ABSTRACT

OBJECTIVE@#This study aimed to investigate whether the VCA0560 gene acts as an active diguanylate cyclase (DGC) in Vibrio cholerae and how its transcription is regulated by Fur and HapR.@*METHODS@#The roles of VCA0560 was investigated by utilizing various phenotypic assays, including colony morphological characterization, crystal violet staining, Cyclic di-GMP (c-di-GMP) quantification, and swimming motility assay. The regulation of the VCA0560 gene by Fur and HapR was analyzed by luminescence assay, electrophoretic mobility shift assay, and DNase I footprinting.@*RESULTS@#VCA0560 gene mutation did not affect biofilm formation, motility, and c-di-GMP synthesis in V. cholerae, and its overexpression remarkably enhanced biofilm formation and intracellular c-di-GMP level but reduced motility capacity. The transcription of the VCA0560 gene was directly repressed by Fur and the master quorum sensing regulator HapR.@*CONCLUSION@#Overexpressed VCA0560 functions as an active DGC in V. cholerae, and its transcription is repressed by Fur and HapR.

ABSTRACT

Resumen Presentamos el registro de un individuo de oso hormiguero norteño (Tamandua mexicana) con color de pelaje anormalmente blanco. Este individuo fue fotografiado con una cámara trampa en el bosque seco ecuatorial en el departamento de Lambayeque, a 560 m de altitud, en del noroeste del Perú. No pudimos diferenciar si la causa de esta aberración cromática correspondió a un caso de albinismo o de leucismo, sin embargo, destacamos este primer registro excepcional para la especie en Perú. Adicionalmente, hacemos una revisión de su presencia en el extremo sur de su distribución global.


Abstract We present the record of an individual of northern tamandua (Tamandua mexicana) with abnormal white coloration that was photographed with a camera trap in the equatorial dry forest in the department of Lambayeque, at elevation of 560 m, northwest Peru. We could not determine if this chromatic aberration was a case of leucism or albinism, but we highlight this uncommon record as the first for Peru. We also revised the information about its presence in the southern portion of its global range.

ABSTRACT

A clinical case in which skin and hair color change occurred after sacrococcygeal epidural anesthesia in a nine-month-old Siamese cross queen undergoing ovariohysterectomy (OHE) is described. Six weeks after surgery, during a re-check, it was noted that in the sacrococcygeal region the color of the skin and new hair growth was dark with a color comparable to the color present on the body extremities (muzzle, pinnae, legs, and tail). The skin and new hair growth of the shaved abdomen presented a standard color. The key enzyme of the melanogenic pathway in mammals is tyrosinase (TYR), and the Siamese temperature-sensitive phenotype is the result of genetic mutations that makes TYR function thermolabile. The activity of TYR in these cats is limited to the extremities where the temperature is lower while pigment production is impaired in the other body areas. The trichotomy of the sacrococcygeal region performed during wintertime in an outdoor cat was probably the trigger for increased activity of TYR in this area promoting pigment production. The absence of the same alterations in the abdominal area may be justified by less exposure of that region to the external environment, as well as to the feline habits of sedentarism, that avoid significant cooling in these regions. This report highlights the importance of taking this type of occurrence into account when performing an epidural in the Siamese cat breed. Also, to avoid skin color change in this breed, the authors recommend a midline abdominal instead of a flank approach to perform OHE.(AU)


Descreve-se um caso clínico no qual ocorreu mudança na cor da pele e do pelo após anestesia epidural sacrococcígea numa gata cruzada de raça Siamês de nove meses submetida à ovariohisterectomia (OVH). Seis semanas após a cirurgia, durante uma avaliação pós-operatória, notou-se que na região sacrococcígea, a cor da pele e o crescimento do pelo apresentavam uma cor escura, comparável à das extremidades do corpo (face, orelhas, membros e cauda). A pele e o crescimento do pelo do abdómen, que também havia sido tosquiado, apresentavam uma cor padrão. A enzima chave da via melanogênica em mamíferos é a tirosinase (TYR) e o fenótipo siamês sensível à temperatura é o resultado de mutações genéticas que tornam a função TYR termolábil. A atividade da TYR nestes gatos é limitada às extremidades onde a temperatura é mais baixa, enquanto a produção de pigmento é prejudicada em outras áreas do corpo. A tricotomia da região sacrococcígea realizada durante o inverno nesta gata com acesso livre ao ambiente externo, provavelmente determinou um aumento da atividade da TYR nesta área, promovendo a produção de pigmento. A ausência das mesmas alterações na região abdominal pode ser justificada pela menor exposição desta área do corpo ao ambiente externo, também devido aos hábitos felinos de sedentarismo, que evitam resfriamento significativo nestas regiões. Este relato destaca a importância de se levar em consideração a possibilidade deste tipo de ocorrência quando da realização de uma anestesia epidural nesta raça de gatos. Além disso, os autores recomendam uma abordagem abdominal na linha média ao invés de uma abordagem de flanco para realizar a OVH nesta raça, a fim de evitar a alteração da cor da pele.(AU)

