ABSTRACT

The aim of this study was to evaluate the antifungal susceptibility patterns of three antifungals, methanolic extracts and N-hexane oil of sesame seeds on C. albicans and C. glabrata, isolated from oral cavity of liver transplant recipients. The results were compared with other reports to develop a mini review as well. Candida species were isolated from liver transplant recipients. To evaluate the antifungal activity of sesame seed oil and methanolic extract, fluconazole, caspofungin and nystatin, the corresponding minimum inhibitory concentrations were determined by CLSI M27-A3 standard method. Minimum fungicidal concentration was also evaluated. The most prevalent species was C. albicans, followed by C. glabrata. Findings indicated sensitivity to antifungal agents and resistance to methanolic extract and N-hexane oil for all C. albicans and C. glabrata isolates. The rate of Candida colonization in the oral cavity of liver transplant recipients was high. Our results revealed that the methanolic and N-hexan extracts of sesame seeds are not effective on C. albicans and C. glabrata species, isolated from the patients. The sesame seed oil pulling and mouthwash cannot effectively cleanse and remove the Candida species in the mouth. Investigation of other medicinal plants or other parts of sesame like leaves and roots are suggested.

ABSTRACT

Devido ao aumento das infecções causadas por Candida albicans e espécies não-albicans, surge a necessidade de novas estratégias terapêuticas, como a identificação de cepas bacterianas com características probióticas. Assim, o objetivo foi avaliar a ação antimicrobiana de cepas clínicas de Lactobacillus paracasei, Lactobacillus fermentum e Lactobacillus rhamnosus sobre diferentes espécies de Candida (C. albicans, C. glabrata, C. krusei e C. tropicalis). Foram utilizadas cepas de Lactobacillus isoladas da cavidade bucal de indivíduos livres de cárie e cepas de Candida isoladas de lesões de candidose orofaríngea obtidas em estudos anteriores. Para estudo in vitro, foram formados biofilmes monoespécies de Candida (Grupo controle) e biofilmes mistos de Candida e Lactobacillus, nos quais os lactobacilos foram acrescentados antes da cepa de Candida (Grupo Profilático), após a cepa de Candida (Grupo Terapêutico) ou ao mesmo tempo da cepa de Candida (Grupo Simultâneo). Após 48 h de incubação, foi realizada a contagem do número de células viáveis de Candida (UFC/mL). Para estudo in vivo, os micro-organismos foram inoculados em larvas de G. mellonella e o desenvolvimento da candidose foi monitorado pela curva de sobrevivência, estudando-se os grupos experimentais descritos acima. Nos resultados in vitro, foi verificado que os efeitos inibitórios mais significativos de Lactobacillus sobre Candida ocorreram nos grupos profiláticos. As cepas de C. tropicalis e C. krusei foram mais sensíveis a ação de Lactobacillus do que C. albicans e C. glabrata. Além disso, foi verificado que as três espécies de Lactobacillus estudadas (L. paracasei, L. rhamnosus e L. fermentum) tiveram ação inibitória sobre Candida. Em relação ao estudo in vivo, também foi verificado que os grupos profiláticos tiveram maiores efeitos inibitórios sobre a candidose experimental em relação aos grupos terapêuticos, levando a um aumento significativo nas taxas de sobrevivência das larvas. Em relação as cepas estudadas, os efeitos profiláticos de Lactobacillus sobre a candidose, foram encontrados, de forma semelhante, para todas as cepas de Candida e Lactobacillus estudadas. Assim, concluiu-se que as cepas clínicas de L. paracasei 28.4, L. rhamnosus 5.2 e L. fermentum 20.4 foram capazes de inibir a formação de biofilme e desenvolvimento de candidose causados por C. albicans, C. krusei, C. tropicalis e C. glabrata, principalmente quando Lactobacillus foi administrado de forma profilática(AU)


