ABSTRACT

Incidental detection of two primaries call for oncosurgeon’s own judgment to decide the best therapeutic approach as no guidelines exist for the rarity of condition. This is the rare case of incidental renal cell carcinoma in a patient of endometrial carcinoma. Renal cell carcinoma was detected on preoperative MRI in a patient who presented as postmenopausal bleeding and histology proven endometrial carcinoma. Final histology confirmed dual primaries with uterine primary being endometrioid adenocarcinoma, whereas renal primary was clear cell carcinoma stage II. Estrogen receptors (ER) have been identified in Hamster and Mouse kidneys as well as in renal cell carcinoma tissues. High plasma estrogen found in some patients of synchronous renal and endometrial cancer may partly explain the association of these two primaries, though not in all cases. Increased serum leptin levels and a common low penetrance susceptibility gene have been reported in both these cancers.

ABSTRACT

Objective To investigate the effect of the serum leptin and hs-CRP levels by Levetiracetam in patients with epilepsy.Methods 150 patients with epilepsy from our hospital were selected and randomly divided into the control group and the experiment group with 75 cases in each group.The control group were treated by sodium valproate and carbamazepine and the experiment group were treated in the base of the control group with Levetiracetam.The LP,hs-CRP,TC and TG levels were compared before and after treatment.Results Compared with before treatment,there was no significant change in serum LP,TC and TG levels,the hs-CRP were lower(P<0.05),and the indexes of the experimental group were significantly lower than those of the control group(P<0.05).Conclusion Levetiracetam to the pediatric epilepsy patients to good therapeutic effect,reduce the side effects of hazards,speculated that the mechanism may be related to the left B Raschig Tan to maintain patients serum leptin,triglyceride,total cholesterol levels,reduce the levels of high sensitivity C reactive protein related.

ABSTRACT

Objective To explore the levels of leptin(LEP) and soluble leptin receptor (sOB‐R) in breast cancer patients′ser‐um ,and to discuss the relationship between LEP ,sOB‐R ,free leptin index(FLI) and the patients′clinical pathologic features .Meth‐ods 70 serum specimens of breast cancer patients who confirmed by pathological diagnosis were collected before the surgery ,the serum of benign breast disease group(n=50) and normal group(n=50) were also collected as control .The levels of LEP and sOB‐Rinserumweredetected,andtheresultafterbodymassindex(BMI)correctionwereanalyzed.Finally,therelationshipbetweenthe expression of LEP ,sOB‐R and clinical pathologic features were analyzed .Results The serum level of LEP in breast cancer group was highly increased than the other two groups (P0 .05) .The level of LEP in the postmenopausal breast cancer patients were highly increased than menopause patients (P>0 .05) ,and the expression of LEP in cancer group were signifi‐cant higher than two control groups by both postmenopausal and menopause (P>0 .05) .The result suggested the serum level of LEP and FLI are associated with pTNM stages and lymph node metastases (P<0 .05) ,and elevated level of LEP in postmenopausal patients had differences in pTNM stages and lymph node metastases (P< 0 .05) .Conclusion The serum LEP level and the FLI may be potential indicators to evaluate the prognosis of the patient with breast cancer .The serum LEP may promoted the process of the postmenopausal patients of breast cancer .

