ABSTRACT

ObjectiveTo screen the preparation technology of Baoyuan chewable tablets and to preliminarily elucidate its anti-fatigue effect and mechanism. MethodTaking encapsulation rate of volatile oil, extract rate and extraction rate of active ingredients as indexes, single factor test and orthogonal test were used to optimize the volatile oil inclusion, aqueous decoction and formulation molding processes of Baoyuan chewable tablets. ICR rats were randomly divided into the blank group, model group, Gaoshan Hongjingtian oral liquids group(6.01 mL·kg-1) and and Baoyuan chewable tablets low, medium, and high dose groups(2.1, 4.2, 8.4 g·kg-1), 8 mice in each group, and were administered by gastric gavage at the corresponding dose once a day, the blank and model groups were given equal volume of saline for 15 d. After the last administration for 30 min, the mice were loaded with 5% of the body mass of lead at the tail and swam until exhaustion to establish the fatigue model, and the weighted swimming time of the mice in each group was recorded, meanwhile, the muscle tissues of the mice were sliced, stained by hematoxylin-eosin(HE) and subjected to pathological observation, and the levels of blood urea nitrogen(BUN), lactic acid(LA), liver glycogen(LG), activities of lactate dehydrogenase(LDH) and creatine kinase(CK) in the serum were determined. ResultThe optimal inclusion process of cinnamon oil in Baoyuan chewable tablets was 10∶1 for β-cyclodextrin-volatile oil, and inclusion at 50 ℃ for 2 h with saturated aqueous solution method. The optimal water extraction process was to extract twice, adding 10 times of water to extract for 50 min for the first time, and adding 9 times of water to extract for 40 min for the second time. The ratio of the extract of Baoyuan chewable tablets with microcrystalline cellulose, maltodextrin, mannitol, citric acid, magnesium stearate was 63∶13∶8∶17∶17∶1∶1, the tablets were pressed by wet granulation, the each tablet weight was 1.2 g, and the hardness was 60-80 N. Compared with the model group, Baoyuan chewable tablets low, medium, and high dose groups could significantly prolong the exhaustion time of mice in weight bearing swimming(P<0.05, P<0.01), and improve the exercise endurance of the body, and the results of HE staining showed that all dose groups of Baoyuan chewable tablets could significantly improve the muscle tissue damage caused by exercise, significantly reduce the levels of BUN, LA and the activities of LDH and CK in serum(P<0.01), and significantly increase the content of LG(P<0.05, P<0.01). ConclusionThe optimized preparation process of Baoyuan chewable tablets is stable and feasible, and the preparation can improve exercise endurance by increasing the LG level in liver tissue, and relieve muscle soreness by accelerating the removal of LA from the body, and reduce CK and LDH activities to exert anti-fatigue effects.

ABSTRACT

Objective: To develop Duzhong Butiansu Tablet (DBT) for the second time and add the effect on male infertility, so as to provide an experimental basis for the treatment of male infertility. Methods: The male mice were randomly divided into five groups: high, medium, and low dose groups of DBT, control group, and positive drug group. The immune and gonadal organ index, anti-fatigue effect and fecuudity of each group were observed. The male SD rats were randomly divided into six groups: high, medium, and low dose groups of DBT, control group, model group, and positive drug group. The changes of organ index, testicular pathological structure, levels of serum sex hormones and effects of serum oxidative stress and fecuudity were measured. The morphological structure of testes was observed by HE staining. Results: By improving the immune function and sex organ index of young male mice, DBT had the anti-fatigue effect on extending the swimming survival time, which would promote the increase of pregnancy rate of female mice. By increasing the weight of model rats, DBT could increase sexual organs and kidney indexes, improve contents of serum sex hormones FSH, LH, T, and E2 and reduce content of MDA in serum, and has anti-oxidant effect of increasing serum SOD and GSH-Px levels, and improves the testicular pathological damage, thus contributs to increase the pregnancy rate of female rats and the average litter size and production of anti-rat spermatogenic dysfunction induced by adenine effect. Conclusion: DBT can improve the growth and reproductive capacity to reverse the impairment effects on male animal, and provide experimentalbasis for the clinical research.

ABSTRACT

Although a number of studies have reported the anti-fatigue effect of anserine, its evidence seems to be still insufficient except the subjective evaluations by questionnaires. The aim of the present study was to propose a method to evaluate the anti-fatigue effect of anserine by a simple protocol and to verify the effectiveness of the method. Subjects were 17 healthy male volunteers (35.5 ± 5 yr., 75.5 ± 5.0 kg). They performed the isometric exercise tolerance test (ETT) on the rectus femoris muscle twice. Median frequencies (MDFs) of electromyogram and their regression curves were calculated for two ETTs. The angles between these curves were defined as a muscle fatigue index and were compared between anserine and water (control) intakes. The results indicate that anserine can alleviate muscle fatigue in humans. To the best of our knowledge, this is the first direct demonstration of the anti-fatigue effect of anserine on muscle in humans.

ABSTRACT

This study was conducted to investigate the anti-fatigue effect of walnut extract (WE) on forced swimming capacity in mice. Twenty-eight male ICR mice were randomly divided into four groups, a vehicle control (VC) or one of three WE administered groups (300, 600 and 900 mg/kg/day). WE was orally administered to mice once a day for 4 weeks, during which time a forced swimming test was conducted once a week. The vehicle control group was given a corresponding volume of sterile distilled water. After 4 weeks, the forced swimming capacity and levels of blood lactate, glucose, glutamine, ammonia and triacylglycerol, and liver glycogen were measured. In the WE administration group (600 and 900 mg/kg) the maximum swimming time increased significantly when compared with the vehicle control group. WE (600 and 900 mg/kg) significantly decreased the levels of lactate andammonia and increased the blood glutamine levels and liver glycogen content after forced swimming relative to the vehicle control group. The results of this study demonstrated the anti-fatigue effects of WE in a dose-dependent manner. The effects of WE at 600 and 900 mg/kg were similar. Overall, these results suggest that walnut has anti-fatigue activity and could elevate exercise tolerance.