Due to the increase of infections caused by Candida albicans and non-albicans species, there is a need for new therapeutic strategies, such as the identification of bacterial strains with probiotic characteristics. The objective of this study was to evaluate the antimicrobial action of Lactobacillus paracasei, Lactobacillus fermentum and Lactobacillus rhamnosus strains on different Candida species (C. albicans, C. glabrata, C. krusei and C. tropicalis). We used Lactobacillus strains isolated from the oral cavity of caries-free and Candida strains isolated from lesions of oropharyngeal candidiasis. For in vitro study, monospecies of Candida (Control Group) and mixed biofilms of Candida and Lactobacillus were prepared, in which the lactobacillus were attached before the Candida strain (Profile Group), after a Candida (Therapeutic Group) strain or same time as the Candida strain (Simultaneous Group). After 48 h of incubation, we counting the number of viable Candida cells (CFU/mL). For in vivo study, we inoculate the microorganisms in larvae of G. mellonella and the development of the application for monitoring by survival curve, studying the experimental groups described above. In the in vitro results, we find that the most significant inhibitory effects of Lactobacillus on Candida occurred in the prophylactic groups. The strains of C. tropicalis and C. krusei were more sensitive to the action of Lactobacillus than C. albicans and C. glabrata. In addition, it we verified that the three species of Lactobacillus studied (L. paracasei, L. rhamnosus and L. fermentum) had an inhibitory action on Candida. In relation to the in vivo study, we verified that the prophylactic groups had more inhibitory effects on an experimental candidose in relation to the therapeutic groups, leading to a significant increase in the survival rates of the larvae. In relation to the studied strains, the prophylactic effects of Lactobacillus on candidiasis were similarly for all Candida and Lactobacillus strains studied. Thus, it concluded that the clinical strains of L. paracasei 28.4, L. rhamnosus 5.2 and L. fermentum 20.4 were able to inhibit a biofilm formation and development of Candida caused by C. albicans, C. krusei, C. tropicalis and C. glabrata, especially when Lactobacillus was given prophylactically(AU)

ABSTRACT

Este trabalho teve como objetivo isolar e identificar leveduras do gêneroCandida isoladas de amostras bucais de crianças na faixa etária de 2 a 8 anos com paralisia cerebral, atendidas na associação de pais e amigos dos excepcionais (APAE) em um município do interior da Bahia e atendidas em unidade de saúde da família do mesmo município. Amostras de saliva de 20 crianças (2-8 anos) foram coletadas com auxílio de swabe encaminhadas para processamento no laboratório de Microbiologia onde foram semeadas em Agar Sabouraud Dextrose com cloranfenicol em duplicata e, em seguida, foram incubadas a 35°C±2/ 24h e mais 5 dias à temperatura ambiente para verificação de crescimento de unidades formadoras de colônia por mililitro (UFC.mL-1). Os dados obtidos em UFC.mL-1 foram transformados em logaritmo (Log) e submetidos tanto à análise descritiva quanto ao teste estatístico ANOVA (5%) para observação de diferenças de relevância estatística. Todos os isolados de amostra bucal de crianças do grupo com paralisia cerebral apresentaram positividade para espécies de leveduras do gênero Candida. No grupo de crianças com paralisia cerebral houve um maior isolamento de leveduras de gênero Candida (médialog = 1,8 UFC.mL-1) nas amostras bucais em relação ao grupo de crianças controle (médialog = 0,6 UFC.mL-1). Crianças portadoras de paralisia cerebral apresentam um índice aumentado de espécies de Candida na cavidade bucal que, juntamente com outras condições odontológicas peculiares, predispõem ao aparecimento de candidíases e requerem preparo da equipe de saúde bucal para obtenção de melhores resultados no tratamento direcionado a essa população...


This study aimed to isolate and identify Candida species from oral samples of 2-8 years children with cerebral palsy from the Association of Parents and Friends of Exceptional (APAE) and assisted at the family health unit at a town in the interior of Bahia. Saliva samples from 19 children were collected with a swab and sent for processing in the microbiology laboratory where they were plated on Sabouraud Dextrose Agar with chloramphenicol in duplicate and then incubated at 35°C±2 for 24h and then 5 days at room temperature to check the growth of colony-forming units per milliliter (UFC.mL-1). Data from UFC.mL-1 were transformed into logarithm (Log) and submitted to descriptive analysis and ANOVA (5%) to observe statistic differences. All oral isolates of children with cerebral palsy were positive for Candida yeast species. Greater isolation of Candida yeasts (medialog = 1.8 UFC.mL-1) was observed in the group of children with cerebral palsy when compared to control group (medialog = 0.6 UFC. mL-1). Children with cerebral palsy have an increased rate of Candida species in the oral cavity, along with other peculiar dental conditions that predispose to candidiasis. Thus proper training of the oral health team is required in order to achieve better results in the treatment of this population...

