ABSTRACT

In order to explore the pathological mechanism of perimenopausal syndrome and seek prevention and treatment measures, it is necessary to establish animal models that similar to human perimenopausal syndrome, so as to provide reference for drug research, new drug development and clinical application. In this paper, the keywords of "perimenopausal period" "perimenopausal syndrome" "menopause" "menopausal syndrome""menopausal period" "menopausal syndrome" and "animal" were searched in China National Knowledge Infrastructure (CNKI), Chongqing Weipu, China Biomedical Literature Database (CBM) and Pubmed. In addition, the selection of domestic peripheral menopausal syndrome model animals in recent years and the advantages and disadvantages of corresponding models were summarized. A total of 673 studies were identified, of which 61 were included in the analysis. The most common animal model of perimenopausal syndrome is castration model, while the immunodeficiency model is less used. With the aging of the population and the rapid increase of psychosocial stress, the incidence of perimenopausal syndrome is high. Therefore, it is particularly important to explore the mechanism of perimenopausal syndrome. According to the experimental purpose, experimental period, experimental technology and other factors, the selection of appropriate model animals and modeling methods is the key of the success of the experiment of perimenopausal syndrome.

ABSTRACT

At present, most of the scholars of traditional Chinese medicine (TCM) considered that climacteric syndrome was caused by yin-deficiency,as the therapy of nourishing yin and clearing heat was the most common treatment, neglecting the pathologic role of yang-deficiency in climacteric syndrome. This paper redefined the pathogensis of climacteric syndrome and its syndromes based on differentiation in Gynecology of Traditional Chinese Medicine of various versions and in accordance with previous syndrome epidemiological results over it.The emerge of hot flushes and sweating, forgetful and hyposexuality was prevalent. The inaccurate description of hot flash and sweating in climacteric syndrome was rectified. The therapeutic effects of nourishing yin therapy on vasomotion were discussed based on the understanding of etiology, pathology and treatment of climacteric syndrome in ancient Chinese medical books. The phytoestrogens action of warming kidney-yang drugs was summarized from the aspect of modern pharmacology for exploring the significance of the role of yang-deficiency in climacteric syndrome. The treatment against yang-deficiency would be an innovative clinical pathway of climacteric syndrome.

ABSTRACT

OBJECTIVE: During the transition to menopause, various symptoms including vasomotor symptoms and depressed mood lead to low quality of life. We investigated the effect of low-dose venlafaxine hydrochloride extended release on depressed mood and vasomotor symptoms of perimenopausal women. METHODS: 33 perimenopausal women fulfilling Diagnostic and Statistical Manual of Mental Disorders, fourth edition criteria for a depressive episode were enrolled between January 2014 and May 2014. Subjects were prescribed 37.5 mg/day or 75 mg/day venlafaxine hydrochloride according to clinician's judgement, and the dosages were maintained for 8 weeks. Depressed mood and other psychological difficulties were evaluated by using Hamilton Rating Scale for Depression (HAMD), Hamilton Rating Scale for Anxiety (HAMA), Clinical Global Impression-Severity. Climacteric symptoms including vasomotor symptoms were evaluated by using Greene Climacteric Scale (GCS). For statistical analysis, paired t-test was used. RESULTS: Significant decreases in HAMD, HAMA, GCS scores were observed after 2 weeks of treatment and the trends continued until the end of the study. The scores of HAMD significantly decreased, 28 of them reached remission (HAMD < or =7). The scores on vasomotor symptoms of GCS after 8 week treatment significantly decreased compared to baseline (13.1+/-5.0, p<0.01). CONCLUSION: These results suggest that venlafaxine hydrochloride is effective and tolerable treatment option for vasomotor symptoms and depressed mood in perimenopausal women. To validate our results, furthur studies with double-blind, placebo controlled will be needed.

ABSTRACT

This study was aimed to explore the clinical effect on treatment of climacteric syndrome by the combina-tion of Yi-Shen Yang-Xin Shu-Gan (YSYXSG) decoction and acupuncture therapy. A total of 78 female patients suffering from climacteric syndrome were selected and randomly divided into two groups, with 39 cases in each group. The control group was treated with oral administration of estradiol valerate. The treatment group was treated with the combination of YSYXSG decoction and acupuncture therapy on basis of western medicine treatment. Four-week was one treatment course. The treatment was continued for three treatment courses. Improvement state of both groups were compared by the integration of clinical symptoms before and after treatment as well as the Hamilton's Depression Scale (HAMD)/(HAMA). The results showed that according to the clinical symptoms, both groups had im-provements in varying degrees (P< 0.05, P< 0.01), in which HAMD, HAMA, FSH decreased (P< 0.05), and E2 in-creased (P< 0.05). The treatment group had more obvious superiority compared to the control group. There was sta-tistical significance between two groups (P < 0.05). The total efficiency rate of the treatment group was obviously higher than the control group with statistical significance (P < 0.05). It was concluded that combination of YSYXSG decoction, acupuncture therapy with hormonotherapy had more superiority in improving clinical symptoms, remitting emotion of anxiety-depression and controlling patient's condition compared with pure hormonotherapy, which had su-periority.