ABSTRACT

Introduction: Serum leptin levels are increased in chronic kidney disease (CKD) patients primarily due to decreased clearance by kidneys. As leptin is a 16 Kda protein, it is also not cleared even by dialysis using conventional dialyzers or by continuous ambulatory peritoneal dialysis (CAPD). Studies have shown that elevated leptin levels are corrected after successful renal transplantation. With this intention, we determined if restoration of renal function with kidney transplantation can reduce serum leptin concentration in CKD patients. Materials and Methods: A total of 21 Patients undergoing living donor kidney transplantation were studied. There were 13 men and 8 women, from 16 to 45 years of age. All patients were receiving Hemodialysis prior to transplant. All patients received triple immunosuppressant therapy after the surgery. There were no graft rejections. Blood samples were collected under fasting conditions before and 6 days after transplantation. Results: The mean age of the patients was 28.38±9.38 years. Pre transplantation leptin concentration was 9.96 + 3.48 ng/ml and this decreased to 4.07±1.7 ng/ml within six days of transplantation (p<0.0001). However there was no concomitant change in Body Mass Index (BMI) as the follow-up was too short. Plasma Creatinine level declined from 7.5±1.6 mg/dl to 1.1±0.7 mg/dl within six days after transplantation. Conclusion: Successful renal transplantation immediately reduces serum leptin levels along with serum creatinine. The reduction in serum leptin levels after renal transplantation is likely due to reversal of renal function. Neither pre nor post transplant plasma Leptin levels correlated significantly with BMI in our study.

ABSTRACT

Objective To explore the effect of big strain rhodiola injection on levels of leptin and leptin receptor mRNA in serum and liver of rabbit with fatty liver.Methods 120 healthy adult male New Zealand white rabbits were grouped with a random number table method.30 rabbits with normal diet for eight weeks in normal group;90 rabbits in fat group were fed with high fat diet for four weeks to prepare fatty liver model in which 30 rabbits were fed with high-fat diet for four weeks as model group,30 rabbits with high-fat diet and glyburide metformin tablets [150 mg/(kg·d)]for 4 weeks as control group,30 rabbits with high-fat diet and big strain rhodiola injection [40 mg/(kg·d)]for four weeks as experimental group.After the test, levels of serum leptin,hepatic leptin,expression levels of leptin receptor mRNA and levels of liver index were compared among groups.Results Serum leptin and liver leptin in experimental group and control group were all increased to varying degrees,and the serum leptin level [(4.51 ±1.17)ng/mL],liver leptin level [(6.43 ±1.93)ng/mL]of experimental group was lower than that of control group respectively,in which the serum leptin was (7.36 ±1.26)ng/mL and liver leptin was (12.33 ±3.27)ng/mL (P<0.05).Serum leptin and leptin liver receptor mRNA in experimental group and control group were all increased to varying degrees,and the serum leptin mRNA level (0.98 ±0.15),liver mRNA level (1.78 ±0.33)of experimental group was lower than that of control group,in which the mRNA level of serum leptin was (2.57 ±0.29)and liver leptin mRNA levels was (3.31 ±0. 26)(P<0.05).Levels of liver index in experimental group and control group were increased,and liver index of experimental group [(3.98 ±0. 13)%]was lower than that in control group [(4.77 ±0.20)%],and the difference was statistically significant (P<0.05).Conclusion Big strain rhodiola injection could significantly reduce leptin levels in serum and liver of rabbits,reduce expression the leptin receptor mRNA levels and improve liver index,which has guiding significance for the treatment of fatty liver.