La infección por Candida a nivel orofaríngeo, es la infección oportunista más frecuente en pacientes VIH/SIDA,al menos un episodio de candidiasis desarrollan estos pacientes en el curso de la evolución de su enfermedad. La determinación de especies de Candida, permite orientar de manera adecuada el tratamiento. El objetivo de la presente investigación fue identificar las especies de Candida aisladas en 60 pacientes VIH/SIDA con candidiasis orofaríngea, asistentes al Centro de Atención Integral - Enfermedades de Transmisión sexual y SIDA. (CAI/ETS/SIDA) en el período Junio-Diciembre 2006. Se les tomó muestra de raspado lingual y se cultivó en Agar Sabouraud-Cloranfenicol para el aislamiento primario. La identificación de las especies de Candida, se realizó por medio de la producción de: tubos germinativos, clamidosporas en Agar - Maiz Tween 80 según la técnica de Dalmau y el sistema Api 20CAUX bioMérieux. Se observó en estos pacientes predominio del sexo masculino (48) 80%, con edades comprendidas entre 20 y 60 años Se aislaron en orden de frecuencia C. albicans 66,6 % (40), C. tropicalis 13,33 % (8), C. glabrata 11,6 % (7), C. parapsilosis 6,66 % (4), C. krusei, 66% (1). Se recomienda la identificación de especies de Candida como prueba de rutina en pacientes VIH/SIDA


Oral candidiasis has been recognized as the most common opportunistic infection in HIV/AIDS patients. These patients present at least one episode of oral candidiasis in the development of the disease. The isolation of Candida species has let to apply the correct treatment to these patients. The purpose of the investigation was to identify the species of isolated Candida in 60 HIV/AIDS patients with oropharyngeal candidiasis that assist to the Center of Integral Attention of Sexual Transmission Diseases and AIDS from June to December 2006. The samples were taken from the mucosal surface of the tongue and laid in Sabouraud-Cloramphenicol agar for the first isolation; the identification of Candida species were done by forming germ tubes, Terminal Chlamydospore formation on corn meal agar with Tween-80 following the Dalmau technique and the Api 20CAUX bioMerieux. Most of the patients were males (48) 80% and age ranging 20 to 60 years old. The most frequent yeast specie found was : C, albicans 66,6 % (40) , C. tropicalis 13,33 % (8) , C. glabrata 11,6 % (7) ,C. parapsilosis 6,66 % (4) , C. krusei 1,66% (1). It is recommended the isolation of the species as a rutin test in the HIV/AIDS patients

ABSTRACT

Candidemia is a disease with high morbidity and mortality especially in critical care patients. Early diagnosis enables early treatment. Objectives: To conduct a systematic review of the literature in order to establish the best laboratory tests for the diagnosis of candidemia in critical patients. Materials and Methods: We conducted a systematic review of available literature in PubMed. Serological studies were subjected to meta-analysis in metadisk-Beta 1.1.1. Results: 4 studies of 1286 reviewed were included. Three were about serological tests and one about molecular testing (RT-PCR). The sensitivity and specificity for RT-PCR, antibody testing and antigen and antibody tests were 87 percent and 100 percent, 47.5 percent and 82.6 percent, 96 percent and 81 percent, respectively. Diagnostic Odds Ratio of antigenemia was 1.51 (95 percent CI = 0,032-70,964, p = 0.001). Conclusions: RT-PCR has better diagnostic performance, measuring antigenemia plus antibodies improves sensitivity, specificity, LR + and LR-- . There is insufficient evidence to support this.