ABSTRACT

Members of the ferric uptake regulator (Fur) protein family are bacterial transcriptional repressors that control iron uptake and storage in response to iron availability, thereby playing a crucial role in the maintenance of iron homeostasis. The fur null mutants of Pseudomonas aeruginosa could not be obtained because fur is an essential gene. In this study, We constructed a Fur inducibly expression strain Δfur/attB::PBAD-fur in order to study the effect of fur on the growth, biofilm formation, motilities and oxidative stress response of P. aeruginosa. The results showed that a low level of fur expression retarded the growth of P. aeruginosa at an iron-depleted condition, or under high concentration of iron, or in the presence of H2O2. Fur affected the biofilm formation and the motilities (swimming, twitching, and swarming) of strain PAO1. The production of pyoverdine is regulated by Fur. Interestingly, proteins from Magnetospirillum gryphiswaldense MSR-1, which shares homology with Fur, can partially recover the pyoverdine production of strain Δfur/attB::PBAD-fur. This study provides new clues for the prevention and treatment of P. aeruginosa infections.

ABSTRACT

@#<p style="text-align: justify;"><strong>Background.</strong> Anxiety and depression are becoming increasingly prevalent today and are often aggravated by day-to-day stresses. Because current management strategies are usually accompanied by unpleasant side effects, there is a need to look into alternative treatment regimens - such as prebiotics - that may provide equally effective anxiolytic and antidepressant effects.</p><p style="text-align: justify;"><strong>Objective.</strong> Therefore, the study aims to determine the effect of a combined fructooligosaccharide (FOS) and galactooligosaccharide (GOS) supplemented diet on anxiety and depression levels in mice subjected to Unpredictable Chronic Mild Stress (UCMS).</p><p style="text-align: justify;"><strong>Methods.</strong> Forty male BALB/C mice were subjected to UCMS under a pretest-posttest control group design where the treatment group received prebiotic supplementation throughout the study. Repeated measures ANOVA was run to evaluate between, within, and time interactions of the measured anxiety parameters using the light-dark box test, and depression parameter using the fur coat state assessment.</p><p style="text-align: justify;"><strong>Results.</strong> Results show that (1) the FOS + GOS treatment did not give the treatment group an advantage over the control group during UCMS, (2) both groups grew more anxious and depressed over time, and (3) the treatment group grew more anxious with time in relation to control in terms of the total time spent in the light side.</p><p style="text-align: justify;"><strong>Conclusion.</strong> These imply that the UCMS protocol was successful in inducing stress in mice, but the FOS + GOS regimen failed to provide anxiolytic and antidepressant effects on male BALB/C mice exposed to UCMS.</p>

ABSTRACT

This is the first study to investigate mercury (Hg) contamination in felid species in the Brazilian Amazon. We collected 26 fur samples from wild felids of four species (Puma concolor, Panthera onca, Leopardus pardalis and Leopardus wiedii) occurring in the Mamirauá and Amanã sustainable development reserves, in the state of Amazonas. Samples were from museum specimens, except for five P. onca samples collected from free-living individuals. Total Hg concentrations ranged from 0.12 to 48.1 µg g-1. Concentrations of Hg did not differ significantly between museum specimens and live individuals of P. onca, but varied significantly among species, with significantly higher concentrations for P. onca and L. pardalis, which could be related to factors such as diet and habitat. ABSTRACT

Se reporta el primer caso de melanismo en el roedor Phyllotis limatus, el cual incrementa el número de especies de mamíferos silvestres con melanismo a 113 especies, a nivel mundial. Los órdenes Rodentia y Carnivora agrupan la mayor cantidad de especies con melanismo, siendo reportadas con mayor frecuencia en países del continente americano, seguidos de Asia, África, Europa y Oceanía. En Sudamérica, Brasil es el país con la mayor cantidad de especies con melanismo, con ocho especies, seguido de Perú, con cinco. El listado de mamíferos silvestres melánicos aquí presentado sugiere que la coloración negruzca es una mutación relativamente frecuente para algunas especies; por consiguiente, es probable que el número de registros de especies con melanismo se incremente en futuras investigaciones.


Here, we report the first case of melanism in the rodent Phyllotis limatus, which increase the number of wild mammal species with melanism to 113 species, worldwide. The orders Rodentia and Carnivora group the largest number of species with melanism, being reported more frequently in countries of America, followed by Asia, Africa, Europe and Oceania. In South America, Brazil is the country with the highest number of species with melanism, with eight species, followed by Peru with five. The melanic checklist of wild mammals presented here suggests that blackish coloration is relatively frequent mutations in some species, therefore, we hope the register numbers of species with melanism will be increased in future investigations.