ABSTRACT

Objective To investigate the effect of Glycyrrhizin combined with puerarin on serum Leptin and insulin resistance in non-alcoholic fatty liver disease(NAFLD) patients.Methods One hundred and twenty patients with NAFLD were randomized into 4 groups,which were control group,compound Glycyrrhizin group,puerarin group,combined group,and each group was 30 cases.Patients in control group were received the regular liver protecting therapy including vitamins,amino acids,glucurolactone,in compound Glycyrrhizin group were given 60 ml compound glycyrrhizin solution (iv),in puerarin group were given puerarin at dose of 400 mg by intravenous infusion,and in combined group were given both compound glycyrrhizin and puerarin combination.All treatment period was 4 weeks.The levels of serum serum alanine aminotransferase (ALT),aspartate aminotransferase (AST),total cholesterol (TC),triglyceride (TG),leptin (LP),fasting blood glucose (FBG) and insulin(INS) were measured,and the insulin resistance index(IRI) was calculated.The liver CT image of patients were performed by Germany Siemens dual source CT instrument.Results The levels of serum ALT,AST,TC,TG,LP and IRI in control group at before and after treatment were ((83.08 ± 115.68) U/L vs.(43.32 ±11.72) U/L,(52.12±15.62) U/Lvs.(36.08 ±7.28) U/L,(6.20±1.30) mmol/Lvs.(5.60 ±0.70) mmol/L,(2.70 ±0.50) mmol/L vs.(2.10 ±0.40) mmol/L,(14.63 ±3.26) μg/L vs.(7.61 ± 2.46) μg/L,(7.9 ± 1.8) vs.(7.0 ± 1.2)),and the difference were statistically significant (t =12.828,4.244,16.648,21.442,3.341,16.152 respectively,P < 0.01).The levels of serum ALT,AST,TC,TG,LP and IRI in compound glycyrrhizin group after treatment were ((43.28 ± 11.06) U/L,(37.28 ± 7.22) U/L,(5.70± 0.80) mmol/L,(2.20 ± 0.50) mmol/L,(7.89 ± 2.26) μg/L,(7.1 ± 1.6) respectively,significant different from before treatment ((83.06 ± 14.38) U/L,(51.68 ± 15.48) U/L,(6.30 ± 1.50) mmol/L,(2.60 ± 0.40) mmol/L,(15.13 ± 3.87) μg/L,(7.8 ± 2.2) respectively,t =8.893,4.225,16.520,24.708,6.353,21.137 respectively,P < 0.01).The levels of serum ALT,AST,TC,TG,LP and ISI in puerarin group after treatment were (44.26 ± 9.68) U/L,(36.86 ± 6.88) U/L,(5.60 ± 0.70) mmol/L,(2.26 ± 0.48) mmol/L,(6.89 ± 2.18) μg/L,(7.0 ± 1.8) respectively,significant different from that before treatment ((82.68±14.36) U/L,(50.06±15.23) U/L,(6.20±1.60) mmol/L,(2.70±0.52) mmol/L,(15.68 ±3.26)μg/L,(7.7 ±2.8) respectively,t =7.087,8.138,18.159,7.244,7.470,32.283 respectively,P < 0.01).The levels of serum ALT,AST,TC,TG,LP and ISI in combined treatment group after treatment were (22.28 ± 9.38)U/L,(28.48 ± 9.06) U/L,(5.00 ± 0.60) mmol/L,(1.70 ± 0.40) mmol/L,(4.63 ± 2.36) μg/L,(6.20± 1.6) respectively,significantly different from that before treatment ((84.62 ± 14.88) U/L,(49.12 ± 16.56)U/L,(5.70 ± 1.60) mmol/L,(2.78 ± 0.50) mmol/L,(14.78 ± 3.68) μg/L,(7.6 ± 2.1),t =14.255,11.272,8.371,9.941,8.102,37.626,P < 0.01).The levels of serum ALT,AST,TC,TG,LP and ISI of patient were no significant difference before treatment,but after treatment,these indexes in combined therapy group were the lowest among 4 groups (P < 0.05).And there were no significant difference among control group,compound glycyrrhizin group,puerarin group (P > 0.05).Conclusion Compound glycyrrhizin combined with puerarin is proved to be an effect treatment method for NAFLD through decreasing serum leptin,improving insulin resistance.

ABSTRACT

The present study was designed to evaluate the relationship between serum leptin and sex hormones in obese infertile men and women. Sex hormones (LH, FSH and testosterone) and serum leptin of infertile obese men (n=66) and women (n=30) compared with control fertile men (n=60) and women (n=30) with same ages to find the relation and contribution of serum leptin and sex hormones in causation of fertility or infertility. The results revealed that Serum leptin level in obese (BMI>240 fertile and infertile male and female were significantly (p<0.01) higher than normal (BMI<24) fertile male but in infertile normal subjects it was significantly decreased. Values of LH, FSH and Testosterone were significantly (p<0.01) higher in obese (BMI<24) fertile and infertile men and women than normal (BMI<24) subjects. Serum leptin has strong negative correlation with LH, FSH and testosterone in fertile obese and normal male and female which were statistically significant at the level of p<0.05. This study has concluded that hyperleptinemia is associated with infertility in men and women. Sex hormonal imbalance may also be associated with BMI and serum leptin in infertility.

