ABSTRACT

Objective To explore the effects of recombinant human hormone(rhGH) on the plasma total protein,plasma albmin,healing of Wound surfaces in patients with the second degree burns wound.Methods 38 pa- tients with the second degree burns wound were divided into treatment group and control group randomly.All the patients were subject general.19 patients in the treatment group were given rhGH in a dose of 0.2U/kg for 14 days beginning from postoperative 5 days.The plasma total protein concentration,plasma albumin concentration,healing rat of wound surface and scar of patients of the two group were compared.Results The plasma total protein concen- tration plasma albumin concentration of the treatment group were significantly in creased,the scar hyperplasia of the treatment group were significantly mitigated and the healing time of wound surfaces of the treatment group were sig- nificantly shortened.Conclusion rhGH is found to promote protein anabotism and shorten the healing time of wound surfaces and mitigate the scar hyperplasia patients with the second degree burns wound.

ABSTRACT

Objective To explore the effects of recombinant human growth hormone(rhGH) on the serum levels of cytokines in severely burned patients.Methods Thirty-six burn patients were en-rolled in the study and were randomly divided into 3 groups according to the rhGH dosage used,i.e. small(0.3IU?kg -1 ?d -1 ,A),large(0.6IU?kg -1 ?d -1 ,B) dose groups and control group(C,with normalsaline).The rhGH was administered beginning from 3 postburn days(PBDs) and lasted for 20 days.The dynamic changes in the serum levels of TNF-?,IL-6,IL-8 and LPS at different time points were observed.Results When compared with these in C group,the serum levels of TNF-?,IL-6 in A,B groups were decreased,especially in B group with earlier decrease and bigger range(P0.05).Conclusions rhGH might decrease the production of postburn inflammatory mediators.The clinical application of rhGH might be a supplementary measure in preventing and ameliorating postburn SIRS and MODS in severely burned patients.

ABSTRACT

Objective To discuss the effect of growth hormone(GH) on acute gastric mucosal lesion(AGML) in rats.Methods 120 Wistar rats were divided into four groups randomly: blank control group(n=8),simple model group(n=32),model+rhGH group(n=40) and model+cimetidine group(n=40).Animal models of AGML were set up through soak and tie of rats.GH was injected to the rats in experimental groups,while cimetidine was used in the positive control group.Morphologic changes of rat gastric mucosa in each group were observed and compared on different days(0,4,8,12 d) after stress through gross looking,light microscope,and electron microscope.Ulcer index(UI),secretion amount of gastric acid,pH value and gastric mucosal were compared.Results Compared with blank control and model+cimetidine groups,the histomorphologic changes of gastric mucosa were obviously alleviated in model+rhGH group: only part of epithelial cells swelled,red blood cells were seldom seen,neutrophils infiltration reduced obviously,no remarkable changes were observed in the submucosal tissue.Mucosal cells were in good state and degeneration were seen in only a few cells under the electronic microscope.In addition,UI and the degree of gastric mucosal atrophy in model+rhGH group were lower than that in control group significantly(P

ABSTRACT

Objective To explore the effects of recombinant human growth hormone(rh-GH) on immune function in children with tetralogy of Fallot (TOF) after radical operation. Methods Thirty children with TOF were divided into two groups: conventional (n=20) and rh-GH (n=10, 0.2U/kg rhGH, sc., three times per week for 4 weeks). The immunoglobulin, complement fraction, lymphocyte subsets, and interleukins were determined. Results After treatment, the abnormal elevation of IgG, IgM, C_3, C_4, CD8~+ and CD19~+ in rhGH group decreased significantly at 1 week to 3 weeks; moreover, the abnormal descend of IgA, CD3~+, CD4~+, CD4~+/CD8~+, CD3~+/HLA-DR~+, and CD3~+/CD~ (16+56) in rh-GH group increased significantly at 1 week to 2 weeks compared to those of conventional group; and all recovered to the levels of control at 4 weeks. The interleukin-6 and tumor necrosis factor-? levels in the plasma and supernatant of peripheral blood mononuclearcytes decreased gradually at 1 week to 2 weeks after treatment compared to those of conventional group, and also recovered to the levels of control at 4 weeks. Conclusion rh-GH therapy could significantly improve immune function of children with TOF after radical operation.

Objective This study was to evaluate the effects of growth hormone (GH) on cell apoptosis of intestinal epithelium in acute necrotizing pancreatitis (ANP) rats.Methods Seventy-two SD rats were divided into three groups randomly: sham operation (SO) group, ANP group, and GH treatment group. GH-treatment group received subcutaneously 0 75?U/kg of GH. At 3?h, 6?h, 12?h, and 24 ?h after induction of ANP, the small intestinal specimens were harvested and apoptosis of intestinal epithelium was studied by DNA gel electrophoresis, FCM and TUNEL method. FasL and Bax protein expressions were detected by immunohistochemistry.ResultsThe ladder pattern on DNA gel electrophoresis presented in all time points of ANP group, but only seen in GH-treated group at 3h. ABSTRACT

Objective:To evaluate the effect of enteral rehabilitative therapy in improving graft structure recovery in rat small bowel transplantation.Methods:Fourty eight recipients of rat allogeneic heterotopic small bowel transplantation(SD→Wistar rat)were dvided into 4 groups randomly according to the presence or absence of glutamine or rhGH in TPN regimen.Cyclosporine A was used as the immunosuppressant.The morphological mucosal indices of transplanted gut were observed and compared.Results:The application of rhGH and glutamine-enriched TPN,especially the enteral rehabilitation therapy which composed both these two agents,can promote the recovery of graft structure significantly compared with standard TPN support.On the postoperative day 14,in the enteral rehabilitation therapy group,all the morphological indexes of transplanted gut recovered to the preoperative state.Conclusion:Enteral rehabilitation therapy is appropriate to the host metabolic status,so it can improve the graft structure recovery more effectively.