ABSTRACT

Trata-se de uma pesquisa qualitativa baseada em narrativas produzidas por meio de entrevistas e oficinas. O estudo pretende demonstrar como profissionais de saúde de sete especialidades de uma UTI neonatal e pediátrica de um Hospital Público Municipal, no interior do Rio de Janeiro, identificam sinais de sofrimento no trabalho. Esses profissionais lidam com nascimento, dor e morte de crianças, mas não incluem essa experiência como parte integrante de seus cotidianos de trabalho. Aposta-se que o uso de narrativas seja um dispositivo capaz de incluir as dimensões da experiência dos quais tenta-se afastar, bem como produzir conhecimentos sobre a vida e a morte nos processos de trabalho.


This paper describes a qualitative research based on narratives collected by means of interviews and workshops. It aims to reveal in what way health professionals from seven specialties of a neonatal and pediatric ICU of a Municipal Public Hospital in the interior of Rio de Janeiro identify signs of suffering at work. These professionals deal with the birth, pain and death of children, but do not include this experience as an integral part into their daily work. We believe that the use of narratives is a device that allows to incorporate the dimensions of these denied experiences and to produce knowledge about life and death in work processes.


Cette recherche qualitative est basée sur des récits produits à travers d'entretiens et d'ateliers. L'étude a pour but de démontrer de quelle façon des professionnels de la santé de sept spécialités d'une unité de soins intensifs néonatals et pédiatriques d'un hôpital public municipal à l'intérieur de l'état de Rio de Janeiro identifient les signes de souffrance au travail. Ces professionnels sont confrontés à la naissance, la douleur et au décès d'enfants, mais ils n'incluent pas cette expérience dans leur travail quotidien. Nous concluons que la production de récits leur permet d'intégrer les dimensions de l'expérience desquelles ils essaient de se distancer, ainsi que de produire des connaissances sur la vie et la mort au travail.


Se trata de una investigación cualitativa basada en narrativas, producidas mediante encuestas y talleres. El estudio pretende demostrar de qué forma los profesionales de salud de siete especialidades de una UCI neonatal y pediátrica de un hospital público municipal, en el interior de Río de Janeiro, identifican señales de sufrimiento en el trabajo. Estos profesionales se enfrentan con el nacimiento, el dolor y la muerte de niños, pero no incluyen esta experiencia como parte integrante de su vida cotidiana de trabajo. Se apuesta a que el uso de narrativas sea un dispositivo capaz de incluir las dimensiones de la experiencia de las que intenta alejarse, así como producir conocimientos sobre la vida y la muerte en los procesos de trabajo.


Dieser Artikel beschreibt eine qualitative Studie, die auf Erzählungen basiert, die während Interviews und Workshops gesammelt wurden. Die Studie stellt dar, wie Gesundheitspersonal von sieben Fachgebieten einer neonatalen und pädiatrischen Intensivstation eines städtischen Krankenhauses im Inland des Staates von Rio de Janeiro Anzeichen von Leiden bei der Arbeit erkennen. Dieses Personal ist der Geburt, den Schmerzen und dem Tod von Kindern ausgesetzt, berücksichtigt diese Erfahrungen jedoch nicht als wesentlichen Bestandteil seiner täglichen Arbeit. Die Verwendung von Erzählungen ist hier ein Mittel, um die Dimensionen dieser Erfahrungen, die man zu vermeiden versucht, zu verarbeiten (Bruner, 1991) und Wissen über Leben und Tod anhand von Arbeitsprozessen zu produzieren.

ABSTRACT

Objetivou-se neste estudo padronizar um protocolo de reação em cadeia da polimerase (PCR) para detecção de Microsporum canis em amostras de pelos e/ou crostas de cães e gatos. Foram selecionadas 48 amostras previamente identificadas por meio de cultura. Destas, 23 foram positivas para dermatófitos no cultivo. Padronizou-se a PCR a partir de primers desenhados para o alvo M. canis. Sessenta e um por cento (14/23) das amostras positivas para dermatófitos foram identificadas como M. canis em cultura. Desse total, 71,4% (10/14) apresentaram um fragmento de 218pb compatível com o esperado para a espécie fúngica alvo dessa reação. Observou-se uma sensibilidade de 71,4% e especificidade de 100% na PCR, além de uma boa concordância entre essas técnicas de diagnóstico (Kappa: 0,78; P<0,0001). O protocolo utilizado neste estudo apresentou alta especificidade na detecção de M. canis diretamente de amostras de pelos e/ou crostas de cães e gatos, viabilizando um diagnóstico mais rápido e específico, podendo esse protocolo ser empregado como um método confirmatório para agilizar a detecção de M. canis.(AU)


