ABSTRACT

SUMMARY AIMS Omentin is an adipokine primarily produced by visceral adipose tissue and its reduced levels have been shown to be associate with worse metabolic outcomes. We aimed to study the effects of preoperative ibuprofen on postoperative omentin levels in rats after surgery. METHODS Forty-eight albino Wistar rats, 6 in each of 8 groups according to the surgical procedure (laparotomy, laparotomy plus ibuprofen (IBU), nephrectomy, nephrectomy plus IBU, hepatectomy, hepatectomy plus IBU, splenectomy and splenectomy plus IBU). The Omentin levels of the groups were postoperatively analyzed. RESULTS The mean omentin was significantly higher in the laparotomy plus IBU group compared to the laparotomy group (p<0.001). Mean Omentin was significantly higher in the hepatectomy plus IBU group compared to the hepatectomy group (p=0.01). Mean Omentin was significantly higher in the nephrectomy plus IBU group compared to the nephrectomy group (p=0.001). CONCLUSION We suggest that preoperative ibuprofen may enhance circulating levels of Omentin, which has beneficial effects in trauma and inflammation settings in subjects that undergo minor or major abdominal surgery.


RESUMO OBJETIVOS A omentina é uma adipocina produzida principalmente pelo tecido adiposo visceral e níveis reduzidos dela foram associados a piores desfechos metabólicos. Nosso objetivo foi estudar os efeitos do uso pré-operatório do ibuprofeno nos níveis pós-operatórios da omentina em ratos. METODOLOGIA Quarenta e oito ratos Wistar albinos foram divididos em 8 grupos (6 em cada), de acordo com o procedimento cirúrgico: laparotomia, laparotomia e ibuprofeno (IBU), nefrectomia, nefrectomia e IBU, hepatectomia, hepatectomia e IBU, esplenectomia, e esplenectomia e IBU. Os níveis de omentina dos grupos foram analisados após a cirurgia. RESULTADOS A omentina média foi significativamente maior no grupo de laparotomia e IBU do que no grupo de laparotomia (p<0,001). A omentina média foi significativamente maior no grupo de hepatectomia e IBU do que no grupo de hepatectomia (p = 0,01). A omentina média foi significativamente maior no grupo de nefrectomia e IBU do que no grupo de nefrectomia (p = 0,001). CONCLUSÃO Sugerimos que o uso pré-operatório de ibuprofeno pode aumentar os níveis circulantes de omentina, que têm efeitos benéficos em um contexto de trauma e inflamação em indivíduos submetidos cirurgia abdominal.

ABSTRACT Objective Adipokines are mediators of body composition and are involved in obesity complications. This study aimed to assess the association of circulating omentin-1, vaspin, and RBP-4 with body composition indices and metabolic health status (MHS) in different phenotypes of body size. Subjects and methods A total of 350 subjects were included in the current cross-sectional study. Body composition was measured using a body composition analyzer, and serum concentrations of omentin-1, vaspin, and RBP-4 were assessed by ELISA kits. Results Circulating omentin-1 was significantly (OR = 1.81, 95% CI: 1.00-1.91, P = 0.01) and marginally (OR = 1.63, 95%CI: 1.00-1.75, P = 0.06) associated with MHS in the overweight and obese subjects, respectively. But no association was seen between omentin-1 and MHS in normal-weight subjects. Serum levels of vaspin and RBP-4 were not correlated with MHS. Furthermore, a significant positive correlation was observed between circulating omentin-1 and body mass index (BMI) as well as fat percentage (P = 0.02) in the MHS group. Serum vaspin concentrations were not related to body composition components in both groups. In addition, in the MHS group, circulating RBP-4 was positively correlated with fat percentage and fat mass (FM) (p < 0.0001) and was negatively correlated with fat-free mass (FFM) and total body water (TBW) (p < 0.0001). In contrast, in the metabolically unhealthy group, RBP-4 was negatively correlated with fat percentage, FM, and BMI (p < 0.0001) and was positively correlated with FFM and TBW (p < 0.0001). Conclusions This study showed that circulating levels of omentin-1 are useful predictors of metabolic health status in overweight and obese people.

