ABSTRACT

Abstract The objective of this work was to perform the phytochemical characterization, to determine total phenols, antioxidant (AAO%) and antimicrobial potential of the ethanolic extracts of carambola. The phytochemical study was carried out through a qualitative analysis of the chemical constituents and quantitative determination of the phenol content By the Folin-Ciocalteu test. Qualitative and quantitative antioxidant tests were performed using the DPPH method (2,2 diphenyl-1-picryl-hydrazila) and iron reduction (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The presence of pyrogallic tannins, steroids and saponins has been identified. The highest total phenol content, quantified in the samples, was found in the stem bark (0.0866 mgEAG/g) and in the fruit (0.0734 mgEAG/g). In the antioxidant evaluation, the extracts of the green fruit bagasse (AAO% 71.9%,) and stem bark at 50 μg/mL (AAO% 94%) with CE50 23.7 μg/mL. Leaf extracts, stem bark, ripe fruit bagasse and green fruit bagasse presented MICs of 100 μg/mL against multiresistant pathogenic bacteria and fungi.


Resumo O objetivo desse trabalho foi realizar a caracterização fitoquímica, determinar fenóis totais, potencial antioxidante (AAO%) e antimicrobiano dos extratos etanólicos de carambola O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. Os testes antioxidantes qualitativos e quantitativos foram realizados pelo método do DPPH (2,2 difenil-1- picril-hidrazila) e redução do ferro (FRAP). A concentração inibitória mínima (CIM) foi determinada por microdiluição em placas de 96 poços. Foi identificada a presença de taninos pirogálicos, esteroides e saponinas. O maior teor de fenóis totais, quantificado nas amostras, foi encontrado na casca do caule (0,0866 mg EAG/g) e no fruto (0,0734 mg EAG/g). Na avaliação antioxidante destacaram-se a 500 µg/mL os extratos do bagaço do fruto verde (AAO% 71,9%,), e casca do caule a 50 µg/mL (AAO% 94%) com CE50 23,7 µg/mL. Os extratos das folhas, casca do caule, bagaço do fruto maduro e bagaço do fruto verde apresentaram CIM de 100 µg/mL contra bactérias e fungos patogênicos multirresistentes.

ABSTRACT

Background: Asparagus (Asparagus officinalis L.) green is a vegetable with a great demand worldwide, and likewise, it is highly perishable, due to its high respiration rate that accelerates its senescence. Disinfection of vegetables after their harvest is an obligatory practice that can reduce losses by decomposition due to the attack of microorganisms. Therefore, it is vital to preserving its microbiological and sensory characteristics to reach the final consumer. Objective: to evaluate the effect of gaseous ozone (0 to 10 ppm) and storage time (0 to 30 days) on phenol content, overall appearance, count of molds, psychrophilic bacteria, and viable mesophilic aerobes. Methods: the response surface methodology was used, applying a rotatable central composite design. Results: the results indicated that there was a significant influence (p <0.05) of the independent variables on the characteristics studied, as well as an adequate lack of fit of the quadratic regression model (p> 0.05). By means of the contour superposition technique, it was determined that the optimal conditions for the highest retention of phenol content (16.99 mg/g) and overall appearance (7.61 points) and lower counts of viable aerobic mesophilic bacteria (5.3 x 103 CFU/g) they corresponded to 10 ppm of gaseous ozone up to 25.91 days of storage, with adequate quality characteristics in the spears. Conclusion: the region of interest was determined for optimal retention of phenol content and overall appearance, and a lower count of viable aerobic mesophilic bacteria in green asparagus during postharvest, suggesting to use the initial application of ozone gas at 10 ppm allowing 25.9 days storage at 1 °C. The results indicate that this technology is a good alternative in the conservation of fresh vegetables


Antecedentes: El espárrago (Asparagus officinalis L.) verde; es una hortaliza con una gran demanda a nivel mundial, y, asimismo, es altamente perecible, por su elevada velocidad de respiración que, acelera su proceso de senescencia. La desinfección de los vegetales después de su cosecha es una práctica obligada que puede disminuir las pérdidas por descomposición debido al ataque de microrganismos. Por lo tanto, es muy importante conservar sus características microbiológicas y sensoriales para llegar al consumidor final. Objetivo: evaluar el efecto del ozono gaseoso (0 a 10 ppm) y tiempo de almacenamiento (0 a 30 días) sobre el contenido de fenoles, apariencia general, recuento de mohos, bacterias psicrófilas y aerobias mesófilas viables. Métodos: se utilizó la metodología de superficie de respuesta, aplicando un diseño compuesto central rotable. Resultados: los resultados indicaron que existió influencia significativa (p<0.05) de las variables independientes sobre las características estudiadas, así como, una adecuada bondad de ajuste del modelo de regresión cuadrático (p>0.05). Mediante la técnica de superposición de contornos se determinó que las condiciones óptimas para la mayor retención de contenido de fenoles (16.99 mg/g) y apariencia general (7.61 puntos) y menor recuentos de bacterias aerobias mesófilas viables (5.3 x 103 UFC/g) correspondieron a 10 ppm de ozono gaseoso hasta los 25.91 días de almacenamiento, con adecuadas características de calidad en los turiones. Conclusión: se determinó la región de interés para una óptima retención de contenido de fenoles y apariencia general, así como, menor recuento de bacterias aerobias mesófilas viables en el espárrago verde durante la postcosecha, sugiriendo utilizar la aplicación inicial de ozono gaseoso a 10 ppm permitiendo 25.9 días de almacenamiento a 1 °C. Los resultados indican que esta tecnología es una buena alternativa en la conservación de hortalizas frescas

