ABSTRACT

Objective To explore the therapeutic mechanism of kidney-reinforcing,blood-activating and phlegmresolving therapy for left ventricular fibrosis of spontaneously hypertensive rats (SHR).Methods Twenty SHR were randomly assigned to Chinese medicine group and model group.Additionally,ten Wistar-Kyoto(WKY) rats served as normal control group.After 12-week prevention,Masson staining method was used to measure the degree of fibrosis of left ventricular myocardial tissues,reverse transcription-polymerase chain reaction(RT-PCR) was used to detect Smad3 mRNA expression,and Western blotting method was used for the detection of Smad3 protein expression.Results The degree of left ventricular fibrosis myocardial tissue in Chinese medicine group was milder than that in the model group,and Samd3 protein and mRNA expression levels in Chinese medicine group were lower than those in the model group (P < 0.05).Conclusion Kidney-reinforcing,blood-activating and phlegmresolving therapy can improve left ventricular fibrosis in SHR by inhibiting Smad3 expression.

ABSTRACT

Objective To investigate the effect of Tianma Gouteng Decoction(TGD) on myocardial fibrosis in spontaneously hypertensive rats(SHR) and to explore its possible mechanism. Methods Twenty-four male SHR with 8 weeks old were equally randomized into three groups: TGD(20g/kg) group,captopril(100mg/kg)positive group and the model group.Eight Wistar Kyoto rats(WKY) with the same age served as the controls.The systolic blood pressure(SBP),left ventricular mass(LVM),left ventricular mass index(LVMI),myocardial collagen(MC) content in left ventricle of three groups were determined after treatment for 24 weeks.Meanwhile,the expression of myocardial insulin-like growth factor 1(IGF-1) was determined by reverse transcription-polymerase chain reaction(RT-PCR) assay.Results TGD had an effect on decreasing the SBP,LVM,LVMI,MC content and IGF-1 expression(P

ABSTRACT

Objective To observe the effect of Compound Fuling Gancao Decoction(CFGD) on transforming growth factor ?1(TGF-?1) in pulmonary artery of rats with pulmonary hypertension(PH),and to explore its therapeutic mechanism for PH and pulmonary vascular remodeling.Methods Thirty SD male rats were randomized into the normal control group,model group,and CFGD(5.46g?kg-1?d-1) group.PH rats models were induced by intraperitoneal injection of monocrotaline(50mg/kg).The mean pulmonary artery pressure(MPAP) in rats was detected with the method of right cardiac catheterization,right ventricular hypertrophy index was calculated,histological changes of lung small artery were analyzed with pathological image analysis,and TGF-?1 expression in pulmonary artery was detected by immunohistochemical method.Results MPAP and right ventricular hypertrophy index were enhanced obviously,the number of muscular arteries and partially-muscular arteries was increased,ratio of media area and luminal thickness in the muscular arteries became bigger,and TGF-?1 expression in pulmonary artery of the model group was increased.The above indexes were improved in CFGD group.Conclusion CFGD is effective on decreasing pulmonary pressure and improving pulmonary vascular remodeling,and its mechanism is probably related with the inhibition of TGF-?1 expression in pulmonary artery.

ABSTRACT

[Objective] To observe the therapeutic effect of Qianyang Dingxuan Decoction (QDD) for hypertension with yin deficiency and yang hyperactivity. QDD is mainly composed of Concha Haliotidis, Ostrea, Os Draconis, Concha Margaritifera Usta, Caulis et Folium Polygoni Multiflori, Radix paenonicae Alba, etc.. [Methods] Sixty cases (Group A) of essential hypertension were treated with QDD and 30 were in positive control group (Group B). Therapentic effect in the two groups were compared. ABSTRACT

[Objective] To evaluate the effect of Bushen Yixin Tablet (BYT) on blood pressure (BP) and hemorrheology in rats with hypertension. [Methods] Rat models of hypertension were established by constricting either of the bilateral renal arteries with one clip. Sixty rats were randomly allocated to high - dose BYT group (Group A), low - dose BYT group (Group B), nitrendipine (Group C) and normal saline group (Group D) . Ten rats served as mimic group (Group E) . BP of tail artery was examined in various groups and after a 4 - week treatment, whole blood viscosity and indexes of erythrocyte aggregation (EA) and erythrocyte deformation (ED) were detected. [Results] BP was increased, high- , middle - and low - shear whole blood viscosity and EA index were elevated and ED index was decreased in model rats as compared with those of the normal. After treatment, BP and whole blood viscosity were reduced and ED index was increased in Group A, Group B and Group C, and EA index in Group A was lowered as compared with those in Group D (P