La candidemia es una patología con alta morbilidad y mortalidad, especialmente en los pacientes sometidos a servicios de cuidado crítico. El diagnóstico precoz permite realizar tratamiento temprano. Objetivos: Realizar una revisión sistemática de la literatura para establecer cuáles son las pruebas de laboratorio con mejor rendimiento diagnóstico y operativo para el diagnóstico de candidemia en cuidado intensivo. Materiales y Métodos: Se realizó una revisión sistemática de la literatura disponible en PubMed, se sometieron a meta-análisis estudios de pruebas serológicas en MetaDisc-Beta 1.1.1. Resultados: Se incluyeron 4 estudios de 1.286 revisados, 3 de pruebas serológicas y 1 de RPC-RT. La sensibilidad y especificidad fue de 87 y 100 por ciento para RPC-RT, 47,5 y 82,6 por ciento para pruebas de anticuerpos, 96 y 81 por ciento para pruebas de antígeno y anticuerpo. La ORD de antigenemia 1,51(IC95 por ciento = 0,03270,964; p = 0,001). Conclusiones: RPC-RT tiene mejor rendimiento diagnóstico, la medición de antigenemia más anticuerpos mejora la sensibilidad, especificidad, LR+ y LR-. No hay suficiente evidencia que soporte esto.

ABSTRACT

Objetivo: avaliar a presença de espécies Candida na cavidade bucal e no tubo orotraqueal de pacientes internados em Unidade de Terapia Intensiva (UTI), além da prevalência de alterações bucais. Materiais e Métodos: A condição bucal de 44 indivíduos internados na Santa Casa de Misericórdia de Barretos foi avaliada durante o período de 30 dias consecutivos. Amostras de saburra lingual, do biofilme dental e do tubo orotraqueal foram coletadas em três dias consecutivos após intubação para posterior contagem das unidades formadoras de colônias de Candida spp. (UFC!mL). A identificação das espécies foi feita por meio de utilização do meio de cultura cromogênico. Resultados: Os achados mostraram que as alterações bucais observadas foram: úlceras traumáticas (68,2%), hematoma de lábio e assoalho da boca (6,8%), hiposalivação (84,1%), acúmulo de biofilme (90,9%). saburra lingual (81,8%), ressecamento labial (86,3%), queilite angular (34%) e outras lesões associadas à Candida spp. (27,3%). Todos os 31 indivíduos intubados apresentaram aumento da colonização por Candida spp. com o tempo de internação, sendo que a saburra lingual apresentou maior número e diversidade de leveduras (UFC!ml.x106) no primeiro dia (2,0) se comparado com a colonização do tubo orotraqueal (0,9) e do biofilme dental (0,6). O tubo orotraqueal apresentou colonização estatisticamente maior no terceiro dia (67,0), se comparado à saburra lingual (9,0) e biofilme dental (4,0). Conclusão: Conclui-se que pacientes internados em UTI apresentam risco elevado de desenvolverem alterações bucais e apresentarem colonização por leveduras do gênero Candida na saburra lingual e no tubo orotraqueal, podendo favorecer o desenvolvimento de pneumonia nosocomial.


Objective: to assess the presence of Candida species in the oral cavity and in the endotracheal tubeof patients admitted to the Intensive Care Unit (lCU), besides, prevalence of oral alterations. Materiais and Methods: The prevalence of oral alterations was verified in forty-four patients admitted to the Santa Casa of the Misericordia of Barretos during the period of 30 days. Samples of tongue coating, dental biofilm and the endotracheal tube were collected on three different days after intubation, for subsequent counting of colonies of Candida spp (CFU! mL). Species identification was performed by use of chromogenic culture medium. Results: The findings showed that the oral features observed were traumatic ulcers (68.2%), hematoma lip and mouth floor (6.8%), hyposalivation (84.1%), accumulation of biofilm (90.9 %), tongue coating (81.8%), dry lips (86.3%), angular cheilitis (34%) and other lesions associated with Candida spp. (27.3%). Ali 31 individuais had been intubated showed increased colonization by Candida spp over time. The tongue coating had the greatest number and diversity of Candida yeast (UFC! ml.x106) at the first day after intubation (2.0), if compared with endotracheal tube (0.9) and dental biofilm (0.6) colonization. The endotracheal tube had the highest statistically colonization at third day (67.0), if compared with tongue coating (9.0) and dental biofilm (4.0). Conclusion: it can conclude that ICU patients have high risk to develop oral abnormalities and colonization by Candida yeasts on the tongue coating and the endotracheal tube, which may favor the development of nosocomial pneumonia.