ABSTRACT

Objective:To investigate the antioxidant activity and chemical composition of 75% ethanol extract of <italic>Rosa cymosa</italic> roots and its different polar parts. Method:The 75% ethanol extract of <italic>R. cymosa</italic> roots was divided into dichloromethane, ethyl acetate, <italic>n</italic>-butanol and water parts by organic solvent extraction. <italic>In vitro</italic> antioxidant activity of each fraction was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging assays, as well as ferric reducing antioxidant power (FRAP) test. The contents of total triterpenes, total phenols, total tannins and condensed tannins in each fraction were determined by spectrophotometry. SPSS 24.0 software was used to conduct Pearson correlation analysis between the antioxidant activity of each fraction and the content of the main components, and then the main active fraction and the main active components were determined. The chemical constituents of the active fraction was analyzed by ultra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS), and the structures of the main chromatographic peaks were predicted. Result:Each fraction of <italic>R. cymosa</italic> roots had certain antioxidant activity, and there was a significant dose-effect relationship within a certain concentration range, but the antioxidant activity of different polar parts was different. In DPPH and ABTS free radical scavenging tests, the antioxidant activity of each fraction and vitamin C (VC, the positive drug) was ranked as ethyl acetate fraction>VC><italic>n</italic>-butanol fraction>ethanol extract>water fraction>dichloromethane fraction. In FRAP test, the activity of ethyl acetate fraction was weaker than that of VC, and the other order was unchanged. The contents of total triterpenes, total phenols, total tannins and condensed tannins in ethyl acetate fraction were 3.81%, 50.33%, 3.32%, and 39.79%, in <italic>n</italic>-butanol fraction were 0.88%, 41.42%, 2.25% and 23.55%, in ethanol extract were 2.90%, 41.95%, 3.43% and 20.14%, in water fraction were 0, 26.80%, 16.90% and 7.57%, and in dichloromethane fraction were 21.23%, 12.90%, 1.59%, and 6.17%, respectively. Correlation analysis results showed that the contents of total phenols and condensed tannins were positively correlated with the antioxidant activity, the contents of total triterpenes were negatively correlated with the antioxidant activity, and the correlation between total tannins and antioxidant activity was not obvious. A total of 26 compounds were identified from the ethyl acetate fraction by UPLC-Q-TOF-MS/MS, including 11 condensed tannins, 4 hydrolysable tannins, 6 triterpenes, 3 flavonoids, 1 benzoic acid derivative and 1 chlorogenic acid analogue. Conclusion:Ethyl acetate fraction is the main antioxidant active site of <italic>R. cymosa</italic> roots, and phenols mainly composed of condensed tannins are the main active components. The results can provide experimental basis for the development of natural antioxidants.

ABSTRACT

Objective: To explore the regulatory effect of malt alkaloid on prolactin (PRL) secretion in the model rat with postpartum hypogalactia induced by bromocriptine based on dopamine D2 receptor, and determine the active fraction of the malt with galactogogue effect. Methods: The postpartum hypogalactia model was established by intragastric administration of bromocriptine mesylate. After the model was successfully established, all groups were given corresponding drug treatment. The concentration of serum PRL, estradiol (E2) and progesterone (P) in each group was detected by ELISA kits. HE staining was used to observe the pathologic changes of breast tissue. RT-PCR was used to determine the mRNA levels of prolactin receptor (PRLR) and dopamine D2 receptor (DRD2) in pituitary gland of rats. Results: Compared with the control group, the levels of serum PRL, P, and E2 were significantly decreased in the model group as well as the mRNA expression of the pituitary PRL cells. But the mRNA expression of the pituitary DRD2 in the model group was significantly increased compared with the control group. Compared with the model group, the malt total alkaloid significantly increased the volume of mammary lobule and dilated the duct. There was a lot of milk in the duct and acinar in the malt total alkaloid group. Besides, the total alkaloids increased the concentration of serum PRL, P, and E2 and the mRNA expression of the pituitary PRL cells, and decreased the mRNA expression of the pituitary DRD2. Conclusion: The primary the active fraction of malt for galactogogue action is total alkaloids, and its mechanism may be related to promoting PRL secretion, increasing serum PRL receptor level and decreasing the mRNA expression of dopamine D2 receptor.

ABSTRACT

To reveal the main nutrients and functional ingredients in the flowers of Polygonatum cyrtonema and P. filipes, the content of the polysaccharides, saponins, amino acids, total phenols, mineral elements, and the DPPH free radical scavenging rates were determined. The flowers and rhizomes of P. cyrtonema were collected from Qingyang in Anhui and Qingyuan in Zhejiang, while the flowers and rhizomes of P. filipes were collected from Longyou in Zhejiang, respectively. The results showed that the polysaccharides content in flowers varied from 60.88 to 97.00 mg·g~(-1), about half of that in rhizomes. The saponins content in flowers varied from 32.55 to 40.93 mg·g~(-1), which was close to the content in rhizomes. The content of total phenols ranged from 40.79 to 50.95 mg·g~(-1), approximately 4.5 times of that in rhizomes. The total amino acids content in flowers was 111.85 to 131.03 mg·g~(-1), about 2.3 times of the content in rhizomes. The essential trace element content was abundant in flowers. The contents of heavy metal elements were all within the limits set by the standards. The DPPH free radical scavenging rate IC_(50) varied from 1.77 to 3.25 mg·mL~(-1), less than one-fifth of that in rhizomes, showing a significant superiority of antioxidant activity compared to rhizomes. The results initially revealed the fundamental of "the flowers exceed the rhizomes in effect", the common saying about the traditional Chinese medicinal herbs over the years, indicating a great developing potential of the flowers. Besides, as polysaccharides, saponins, amino acids, total phenols and other nutritive substances in flowers differ widely among species and provenances, it's important to develop variety breeding to improve the quality and yield of flowers.