The aim of this study was to standardize a Polymerase Chain Reaction protocol (PCR) for the detection of Microsporum canis in fur and/or crusts of dogs and cats. 48 samples previously identified by culture were selected. Of these, 23 were positive for dermatophytes in culture. PCR was standardized from drawn primers whose target is M. canis. A total of 61% (14/23) of the dermatophyte positive samples were identified as M. canis in culture. Of this total, 71.4% (10/14) presented a fragment of 218bp compatible with that expected for the fungal species target of the reaction. A sensitivity of 71.4% and specificity of 100% in the PCR were observed, in addition to a good agreement between the techniques (Kappa: 0.78; P<0.0001). The protocol used in this study showed high specificity in the detection of M. canis directly from fur and/or crusts of dogs and cats, making possible a faster and more specific diagnosis. This protocol could be used as a confirmatory method, speeding the detection of M. canis.(AU)

OBJECTIVES: This study aimed to evaluate several methods to estimate glucose consumption in the male Wister rat brain as measured by PET. METHODS: Fourteen male Wistar normoglycemic rats were studied. The input function consisted of seventeen blood samples drawn manually from the femoral artery. Glucose uptake values were calculated using the input function resulting from the arterial blood samples and the tissue time-activity curve derived from the PET images. The estimated glucose consumption rate (Ki) based on the 2-tissue compartment model (2TCM) served as the standard for comparisons with the values calculated by the Patlak analysis and with the fractional uptake rate (FUR), standardized uptake value (SUV) and glucose corrected SUV (SUVglu). RESULTS: No significant difference between the standard Ki and the Patlak Ki was observed. The standard Ki was also found to have strong correlations and concordance with the Ki value estimated by the Patlak analysis. The FUR method presented an excellent correlation with the Ki value obtained by the 2TCM/Patlak analyses, in contrast to the SUV or SUVglu. CONCLUSIONS: From a methodological point of view, the present findings confirm the theoretical limitations of the cerebral SUV and SUVglu as a substitute for Ki in the estimation of glucose consumption in the brain. Our data suggest that the FUR is the surrogate to Ki.

ABSTRACT

O objetivo desta pesquisa foi verificar a prevalência de dermatófitos em tegumento de bovinos e ovinos hígidos e a sua capacidade de transmissão e desenvolvimento da doença. Para isso, foram coletadas amostras de pelo e descamações de 90 bovinos e 90 ovinos hígidos. Essas amostras foram semeadas em meio DTM e ágar Sabouraud Dextrose enriquecido com extrato de levedura, tiamina, antibióticos (estreptomicina e cloranfenicol) e suplementado de cicloheximida, sendo essas incubadas a 35 oC por 10 dias. As culturas positivas foram avaliadas macro e microscopicamente e, os fungos foram identificados por métodos bioquímicos. Verificou-se que as espécies isoladas com maior frequência, nos bovinos, foram T. mentagrophytes e M. gypseum. Nos ovinos, foi constatada maior ocorrência do agente T. verrucosum, seguido por T. mentagrophytes e M. gypseum. Houve também isolamento de M. canis, porém, em um número reduzido de amostras. Assim, concluiu-se que o tegumento de bovinos e de ovinos hígidos apresentou incidência elevada de dermatófitos de diferentes espécies, em amostras coletadas durante o período chuvoso do ano. Por isso, em animais jovens ou naqueles submetidos a elevados níveis de estresse e, consequente queda da resposta imunológica, o risco de desenvolvimento da dermatofitose e da transmissão dos dermatófitos neste período são eminentes.(AU)


The purpose of this study was to determine the prevalence of dermatophytes in healthy cattle and sheep teguments, and their ability to transmit and develop diseases. Samples of fur and flaking from 90 healthy cattle and 90 healthy sheep were collected. These samples were plated on DTM medium and Sabouraud Dextrose agar supplemented with yeast extract, thiamine, antibiotics (streptomycin and chloramphenicol) enhanced with cycloheximide. They were then incubated at 35 °C for 10 days. Positive cultures were macroscopically and microscopically evaluated, and fungi were identified by biochemical methods. It was found that the most frequent species isolated in cattle were T. mentagrophytes and M. gypseum. In sheep, a higher occurrence of the agent T. verrucosum could be observed, followed by T. mentagrophytes and M. gypseum. There was also the isolation of M. canis, but in a small number of samples. Thus, it could be concluded that the tegument of healthy cattle and sheep showed high incidence of dermatophytes from different species in samples collected during the rainy season of the year. Therefore, in young animals, in those subjected to high levels of stress and consequent drop in immune response, there is eminent risk of developing dermatophytosis and transmission of dermatophytes in this period.(AU)


