ABSTRACT

Aim The present study is designed to elucidate the protective effect of Polysaccharide from Spirulina Platensis(PSP) on dopaminergic neurons of MPTP-induced Parkinsons disease model in C57BL mice.Methods C57BL mice were divided into 5 groups:control group,MPTP group,PSP 200 mg?kg-1 plus MPTP group,PSP 400 mg?kg-1 plus MPTP group,and PSP 800 mg?kg-1 plus MPTP group.Immunohistochemistry assay,RT-PCR,and HPLC-ECD were used to detect the immunoreactivity of tyrosine hydroxylase(TH) and dopamine transporters(DAT),the expression of TH mRNA and DAT mRNA in substantia nigra(SN),the content of dopamine(DA) and its metabolites,i.e.3,4-dihydroxyphenyl acetic acid(DOPAC) and homovanilic acid(HVA) in striatum(Str).Results The result showed that,compared with control group,the TH and DAT immunoreactive stain in MPTP group was significantly decreased(P

ABSTRACT

Objective: To evaluate the antitumor effect of polysaccharide from spirulina platensis(PSP), and its synergic and attenuated effects on chemotherapeutic drugs in tumor-bearing mice. Methods: Mouse models of transplanting tumor were established. Antitumor effects of PSP and PSP combined with chemotherapeutic drugs were observed and the effect of PSP on leukopenia induced by chemotherapeutic drugs was also investigated. Results: PSP significantly inhibited S180 and H22, the inhibitory rate being 50 %-58 %and 43 %-51 %respectively. PSP evidently increased the antitumor action of cytoxan, PDD and 5-Fu, and elevated the number of white blood cells in leukopenia. Conclusion: PSP can obviously inhibit the tumor growth, also has synergic and attenuated effects on chemotherapeutic drugs.

ABSTRACT

Objective To investigate the effects of polysaccharide from Spirulina platensis (PSP) on the growth of HeLa cells. Methods The antiproliferation activities of PSP on HeLa cells were evaluated by MTT assay. The morphologic changes of HeLa cells were observed under light microscope. Cells division cycle was measured by flow cytometry. Results It was showed that with the increase of PSP concentration, the inhibited rate on HeLa cells increased and the survived rate declined; The HeLa cells exposed to PSP for 24~48h displayed significant morphologic changes; PSP had a specific effect on the HeLa cell division cycle, increased the number of cells in phase G 1. Conclusion These results indicated that the effects of PSP on HeLa cells may probably be attributed to increase of the number of cells in phase G 1.

ABSTRACT

Objective To observe the influence of polysaccharide from spirulina platensis(PSP) on human embryo lung diploid fibroblastic DNA damage and repair after UV irradiation. Methods After UV irradiation for 30min, the single cell gel electrophoresis(SCGE) was used to investigate the level of DNA damage, the level of DNA damage and repair at 30, 60, 90min were also investigated by SCGE respectively. Results The level of DNA damage induced by UV irradiation was prevented by PSP, and it also promote DNA repair capacity. Conclusion PSP can increase the activity of DNA endonuclease and DNA ligase in dose-de- pendrntly.
