill acquisition was relevant and sustained. Conclusion: Training in endoscopic nasal surgery on a lamb's head model improves surgical skills and handling of surgical instruments. Our results showed the relevance of the lamb model for training in otolaryngology surgery, impacting on patient safety. Level of evidence: 2.

ABSTRACT

ObjectiveTo investigate the value of remote consultation of heart sound acquisition in screening and referral of neonates with congenital heart diseases (CHD) in primary hospitals. MethodsA total of 4 030 neonates with non-critical diseases were selected. They were born in Shanghai Pudong New Area Maternal and Child Health Hospital from November 5, 2019 to March 31, 2021. After birth, routine cardiac auscultation was performed and remote consultation of heart sound collection were performed at the same time in combination with percutaneous oxygen saturation measurement to screen CHD. The children with any positive screening index were advised to verify the diagnosis by cardiac ultrasound examination in Shanghai Children's Medical Center. The diagnostic value of different screening methods was compared. ResultsA total of 110 cases were detected positive by routine screening. Among them, 16 cases were lost to follow-up, and 46 cases were confirmed by cardiac ultrasound examination, with a positive diagnosis rate of 48.94% (46/94). A total of 51 cases were detected positive by routine screening and remote consultation of heart sound collection simultaneously. Among them, 42 cases were confirmed by cardiac ultrasound examination, with a positive diagnosis rate of 82.35% (42/51). The difference between the two positive diagnosis rates was statistically significant (P<0.001). ConclusionRemote consultation of heart sound acquisition on the basis of routine neonatal CHD screening can effectively improve the positive diagnosis rate of CHD screening in primary hospitals, and reduce unnecessary referrals. This method is simple and feasible. It has practical value in primary hospitals that lack professional technicians for the diagnosis and treatment of CHD.

ABSTRACT

A high-precision human metabolic measurement system is designed. The system uses STM32F103 as the main control chip to acquire oxygen, carbon dioxide and flow signals to calculate four quantitative indicators: oxygen consumption(VO2), carbon dioxide production(VCO2), respiratory entropy(RQ) and resting energy metabolism(REE), and finally uses an upper computer to display the calculation results.In this paper, the signal acquisition circuit design was carried out for the oxygen sensor, carbon dioxide sensor and flow sensor, and the validity of the device was verified with the American machine MGCDiagnositcs using Bland-Altman analysis method, and the results showed that the four parameters of VO2,VCO2, RQ and REE of both devices fell in the agreement interval of more than 95%. The device thus provides accurate metabolic measurements and offers an effective tool for the field of general health and clinical nutrition support in China.

ABSTRACT

Objective To identify the differentially expressed proteins in different liver tissues in the mouse model of alveolar echinococcosis using high-resolution mass spectrometry with data independent acquisition (DIA), and to identify the key proteins contributing to the pathogenesis of alveolar echinococcosis. Methods Protoscoleces were isolated from Microtus fuscus with alveolar echinococcosis and the experimental model of alveolar echinococcosis was established in female Kunming mice aged 6 to 8 weeks by infection with Echinococcus multilocularis protoscoleces. Mice were divided into the experimental and control groups, and animals in the experimental group was injected with approximately 3 000 protoscoleces, while mice in the control group were injected with the same volume of physiological saline. Mouse liver specimens were sampled from both groups one year post-infection and subjected to pathological examinations. In addition, the lesions (the lesion group) and peri-lesion specimens (the peri-lesion group) were sampled from the liver of mice in the experimental group and the normal liver specimens (the normal group) were sampled from mice in the control group for DIA proteomics analysis, and the differentially expressed proteins were subjected to bioinformatics analysis. Results A total of 1 020 differentially expressed proteins were identified between the lesion group and the normal group, including 671 up-regulated proteins and 349 down-regulated proteins, and 495 differentially expressed proteins were identified between the peri-lesion group and the normal group, including 327 up-regulated proteins and 168 down-regulated proteins. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis revealed that these differentially expressed proteins were involved in peroxisome, peroxisome proliferator-activated receptor (PPAR) and fatty acid degradation pathways, and the peroxisome and PPAR signaling pathways were found to correlate with liver injury. Several differentially expressed proteins that may contribute to the pathogenesis of alveolar echinococcosis were identified in these two pathways, including fatty acid binding protein 1 (Fabp1), Acyl-CoA synthetase long chain family member 1 (Acsl1), Acyl-CoA oxidase 1 (Acox1), Enoyl-CoA hydratase and 3-hydroxyacyl CoA dehydrogenase (Ehhadh) and Acetyl-Coenzyme A acyltransferase 1B (Acaa1b), which were down-regulated in mice in the experimental group. Conclusion A large number of differentially expressed proteins are identified in the liver of the mouse model of alveolar echinococcosis, and Fabp1, Acsl1, Acox1, Ehhadh and Acaa1b may contribute to the pathogenesis of alveolar echinococcosis.

