ABSTRACT

OBJECTIVE:To establish a method for simultaneous determination of ferulic acid(FA)and TMP in serum sample.METHODS:After deprotein with boiling water-bath,RP-HPLC was performed on ODS C 18 column(250mm?4.6mm)with methanol-water(containing0.1%acetic acid)=8∶17as the mobile phase and UV detection at280nm.RESU LTS:The calibration curve of FA was linear within the range of30.2~70.8?g/ml,Y=57.51+413.12X,r=0.9974;the curve of TMP was linear within the range of10.1~50.7?g/ml,Y=41.32+308.64X,r=0.9922.CONCLUSION:Compared with the traditional extraction,the method is simple,rapid and sensitive,and can be used for simultaneous determination of the contents of active compositions in Chinese materia medica.

ABSTRACT

OBJECTIVE: To study the relationship among the dosage, serum concentration and clinical effect of phenytoin(PHT) in treating epileptic patients .METHODS: 130 steady -- state serum concentrations of PHT were determined by fluores-cence polarization immunoassay(FPIA) in 70 patients with or without other drugs used in combination .RESULTS: The indi-vidual difference among the patients who received routine dosage of PHT was very significant. Intoxication occurred ex-tremely often in patients whose PHT serum concentrations were over 30?g/ml .CONCLUSION: It is necessary to perform TDM(Therapeutic Drug Monitoring) and individualize the use of PHT,so that the safe and effective use of drug can be ensured.

ABSTRACT

OBJECTIVE:To develop a RP-HPLC method for the determination of salicylic acid in serum and to apply this method to the pharmacokinetic and bioavailability study of salycylic acid in compound aspirin preparations.METHODS:Waters2690HPLC instrument was used with Diamonsil C 18 column(5?m,250mm?4.6mm)as stationary phase and methanol-ace?tonitrile-0.2%phosphoric acid(18∶32∶50)as mobile phase at a flow rate of1.0ml/min,and the detective wavelength was237nm.RESULTS:Calibration curve of salicylic acid was linear in the range of0.40~101.00?g/ml(r=0.9999).CONCLUS_ ION:The method is simple,sensitive,rapid and suitable for pharmacokinetic and bioavailability study of salicylic acid.

ABSTRACT

OBJECTIVE:To study the effects of diltiazem on the blood concentration and the renal toxicity of cyclosporin A(CsA) in patients with renal transplantation.METHODS: 25 patients with renal transplantation were divided into two groups,the therapeutic group were treated with diltiazem plus CsA and control group CsA alone and the valley blood concentration of CsA was detected by FPIA assay.RESULTS:The dosage of CsA in therapeutic group was less than that in control group significantly,the levels of UA,Scr and BUN in patients were decreased by diltiazem.CONCLUSION:The diltiazem therapy can decrease the dosage and toxic effect of CsA,then save the patient's medical costs.
