ABSTRACT

Objective:To explore the changes of blood viscosity in high-intensity alternating magnetic field and the mechanisms.Methods:Five adult sheep were randomly selected and the blood samples were placed in highintensity alternating magnetic field.Before and after exposure,the blood samples were taken and divided into 2 groups:a control group and a magnetic field group.The blood rheology and transmission electron microscopy (TEM) were performed.Results:Compared to the control group,the high shear viscosity of whole blood was decreased in the magnetic field group (P<0.05);the whole blood low shear viscosity and plasma viscosity were also decreased (both P<0.01).TEM showed the changes in red blood cell morphology and the double concave disc curvature.The radian of double concave disc and cell volume in the magnetic field group was larger than those in the control group.Conclusion:The high intensity alternating magnetic field may affect the distribution of surface charge and molecular current in blood cells,which in turn decrease the aggregation of cells and the blood viscosity.

ABSTRACT

Objective To evaluate the quality and application value of improved sheep blood chocolate medium.Methods The ATCC 10211 of Haemophilus influenzae was inoculated into the modified medium and unmodified medium,the average growth index(GI)of Haemophilus influenzae in two kinds of culture medium was compared.Based on 352 selected qualified sputum specimens for detection of Haemophilus influenzae,the positive isolation rate of medium was compared between the two groups.Results GI value of the traditional blood chocolate culture medium was (3.69 ±0.58),which was significantly lower than (15.08 ±1.34)of the improved sheep blood chocolate culture medium,the difference was significant (t =25.31,P <0.01 ).352 sputum specimens in the improved sheep blood chocolate culture medium,Haemophilus influenzae detected in 41 cases,the positive rate was 11.65%.And 352 sputum specimens in traditional sheep blood chocolate culture,Haemophilus influenzae detected in 18 cases,the positive rate was 5.54%.There was significant differenceof Haemophilus influenzae in separation based on 2 kinds of chocolate culture medium (χ2 =21.04,P <0.05).Conclusion Haemophilus influenzae significantly improved sheep blood colony in chocolate culture medium,grows well,easily to be identified,which helps to detect sputum specimens of Haemophilus influenzae.

ABSTRACT

Bartonella henselae was detected in defibrinated sheep blood employed in supplementing a selective bacteria culture medium by nested PCR. We recommended that highly sensitive technical tests be run to ensure a sterile culture medium for Bartonella spp. isolation, since infected blood samples used in preparation could lead to false-positive results.

ABSTRACT

Hemoderivados de origem animal säo amplamente empregados no preparo de meios de cultura utilizados em práticas laboratoriais de interesse em Saúde Pública. Para avaliaçäo da qualidade deste produto foram testados dois sistemas de coleta de sangue em carneiro. O sistema tradicional, utilizando seringa, e um sistema em bolsa confeccionada em cloreto de polivinila (PVC) atóxico. Foram utilizados 40 carneiros adultos, machos e fêmeas, distribuídos de forma aleatória em dois grupos compostos por vinte animais cada. O sangue coletado para o estudo foi manipulado com assepsia em ambiente estéril, sendo posteriormente incubado a 37 graus C durante 24 horas e semeado em placas com ágar Mueller Hinton sangue (MHS) e em tubos com caldo de infusäo cérebro coraçäo (BHI). Os dados obtidos demonstraram a conveniência do uso da bolsa em PVC, por näo apresentar crescimento bacteriano em nenhuma das amostras testadas


Animal-origin hemoderived products are largely employed for culture media that accountfor laboratory practice of interest for Public Health. In order to evaluate the quality of this product, twosystems of blood collection were tested, the traditional system that makes use of syringe, and a bagsystem made of atoxic polyvinyl chloride (PVC). Forty adult sheep, males and females, distributed atrandom in two groups of twenty animals each, were used. Blood used in this study was collected withasepsis, in a sterile environment, incubated at 37°C for 24 hours, and then streaked out on Mueller Hintonblood agar plates and seeded in tubes with Brain Heart Infusion Broth (BHI). Our data showed that thePVC bags did not present any bacterial growth in the tested samples, indicating to be better than thetraditional system