ABSTRACT

Objetivo: O objetivo deste estudo foi avaliar in vitro a ação antifúngica do extrato hidroalcóolico da Uncaria tomentosa L. (unha de gato) sobre cepas padrão de Candida albicans ATCC 36.232, Candida guilliermondii ATCC 6.260, Candida krusei ATCC 34.135 e Candida tropicalis ATCC 13.803, responsáveis por afecções bucais conhecidas por candidíases, ou candioses. Métodos: O material botânico utilizado para os testes foram o micropulverizado do caule da Uncaria tomentosa L., que em seguida foi submetido ao processo de maceração para preparação do extrato. Foram utilizadas linhagens padronizadas de microrganismos Candida (ATCC), obtidas mediante solicitação na Fundação Oswaldo Cruz, Rio de Janeiro. A atividade antimicrobiana e determinação da Concentração Inibitória Mínima (CIM) do extrato da Uncaria tomentosa L. foi determinada através da técnica de ágar-difusão em placas de Petri, meio sólido Agar Mueller Hington - DIFCO. Os mesmos procedimentos foram realizados com a clorexidina em concentração 0,12%, um anti-séptico oral utilizado como controle positivo nos experimentos. Resultados: Todas as espécies avaliadas apresentaram-se sensíveis ao extrato da Uncaria tomentosa L., atuando de forma semelhante à ação da clorexidina (controle positivo). As cepas apresentaram halos de inibição de crescimento até a diluição de 1:16 do extrato. Candida albicans foi o microorganismo que apresentou maior sensibilidade ao extrato da Uncaria tomentosa L., seguido de Candida guilliermondii, Candida krusei e Candida tropicalis. Conclusão: Os resultados deste estudo demonstram que a Uncaria tomentosa L., conhecida popularmente como unha de gato, possui compostos bioativos com atividade antimicrobiana sobre fungos do gênero candida, responsáveis pelas manifestações de candidíase bucal.


Objective: The aim of this study was to evaluate in vitro the antifungal effect of the hydroalcoholic extract of Uncaria tomentosa L. (Cat's Claw) on strains of Candida albicans ATCC 36232, Candida guilliermondii ATCC 6260, Candida krusei ATCC 34135 and Candida tropicalis ATCC 13 803, responsible for oral diseases known as candidiasis or candioses. Methods: The botanical material used in the tests were sputtered from the stem of Uncaria tomentosa L., which was then subjected to the maceration process for preparing the extract. We used standardized strains of Candida microorganisms (ATCC), obtained upon request at the Oswaldo Cruz Foundation, Rio de Janeiro. The antimicrobial activity and determination of Minimum Inhibitory Concentration (MIC) of the extract of Uncaria tomentosa L. was determined by agar-diffusion technique in Petri dishes, solid Mueller Hinton Agar - DIFCO. The same procedures were performed with the 0.12% chlorhexidine, an antiseptic used as positive control in the experiments. Results: All species evaluated were sensitive to the extract of Uncaria tomentosa L., operating similarly to the action of chlorhexidine (positive control). The strains showed inhibition zones of growth until the 1:16 dilution of the extract. Candida albicans was the microorganism that showed greater sensitivity to the extract of Uncaria tomentosa L., followed by Candida guilliermondii, Candida krusei and Candida tropicalis. Conclusion: The results of this study show that Uncaria tomentosa L., popularly known as cat's claw, has bioactive compounds with antimicrobial activity against fungi of the genus Candida are responsible for the manifestations of oral candidiasis.