ABSTRACT

Objective To study the effect of growth hormone (GH) and PRL (prolactin) on the secretion of Th1/Th2 type cytokines in the patients with systemic lupus erythematosus (SLE). Methods The secretion of cytokines in PBMCs stimulated by GH and PRL was detected by ELISA in vitro. Results The results showed that the secretion levels of IL-6 and IL-10 from PBMCs of the SLE patients in active stage were higher than those of the normal controls, and the secretion level of IFN-? was lower than that of SLE patients in resting stage and the normal controls, but it is of no statistical significance. The secretion levels of IL-6 and IL-10 were significantly increased in PBMC stimulated by GH and PRL, but GH and PRL had no effect on the secretion of IFN-?. There was no difference on the secretion level of IL-6, IL-10 and IFN-? when the PBMC was stimulated by GH and PRL. Conclusion GH and PRL might play an important role in the pathogenesis of SLE.

ABSTRACT

ObjectiveTo observe the efficacy of rehabilitation therapy for patients and experimental animals with short bowel syndrome.MethodsThirty SD rats were randomly divided into control group,short bowel group and growth hormone (GH) treatment group.Short bowel was produced by resecting 80% of the rats′ small intestine, growth hormone at a dosage of 1?U/kg?d was delivered to the rats from POD1 to POD28.Nine patients with short bowel syndrome were enrolled in the study,the average length of the small intestine was (44?24)?cm, 3 cases had also suffered a short colon.ResultsIncreased ratio of S phase cells, proliferation index and expression of proliferative cell nucleus antigen (PCNA) were observed, indicating the putative effects of exogenous GH on residual small intestinal mucosa. The mechanism of action was probably through the alteration of protooncogene C-jun. The nutritional state of the patients and the absorptive capacity of the residual intestine improved markedly. Follow up were made to 8 patients,75% of the patients have weaned TPN,25% of them need intermittent TPN supplementation.ConclusionsRehabilitation therapy provides a new and effective alternative for short bowel patients.

ABSTRACT

Objective To study the effect of growth hormone (GH) combined with parenteral nutrition on skeletal muscle proteolysis for surgical critical patients. Methods Twenty surgical patients with APACHE II scores from 10 to 20 were divided randomly into two groups: parenteral nutrition group (PN group,n = 10) ; PN + GH ( GH group,n = 10). At the 5th day and 8th day during the study, urinary output of 3-methylhistidine ( 3-MH ) in 24 h urine were assessed with high performance liquid chromatography; at the 8th day the protein expressions of ubiquitin in quadriceps femoris muscle were determined by immunohistochemistry and the expressions of ubiquitin and C2 mRNA were determined by Northern blot analysis, the ultrastructure of quadriceps femoris muscle was observed under electronic microscope. All data were collected down in the form as mean ? standard deviation and analyzed with SPSS 11.5 for Windows. ABSTRACT

Objective To investigate the effects of growth hormone on illness severity and prognosis of septic patients. Methods Thirty septic shock patients were randomized into control group (10 cases) and growth hormone treatment group (20 cases) , to receive 8 U recombinant human growth hormone every day for 7 consecutive days, control group received NS. Serum concentration of procalcitonin ( PCT) , C-reactive protein ( CRP) , tumor necrosis factor-? ( TNF-?) and interleukin-6 ( IL-6 ) were measured on the 1st,4th,and 7th day. The severity of illness was assessed daily with the Acute Physiology and Chronic Health Evaluation-Ⅱ (APACHE-Ⅱ ) scoring system and the Elebute & Stoner's Sepsis scoring system. The efficacy also were evaluated on day 15 postoperation. Results The serum concentration of PCT, CRP, TNF-?, IL-6 decreased gradurally in both group after study starting. ABSTRACT

In order to investigate if the recombinant human growth hormone(rhGH) is suitable for hepatocellular carcinoma(HCC) treatment,we used radioreceptor assays to detect if growth hormone receptor(GHR) was presented in 32 HCC tissues.The results showed that the growth hormone specific singular binding site,namely GHR,was detected in 28 cases. The binding capacity(R T ) of GHR was 18 4300?4 1633fmol/mg.protein and affinity constant(Kd) was 0 6432?0 1961nmol/L R T was lower ( P 0 05). The R T of the GHR was negatively correlated to the tumor's size,but was not correlated with differentiation of tumor, or the co existence of cirrhosis,GHR was absent in 4 cases. This study showed that the majority of the HCC tissue still express a low level of GHR. Before their functions are well understood, rhGH should be very carefully used in HCC patients.

ABSTRACT

To determine the effect of recombinant human growth hormone (rhGH) on postburn hypermetabolism postburn, sixteen male minipigs were inflicted with 35% TBSA full thickness burns, and the burned minipigs were then randomly divided into two groups: rhGH group and control group. REEs were monitored by means of Cardiorespiratory Diagnostic Systems (Medical Graphics Corporation, USA). The sequential changes in serum levels of GH, IGF 1,insulin,glucagon,cortisol,albumin were analyzed. The level of REE is significantly lower in rhGH treated groupas as compared with that of control, while the serum concentrations of GH, IGF 1, albumin are higher in the former, Body weighf gain is observed in rhGH treated group with no significant effect on serum concentration of insulin,glucagon and cortisol. RhGH therapy is thus helpful in the control of hypermetabolism after extensive burn.