ABSTRACT

In order to reveal the main nutrients and functional ingredients in the shoots of Polygonatum cyrtonema, the polysaccharides, proteins, amino acids, and total phenols were determined. The tested samples cultured in Ma'nijiaonong, Hengtang village, Tianmushan town, Lin'an, Zhejiang, which were collected from three provenances(Pan'an and Longquan in Zhejiang and Qingyang in Anhui). The results showed that the polysaccharide content of the shoots varied from 2.34% to 12.73%, roughly one-third of rhizomes. The protein content varied from 107.75 to 192.49 mg·g~(-1), nearly 5.50 times more than rhizomes. Moreover, the average of total amino acid content was 193.13-248.74 mg·g~(-1), approximately 4.16 times of rhizomes. And the essential amino acids account for 35.57%-39.44% of the total amino acids content, which was close to the standard of the ideal protein proposed by FAO/WHO(the essential amino acid/total amino acid is about 40%). In addition, the taste amino acids(TaAA) changed from 160.12 to 208.29 mg·g~(-1), revealing the material basis of "shoots were extremely delicious" in Chinese ancient herbal medicine. Additionally, the total phenols varied from 51.21-58.76 mg·g~(-1), about 2.96 times of rhizomes. The DPPH free radical scavenging rate of tested shoots was over 95%, which obviously superior to rhizomes. Therefore, the shoots of P. cyrtonema is a very high-quality vegetable and functional food with good development potential. Furthermore, the main nutrients and functional substances in P. cyrtonema shoots are closely related to the provenances and harvesting seasons. It is important to improve the quality and yield of the shoots by strengthening the variety of breeding and cultivation techniques.

ABSTRACT

To compare the polysaccharides( LBLP),total flavonoids( LBLTF) and total phenols( LBLTP) of Lycium barbarum leaves from 14 production areas from Ningxia,Qinghai and Gansu at different harvesting time. Our results showed that there were significant difference of LBLP,LBLTF and LBLTP among different production areas. The LBLP collected from Xiangride township production area,Dulan,Qinghai was as high as 15. 02%. The LBLTF collected from Dagele township production area,Geermu,Qinghai was as high as28. 63%. The LBLTP collected from Keluke township production area,Delingha,Qinghai was as high as 16. 7%. There were also significant difference of these 3 components between different harvest periods. The average LBLP in May( 10. 20%) was significantly higher than that in June( 8. 49%). However,the average LBLTF( 17. 71%) and LBLTP( 12. 77%) in June was significantly higher than that in May( 14. 15%,11. 19%). The LBLTF and LBLTP have a positive correlation with the altitude of production areas,but there was no significant association between LBLP and altitude. The LBLP,LBLTF and LBLTP in different production areas and harvesting time was significant difference,the results provide some references for furthermore development and utilization of L. barbarum leaves.

ABSTRACT

Objective: Maijunan (MJA) Tablets is a protected variety of traditional Chinese medicine (TCM) consisted of Pueraria lobata, hydrochlorothiazide (HTCZ), Uncaria rhynchophylla (366:1:980) and excipient. In the present work, MJA was consisted of the total flavones of P. lobata, HCTZ and total alkaloids of U. rhynchophylla (40:11:75). The combination of MJA and the total phenols of Magnolia officinalis (M-MJA) was consisted of the total flavones of P. lobata, the total phenols of M. officinalis, HCTZ and the total alkaloids of U. rhynchophylla (40:40:11:75). The aim of this work was to examine the effect and mechanism of M-MJA on the blood pressure of spontaneous hypertensive rats (SHRs). Methods: Adult male SHRs were randomly divided into control group, MJA group (180 mg/kg·d), and the M-MJA group (218 mg/kg·d) (n = 5). SHRs were orally administered with M-MJA and MJA respectively once a day for 8 weeks, the blood pressure of SHRs was measured every two weeks, and the biochemical indicators related to blood pressures were detected at the last dosing. Results: After oral administration of M-MJA to SHRs once a day for 8 weeks, the systolic and diastolic blood pressures of SHRs were deceased significantly. M-MJA affected renin-angiotensin-aldosterone system by decreasing the levels of Ren, Ang II and ALD, affected the endothelial function by decreasing the levels of ET-1 and 20-HETE, and increasing the level of eNOS, affected the oxidative stress by increasing the protein expression of Nrf2 and the activities of HO-1 and GSH-Px, and decreasing the protein expression of CYP2E1 and CYP4A, as well as the content of MDA. Conclusion: These results indicated that M-MJA could regulate the renin-angiotensin-aldosterone system, improve endothelial function, and inhibit CYP4A activity to reduce the production of 20-HETE, alleviate the oxidative stress disorder of the visceral organs, and eventually exert antihypertensive effect. Additionally, the anti-oxidant ability, regulating the renin-angiotensin-aldosterone system and improving endothelial function of M-MJA are more powerful than that of MJA, suggesting that M-MJA may have a better anti-hypertensive effect than MJA.