El objetivo de esta investigación ha sido verificar la prevalencia de dermatofitos en tegumento de bovinos y ovinos hígidos y su capacidad de transmisión y desarrollo de la enfermedad. Para ello, se recolectó muestras de pelo y descamaciones de 90 bovinos y 90 ovinos hígidos. Esas muestras fueron sembradas en medio DTM y agar Sabouraud Dextrosis enriquecida con extracto de levadura, tiamina, antibióticos (estreptomicina y cloranfenicol) y suplementado de cicloheximida, siendo esas incubadas a 35ºC durante 10 días. Los cultivos positivos fueron evaluados macro y microscópicamente y los hongos fueron identificados por métodos bioquímicos. Se verificó que las especies aisladas con mayor frecuencia, en los bovinos, fueron T. mentagrophytes y M. gypseum. En los ovinos, se constató mayor ocurrencia del agente T. verrucosum, seguido por T. mentagrophytes y M. gypseum. También hubo aislamiento de M. canis, sin embargo, en un número reducido de muestras. Así, se concluyó que el tegumento de bovinos y de ovinos hígidos presentó una elevada incidencia de dermatofitos de diferentes especies, en muestras recogidas durante el período lluvioso del año. Por lo tanto, en animales jóvenes o en aquellos sometidos a altos niveles de estrés y, consecuente caída de la respuesta inmunológica, el riesgo de desarrollo de la dermatofitosis y de la transmisión de los dermatofitos en este período son eminentes.(AU)

ABSTRACT

Objetivou-se padronizar uma reação do tipo multiplex PCR (mPCR) para detectar Microsporum canis, Microsporum gypseum e o complexo Trichophyton mentagrophytes em amostras de pelos e/ou crostas de cães e gatos. 250 amostras de pelos e/ou crostas de cães e gatos foram analisadas por meio de exame direto e cultura, o DNA das mesmas foi extraído para mPCR. Primers foram desenhados e como controle positivo da reação utilizou-se o DNA extraído de colônias de M. canis (URM 6273), M. gypseum (URM 6921) e T. mentagrophytes (URM 6211), provenientes da Coleção de Culturas (Micoteca URM), Departamento de Micologia, Centro de Ciências Biológicas da Universidade Federal de Pernambuco (CCB/UFPE). Como controles negativos de reação, utilizou-se água destilada esterilizada e DNA extraído de Alternaria sp. para verificar a especificidade dos primers. Do total de amostras analisadas, 15 (6%) foram identificadas, em cultura, como dermatófitos, e destas, 10 foram M. canis, três M. gypseum e dois T. mentagrophytes (complexo). Destas 15 amostras positivas, 11 (73,3%) foram detectadas por meio da mPCR. Além destas, seis outras, negativas em cultura, foram identificadas como M. gypseum. Verificou-se uma boa concordância entre os resultados da cultura e mPCR (Kappa: 0,66). O protocolo padronizado neste estudo pode ser utilizado como um método de triagem, por apresentar uma sensibilidade maior que a da cultura, usado paralelamente aos exames de rotina, permitindo um diagnóstico em menor tempo.(AU)


The aim of this study was to standardize a multiplex PCR (mPCR) reaction to detect Microsporum canis, Microsporum gypseum and the Trichophyton mentagrophytes complex in dog and cat fur and/or crusts. 250 fur and/or crusts samples from dogs and cats were analyzed by direct examination and culture, DNA from them was extracted for mPCR. Primers were designed and the DNA extracted from colonies of M. canis (URM 6273), M. gypseum (URM 6921) and T. mentagrophytes (URM 6211) from the Collection of Cultures - URM Micoteca - Department of Mycology, Biological Sciences Center of the Federal University of Pernambuco (CCB / UFPE). As negative controls, sterile distilled water and DNA extracted from Alternaria sp., were used to verify the specificity of the primers. Of the total samples analyzed, 15 (6%) were identified in culture as dermatophytes, and of these, 10 were M. canis, three M. gypseum and two T. mentagrophytes (complex). Of these 15 positive samples, 11 (73.3%) were detected by mPCR. Besides these, six others, negative in culture, were identified as M. gypseum. There was good agreement between culture results and mPCR (Kappa: 0.66). The protocol standardized in this study can be used as a screening method, because it has a sensitivity greater than that of the culture, used in parallel to the routine exams, allowing a diagnosis in a shorter time.(AU)