ABSTRACT

Objective To identify the differentially expressed proteins in different liver tissues in the mouse model of cystic echinococcosis (CE), so as to provide insights into the research and development of therapeutic drugs targeting CE. Methods Female Kunming mice at ages of 6 to 8 weeks were randomly assigned into the CE group and the control group. Mice in the CE group were intraperitoneally infected with 2 000 Echinococcus multilocularis protoscoleces, while mice in the control group were injected with the same volume of physiological saline. All mice in both groups were sacrificed after breeding for 350 d, and the lesions (the lesion group) and peri-lesion specimens (the peri-lesion group) were sampled from the liver of mice in the CE group and the normal liver specimens (the normal group) were sampled from mice in the control group for data independent acquisition (DIA) proteomics analysis, and the differentially expressed proteins were subjected to Gene Ontology (GO) term enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis. Results A total of 26 differentially expressed proteins were identified between the lesion group and the normal group and between the peri-lesion group and the normal group, including 8 up-regulated proteins and 18 down-regulated proteins. GO term enrichment analysis showed that these differentially expressed proteins were predominantly enriched in endoplasmic reticulum membrane (biological components), oxidoreductase activity (molecular function) and oxoacid metabolic process and monocarboxylic acid metabolic process (biological processes). KEGG pathway enrichment analysis revealed that the differentially expressed protein Acyl-CoA oxidase 1 (Acox1), which contributed to primary bile acid biosynthesis during the fatty acid oxidation, was involved in peroxisome signaling pathway, and the differentially expressed protein fatty acid binding protein 1 (Fabp1), which contributed to fatty acid transport, was involved in the peroxisome proliferator-activated receptor (PPAR) signaling pathway. Conclusion Differentially expressed proteins are identified in the liver specimens between mouse models of CE and normal mice, and some differentially expressed proteins may serve as potential drug targets for CE.

RESUMO Objetivo Analisar a validade de conteúdo e de construto de sinais enunciativos de aquisição da linguagem para crianças de 13 a 24 meses. Método Os sinais criados foram submetidos à avaliação de clareza e pertinência por seis juízes experts em aquisição da linguagem na perspectiva enunciativa. A partir do seu julgamento foi produzida uma versão experimental que foi aplicada na análise de vídeos de interações mãe-bebê/examinador, com duração de 15 minutos, de 77 díades mãe-bebê na faixa etária de 13 a 18 meses e 89 díades na faixa etária de 19 a 24 meses. A validação de fidedignidade e consistência interna foi realizada pela análise de 10% da mostra por dois juízes fonoaudiólogos. A validação do construto foi realiza pela análise fatorial efetivada sobre a amostra total. Os dados foram analisados no Statistica 9.1 e PASW 17.0. Resultados Todos os sinais foram considerados claros e pertinentes pelos juízes experts. A análise de fidedignidade apontou a concordância quase perfeita (0,8 ≤ Kappa ≥ 1,0) para a maior parte dos sinais. A consistência interna para a Fase 3 apresentou alpha=0,771 considerada alta e Fase 4 apresentou alpha=0,917 limítrofe a muita alta. A análise fatorial da fase 3 revelou 2 fatores, explicando 59,1% da variância total e a fase 4 revelou 1 fator, explicando 75,7%. Conclusão Observou-se validade de conteúdo e de construto para cinco dos doze sinais na Fase 3 e para todos sinais na fase 4.


