ABSTRACT

Background Resveratrol has been unanimously recognized as an cardiovascular protective substance in red wine. It has been speculated that the anti-atherosclerosis effect of resveratrol is ascribed to its powerful anti-inflammatory effect. Objective To investigate the effects of resveratrol on injured human umbilical veno-endothelial cells (HUVEC) and the reactive oxygen species(ROS) production induced by TNF-? or soluble CD40L (sCD40L). Methods Cultured HUVEC were pre-incubated with resveratrol(1-50 ?mol/L) for 2 hours and then treated with TNF-?(10 ?g/L) or sCD40L?(10 ?g/L) for another 4 hours. MTT assay was used to detect proliterative activity of HUVEC. Immunofluorescence microscopy was used for determination of ROS expression. @#Hepatocyte growth factor(HGF) is a mesenchyme-derived pleiotropic factor which regulates cell growth,cell motility,and morphogenesis of various types of cells,and is thus considered a humoral mediator of epithelial-mesenchymal interactions responsible for angiogenesis,apoptosis and oxidative stress,etc.This article reviewed the relationships between HGF and cardiovascular diseases.

ABSTRACT

@#ObjectiveTo observe the ultrastructures of vessel and endotheliocyte in the early stage(2 h after stimulation finished) after acute mechanical middle cerebral artery(MCA) vasospasm in cats.MethodsThe right MCA of 6 cats were mechanically stimulated using a small smooth stainless steel nail in the field acrossing the olfactory tract for 30 min.The diameter of MCA was recorded by metrical ocular of microscope.The changes of the perfusion index of brain tissue were observed through the Laser Doppler flowmetry monitor fixed on the skull.The ultrastructure of vessel and endotheliocyte were investigated through the electron microscope.ResultsThe diameter of MCA decreased to 68.78% of normal.2 h later,the diameter of MCA recovered.The perfusion index of the cortex surface decreased to 42.59% of normal and up to 61.87% 2 h later.The chromatin of endotheliocyte congregated to the side and formed the lunular body.The heaves of mitochondria(MIT) are blurry 2 hours after the mechanical stimulation.ConclusionThe persistent mechanical stimulation can cause acute cerebral vasospasm,which may involve the apoptosis of endotheliocyte of MCA.It is important to reduce the mechanical stimulus to the cerebral vessel in order to avoid acute cerebral vasospasm during clinical and basic craniotomy.

ABSTRACT

Objective To establish the expression of exogenous tissue plasminogen activator (TPA) gene in human umbilical vein endotheliocytes to provide both theoretical basis and new method for gene therapy of ischemic heart disease and prevention of postoperative vessel re-stenosis. Methods Expression vector pcDNA3 1(+)TPA was constructed, and transfected into human umbilical vein endotheliocytes cultivated in vitro by lyposome. The exogenous TPA expression was observed. Results The expression vector pcDNA3 1(+)TPA could efficaciously express TPA in human umbilical vein endotheliocytes. The protein quantity of TPA in the transfected cells was 568 6ng/10 6cell/24h, when measured by enzyme-linked immunosorbent assay, while that in the non-transfected cells was 17 8ng/10 6 cell/24h. The exogenous TPA's activity in the transfected cells was 108 8IU/10 6cell/24h, when measured by chromogenic substrate assay, while that in the non-transfected cells was 5 6 IU/10 6cell/24h. Conclusion When pcDNA3 1(+)TPA was transfected into human umbilical vein endotheliocytes, exogenous TPA could be expressed efficaciously, which provides theoretical basis and new method for TPA gene therapy of ischemic heart disease.

ABSTRACT

Objective To search for the application of trehalose as cryoprotectant for keeping aortic valve homograft in liquid nitrogen.Methods The aortic valve homografts of Wistar rat were kept in 10%DMSO and divided into 2 groups(control and test group).The test group was added a supplement of trehalose 0.1mol?L~(-1) and all the hemografts were gradually hypothermied and then cryopreserved in-196℃ liquid nitrogen.After 6 months all the cryopreserved valves were thawed.The aortic valve homografts were observed by light and electron microscope.At the same time,the viability of endotheliocyte was measured.Results The viability of endotheliocyte in test group was higher than that in the control group and the structural damage was also much lighter.Conclusion The cryoprotective effect of combined use of 10%DMSO+0.1mol?L~(-1) trehalose was much better than that of 10%DMSO alone.

ABSTRACT

Purpose:To explore the effects and mechanisms o f anti-invasion and anti-metastasis of ginsenoside Rg3 on human hepatocellular carcinoma cells. Methods:To select human hepatocellular carcinoma cell line SMMC -7721 and the human allantoic veins endothelial cell line ECV304 as the study objects. We observed the effet of ginsenoside Rg3’s effect on the growth of SM MC-7721 and ECV304 by MTT methods,the adhesion of SMMC-7721 and Fibronectin by cell adhesion experiment, the expression of gene protein of nm23,CD44 and VEGF by immunohisto-chemical method. Results:Ginsenoside Rg3 could inhibit not only the growth of SM MC-7721 and ECV304 significantly, but also the adhesion of SMMC-7721 and FN. I t might also down-regulate the expression of CD44, VEGF and up-regulate the nm 23 gene expression. Conclusions:Ginsenoside Rg3 can inhibit invasion and metastasis of the hepatic carcinoma cell. The mechanisms are probably that Ginsenoside Rg3 can inhibit the hepatic carcinoma cells’ invasion activity,regulate the expres sion of the gene proteins which are closely related with invasion and metastasis ,inhibit neovasularization.

Angiogenesis is a fundamental physiological and pathological process in human body.Angiogenesis is regulated by integrins,a family of cell surface receptors.After binding with their ligands such as extracellular matrix proteins and immunoglobulin superfamily molecules,integrins can promote the proliferation and migration of endothelial cells during angiogenesis.Several integrin-targeted therapeutic agents are currently used in clinical trials for the therapy of angiogenesis related diseases.Here,there will show the roles of integrins in angiogenesis and clinical application.

ABSTRACT

Objective To determine an optimal method of in vivo endothelialization when homograft valve (HV) leaflets are treated with different methods. Methods Twenty five fresh HV leaflets were divided into five groups randomly. The leaflets of GroupⅠ were fresh HV ones without any treatment; GroupⅡwere cryopreserved in liquid nitrogen for 1 month; Group Ⅲ was treated by DOA; GroupⅣ was treated by 25 g?L -1 glutaraldehyde; Group Ⅴ was first treated by hypotonic/hypertonic, then with an enzyme-based solution. Each HV leaflet was trimmed to 2 mm one which was cultured with lymphocyte from the recipient. In order to compare the antigenicity of HV leaflets, 3H-TdR incorporation was observed after tissue-cell coculture and stimulous index (SI) was calculated. The HV leaflets of different groups were implanted into the sheep's femorofemoropopliteal for three months; then the following procedures were done. Immunohistochemistry of Ⅷ factor correlative antigen was used to identify the cells property; observation of growth condition of EC and superficial morphosis of HV leaflets was made with scanning election microscope (SEM). Results Multiple comparison showed that SI of Groups Ⅱ, Ⅲ, Ⅳ, and Ⅴ differed significantly from Group Ⅰ(P0.05), but Group Ⅲ, Ⅳ, and Ⅴ all had significant differences from GroupⅡ (P