ABSTRACT

This study evaluated the concentration of Der p 1, Der f 1 and Blo t 5 in the fur and households of 20 dogs with atopic dermatitis (AD) and 20 healthy dogs. The diagnosis of AD was clinical based on Favrot's criteria. Dust samples were collected with a domestic vacuum cleaner. For each site, 1m2 was vacuumed for 2 min. The samples were collected in separate filters, transferred into plastic containers, sealed and kept frozen until ELISA analysis. In the fur of atopic dogs the average concentration of Der p 1 was 0.25µg/g compared to 0.03µg/g in healthy dogs. In households with atopic dogs the highest concentrations of Der p 1 were found in carpets (2.18µg/g), followed by couches (1.53µg/g), beds (1.14µg/g), dogs' bed linen (0.64µg/g) and floors (0.14µg/g). The concentrations of Der p 1 on carpets, couches and beds were significantly higher than in atopic dogs' fur (p<0.05). There was no statistical difference when comparing the concentrations of Der f 1 and Blo t 5 in different environments of atopic dogs (p>0.05). The concentrations of Der p 1, Der f 1 and Blo t 5 were equivalent in atopic and non-atopic dog's households. Among the allergens studied, Der p 1 was the most commonly found, predominantly in carpets and couches.(AU)


O presente estudo avaliou a concentração de Der p 1, Der f 1 e Blo t 5, na pelagem e no ambiente domiciliar de cães com dermatite atópica (DA). Para tal, foram selecionados 20 cães com DA e 20 cães saudáveis, e seus domicílios. O diagnóstico de DA foi baseado nos critérios de Favrot. As amostras de poeira foram colhidas com um aspirador de pó doméstico. Para cada local de colheita, foi aspirado 1m2 por dois minutos. As amostras foram recolhidas em filtros separadamente, transferidas para envelopes plásticos, seladas e mantidas congeladas até serem analisadas pelo método de ELISA. Na pelagem dos cães com DA, a concentração média de Der p 1 foi de 0,25µg/g de poeira e nos cães saudáveis foi de 0,03µg/g. No ambiente de cães com DA, o Der p 1 foi encontrado em maior concentração média no tapete (2,18 µg/g), seguido pelo sofá (1,53g/g), cama (1,14µg/g) e roupa de cama dos cães (0,64µg/g) e chão (0,14µg/g). As concentrações de Der p 1 no tapete, sofá e cama dos cães foram significativamente maiores que na pelagem de cães com dermatite atópica (p<0,05). Enquanto que, não houve diferença estatística quando comparadas as concentrações de Der f 1 e Blo t 5 nos diferentes ambientes avaliados (p>0,05). As concentrações de alérgenos Der p 1, Der f 1 e Blo t 5 se equivaleram em ambientes de cães com DA e saudáveis. Entre os alérgenos estudados, o Der p 1 foi o mais comumente encontrado, prevalecendo no tapete e no sofá.(AU)

ABSTRACT

We investigated the role of autophagy in SNUC5/5-FUR, 5-fluorouracil (5-FU) resistant SNUC5 colon cancer cells. SNUC5/5-FUR cells exhibited low level of autophagy, as determined by light microscopy, confocal microscopy, and flow cytometry following acridine orange staining, and the decreased level of GFP-LC3 puncta. In addition, expression of critical autophagic proteins such as Atg5, Beclin-1 and LC3-II and autophagic flux was diminished in SNUC5/5-FUR cells. Whereas production of reactive oxygen species (ROS) was significantly elevated in SNUC5/5-FUR cells, treatment with the ROS inhibitor N-acetyl cysteine further reduced the level of autophagy. Taken together, these results indicate that decreased autophagy is linked to 5-FU resistance in SNUC5 colon cancer cells.

ABSTRACT

Abstract The occurrence of anomalous coloration (albinism, leucism and melanism) in mammals is a rare phenomenon in nature, but this phenomenon has been reported for several species of mammals. In this study, we report on the occurrence of leucism in Eira barbara by examining three road-killed individuals and two sightings of live animals in Reserva Particular do Patrimônio Natural Santuário do Caraça, southeastern Brazil. In addition, we examined tayra specimens housed in mammal collections from Brazil and USA. The animals found dead and those sighted had a whitish yellow fur on the body and head, resulting in lighter coloration than the coloring pattern commonly observed in tayras. Despite these lighter color pattern, the specimens showed parts of soft tissue, such as iris and the skin, with pigmentation very similar to that present in individuals with the typical color pattern. This set of factors indicates the specimens recorded were in fact leucistic and not albino. Among the specimens examined in the scientific collections, we found nine individuals from different localities that presented the whitish yellow color pattern. Some studies attribute the higher frequency of cases of leucism due to small populations and / or with some mechanism of reproductive isolation. Thus, analysis of the genetic variability of populations containing individuals with such characteristics should be considered. On the other hand, the occurrence of polymorphic color phenotype in tayras indicates that hypotheses related to the fixation of recessive characteristics, or on possible environmental adaptive advantages of these phenotypes can be tested.


