@#Objective To compare the immunogenicity of varicella-zoster virus(VZV)Oka-7S strain and VZV Oka strain.Methods Female BALB/c mice were subcutaneously immunized with VZV Oka-7S and Oka strains at high,medium and low doses(1 000,250 and 62.5 PFU/dose)for total two doses at an interval of 14 d,separately,with 12 mice in each group.The VZV-specific humoral immunity was evaluated based on fluorescent antibody to membrane antigen assay(FAMA),and the VZV-specific cellular immunity was evaluated by enzyme-linked immunospot assay(ELISPOT).Results Both VZV Oka-7S and VZV Oka strains induced specific humoral and cellular immune responses,and showed considerable immune effects;The level of anti-VZV specific IgG in serum of mice in Oka-7S-2.2 group increased significantly with the increase of immunization times and immunization doses.Conclusion The VZV Oka-7S strain had comparative immunogenicity with VZV Oka strain.

【Objective】 To establish and evaluate a fluorescent antibody to membrane antigen (FAMA) method for detecting antibodies against varicella-zoster virus (VZV) based on Vero E6 cells. 【Methods】 Based on the adapted VZV-Oka-E6 strain that VZV-Oka live attenuated varicella vaccine strain grew in Vero E6 cells, Vero E6 cells were infected with VZV-Oka-E6 of three different doses (104.65, 104.95 and 105.25 TCID50), and the cytopathic effect was observed under a microscope to determine the optimal infection dose of VZV-Oka-E6 strain. Then the detectable sensitivity of the infected cell antigen slides prepared after fixing the infected cells with different fixatives was compared to determine the optimal fixative. As a result, the FAMA method based on Vero E6 cells for the determination of neutralizing anti-VZV has been developed. The established FAMA assay was used to detect the international standard for varicella zoster immunoglobulin with different titers to determine the sensitivity of the assay. The specificity of the assay was evaluated by detecting specific antibodies against human herpes simplex virus (HSV) type 1 and type 2. The neutralizing anti-VZV antibodies of the international standard for varicella zoster immunoglobulin were detected using VZV-infected cell antigen slides prepared in the same batch and four different batches, respectively, to determine the intra-assay repeatability and inter-assay repeatability. The international standard for varicella zoster immunoglobulin with three known titers was detected to evaluate the relative accuracy of this assay. The anti-VZV titers in 20 apheresis plasma samples were determined with the newly established FAMA test and ELISA test, respectively, and the detection results of the two methods were compared using Spearman’s correlation test. 【Results】 The optimal infection dose of the VZV-Oka-E6 strain in FAMA assay was determined to be 105.25 TCID50, and acetone precooled at -20℃ was used as the fixative. The FAMA test has a high sensitivity with a detecting limit of 31.25 mIU/mL for neutralizing anti-VZV titers. The negative result was observed when detecting herpes simplex virus (HSV) type 1 and 2 specific antibodies. The international standard for varicella zoster immunoglobulin was detected by VZV infected cell antigen slides prepared in the same batch and 4 different batches, with the coefficient of variation being 29.95% and 26.71%, respectively. The detection value of the international standard for varicella zoster immunoglobulin with three different titer levels was consistent with their theoretical value. The correlation coefficient of the detection results of 20 apheresis plasma samples by the FAMA test and ELISA test was 0.268. 【Conclusion】 The VZV FAMA assay has good sensitivity, specificity, repeatability, and relative accuracy in detecting neutralizing anti-VZV titers. It can be applied for detecting neutralizing anti-VZV titers in apheresis plasma samples as well as the varicella-zoster immunoglobulin (VZIG) preparations.

Este artigo apresenta resultados de uma pesquisa que investigou a produção de memórias e esquecimentos relacionada ao extinto "Abrigo Municipal de Alienados Oscar Schneider", instituição que funcionou em Joinville/SC entre 1923 e 1942 e teve seu prédio demolido cerca de 50 anos depois da sua construção. A inauguração da instituição marcou o início do projeto de "modernização-urbanização" de Joinville, pautado em normas higiênicas centradas nas novas prerrogativas da ciência moderna. Para a realização da pesquisa, rastreamos documentos que contam a história do Abrigo de Alienados: encontramos fotografias, um cartão-postal, trechos de documentos oficiais e notícias de jornal. Com esses documentos, problematizamos o campo de produção da fama e da infâmia em relação a determinados personagens que compuseram a cena dos acontecimentos dessa instituição. O olhar para os registros do Abrigo de Alienados procurou tensionar as diversas vozes que ali se encontram para seguir os rastros insignificantes de uma história, os quais correspondem a certos fatos e sujeitos que não foram considerados dignos de serem memorados: as vidas que sofreram a experiência do confinamento no Abrigo. This article presents results of a research that investigated memories and forgetfulness production related to the already extinct "Abrigo Municipal de Alienados Oscar Schneider", an institution that operated in Joinville, state of Santa Catarina, between 1923 and 1942, whose building was destroyed 50 years after its construction. The institution inauguration marked the beginning of Joinville's "modernization-urbanization" project, based on hygienic standards centered on the new prerogatives of modern science. To conduct this research, we traced documents that narrates the Alienated People's Shelter history: we found photographs, a postcard, oficial documents excerpts, and newspaper news. With these documents, we problematized fame and infamy production field in relation to some characters that were part of the institution happenings scene. Our vision to the Alienated People's Shelter aimed to tense up several voices that exist there to follow the insignificant trails of a history which corresponds to certain facts and subjects not considered dignified to be remembered: the suffering lives of the Shelter confinement experience. PURPOSE: To evaluate a recently marketed commercial glycoprotein enzyme-linked immunosorbent assay (gpEIA) kit, the VaccZyme(TM) VZV gpEIA, for measuring the immunity of varicella-vaccinated children. MATERIALS AND METHODS: We investigated the accuracy and reproducibility of the VaccZyme(TM) VZV gpEIA kit for the detection of antibodies to VZV. We also examined the sensitivity, specificity, and correlation between antibody titers calculated with gpEIA versus fluorescent antibody to membrane antigen (FAMA) by using sera of 349 children, ranging from 1 to 6 years old. RESULTS: VaccZyme(TM) VZV gpEIA gave precise and reproducible intra- and inter-assay results. FAMA and gpEIA titers showed a linear correlation (Pearson correlation coefficient=0.987). The sensitivity and specificity of the VaccZyme(TM) gpEIA was 31.4% and 100%, respectively, when the guidelines of the gpEIA (<100 mIU/mL) and FAMA 1:4 were adopted as cutoff values. However, the maximum sensitivity and specificity were 88.9% and 95.1%, respectively, with the highest correlation (kappa=0.840), if the cutoff values were set with gpEIA at 49.7 mIU/mL and FAMA 1:16. CONCLUSION: These results demonstrate that the VaccZyme(TM) VZV gpEIA kit gave precise and reproducible data for measuring antibody titer after varicella vaccination. The results also showed that the antibody titer calculated with the VaccZyme(TM) gpEIA kit strongly correlated with the FAMA titer. However, cutoff values should be re-optimized for the evaluation of vaccine immunity.

We reviewedonly randomized clinical trials.Results: Three studies were reviewed.The Gibney basket weave technique iseffective in improving the sensory-motorsystem, but the Mulligan techniqueand the association of three techniques(ankle stirrups, ankle lock and figure of6) didnt show to be effective.