ABSTRACT

Objective To investigate the relationship between leptin receptor(LEPR) gene Gln223Arg in polymorphism and serum leptin level of patients with asthma.Methods A case-control study was conducted.One hundred and ninety-two asthma patients were recruited from asthma gene library,among them 100 patients only with asthma(asthma group) and 92 patients with both asthma and metabolic syndrome(asthma complicated with metabolic syndrome group).And 108 normal people were selected as control group.Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the polymorphism of LEPR gene Gln223Arg locus.Serum leptin levels were measured by enzyme-linked immuno sorbent assay,its relationship with asthma was analyze.Results The frequencies of genotype GG,GA,AA in asthma group,asthma complicated with metabolic syndrome group and normal control group were (83.00% (83/100) vs.83.70% (77/92) vs.70.37% (76/108),15.00% (15/100) vs.15.22% (14/92) vs.27.78% (30/108),and2.00% (2/100) vs.1.08% (1/92) vs.1.85% (2/108) respectively).G and A alleles in asthma group,asthma complicated with metabolic syndrome group and normal control group were 90.50% vs.91.30%vs.84.26%,9.50% vs.8.70% vs.15.74% respectively.Compared with the control group,GG genotype and GA + AA genotype were different between in asthma group and asthma complicated with metabolic syndrome group.The patients with GG genotype showed 2.056 (x2 =4.599,P =0.032,OR =2.056,95% CI =1.057-3.999) and 2.161 folds (x2 =4.907,P =0.027,OR =2.161,95 % CI =1.084-4.311) risk in asthma group and asthma complicated with metabolic syndrome group than that with GA + AA genotype.In terms of leptin levels,in asthma group,female were significant higher than that of control group (P =0.037,P =2.93),while in asthma complicated with metabolic syndrome group,male and female were significantly higher than control group (P =0.001,0.000 ; P =4.530,4.690).Conclusion The polymorphisms of LEPR gene locus Gln223 Arg are related with asthma.Genotype GG carrier is susceptible for the disease.In female,serum leptin is positive relationship with asthma group,especially in asthma complicated with metabolic syndrome.

ABSTRACT

Objective To observe the effect of acupuncture combined with traditional Chinese medicine on serum leptin contents and the expression of leptin receptor in the hypothalamus in simple obesity rats so as to explore the underlying mechanism. Methods A total of 120 male Wistar rats were randomly divided into normal control group and high grease group, which was established by fed with high fat forage for 16 weeks. 60 successful rat models were divided into shilianyin+electro-acupuncture group, fatness model group, and control group with random digits table. Electro-acupuncture was applied to Zusanli and Zhongwan, and Shilian-yin 22.4 g/kg was administered, once daily, for 14 days in the shilianyin + electro-acupuncture group; Fatness model group was lavaged with isotonic Na chloride 2 ml/kg; while the control group was lavaged with sibutramine 1.6 mg/kg. The body weight, body length and Lee's index were measured once every 10 days. Serum leptin contents were measured by radioimmunoassay, and leptin receptor expression was detected with immunohistochemistry. Results In comparison with fatness model group (13.401 ± 6.138) ng/ml, the serum leptin content of shilianyin+electro-acupuncture group decreased significantly (9.467+ 3.587) ng/ml (P<0.01) , and the number of leptin receptor expression increased considerably (leptin-acceptor masculine cell was 52.30±21.35piece/mm2) (P<0.01). Conclusion The benign regulation of acupuncture combined with traditional Chinese medicine on hypothalamus and serum leptin of obesity body is a significant joint of improving leptin-counteract and abnormal internal hormone metabolism.