Objective: To study the effects of salicylic acid (SA) and methyl jasmonate (MeJA) on growth, activity of related enzymes and chemical components in Polygala callus. Methods: The callus of Polygala was used as material. After 30 d of dark culture at different concentrations of SA (0, 4, 8, 12, 16, 20, 24, 28, 32 mg/L) and MeJA (0, 100, 200, 400, 600, 800, 1 000 μmol/L), the growth of callus, anti-oxidant enzyme activity, the content of MDA, total phenolic, total flavonoid, polygalaxanthone III, and 3,6’-disinapoyl sucrose were determined. Results: MeJA inhibited the growth of Polygala callus, and 12 mg/L SA promoted the growth of Polygala callus. SA and MeJA promoted the activity of SOD, CAT, POD, and MDA in the callus of Polygala. With the increase of SA and MeJA concentration, the activities of SOD, CAT and POD increased first and then decreased, and the content of MDA continued to rise. When the concentration of SA was 20 mg/L, the activities of CAT and SOD reached the maximum, which were 248.45 U/mg and 4451.06 U/mg, respectively. When the concentration of SA was 16 mg/L, the activity of POD reached the maximum, which was 7.22 U/mg. When the concentration of SA was 32 mg/L, the MDA content reached the maximum value of 25.09 nmol/mg. When the concentration of MeJA was 600 μmol/L, the activities of CAT, SOD, and POD reached the maximum, which were 273.30, 1451.06 and 15.27 U/mg, respectively. When the concentration of MeJA was 1000 μmol/L, the MDA content reached the maximum value of 27.10 nmol/mg. SA inhibited the accumulation of total flavonoids in Polygala callus, and had no significant effect on total phenols. MeJA promoted the accumulation of total phenols and total flavonoids in Polygala callus. When MeJA concentration was 600 μmol/L, the content of total flavonoids was the highest. When the concentration of MeJA was 400 μmol/L, the total phenolic content was the highest. Both SA and MeJA promoted the accumulation of polygalaxanthone III and 3,6’-disinapoyl sucrose in Polygala callus. When the concentration of SA was 32 mg/L, the concentration of polygalaxanthone III, 3,6’-disinapoyl sucrose was the highest. When the concentration of MeJA was 1 000 μmol/L, the content of polygalaxanthone III, 3,6’-disinapoyl sucrose was the highest. Conclusion: MeJA had an inhibitory effect on the growth of Polygala callus, and 12 mg/L SA can promote this effect. SA and MeJA promoted the activity of SOD, POD, CAT, the content of MDA and the accumulation of polygalaxanthone III and 3,6’-disinapoyl sucrose in Polygala callus. SA inhibited the accumulation of total flavonoids in Polygala callus, and had no significant effect on total phenolics. MeJA promoted the accumulation of total phenolics and total flavonoids in Polygala callus.

ABSTRACT

Colocasia esculenta (L.) Schott, conocida comúnmente como taro o malanga se cultiva en los trópicos húmedos en varios países de África, América y Asia y su tubérculo es un alimento esencial para humanos y animales. El objetivo de este estudio fue caracterizar física y nutricionalmente la harina del tubérculo de malanga cultivada en Actopan, Edo. De Veracruz, México. Se llevaron a cabo análisis químico proximal, aminoácidos, fracciones de fibra, minerales, actividad de agua, color, factores antinutricionales y contenido de fenoles totales. Los resultados indican un contenido de humedad de 6,87 g/100 g, proteína cruda de 5,93 g/100 g, extracto etéreo 1,25 g/100 g, fibra dietaria 12,08 g/100g, cenizas 3,47 g/100 g y extracto libre de nitrógeno 77,27 g/100g. El contenido de fenoles totales fue de 113,57±14 expresado como miligramos de equivalente de ácido gálico (mg EAG/100g). La harina de malanga presentó un alto contenido de K (1743 mg/100 g) y bajo en Na (9,25 mg/100 g). Se concluye que a pesar de no ser una fuente importante de proteína el perfil de aminoácidos esenciales supera los valores de FAO, en relación a la fibra dietética total podría ser un ingrediente en formulaciones alimenticias que además aportaría K, Ca y Mg. De acuerdo a las fracciones de fibra la harina es una alternativa para la elaboración de alimentos balanceados. El contenido de fenoles totales abre la posibilidad de que se complemente con la identificación de su actividad antioxidant(AU)


Colocasia esculenta (L.) Schott, commonly known as taro, malanga or yam, is grown in the humid tropics in several countries in Africa, America and Asia and its tuber is an essential food for humans and animals. The objective of this study was to characterize malanga meal physicochemically and content of phenolic compounds cultivated in Actopan, Edo. Veracruz, Mexico. The proximal chemical analyzes, aminoacid, fiber fractions, minerals, water activity, color, antinutritional factors and total phenol content were carried out. The results indicate a moisture content of 6.87g /100 g, crude protein of 5.93 g /100 g, ether extract 1.25g /100g, dietary fiber 12.08 g /100 g, ash 3.47g/100g and nitrogen-free extract 77.27g/100g The total polyphenol content in this study was 113.57 ± 14 expressed as milligrams of gallic acid equivalent (mg EAG/100g). It concluded that despite not being an important source of protein the profile of essential amino acids exceeds the values of FAO, in relation to total dietary fiber could be an ingredient that in food formulations that also provide K, Ca and Mg. According to fiber fractions, meal is an alternative for the elaboration of balanced foods. The content of total phenols opens the possibility that it complements with the identification of its antioxidant activity(AU)