El presente trabajo se llevó a cabo para evaluar la eficiencia del medio de cultivo a partir de guayaba agria (Psidium araca) frente a medios comerciales en el crecimiento de tres cepas nativas: Candida guillermondii, Candida famita y Candida sp. Se evaluó el crecimiento microbiano a diferentes concentraciones de fruta, 5, 10, 25 y 50% p/v, tomando como control los medios comerciales: Malta, Sabouraud y agar papa dextrosa (PDA). La productividad y selectividad del medio de guayaba agria fue determinada mediante el método Ecométrico en un tiempo de 48 horas. Los análisis estadísticos aplicados para evaluar y comparar el crecimiento de las cepas en los medios comerciales y en el medio de guayaba agria a diferentes concentraciones demostraron lo siguiente: Candida guillermondii presentó crecimiento mayor o igual a 25 y 50% p/v comparado con los medios comerciales; Candida famata y Candida sp presentaron mejores crecimientos al 5% p/v, con respecto a los diferentes medios comerciales. Los resultados demostraron que el medio de cultivo es altamente productivo y no selectivo, lo que representa una alternativa en la conservación, el mantenimiento y el desarrollo de las levaduras estudiadas.


This work was carried out to evaluate the efficiency of the culture medium from sour guava (Psidium araca) against commercial media in the growth of three native strains: Candida guillermondii, Candida famata and Candida sp. Microbial growth was evaluated at different concentrations of fruit, 5, 10, 25, 50% w /v, using as control the commercial media: Malta, Sabouraud and PDA (Potato Dextrose Agar). The productivity and selectivity of the sour guava medium was determined by the Ecometric method in a time of 48 hours. The applied statistical analysis to evaluate and compare growth of strains in commercial culture medium and in the medium from sour guava at different concentrations showed: Candida guillermondii grew greater than or equal to 25 and 50% w / v compared with commercial medium, Candida famata and Candida sp showed better growth at 5% w / v, with respect to commercial medium. The results showed that the medium is highly productive and non-selective representing an alternative to the conservation, maintenance and development of the yeasts.

There is only scanty data on the effects of specific antibody, with or without complement, on Candida albicans or Candida krusei in cell-free systems in vitro, although previously published work has shown that specific antibody mediates anti- Candida immunity in vivo by inhibition of adherence to host cells or surfaces and by the promotion of phagocytosis and intra-phagocytic killing. The MTT (3-[4, 5-dimethyl-2-thiazolyl] -2, 5-diphenyl -2H- tetrazolium bromide)-reduction method as a test of the viability of fungi was used to investigate the effect of complement, normal serum and immune serum on these two species of Candida that are of increasing importance as opportunistic pathogens. We report that normal rabbit serum or strain-specific, polyclonal anti- Candida rabbit antibody, with or without guinea pig complement, did not cause the reduction of total cell-mass or of the viability of either C. albicans or C. krusei, in vitro as determined by the MTT-reduction test. Complement alone without specific antibody, also, had no such effect on these two Candida species.

ABSTRACT

Se identificaron mediante métodos tradicionales y automatizados las especies de Candida provenientes de una cohorte de 289 pacientes VIH seropositivos y con SIDA con o sin orofaringitis y con o sin tratamiento antifúngico. Se evaluó la susceptibilidad de las diferentes cepas aisladas a las drogas antifúngicas con el método de la NCCLS y el método de los pozos de difusión. Los aislados obtenidos se tipificaron a través de la reacción en cadena de la polimerasa (RAPD), principalmente las que fueron resistentes a diferentes antifúngicos imidazólicos con el fin de buscar los factores que modulan esta conducta en nuestro medio. Nuestros resultados demostraron que en esta cohorte de pacientes VIH/SIDA evaluados, las características clínicas y demográficas son similares a las reportadas mundialmente, y desde la introducción de el TARGA, la frecuencia de las infecciones por Candida spp disminuyó. El incremento del uso de TARGA en estos pacientes, resultó inversamente proporcional al uso de antimicóticos. La distribución y frecuencia de las especies de Candida aisladas, en este estudio fue: C. Albicans 394 (80,4 por ciento), C. tropicalis 43 (8.78 por ciento), C. Glabrata 24 (4,89 por ciento), C. dubliniensis 16 (3,26 por ciento), C parapsilosis 8 (1,63 por ciento), C. guilliermondii 2 (0,4 por ciento), C. krusei 2 (0,4 por ciento) y C. lipolytica 1 (0,2 por ciento). El uso indiscriminado principalmente de fluconazol, podría ser responsable de la presión selectiva sobre Cándida spp y podría estar impactando la ecología oral de la misma. Hasta presente fecha, en Venezuela no se había reportado el hallazgo de C. dublimensis. (3,26 por ciento). El 37,5 por ciento C. albicans provenientes de pacientes con tratamiento antimicótico fueron resistentes al fluconazol (resistencia secundaria) y 23,95 por ciento mostraron también resistencia al itraconazol (resistencia cruzada).