ABSTRACT

During pregnancy and protein restriction, changes in serum insulin and leptin levels, food intake and several metabolic parameters normally result in enhanced adiposity. We evaluated serum leptin and insulin levels and their correlations with some predictive obesity variables in Wistar rats (90 days), up to the 14th day of pregnancy: control non-pregnant (N = 5) and pregnant (N = 7) groups (control diet: 17 percent protein), and low-protein non-pregnant (N = 5) and pregnant (N = 6) groups (low-protein diet: 6 percent). Independent of the protein content of the diet, pregnancy increased total (F1,19 = 22.28, P < 0.001) and relative (F1,19 = 5.57, P < 0.03) food intake, the variation of weight (F1,19 = 49.79, P < 0.000) and final body weight (F1,19 = 19.52, P < 0.001), but glycemia (F1,19 = 9.02, P = 0.01) and the relative weight of gonadal adipose tissue (F1,19 = 17.11, P < 0.001) were decreased. Pregnancy (F1,19 = 18.13, P < 0.001) and low-protein diet (F1,19 = 20.35, P < 0.001) increased the absolute weight of brown adipose tissue. However, the relative weight of this tissue was increased only by protein restriction (F1,19 = 15.20, P < 0.001) and the relative lipid in carcass was decreased in low-protein groups (F1,19 = 4.34, P = 0.05). Serum insulin and leptin levels were similar among groups and did not correlate with food intake. However, there was a positive relationship between serum insulin levels and carcass fat depots in low-protein groups (r = 0.37, P < 0.05), while in pregnancy serum leptin correlated with weight of gonadal (r = 0.39, P < 0.02) and retroperitoneal (r = 0.41, P < 0.01) adipose tissues. Unexpectedly, protein restriction during 14 days of pregnancy did not alter the serum profile of adiposity signals and their effects on food intake and adiposity, probably due to the short term of exposure to low-protein diet.

ABSTRACT

OBJECTIVE: This study was performed to prove the relationship between serum leptin level and bone mineral density of lumbar spine, femur neck and bone markers in postmenopausal Korean women. METHODS: We measured serum leptin, serum osteocalcin, urine deoxypyridinoline levels and bone mineral density of lumbar spine, femur neck in 88 premenopausal and 118 postmenopausal women who visited St. Vincent Hospital of Catholic University of Korea from March 1st, 2007 to December 31th, 2007. RESULTS: Statistically significant correlation was shown between serum leptin level and body mass index (BMI) in both premenopausal (r= 0.343, P<0.0001) and postmenopausal women (r=0.360, P<0.0001). And no significant correlation was observed between serum leptin level and bone mineral density (BMD) of lumbar spine and femur neck in premenopausal women (r=0.013, P=0.107 and r=0.004, P=0.425, respectively), but in postmenopausal women, there was a positive correlation between serum leptin and lumbar spinal BMD (r=0.085, P=0.02). But after the adjustment with age and BMI, the serum leptin and BMD of lumbar spine did not showed a significantly correlation in the same group (r=0.088, P=0.939). Also, no significant correlations were observed between serum leptin level and serum osteocalcin and urine deoxypyridinoline in premenopausal (r=0.004, P=0.566 and r=0.002, P=0.707, respectively) and postmenopausal women (r=0.026, P=0.096 and r=0.000, P=0.933, respectively). CONCLUSIONS: In our study, there is no significant correlation between serum leptin level and bone mineral density and bone markers in premenopausal and postmenopausal Korean women. Our own data would suggest that leptin has both negative and positive effects in bone mass regulation. Furthermore, larger clinical studies are necessary to clarify leptin's role to assess the contribution of the central and peripheral role of leptin in the overall maintenance of bone turnover.

ABSTRACT

PURPOSE: This study was performed to investigate the correlation between mother's characteristics and obesity index, percentage of body fat, total cholesterol, and serum leptin levels in obese children. METHOD: Sixty two students in 4th, 5th, and 6th grades were selected from two elementary schools located in the suburbs of Seoul. Mother's characteristics were collected by questionnaires. Biophysical indexes were as follows; obesity indexes were calculated by height and weight, percentages of body fat were measured by a bioelectrical impedence fatness analyzer, total cholesterol levels were measured by a Hitachi automatic analyzer, and serum leptin levels were measured by the immunoradiometric assay method. RESULT: The degree of mother's concern for their children's obesity showed a statistically significant positive correlation with biophysical indexes such as obesity index and serum leptin level in obese children. However, there were no significant correlations between mother's economic state, education level, occupation and biophysical indexes. Also, there were no significant correlations between mother's concern for their children's obesity and percentage of body fat, and serum cholesterol levels. CONCLUSION: These results indicate that mother's concern for their children's obesity is one of the most important factors in child obesity.