Aims: To explore the use of Borassus aethiopum fruit as composite flour and bread and determine the phytochemical composition in each product. Place and Duration: Department of Biochemistry, and Department of Food science food and sensory evaluation laboratory, between September, 2016 and April, 2017. Study Design: An experimental study. Methods: The African palmyra fruits were obtained from Sekyere Odumase and processed into Borassus flour and composite bread. Phytochemicals constituents were determined in both products using aqueous and methanol extracts. Results: The results of both aqueous and methanol extraction for the phytochemical testing indicated that the raw B. aethiopum powder (RBAP) contained flavonoids, saponins, phenols, cardiac glycosides, alkaloids, triterpenes, steroids and sterols. Borassus bread contained flavonoids, alkaloid, triterpenes, steroids and sterols in both extracts, whilst cardiac glycosides, saponins and phenols maintained their strong presence in both extracts of composite Borassus bread. Tannins were absent in both extracts of B. aethiopum powder and bread composite. Also, the total phenol content in the composite bread was lower compared with the Borassus flour (P=0.000). The total antioxidant capacity in the Borassus flour (EC50= 2.1±0.24 mg/mL) was significantly higher than the composite bread (EC50=2.24±0.4 mg/mL) (P= 0.000). However, both products had less antioxidants than standard ascorbic acid (EC50=0.12±0.2 mg/mL). Conclusion: Both Borassus flour and composite bread had appreciable phytochemicals present, with some antioxidant capacity which could beneficially help in the management of people with chronic diseases such as diabetes and cardiovascular diseases.

ABSTRACT

Ephedra tweediana (Ephedraceae), conocida como “tramontana”, es empleada en la medicina popular como antiasmático. Se analizaron comparativamente los extractos acuosos de los tallos herbáceos, lignificados y partes subterráneas, provenientes de ejemplares femeninos y masculinos. En los tallos lignificados se determinó la presencia de proapigeninidina (cuya identidad fue corroborada por comparación con un testigo de apigeninidina sometido a estudios de FAB-MS, UV, HPLC, 1H-NMR y espectroscopía IR); mientras que en los órganos subterráneos se determinó la presencia de proapigeninidina y propelargonidina (la identidad de la pelargonidina fue establecida por comparación con un testigo de pelargonidina sometido a estudios de TLC/HPTLC y espectroscopía UV-Visible). Estos compuestos no se observaron en los tallos herbáceos. Los tallos herbáceos presentaron las mayores concentraciones de flavonoides y ácidos hidroxicinámicos totales. Los órganos subterráneos presentaron la mayor concentración de taninos y proantocianidinas. En los tallos herbáceos se detectó una reacción fuertemente positiva para flavonoides. No se observó reacción positiva para proantocianidinas. En el tallo aéreo lignificado se observó una reacción positiva para flavonoides y proantocianidinas a nivel de la peridermis. En los órganos subterráneos, los flavonoides y proantocianidinas se localizaron principalmente en los tejidos más externos. Este trabajo constituye el primer aporte a la dinámica de polifenoles de E. tweediana.


Ephedra tweediana (Ephedraceae), known as "tramontana" is used in folk medicine as antiasthmatic. Aqueous extracts obtained from young stems, woody stems and underground parts were analyzed and compared. In lignified stems was detected proapigeninidin (whose identity was confirmed by comparison with a control apigeninidin subjected to FAB-MS, UV, HPLC, 1H-NMR and IR spectroscopy), while underground organs were detected proapigeninidin and propelargonidin (by comparison with a control pelargonidin whose identity was established by studies of TLC/HPTLC, and UV-visible spectroscopy). These compounds were not observed in the herbaceous stems. The herbaceous stems had the highest concentrations of total flavonoids and hydroxycinnamic acids. Uderground organs had the highest concentration of tannins and proanthocyanidins. In herbaceous stems a strong positive reaction for flavonoids was detected. No positive reaction was observed for proanthocyanidins. In the periderm of woody aerial stem a positive reaction for flavonoids and proanthocyanidins were observed. In the underground organs, flavonoids and proanthocyanidins were located mainly in the more external tissues. This work is the first contribution to the dynamic of E. tweediana polyphenols.

AIM To investigate the correlations between antioxidant activities and contents of total flavonoids and total phenols of chloroform,ethyl acetate,n-butanol and ethanol extracts from Nervilia fordii (Hance) Schltr..METHODS The contents of total flavonoids and total phenols in four extracts from N.fordii were determined by AlCl3-HAc-NaAc coloration method and Folin-Ciocalteu colorimetric method,respectively.The antioxidant activities of extracts were evaluated by determining scavenging activities of DPPH radical,hydroxyl radical and ABTS radical,together with reducing power.Then the correlations between antioxidant activities and contents of two constituents were analyzed by SPSS17.0 software.RESULTS The antioxidant activities of four extracts from N.fordii all showed dose-effect relationships,that of ethyl acetate extract was the strongest,while that of ethanol extract was the weakest.They had some correlations with the content of total flavonoids (0.604 < r < 0.638) and significant correlations with the content of total phenols (r > 0.977).CONCLUSION Ethyl acetate extract from N.fordii can be used for the development of natural antioxidants,and the content of total phenols is the main factor to influence its antioxidant activity.

Abstract The objective of this work was to perform the phytochemical characterization, to determine total phenols, antioxidant (AAO%) and antimicrobial potential of the ethanolic extracts of carambola. The phytochemical study was carried out through a qualitative analysis of the chemical constituents and quantitative determination of the phenol content By the Folin-Ciocalteu test. Qualitative and quantitative antioxidant tests were performed using the DPPH method (2,2 diphenyl-1-picryl-hydrazila) and iron reduction (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The presence of pyrogallic tannins, steroids and saponins has been identified. The highest total phenol content, quantified in the samples, was found in the stem bark (0.0866 mgEAG/g) and in the fruit (0.0734 mgEAG/g). In the antioxidant evaluation, the extracts of the green fruit bagasse (AAO% 71.9%,) and stem bark at 50 g/mL (AAO% 94%) with CE50 23.7 g/mL. Leaf extracts, stem bark, ripe fruit bagasse and green fruit bagasse presented MICs of 100 g/mL against multiresistant pathogenic bacteria and fungi.