ABSTRACT Purpose To analyze the content and construct validity of enunciative signs of language acquisition for children aged 13 to 24 months. Methods The signs created were subjected to an assessment of clarity and relevance by six expert judges in language acquisition from an enunciative perspective. Based on their judgment, an experimental version was produced that was applied to the analysis of videos of mother-baby / examiner interactions, lasting 15 minutes, of 77 mother-baby dyads aged 13 to 18 months and 89 dyads in the age range aged between 19 and 24 months. The validity of reliability and internal consistency was performed by analyzing 10% of the sample by two speech therapists. The construct validation was carried out by the factorial analysis carried out on the total sample. The data were analyzed in Statistica 9.1 and PASW 17.0. Results All signs were considered clear and relevant by the expert judges. The reliability analysis showed an almost perfect agreement (0.8 ≤ Kappa ≥ 1.0) for most signs. The internal consistency for Phase 3 showed alpha = 0.771 considered high and Phase 4 presented alpha = 0.917 bordering on very high. The factor analysis of phase 3 revealed 2 factors, explaining 59.1% of the total variance and phase 4 revealed 1 factor, explaining 75.7%. Conclusion Content and construct validity were observed for five of the twelve signs in Phase 3 and for all signs in Phase 4.

RESUMO Objetivo Apresentar e analisar a curva de aquisição segmental do Português Brasileiro em onset simples, complexo e coda. Método Participaram 857 crianças com aquisição fonológica típica, com idades entre 3:0 e 8:11, divididos em faixas etárias de 6 em 6 meses. Os participantes foram avaliados, por meio do software INFONO de avaliação fonológica. Após a análise dos resultados foram elaboradas curvas de aquisição dos segmentos analisados nas diferentes estruturas (onset simples, complexo e coda). Resultados Observou-se que, em onset simples, alguns segmentos estavam adquiridos antes dos 3:0 (plosivas, nasais e fricativas /f, v, s, z/). Os segmentos /ʃ/ e /l/ foram adquiridos aos 3:0, /ʒ/ e /x/ aos 3:6, /ʎ/ aos 4:0, /ɾ/ aos 4:6; em coda, /N/ e /L/ foram adquiridos antes dos 3:0, /S/ aos 3:6 e /ɾ/ aos 4:6; em onset complexo, as estruturas compostas por Fricativa+/ɾ/ e Plosiva+/ɾ/ foram adquiridas aos 5:0, Plosiva+/l/ aos 5:6 e Fricativa + /l/ aos 6:0. Conclusão Analisar a curva de aquisição é fundamental, pois fornece referência sobre a idade de aquisição dos segmentos nas diferentes estruturas silábicas. A curva de aquisição contribui para a identificação precoce de atrasos no processo de aquisição fonológica possibilitando um encaminhamento oportuno para a intervenção fonoaudiológica.


ABSTRACT Purpose To present and analyze the acquisition segmental curve of Brazilian Portuguese in simple and complex onset position and coda position. Methods 857 children with typical phonological acquisition participated in it, aged between 3:0 and 8:11, divided into age groups every 6 months. Participants were assessed using INFONO phonological assessment software. After analyzing the results, acquisition curves were drawn up for the segments analyzed in the different structures (simple and complex onset position and coda position). Results It was noted that, in simple onset position, some segments were acquired before 3:0 (stops, nasal and fricative, /f, v, s, z/. The /ʃ/ and /l/ segments were acquired at 3:0, /ʒ/ and /x/ at 3:6, /ʎ/ at 4:0, /r/ at 4:6; in coda position /N/ and /L/ were acquired before of 3:0, /S/ at 3:6 and /r/ at 4:6; in complex onset position, the structures composed by Fricative + /r/ and Stop + /r/ were acquired at 5:0, Stop + /l/ at 5:6 and Fricative + /l/ at 6:0. Conclusion Analyzing the acquisition curve is essential, as it provides a reference on the age of acquisition of segments in different syllabic structures. The acquisition curve contributes to the early identification of delays in phonological acquisition process enabling a timely referral for speech therapy intervention.