ABSTRACT

BACKGROUND: Leptin is known to affect bone metabolism both centrally and peripherally. This study was performed to investigate the relationship between leptin and bone mineral density(BMD) in healthy premenopausal and postmenopausal Korean women. METHODS: 140 women were recruited for a routine health check-up. Anthro-pometric and biochemical data were checked as usual. BMDs were measured by dual x-ray absorptiometry of the spine and femur in 67 premenopausal women and 73 postmenopausal women, in addition to their serum leptin levels. RESULTS: Serum leptin level showed no correlation with BMD in premenopausal women, but there was a positive correlation betwen serum leptin and spinal BMD in postmenopausal women(r=0.468, p<0.001). After the correcting for age, body mass index, and duration of menopause, the serum leptin level and BMD still showed a positive correlation(r=0.217, p=0.088) although weak. The women in the lowest quartile of serum leptin level showed significantly lower lumbar and femoral neck BMD. CONCLUSION: Leptin level seems to have a weak relationship with BMD showing different features in premenopausal and postmenopausal women.

ABSTRACT

As obesity is known to be related to hyperlipidemia, insulin and leptin resistance, and other chronic diseases, much recent research has focused on functional food materials and their anti-obesity activity. This study was performed to study the effects of Artemisia Iwayomogi oligosaccharide AIPI on the anti-obesity function in normal rats and diet-induced obese (DIO) rats. F344 male rats were divided into four groups: Normal-control (CONT), normal-AIPI, DIO-CONT and DIO-AIPI. The groups were provided with water (in the CONT groups) or another drink for 4 weeks. The final body weights of rats in the DIO-AIPI group were lower than those in the CONT group. Abdominal adipose tissue weight per kg of body weight in the DIO-AIPI group was significantly lower than that in the DIO-CONT group. Also, the final levels of serum-triglyceride, serum-total cholesterol and serum-low density lipoprotein cholesterol in the DIO-AIPI group were lower than those in the DIO-CONT group. Moreover, the serum-high density lipoprotein cholesterol level in the normal-AIPI group was significantly higher than that in the normal-CONT group. Finally, the serum-leptin concentration was significantly lower in the DIO-AIPI group. Total lipid, triglyceride, and total cholesterol contents in the feces of the DIO-AIPI group were as high as 142%, 199%, and 165% of the respective values of the DIO-CONT GROUP. These results indicate that orally administered Artemisia Iwayomogi oligosaccharide not only has hypotriglycemic and hypocholesterolemic effects, but also has the effect of reducing the body weight and the abdominal adipose tissue weights obese rats. Therefore, we expect that Artemisia Iwayomogi oligosaccharide AIPI may have an anti-obesity function in F344 diet-induced obese rats.

