ABSTRACT

The present study was carried out to see the effect of two zinc salts i.e zinc sulphate and zinc chloride on gastric ulcers induced by stress, pylorus ligation and aspirin in albino rats. The rats were divided into two main groups (zinc sulphate 30, 60, 90 mg/kg i.p and zinc chloride 10 and 20mg/kg i.p). They were further sub-divided into three sub-groups dependant on ulcer model i.e stress, pylorus ligation and aspirin induced ulcers. It was found that zinc sulphate and zinc chloride had a dose dependant reduction in ulcer index in all three models of gastric ulceration. Also, both the salts had anti acid secretory effect, raised pH of gastric secretion and reduced total acidity significantly. Thus zinc salts prevent gastric ulceration. Probably this effect is mediated by anti acid secretory action.

ABSTRACT

Lansoprazole is a new proton pump inhibitor which produces prolonged decrease of gastric acidity. It has been used in severe gastro-oesophageal reflux disease with beneficial effect. We confirmed the efficacy of Lansoprazole in severe [grade IV] reflux esophagitis at the dosage of 30mg daily for 3 months. All patients had symptomatic response after 2 to 6 weeks of therapy. Endoscopically 90% of patients had complete healing of reflux esophagitis at 12 weeks. This indicates that at conventional dosage of 30 mg daily Lansoprazole gives good symptomatic and endoscopic response in patients with gastro-oesophageal reflux disease who failed to improve with H2 receptor antagonists

ABSTRACT

The effect of maprotiline [a tetracyclic antidepressant] in a dose of 5 mg/kg, orally for 21 days on gastric secretions and ethanol induced gastric ulcer was tested in rats. Maprotiline decreased the volume, free and total HCI acid concentration and peptic activity. While it produced an increase in mucin content as compared with the control group. Moreover, Maprotiline pretreatment [5 mg/kg, daily for 21 days orally] decreased the ulcer index of ethanol induced ulcer by 59.2%. The data suggest that Maprotiline in the tested dose had acid-pepsin antisecretory effect and ulcer protective activity against ethanol induced ulcer. These effects may be mediated through anticholinergic, antihistaminergic and the possible antidopaminergic effects of Maprotiline

ABSTRACT

The effect of serotonin on gastric acid secretion and its acidity have been examined by using four hour pylorus ligated rats of different age groups. Serotonin alone decreased the volume of gastric secretion and, free and total acidity in both younger and older animals, while methylsergide alone increased these parameters. Each of these drugs produced a significantly higher response in older animals. Methylsergide when given with serotonin abolished the inhibitory effect of serotonin and the observed secretions were similar to those of basal state. This indicates that serotonin as gastric inhibitor acts in accordance with parietal cell mass which increases with age

ABSTRACT

The efficacy of omeprazol in increasing intragastric pH to more than 2.5 and decreasing the gastric fluid volume to less than 25 ml was studied in 20 obstetric patients undergoing elective cesarean section. Omeprazole capsule [20 mg] was given orally twice, the evening before and the morning of surgery. A gastric tube was inserted and intragastric contents were aspirated before the drug. The volume and pH were measured immediately after induction of anesthesia and repeated at completion the end of surgery. No adverse reactions were noted in the mothers or the neonates. Omeprazole 20 mg orally twice appeared to be effective in reducing intragastric volume and acidity [P < 0.005]

ABSTRACT

Effects of nifedipine and cimetidine on gastric acidity were studied in pylorus ligated rats in doses of 0.25 mg, 0.5 mg and 1.0 per Kg given intraperitoneallv. Both drugs inhibited gastric secretions dose dependently. Nifedipine was found to be slightly more effective than cimetidine in raising gastric pH and reducing acidity titratable to 7.0 pH at all dose levels. However, it was less effective in reducing voume of gastric secretions. It is likely that nifedipine has greater anti-ulcer effect than cimetidine in rats