Resumo A ocorrência de coloração anômala (albinismo, leucísmo e melanismo) em mamíferos é um fenômeno raro na natureza, mas ela tem sido relatada para diversas espécies. Neste trabalho nós relatamos a ocorrência de leucísmo em Eira barbara por meio do exame de três indivíduos encontrados mortos atropelados por veículos e pela visualização direta de indivíduos vivos na Reserva Particular do Patrimônio Natural Santuário do Caraça, sudeste brasileiro. Em adição, nós consultamos coleções de mamíferos em museus do Brasil e dos Estados Unidos da América. Os animais encontrados mortos e os avistados apresentavam pelagem amarela esbranquiçada no corpo e na cabeça, resultando em uma coloração muito mais clara que o padrão de coloração comumente observado em iraras. Apesar deste padrão de coloração mais claro, os espécimes apresentavam partes do tecido mole, tais como a íris e a pele, com pigmentação muito semelhante àquela presente em indivíduos com padrão de coloração típico da espécie. Este conjunto de fatores indicou que os espécimes registrados eram de fato leucísticos e não albinos. Dentre os espécimes examinados nas coleções científicas, nós encontramos nove indivíduos de diferentes localidades que apresentavam o padrão de coloração esbranquiçado. Alguns estudos atribuem a frequência elevada de casos de leucísmo a pequenas populações e / ou com algum mecanismo de isolamento reprodutivo. Dessa forma, análises da variabilidade genética de populações contendo indivíduos com essas características devem ser consideradas. Por outro lado, a ocorrência de fenótipos de coloração polimórficos em iraras indica que hipóteses podem ser testadas tanto com relação à fixação de caraterísticas recessivas, quanto sobre possíveis vantagens adaptativas ambientais desses fenótipos.

A personagem enlouquecida amarrada ao pé do castanheiro de "Cem anos de solidão" serve-nos de metáfora para as amarras que a institucionalização pode propiciar. Como fazer para que uma Residência Terapêutica (RT), por não oferecer as mesmas amarras das instituições psiquiátricas, possa ter a função de um espaço transitório para outro lugar? Para tentar responder esta questão construímos uma aproximação entre o ambiente facilitador e o meio que a RT representa a partir dos conceitos de Winnicott.


The mad man tied to the chestnut tree in One Hundred Years of Solitude can be seen as a metaphor of the ties that institutionalization can engender. What can be done to make a therapeutic residence perform the role of a transitory space for patients, since there they are not tied down as they would be in a psychiatric institution? Based on Winnicott's concepts we tried to answer this question by making a connection between the facilitating environment and the therapeutic residence environment.


Le personnage fou attaché au pied d'un châtaignier dans Cent ans de solitude nous sert de métaphore pour les chaines invisibles que l'institutionnalisation peut représenter. Étant donné que la résidence thérapeutique (RT) ne produit pas les mêmes rapports de dépendance que les institutions psychiatriques, comment la configurer pour qu'elle puisse jouer un rôle d'espace transitionnel? Pour essayer de répondre à cette question, nous avons construit une approximation entre l'environnement facilitateur et celui de la RT à partir des concepts de Winnicott.


El personaje loco atado al castañero del libro Cien años de soledad nos sirve como una metáfora de las amarras que la institucionalización puede propiciar. ¿Como hacer para que una Residencia Terapéutica (RT), por no ofrecer las mismas amarras de las instituciones psiquiátricas, tenga la función de un espacio de transición para otro lugar? ABSTRACT

Aims: To present and discuss the etiology of three distinct episodes of thumb felon in a fur industry worker, in association with the required precautions. Case Presentation: A 61-year-old man working in a fur industry was treated for three distinct episodes of a felon of the right thumb within an 8-month period. He was treated successfully with surgical drainage and antibiotics following each episode. He was symptom-free and returned to work about two months following each episode. Felon formation, in all cases, was due to a Streptococcus constellatus infection secondary to mink hair penetration through the distal nail groove of his thumb. Using protective gloves, no further relapses have occurred during a 5-year follow-up. Discussion: A felon is an abscess of the pulp of a finger or thumb usually due to percutaneous trauma. Streptococcus constellatus, which is included in the Streptococcus milleri group, is often associated with various pyogenic infections. In the reported case, the lack of hand protection during fur processing was the cause of mink hair infiltration into the pulp of the patient’s thumb through the distal nail groove and of the three episodes of felon formation due to the secondary infection by Streptococcus constellatus. Conclusion: Lack of hand protection during fur processing could make hands vulnerable to micro-injuries and secondary infections.

ABSTRACT

The aim of the present study was to evaluate the efficacy of the d-phenothrin/pyriproxyfen association against Psoroptes ovis, Cheyletiella parasitivorax, and Leporacarus gibbus infestations in naturally co-infested rabbits. Twenty crossbreed (New Zealand White x California) rabbits concurrently infested by the three mite species were randomly divided in two groups. All rabbits presented with hyperemia, erythema and formation of crusts in the ear canals caused by P. ovis. Infestations by both C. parasitivorax and L. gibbus were considered asymptomatic in all animals.Ten animals were treated with a 4.4% d-phenothrin and 0.148% pyriproxyfen spray formulation until have their body surface uniformly sprayed, including external ear canals. The other ten rabbits remained untreated, serving as control group. Observations were done on days +7, +14, +21, +28, and +35 post-treatment. The d-phenothrin/pyriproxyfen association showed 100% efficacy against the three mite species and was responsible for the remission of psoroptic mange lesions on treated animals. No signs of intoxication were observed. The results indicate that d-phenothrin/pyriproxyfen spray formulation in a single application is an effective and clinically safe option for the control of different mite infestations in rabbits.