ABSTRACT

Behavior modification is an approach to decrease degree of obesity through changing maladaptive eating habit and life style. It produces small amount of weight loss but it has few side effects and low dropout rate. It also has great effectiveness on maintenance of decreased weight. This study was performed to investigate effects of behavior modification on physical variables, fat and thin habit, and self-esteem in obese elementary school children. Sixty two students of 4th, 5th, and 6th grade were selected from two elementary schools located in the suburbs of Seoul. Thirty four children in one school were assigned to experimental group and twenty eight children in another school were assigned to control group. All subjects were healthy and were not on any type of obesity control. Behavior modification, in this study, was consisted of introduction, self-monitoring and stimuli control, education about diet and physical activities, individual counseling and reinforcement, requesting personal assistance, cognitive restructuring, and closing and long-term planning. Experimental group was received 60 ~ 70 minutes of behavior modification once a week for 8 weeks from October 16 to December 22, 2000. Control group was not received any intervention. Data were collected before and after intervention by measuring physical variables, fat and thin habit, and self-esteem. Physical variables consisted of obesity index, skinfold thickness, body fat(percentage of body fat, fat mass, fat free mass), serum lipids(total cholesterol, HDL-C, LDL-C, triglyceride) and serum leptin. The results were as follows ; 1. physical variables 1)Obesity index of the experimental group was significantly decreased after behavior modification. But there was no significant difference between two groups. 2)The increase of skinfold thickness was significantly low in the experimental group compared to the control group. 3) Percentage of body fat and fat mass were significantly decreased in the experimental group. Fat free mass was significantly increased in the experimental group. 4) Total cholesterol, HDL-C, triglyceride and serum leptin between the experimental group and control group showed no significant difference. LDL-C was significantly decreased in the control group. 2. Thin habit score was significantly increased in the experimental group. In subcategories of habit, thin scores of life style, attitude, social relationship, nutrition were significantly increased in the experimental group. 3. Self-esteem score was significantly increased in the experimental group. These results indicate that behavior modification is effective in decreasing percentage of body fat and fat mass, in less increasing skinfold thickness and in increasing fat free mass, thin habit, and self-esteem. In conclusion, behavior modification can be used as safe and effective strategy for managing obesity in elementary school children.

ABSTRACT

Objective To explore the effect of serum leptin on puberty development in schoolboys.Methods 170 health schoolboys were investigated on the first spermatorrhea time and detected the levels of serum leptin, testosterone,follicle stimulating hormone (FSH), luteinizing hormone (LH). Case-controlled study was carried by multivariable logistic regression in puberty development of schoolchildren with and without spermatorrhea. Meanwhile, follow-up studies on the independent influencing factors on the puberty development were applied to this investigation in continuous two years.Results The levels of serum leptin, testosterone,FSH and LH of schoolboys with spermatorrhea were significantly higher than those of schoolboys without spermatorrhea. Multiple logistic regression analysis demonstrated that the serum leptin,testosterone, FSH, and bodyweight were the independent influence factors in puberty of the schoolboy.The levels of serum leptin,testosterone of pre-spermatorrhea schoolboys were significantly higher than schoolboys without spermatorrhea.Conclusions Serum leptin,testosterone, FSH, and bodyweight were the independent influence factors in puberty of the schoolboy.The levels of serum leptin and testosterone had obviously changed before spermatorrhea.

Objective To observe the changes of serum leptin(LP) levels and serum tumor necrosis tactor-alpha(TNF?) concentrations and to explore the relationships with fasting serum insulin levels and congestive heart failure(CHF).Methods Serum leptin and inslin concentrations were measured by radioimmunoassay and tumor necrosis factor-alpha by ELISA.Body mass index(BMI),mean arterial blood pressure(MBP),fasting serum insulin(FINS),LP and TNF-alpha levels were determined in 40 patients with CHF and in 20 healthy controls matched for gender,age,BMI and MBP.The correlations between leptin and other parameters were analyzed.Results ⑴The fasting serum LP and FINS concentrations of patients with NYHA grade Ⅲ and NYHA grade Ⅳ were significantly lower as compared with the controls(P0 05).⑵The TNF alpha was higher in heart failure patients that in healthy controls(P