Candidíase Mucocutânea crônica é uma imunodeficiência primária que tem como característica a suscetibilidade a infecções em mucosas, no tecido cutâneo e em seus anexos por Candida, na maioria das vezes por Candida albicans. Um defeito na resposta a Candida, mediada por células tem sido documentada em portadores de CMC. Alguns poucos trabalhos sugerem que os pacientes com CMC apresentem uma alteração na produção de citocinas em resposta a Candida. Nós estudamos a produção de citocinas pelas células T de quinze pacientes com CMC. Avaliamos a produção de IL-2,IL-4,IL-10 e IFN-y em ensaios de linfoproliferação estimulados com antígenos de Candida, Toxóide tetânico(TT) e também avaliamos a produção de citocinas destas células,nestas mesmas condições,quando imunomoduladas com IL-2,IL-12 e anticorpo anti-IL-10. Para obter esta analise, utilizamos ensaios de citocina intracelular e avaliação por citometria de fluxo / Chronic Mucocutaneous Candidiasis (CMC) is a primary immune deficiency presenting as an inability to clear fungal infections and consequently as persistent and recurrent infections of the skin and mucous membranes with yeasts, mostly Candida albicans. ABSTRACT

Se estudiaron 18 pacientes con sida, que presentaban síntomas gastrointestinales altos o lesiones en la cavidad oral sugestivos de candidiasis. Se obtuvieron datos clínicos, muestras de la cavidad oral, biopsias y cepillado esofágico, así como suero de todos los pacientes. El síntoma que principalmente se observó en la candidiasis esofágica fue la disfagia; la candidiasis oral se comportó de forma asintomática. Candida albicans fue la especie que más se aisló con predominio del serotipo A. Se analizaron por la técnica de electroforesis en gel de poliacrilamida los perfiles proteicos de las 12 cepas pertenecientes a esta especie, se observó identidad de sus patrones de peso molecular, lo que sugiere que sea la misma cepa la que se halla en la cavidad oral y en el esófago. Se evaluó la respuesta de anticuerpos anti-C. albicans, no resultó útil la inmunodifusión doble para el diagnóstico de candidiasis esofágica.


18 AIDS patients who presented high gastrointestinal symptoms or lesions in the oral cavity suggestive of candidiasis were studied. Clinical data, specimen of the oral cavity, biopsies and esophageal brushing, as well as serum from all patients were obtained. Dysphagia was the main symptom observed in the esophageal candidiasis. Candida albicans was the most isolated species with a predominance of serotype A. The protein profiles of 12 strains belonging to this species were analyzed by the polyacrylamide gel electrophoresis, and it was found that their molecular weight patterns were identical, which indicates that the same strain is in the oral cavity and in the esophagus. The response of the anti-C. albicans antibodies was evaluated. The double immunodiffussion was not useful for the diagnosis of esophageal candidiasis.

ABSTRACT

The role of selenium in the diet of rats has been examined with respect to the neutrophil functions. Feeding of Se-deficient diet for 75 days resulted in reduction in candidacidal activity, superoxide production, oxygen consumption, glucose utilisation and glutathione peroxidase activity. Supplementing the diet with Se for 30 days resulted in partial restoration of all the activities.