Resumo O objetivo desse trabalho foi realizar a caracterização fitoquímica, determinar fenóis totais, potencial antioxidante (AAO%) e antimicrobiano dos extratos etanólicos de carambola O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. Os testes antioxidantes qualitativos e quantitativos foram realizados pelo método do DPPH (2,2 difenil-1- picril-hidrazila) e redução do ferro (FRAP). A concentração inibitória mínima (CIM) foi determinada por microdiluição em placas de 96 poços. Foi identificada a presença de taninos pirogálicos, esteroides e saponinas. O maior teor de fenóis totais, quantificado nas amostras, foi encontrado na casca do caule (0,0866 mg EAG/g) e no fruto (0,0734 mg EAG/g). Na avaliação antioxidante destacaram-se a 500 µg/mL os extratos do bagaço do fruto verde (AAO% 71,9%,), e casca do caule a 50 µg/mL (AAO% 94%) com CE50 23,7 µg/mL. Os extratos das folhas, casca do caule, bagaço do fruto maduro e bagaço do fruto verde apresentaram CIM de 100 µg/mL contra bactérias e fungos patogênicos multirresistentes.

ABSTRACT

In the present study, antioxidant and anticancer activities of different solvent extracts of an Indian medicinal plant, Spathodea campanulata P. Beauv. (leaf) were investigated. Among the different extracts studied, 70% ethanolic extract of S. campanulata leaf registered high antioxidant activity in terms of phosphomolybdate reducing power (955 FAEA), radical scavenging activity against DPPH (84.67%), superoxide (72.69%), hydrogen peroxide (83.20%) and hydroxyl radicals (70%), when compared to ethyl acetate and chloroform extracts. Further, the ethanolic extract illustrated remarkable anticancer activity against EAC cell line (85%) when compared to ethyl acetate and chloroform extracts. Ethanol extract showed higher level of total phenolic concentration (9.21 mg FAE/L), which is correlated with the maximum antioxidant activity exhibited by this extract. Due to remarkable antioxidant and anticancer properties, the ethanolic extract of S. campanulata leaf could be considered as a natural source of plant based drug to prevent / treat oxidative stress induced diseases such as cancer.

Introducción: Bixa orellana L. también conocida como achiote, es una planta con un rico depósito de compuestos bioactivos, tales como, saponinas, alcaloides y flavonoides, a los cuales se les atribuyen propiedades fisiológicas entre las que se encuentran: actividad antimicrobiana, antiinflamatoria, antialérgica, antiviral, anticancerígena, antioxidante, entre otras. Objetivos: evaluar el efecto del tiempo de extracción y la relación solvente/ sobre el contenido de fenoles totales, así como el efecto del contenido de sólidos y el pH de la solución, sobre la actividad antioxidante del extracto de hojas de B. orellana. Métodos: el contenido total de fenoles fue evaluado por el método de Folin-Ciocalteu. La actividad antioxidante se determinó por los métodos espectrofotométricos de reacción con el radical 2,2´-azino-bis (3-etilbenzotiazolin-6- ácido sulfónico) y la medida de la capacidad reductora sobre el Fe+3, los resultados se expresaron como micromoles de equivalentes Trolox por gramo de extracto (µmol ET.g-1). Resultados: las condiciones del proceso que más favorecen la extracción de compuestos fenólicos desde las hojas de Bixa orellana L. son: tiempo de extracción de 60 h y relación solvente/ hojas (v/p) de 4/1. El contenido máximo de fenoles totales fue de 144,77 ± 9,66 mgAT.g-1, que al someterlo a una solución de pH de 8 y 11,7 °Brix, presenta una actividad antioxidante de 4406,83 ± 43,30 µmol ET.g-1 por el método de reacción con el radical 2,2´-azino-bis (3-etilbenzotiazolin-6- ácido sulfónico) y 4547,22 ± 53,19 µmolET.g-1 por el método de medida de la capacidad reductora sobre el Fe+3. Conclusiones: se demostró que la cantidad de fenoles totales extraídos de hojas de B. orellana dependen de la relación solvente/material vegetal y del tiempo de extracción; asimismo, se encontró que el pH tiene efecto sobre la actividad antioxidante determinada por la reacción con el radical 2,2´-azino-bis (3-etilbenzotiazolin-6- ácido sulfónico) y la medida de la capacidad reductora sobre el Fe+3(AU)


Introduction: Bixa orellana L. also known as annatto, is a plant with a high content of bioactive compounds, such as saponins, alkaloids and flavonoids that have physiological properties as antimicrobial, anti-inflammatory, anti-allergenic, antiviral, anticarcinogenic and antioxidant properties, among others. Objectives: To evaluate the effect of extraction time and the solvent/ leaf ratio over the total phenolic content and the effect of solids and pH of the solution on the antioxidant activity in extracts from Bixa orellana leaves. Methods : The ethanol extract of leaves of Bixa orellana was obtained by percolation and filtering. Total phenol content was evaluated by the Folin-Ciocalteu. The antioxidant activity was determined by spectrophotometric methods as reaction with 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) and ferric reducing antioxidant power, the results were expressed as micromoles of Trolox equivalents per gram of extract (µmolTE.g -1). Results: Process conditions that improve the extraction of phenolic compounds from Bixa. orellana extract are: extraction time of 60 h and solvent/ leaves ratio (v/w) of 4/1. The maximum total phenol content was 144.77 ± 9.66mgAT.g-1, which when subject to a solution of pH 8 and 11.7 °Brix, has an antioxidant activity of 4406.83 ± 43.30 µmolTE.g-1 by the 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) reaction and 4547.22 ± 53.19 µmolET.g-1 by ferric reducing antioxidant power. ABSTRACT