ABSTRACT

The ob gene product leptin is thought to be an adipostatic hormone through the regulation of food intake and energy expenditure. There are many reports that serum leptin concentration was increased in CRF patients, especially CAPD patients. The causes of elevated serum leptin concentration in CRF are believed to be multifactorial. Increased body fat mass, decreased residual renal function, active inflammation and hyperinsulinemia all are suggested to influence serum leptin concentration in CAPD patients. In this study, in order to investigate the pathogenic mechanism of increased serum leptin level in CAPD patients, we observed the changes of serum leptin concentration, leptin expression in the abdominal subcutaneous fat tissue, body fat composition, residual renal function, serum insulin concentration and CRP. Thirteen patients(7 men and 6 women, mean age 53+/-14 years) were enrolled in this study. Serum leptin concentration was measured by RIA before start of CAPD(baseline data), 5 days and 1, 3 months after start of CAPD. Simultaneously, fat tissues were aspirated from the abdominal subcutaneous fat tissues for analysis of ob gene expression. Ob mRNA expression was measured by semiquantitative RT-PCR method. The changes of serum insulin concentration, C-reactive protein, residual renal function were measured. All studies were done immediately before starting CAPD, 5 days, 1 month and 3 months after starting CAPD. Total body fat was estimated by dual energy X-ray absorptiometry and abdominal visceral and parietal fat area measured by computed tomography were done at 1-3 days(baseline data), 1 month, 3 months after start of CAPD. Serum leptin concentration increased significantly as early as 5 days after start of CAPD and maintained high up to 3 months(4.3+/-2.6->8.2+/-7.6->7.4+/-6.5->10.8+/-13.8ng/mL), while leptin expression in the abdominal subcutaneous fat tissue did not change during the study period(0.24+/-0.06->0.25+/-0.08->0.20+/-0.07->0.34+/-0.21ng/mL). No significant changes of body fat composition, residual renal function and serum insulin concentration were observed during the study period. These results suggest that increase of serum leptin concentration after CAPD may be due to increase of local leptin production, especially from the peritoneum, as has also been suggested by several reports of relatively higher dialysate leptin.

ABSTRACT

PURPOSE: To investigate the effect of growth hormone(GH) secretion on serum concentration of leptin, we evaluated the differences in serum concentration of leptin between patients with growth hormone deficiency(GHD) and GH normal short stature in prepubertal male children, and the relationship between GH and serum leptin level. Also, we analyzed the correlation between serum concentrations of leptin and insulin-like growth factor-I(IGF-I), insulin-like growth factor binding protein-3(IGFBP-3), total cholesterol(TC), free fatty acid(FFA) and body mass index(BMI). METHODS: Eight cases with GHD and 10 cases with GH normal short stature were included in this study. Patients were all male, prepubertal children with mean age 10.6 years. Blood samples were collected in these patients by L-dopa GH provocation test. Statistical analysis of the comparisons of serum leptin level and correlation of multiple variables were performed using PC-SAS program. Significance was defined as P<0.05. RESULTS: The mean serum concentration of leptin in GHD(12.27+/-10.63ng/mL) was significantly higher than in GH normal short stature(5.39+/-3.21ng/mL)(P=0.0344). Serum concentration of leptin was negatively correlated to GH(y=-7.7x+43.3, r=-0.23, P=0.0109). Serum concentration of leptin was positively correlated to BMI(y=2.5x-33.3, r=0.75, P=0.0003). Serum concentration of leptin was not correlated to IGF-I, IGFBP-3, TC and FFA. CONCLUSION: Serum concentration of leptin was siginificantly increased in patients with GHD compared to GH normal short stature and was negatively correlated to GH.

ABSTRACT

Objective:To explore effect of serum leptin level on nutriture in the patient of chronic renal failure(CRF)and the relation with TCM syndrome types of CRF.Methods:110 cases in CRF group were divided into 5 subtypes and 30 normal persons were used as control group.Serum leptin level,body height,body weight,and body mass index(BMI)were determined in both groups;and serum albumin,serum pre-albumin,transferin,serum creatinine,urea nitrogen,C-reaction protein,blood insulin,and nutrition risk index(NRI)were detected in CRF group.Results:Serum leptin level in CRF group was higher than that in the control group,and leptin level and nutritional index level in both the subtype groups of Qi-and Yang-deficiency of spleen and kidney were significantly different with those in other 3 subtype groups.Conclusion:High blood leptin is possibly one of the reasons inducing malnutriture of CRF patient,and serum leptin is possibly a partial basis of substances of Qi-and Yang-deficiency syndrome of CRF.