ABSTRACT

To investigate the association between serum and synovial fluid levels of both resistin and YKL-40 with markers of inflammation, disease activity and radiographic grading and to determine if they have a role in the pathogenesis of RA. This study was conducted on 30 patients with RA and 15 healthy controls with acute post traumatic knee effusion. Serum and synovial fluid levels of both resistin and YKL-40 were measured in patients and controls using ELISA technique. Plain x-ray of hands, wrists and feet were done for all patients and assessed according to Larsen score. Serum levels of resistin and YKL-40 were significantly higher in RA patients than controls and in active RA than in non active patients. Also, their levels significantly correlated with CRP, ESR, RF, disease activity parameters and Larsen score. Furthermore, synovial levels of resistin and YKL-40 were significantly higher than that of the serum, and synovial YKL-40 levels showed significant correlation with CRP, ESR, RF, disease activity parameters and Larsen score. On the other hand, resistin synovial levels significantly correlated with CRP, ESR, RF and synovial leucocytic count. Resistin and YKL-40 were increased in the serum and synovial fluid of rheumatoid arthritis patients. Their serum levels correlated with markers of inflammation, parameters of disease activity and radiographic progression of joint damage. These results suggested the key roles of resistin and YKL-40 in the pathogenesis of RA and they may be important new therapeutic targets

ABSTRACT

Erythrocytes from 30 alcoholic patients as well as age and sex matched healthy control were examined, for hemagglutination titre, against, a purified galactose specific lectin from the latex of Pedilanthus tithymalodies. Mean hemagglutinin titre was significantly elevated (P < 0.001) in alcoholics as compared to controls. The results indicate that alcohol abuse is associated with adoptive cell surface changes on erythrocytes, measurable by its lectin binding capacity.

ABSTRACT

Os extratos de 176 espécies de sementes de plantas colombianas, correspondentes a 49 famílias e 147 gêneros foram testados para detectar a presença de aglutininas frente a hemácias humanas dos grupos A+, B+, O+ e de cachorro, cavalo e coelho. Os extratos que apresentaram alguma atividade hemaglutinante, foram usados para testar a aglutinaçao de Trypanosoma cruzi e T. rangeli. Além disso, a hemolinfa de 16 espécies nativas de invertebrados foram testadas nas mesmas condiçoes. Diluiçoes seriadas dos extratos foram usadas para as aglutinaçoes. Ambas epimastigotas de T. cruzi e T. rangeli foram aglutinadas com os extratos de sete sementes de plantas diferentes e com dois tipos de hemolinfa de invertebrados. As sementes de cinco plantas aglutinaram exclusivamente as epimastigotas de T. cruzi podendo assim ser usadas para a diferenciaçao entre as formas de cultura desses tripanossomos. A secreçao do pulmao do caramujo (Bulimus sp.) lisou completamente as epimastigotas de T. cruzi mas nao afetou as formas de T. rangeli. Nao foram encontrados extratos que aglutinaram ou lisaram exclusivamente as epimastigotas de T. rangeli.

ABSTRACT

In order to identify the forces involved in the binding and to understand the mechanism involved, equilibrium and kinetic studies were performed on the binding of the winged bean acidic lectin to human erythrocytes. The magnitudes of delta S and delta H were positive and negative respectively, an observation differing markedly from the lectin-simple sugar interactions where delta S and delta H are generally negative. Analysis of the sign and magnitudes of these values indicate that ionic and hydrogen bonded interactions prevail over hydrophobic interactions resulting in net -ve delta H (-37.12 kJ.mol-1) and +ve delta S (14.4 J.mole-1 K-1 at 20 degrees C), thereby suggesting that this entropy driven reaction also reflects conformational changes in the lectin and/or the receptor. Presence of two kinds of receptors for WBA II on erythrocytes, as observed by equilibrium studies, is consistent with the biexponential dissociation rate constants (at 20 degrees C K1 = 1.67 x 10(-3) M-1 sec-1 and K2 = 11.1 x 10(-3) M-1 sec-1). These two rate constants differed by an order of magnitude accounting for the difference in the association constants of the two receptors of WBA II. However, the association process remains monoexponential suggesting no observable difference in the association rates of the lectin molecule with both the receptors, under the experimental conditions studied. The thermodynamic parameters calculated from kinetic data correlate well with those observed by equilibrium. A two-step binding mechanism is proposed based on the kinetic parameters for WBA II-receptor interaction.(ABSTRACT TRUNCATED AT 250 WORDS)

ABSTRACT

A hemolinfa de Panstrongylus megistus mostrou uma atividade lectínica natural para eritrócitos de vários vertebrados e näo mostrou especificidade para os diversos tipos de eritrócitos testados (humano ABO, pato, coelho,c amundongo,carneiro, galinha e boi). Com relaçäo aos eritrócitos humanos a atividade lectínica foi similar nos tipos A+, B+ e AB+ enquanto a atividade mais alta foi observada no tipo O+. O título de aglutinaçäo entre eritrócitos animais näo mostrou diferença apreciável, excluindo eritrócitos de boi, que apresentaram o título mmais baixo. A determinaçäo da concentraçäo mínima de inibiçäo foi realizada com eritrócitos humanos O+. A aglutinaçäo foi inibida por vários carboidratos (ramnose, D-dalactose, rafinose, D-lactose e D-fucose). A ramnose foi o inibidor mais potente (0,78 mM). Os resultados sugerem a presença de mais de uma lectina na hemolinfa de P. megistus