ABSTRACT

A total of one hundred and fifty-two strains of different Candida species was obtained from five hundred and ten human aces of candidiasis. The isolates could be identified as 67 Candida albicans, 20 C. Krusei, 18 C. tropicalis, 10 C. utilis, 9 C. pseudotropicalis, 8 C. parapsilosis, 6 C. lipolytic; 5 each of C. guilliermondii and C. stellatoidea and 4 C. tenusi. Experimental infection in mice showed that C. albicans and C. Krusei were most pathogenic than other species. The study of immune response after experimental injection in rabbits with the most pathogenic strains of each Candida species by agglutination, double diffusion, passive hemagglutination and ELISA was carried out. All the techniques were done using the whole cell antigen and extracts with ether, petroleum ether, chloroform and ethanol. Cross antigenic reaction was demonstrated between the six Candida species. Using the agglutination, precipitation and hemagglutination techniques were able to detect specific antibodies in eight out of seventy-five patients serum samples. The number of serologically positive cases was higher [22.6%] by using the ELISA method

ABSTRACT

Twenty diabetic uraemics and twenty two healthy matched controls comprised the material for this study. Cell mediated immunity was assessed by estimation of T-cell rosette percentage and cutaneous response to recall antigens--purified protein derivative, candida antigen and 2:4 dinitrochlorobenzene. Results analysis revealed depressed cell mediated immunity in diabetic uraemics in the form of impaired cutaneous response to recall antigens and reduction in T cell rosette percentage, in comparison to controls.

ABSTRACT

Se estudiaron tres cepas de hongos anemófilos de distintos géneros, que fueron: Alternaria, Rhizopus y Monilia. Encontramos que el medio de cultivo utilizado en esta investigación (Sabouraud líquido) perdía nutrientes a partir de los siete días del crecimiento de las cepas, pero sin afectarse la viabilidad de las mismas. Esto se comprobó por resiembras a los 15 y 60 días. Se elaboraron extractos alergénicos con micelios de esos hongos con siete, 15 y 60 días de crecimiento y se probaron dichos extractos en pacientes supuestamente sensibilizados, por medio de pruebas cutáneas por intradermorreacción, no encontrando diferencias significativas entre los habones obtenidos con estas pruebas. Se recomiendan investigaciones por periodos más prolongados (90,120,180 ó más días) y ver si ello incide en la viabilidad y alergenicidad de los hongos anemófilos.

Forty strains of Cryptococcus neoformans, 30 of Candida albicans, 24 of C. parapsilosis and 10 strains each of C. tropicalis and C. (Torulopsis) glabrata were examined. A 0.2 per cent solution of concanavalin A (Con A) in phosphate buffered saline and commercial anti-A and anti-B blood grouping sera were used in the whole cell slide agglutination test. All the isolates of Candida species strongly reacted with Con A and over 90 per cent were agglutinated by anti-A and -B blood grouping sera. In contrast, except for one strain, Cryptococcus neoformans did not agglutinate with Con A or blood grouping sera. These findings suggest, on one hand, a fundamental difference in the sugar mosaic of the cell surface components of Cryptococcus neoformans and Candida species, and on the other, presence of similar antigenic determinant(s)/receptors on the cell surface of Candida species and human erythrocytes.

ABSTRACT

Se procesaron 268 sueros para detectar la presencia de anticuerpos y antígenos de Candida por inmunodifusión en gel de agar o contrainmunoelectroforesis. Cuarenta y dos sueros dieron resultados positivos: 37 para anticuerpos, 4 para antígeno y 1 para ambos. Fueron comparados los datos clínicos de los pacientes que tuvieron resultados positivos, con los correspondientes al total de los enfermos cuyos sueros fueron analizados. La significación estadística de las diferencias encontradas, fue determinada por el método de X2. Pudo observarse un aumento significativo de la incidencia de resultados positivos en pacientes de edad avanzada (71-80 años), en los materiales procedentes de hospitales oncológicos, de servicios de cirugía o de terapia intensiva. También fue elevada la frecuencia de resultados positivos en pacientes diabéticos, con neumopatías y con tumores sólidos. Por el contrario, la incidencia de serologías positivas fue significativamente más baja en los sueros procedentes de hospitales pediátricos, en niños de 4 a 10 años, en los portadores de leucemias y en los pacientes inmunosuprimidos

Foram investigadas as atividades antibacteriana e antifúngica dos extratos éter de petróleo e alcaloídico de folhas de Aristolochia gigantea Mart e Zucc, pelo método de difusäo em agar. O extrato éter de petróleo mostrou atividade contra uma bactéria Gram-positiva. O extrato alcaloídico apresentou atividade contra duas bactérias Gram-positivas. Uma Gram-negativa, näo apresentando qualquer atividade antifúngica.