Tissue culture seedlings of Bletilla striata were treated with MeJA, SA and two kinds of endophytic fungi in order to study the effects of those treatments on the physiology and total phenols content. The method of tissue culture was used to culture seeds into seedlings, and then different treatments were applied on them to observe and measure the changes of physiology and total phenols content. We find that the growth of seedlings treated with SA was poor, which treated with 40 μmol•L⁻¹ MeJA, 50 mL•L⁻¹ Hypocrea koningii and 10 mL•L⁻¹ Trichoderma koningiopsis showed better. The activity of SOD, POD and CAT was at a high level under SA treatment of each concentration. The activity of SOD and POD increased as the rise of MeJA concentration, while CAT was highest at 80 μmol•L⁻¹. The activity of SOD and POD increased with the increasing of the concentration of H. koningii treatment, while CAT reached the highest at 1 mL•L⁻¹. The activity of SOD, POD and CAT increased first and then declined with the concentration of T. koningiopsis increasing, and the highest activity was at 10 mL•L⁻¹. The contents of MDA, soluble protein and proline were increased more or less under the four treatments. The content of polysaccharide was at a high level under 60 μmol•L⁻¹ of MeJA. The total phenols content was at a high level under 40 μmol•L⁻¹ of MeJA, 60 μmol•L⁻¹ of SA, 1 mL•L⁻¹ of H. koningii and 10 mL•L⁻¹ of T. koningiopsis. The results indicated that the addition of exogenous MeJA, SA and endophytic fungi under certain concentrations could improve the resistance of B. striata and increase the content of total phenols at some degree and the trearment of MeJA, H. koningii and T. koningiopsis could promote the growth of seedlings under certain concentrations.

ABSTRACT

RESUMO Extratos etanólicos de Luehea paniculata, Anadenanthera colubrina, Cariniana rubra, Hancornia speciosa, Qualea grandiflora, Kielmeyera coriaceae e Palicourea rigida, coletadas no Cerrado de Mato Grosso foram submetidos à análise fitoquímica preliminar para identificação das principais classes de metabolitos secundários e à quantificação do teor de compostos fenólicos totais, flavonoides e cumarinas. O estudo também descreve a atividade antimicrobiana, antioxidante e a toxicidade preliminar frente a larvas de Artemia salina. Os extratos de C. rubra e Q. grandiflora foram os mais ativos contra as cepas fúngicas de C. neoformans (CIM 3,91 µg/mL), C. krusei(CIM 15,63 µg/mL), C. parapsilosis (CIM 31,25 µg/mL) e C. albicans (CIM 62,5 µg/mL), ambos com os mesmos resultados para estes microrganismos. Em relação às cepas bacterianas a mais sensível foi S. aureus com CIM de 250 µg/mL para todos os extratos testados. Luhea paniculata e Q. grandiflora foram as espécies vegetais mais ativas contra E. coli (CIM 500 e 250 µg/mL, respectivamente) e E. faecalis (CIM 250 e 500 µg/mL, respectivamente). No teste do DPPH, merece destaque as espécies K. coriaceae (Ca) (IC50 = 0,72 ± 0,51 μg /mL), K. coriaceae (Fo) (IC50 = 1,34 ± 0,53 μg /mL), H. speciosa (Fo)(IC50 = 1,03 ± 0,52 μg /mL), Q. grandiflora (Fo) (IC50 = 1,31 ± 0,19 μg /mL) e P. rigida (Fo) (IC50 = 1,39 ± 0,53 μg /mL). Com exceção de L. paniculata(Fo), que apresentou IC50 de 14,40 ± 0,34 μg /mL e teor de flavonoides de 295,5 mg/g, todas as espécies que tiverem significativa atividade no teste do DPPH, também apresentaram teor de flavonoides semelhante ao padrão, ou seja, H. speciosa (Fo) (280,5 mg/g), Q. grandiflora(Fo) (260,5 mg/g), K. coriaceae (Ca) (250,5 mg/g), K. coriaceae (Fo) (235,5 mg/g). Quanto à determinação de fenóis totais e cumarinas os resultados foram inferiores quando comparados aos padrões.


ABSTRACT Ethanol extracts of Luehea paniculata, Anadenanthera colubrina, Cariniana rubra, Hancornia speciosa, Qualea grandiflora, Kielmeyera coriaceae and Palicourea rigida, collected in the Cerrado of Mato Grosso went through preliminary phytochemical analysis in order to identify major classes of secondary metabolites and also to quantify the total content of phenolics, flavonoids and coumarins. The study also describes the antimicrobial and antioxidant activity and the toxicity of the larva of Artemia salina. The C. rubra extracts and the Q. grandiflora were the most active against fungal strains of C. neoformans (MIC 3.91 µg/ml), C. krusei (MIC 15.63 µg/ml), C. parapsilosis(31.25 MIC µg/ml) and C. albicans (MIC 62.5 µg/ml), both with the same results for these organisms. Regarding the bacterial strains, the most sensitive one was the S. aureus with 250 µg/ml MIC for all the tested extracts. The Luhea paniculata and the Q. grandiflora were the most active against E. coli species (MIC 500 and 250 µg/ml, respectively) and E. faecalis (MIC 250 and 500 µg/ml, respectively). In the test of DPPH, K. coriaceae (Ca) (IC50 = 0.72 ± 0.51 µg/mL), K. coriaceae (Fo) (IC50 = 1.34 ± 0.53 µg/mL), H. speciosa (Fo) (IC50= 1.03 ± 0.52 µg/mL), Q. grandiflora (Fo) (IC50 = 1.31 ± 0.19 µg/ml) and P. rigida (Fo) (1.39 ± 0,53 µg/ml) stood out. With the exception of the L. paniculata (Fo), which showed an IC50 of 14.40 ± 0.34 µg/ml and a flavonoid content of 295.5 mg/g, all species that had significant activity in the DPPH test, also showed flavonoid content similar to the standard one, or H. speciosa (Fo) (280.5 mg/g) Q. grandiflora (Fo) (260.5 mg/g), K. coriaceae (Ca) (250.5 mg/g), K. coriaceae (Fo) (235.5 mg/g).