O objetivo do presente estudo foi avaliar a eficácia da associação de d-fenotrina e piriproxifen no controle de infestações simultâneas por Psoroptes ovis, Cheyletiella parasitivorax e Leporacarus gibbus em coelhos naturalmente co-infestados. Vinte coelhos mestiços (Nova Zelândia Branco x Califórnia) infestados simultaneamente pelas três espécies de ácaros foram divididos aleatoriamente em dois grupos. Todos os coelhos infestados por apresentavam eritema, hiperemia e formação de crostas nas orelhas, causados por P. ovis. Infestações simultâneas por C. parasitivorax e L. gibbus foram considerados assintomáticas em todos os animais. Dez animais foram tratados com uma formulação spray contendo d-fenotrina a 4,4% e piriproxifen a 0,148%, pulverizando toda a superfície corporal de forma uniforme, incluindo a face interna das orelhas. Os outros 10 coelhos não foram t ratados, sendo mantidos como grupo controle. Os animais foram avaliados nos dias 7, 14, 21, 28 e 35 pós-tratamento. A associação de d-fenotrina e piriproxifen foi 100% eficaz no controle das três espécies de ácaros e foi responsável pela remissão das lesões de sarna psoróptica nos animais tratados. Não foram observados sinais de intoxicação. Os resultados indicam que a formulação spray de d-fenotrina e piriproxifen em uma única aplicação é uma opção clinicamente segura e eficaz no controle de infestações por ácaros em coelhos.

Male capacity for spreading genes to a great number of descendents and to determine population dynamics depend directly on the genital organs. Morphological studies in pinnipeds are scarce and the functional meaning of some characteristics has never been discussed. We hypothesized that Arctocephalus australis (A. australis) shows morphophysiological adaptations in order to guarantee the perpetuation of the species in the unique annual mating season. Seven males, dead from natural causes, had their genital organs collected and fixed for morphological description. Some features differ from other described mammalian males and are closely related to the biology and reproductive cycle of this species, as the scrotal epidermis, absence of glandular portion in the ductus deferens and spermatogenic epithelium suggest a recrudescent testis period. The corona glandis exhibits a singular arrangement: its erectile border looks like a formation of petals and its association with the os penis gives a "lily-flower" form to this region. We propose the name margo petaliformis to this particular erectile border of the corona glandis because of its similarity to a flower corola. The male genital organs of A. australis show morphological features compatible with adaptation to environment requirements and reproductive efficiency.(AU)


A capacidade do macho de espalhar seus genes a um grande número de descendentes e determinar a dinâmica populacional depende diretamente dos seus órgãos genitais. Estudos morfológicos em pinípedes são escassos e o significado funcional de algumas de suas características ecológicas ainda foi pouco discutido. Nossa hipótese é que Arctocephalus australis (A. australis) apresenta adaptações morfofisiológicas em seus órgãos genitais capazes de interagir com o meio e garantir a perpetuação da espécie que apresenta apenas uma época de acasalamento que ocorre uma vez a cada ano. Sete A. australis machos, mortos recentes por causas naturais, tiveram seus órgãos genitais coletados e fixados para a descrição macro, micro e ultraestrutural. Algumas características diferem de outros machos já descritos e estão intimamente relacionados com a biologia e ciclo reprodutivo da espécie, dentre elas podemos citar a alta queratinização da epiderme escrotal que pode se relacionar com as rotineiras lesões por atrito desta região nas pedras; a ausência da porção glandular do ducto deferente aqui descrita pela primeira vez, o epitélio espermatogênico sugere um período de testículo recrudescente. A glande apresenta um arranjo singular: a coroa da glande apresenta porção lateral de tecido esponjoso que são bordas livres com capacidade de intumescencia. O osso peniano se encontra no centro destas bordas e representa a extremidade mais distal do penis, levando consigo o óstio uretral externo. As bordas associadas ao osso peniano, dão uma forma de "Flor de lírio" a esta região. Utilizamos o nome margo petaliformis a margem erétil liliforme a particular morfologia da glande, pela sua semelhança a uma corola de flor. Os órgãos genitais masculinos de A. australis mostram características morfológicas compatíveis com uma adaptação aos requisitos ambientais e de eficiência reprodutiva.(AU)