ABSTRACT

RESUMO: O objetivo deste artigo é caracterizar a variabilidade das formas de aquisição da linguagem e analisar os meios de produção verbal, focalizando o papel da interação social. Na contramão de um modelo pautado na descrição de ausências, sintomas e desvios, adotou-se um ponto de vista dinâmico e processual na investigação da linguagem, considerando o sujeito, a sua história, seu meio social e sua condição de desenvolvimento. A pesquisa consistiu no seguimento de cinco crianças com idades entre dois e quatro anos, com queixas relacionadas à aquisição da fala. As sessões foram gravadas em vídeo e os dados foram construídos a partir da descrição do processo aquisicional envolvendo a produção das crianças em situações clínicas. Observou-se que os processos de aquisição da linguagem são entrelaçados com a história de constituição da singularidade dos sujeitos, e que os diversos fatores de caráter orgânico, psíquico e social incidem na trama do desenvolvimento linguístico e na formação da personalidade. Nas análises, chama-se atenção para o mosaico de recursos comunicacionais que se confgura ao confrontarem-se os casos e examinarem-se os componentes aquisicionais que se differenciam e se assemelham. Verificou-se que, para além de alterações de linguagem, se faz necessário focalizar os usos peculiares de recursos da linguagem.


ABSTRACT: The aim of this paper is to characterize the variability in the forms of language acquisition and to analyze the means of verbal production, focusing on the role of social interaction. As opposed to a model based on the description of absences, symptoms, and deviations, we have adopted a dynamic and procedural point of view in the investigation of language, considering the subject, their history, social environment and development condition. The research consisted in the follow-up of fve children aged two to four years with indications related to speech acquisition. The sessions were video-recorded, and the data were constructed based on the description of the acquisition process involving the production of children in a clinical context. We have perceived that language acquisition processes are intertwined with the account in the constitution of singularity of the subjects, and that various organic, psychic, and social factors affect language development and personality formation. In the analyses, we draw attention to the mosaic of communicational resources that is revealed when we confront the cases and examine the acquisition components that are different from each other and that are similar. We have found that, in addition to language alterations, it is necessary to focus on the peculiar uses of language resources.

ABSTRACT

Lipidomics coverage improvement is essential for functional lipid and pathway construction.A powerful approach to discovering organism lipidome is to combine various data acquisitions,such as full scan mass spectrometry(full MS),data-dependent acquisition(DDA),and data-independent acquisition(DIA).Caenorhabditis elegans(C.elegans)is a useful model for discovering toxic-induced metabolism,high-throughput drug screening,and a variety of human disease pathways.To determine the lipidome of C.elegans and investigate lipid disruption from the molecular level to the system biology level,we used integrative data acquisition.The methyl-tert-butyl ether method was used to extract L4 stage C.elegans after exposure to triclosan(TCS),perfluorooctanoic acid,and nanopolystyrene(nPS).Full MS,DDA,and DIA integrations were performed to comprehensively profile the C.elegans lipidome by Q-Exactive Plus MS.All annotated lipids were then analyzed using lipid ontology and pathway analysis.We annotated up to 940 lipids from 20 lipid classes involved in various functions and pathways.The biological in-vestigations revealed that when C.elegans were exposed to nPS,lipid droplets were disrupted,whereas plasma membrane-functionalized lipids were likely to be changed in the TCS treatment group.The nPS treatment caused a significant disruption in lipid storage.Triacylglycerol,glycerophospholipid,and ether class lipids were those primarily hindered by toxicants.Finally,toxicant exposure frequently involved numerous lipid-related pathways,including the phosphoinositide 3-kinase/protein kinase B pathway.In conclusion,an integrative data acquisition strategy was used to characterize the C elegans lipidome,providing valuable biological insights into hypothesis generation and validation.