Atualmente tem crescido a busca por plantas como fontes naturais de antioxidantes. Plantas com esta propriedade podem ser utilizadas para melhorar a qualidade de alimentos, seja para fins nutritivos ou de conservação, como também para a prevenção de doenças relacionadas ao aparecimento de radicais livres. A Pereskia aculeata Mill. (Ora-pro-nóbis) é uma espécie da família Cactaceae e suas folhas são consumidas por muitas pessoas. Neste contexto, este trabalho estudou o teor de fenóis totais (método Folin-Ciocalteau) e de proantocianidinas (vanilina sulfúrica), correlacionando-os com a atividade antioxidante (radical DPPH, sistema b-caroteno/ácido linoleico e voltametria por pulso diferencial) de extratos em acetona 80%, etanol 70% e água das folhas da espécie P. aculeata Mill., apresentando o extrato em acetona 80% maior teor de fenóis totais e proantocianidinas. Com relação à atividade antioxidante frente ao radical DPPH, o extrato em acetona 80% se apresentou ligeiramente melhor que o em etanol 70%. A técnica de voltametria por pulso diferencial mostrou que o extrato em acetona 80% apresenta constituintes com maior potencial redutor que o extrato em etanol 70%. Os dados obtidos a partir da voltametria corroboram com os resultados obtidos pelo método tradicional de DPPH. Os extratos em etanol 70% e em água apresentaram melhor atividade antioxidante pelo sistema b-caroteno/ácido linoleico, o que sugere a presença de outra classe de compostos que atuam na inibição do b- caroteno. Portanto, as folhas da espécie Pereskia aculeata podem ser consideradas como fonte potencial de compostos antioxidantes.


Currently, the search for plants as natural sources of antioxidants has been growing because they can be used to improve the nutritive or preservation properties of food, as soon as for the prevention of diseases related to free radicals. The Pereskiaaculeata Mill. (Ora-pro-nóbis,) is a specie of Cactaceae family and its leaves are consumed by many people. In this context, this paper studied the total phenols (Folin-Ciocalteu) and proanthocyanidins (vanillin sulfuric) correlating with the antioxidant activity (DPPH, b-carotene/linoleic acid system and differential pulse voltammetry) of extracts of 80% acetone, 70% ethanol and water of Pereskiaaculeata Mill. leaves. The 80% acetone extract showed higher total phenols and proanthocyanidins. Regarding to the antioxidant activity by DPPH radical scavenger, the 80% acetone extract was slightly better than 70% ethanol extract. The differential pulse voltammetry technique displayed that the 80% acetone extract presented constituents with greater reducing potential than the ones presented in 70% ethanol extract, what can corroborate the results obtained. The 70% ethanol and water extracts has shown better antioxidant activity by b-carotene/linoleic acid system, what probably suggests the presence of another compounds classes that are responsible for the inhibition the b-carotene. Therefore, the Pereskiaaculeata Mill. showed that its leaves may be considered as a potential source of antioxidant compounds.

ABSTRACT

Green, black and white teas are all produced from leaves and shoots of Camellia sinensis, the only difference is how they are processed. The aim of this study was to compare the total phenols and flavonoid contents and antioxidant capacity of green, black and white tea bags of different brands. The morphodiagnosis of leaves was used to identification of plant material. HPLC-DAD fingerprinting coupled with Principal Component Analysis (PCA) was applied to analyze similarities of the tea samples. The results showed considerable variability between tea brands in both total phenols (30.55 to 60.85 mg of pyrogallol/g) and flavonoids (6.35 to 8.92 mg of quercetin/g). Green and white teas demonstrated the highest ABTS and DPPH radical scavenging activities.


Os chás verde, preto e branco são produzidos de folhas e brotos de Camellia sinensis, diferenciando-se pelo processamento. O objetivo deste estudo foi comparar o conteúdo de fenóis totais, flavonoides e capacidade antioxidante dos chás verde, preto e branco de diferentes marcas na forma de sachês. A análise morfo-anatômica das folhas foi realizada para a identificação do material vegetal. Os perfis químicos (fingerprints) obtidos por Cromatografia Líquida de Alta Eficiência (CLAE - DAD) foram analisados por ferramentas quimiométricas de análise exploratória (PCA) para análise comparativa entre as amostras. Os resultados evidenciaram considerável variação entre as marcas de chás, tanto para fenóis totais (30,55 a 60,85 mg de pirogalol/g), quanto para flavonoides (6,35 a 8,92 mg de quercetina/g). As amostras de chá verde e de chá branco apresentaram maior atividade antioxidante contra os radicais ABTS e DPPH.
